14-and-6 Hz positive bursts in childhood encephalopathies.

14-and-6 Hz positive bursts, a benign epileptiform variant, have also been described in comatose children with liver disease, chiefly Reyes syndrome. To determine the incidence of this pattern in children in stupor or coma, 154 portable EEGs in 111 children with mental status changes were reviewed. Seven children showed 14-and-6 Hz bursts in the setting of moderate to marked background slowing. They were similar to positive bursts seen in normals in location, duration, morphology, frequency and repetition rate. They differed in being readily activated by stimulation. The etiology was toxic/met

Investigating the neural origins of Ear-EEG: A correlation study using scalp EEG source reconstruction.

Ear electroencephalography (Ear-EEG) has emerged as a promising alternative to traditional scalp EEG, offering a noninvasive, discreet, and portable method for neural monitoring. While its ability to capture brain activity is established, the cortical origins of Ear-EEG signals remain unclear. It is uncertain whether these signals primarily reflect activity from nearby cortical regions or volume-conducted signals from distant areas. This study investigates the cortical sources of Ear-EEG signals and their relationship to neural activity across frequency bands, including alpha rhythms, sleep sp

S32212, a novel serotonin type 2C receptor inverse agonist/α2-adrenoceptor antagonist and potential antidepressant: II. A behavioral, neurochemical, and electrophysiological characterization.

The present studies characterized the functional profile of N-[4-methoxy-3-(4-methylpiperazin-1-yl)phenyl]-1,2-dihydro-3-H-benzo[e]indole-3-carboxamide) (S32212), a combined serotonin (5-HT)(2C) receptor inverse agonist and α(2)-adrenoceptor antagonist that also possesses 5-HT(2A) antagonist properties (J Pharmacol Exp Ther 340:750-764, 2012). Upon parenteral and/or oral administration, dose-dependent (0.63-40.0 mg/kg) actions were observed in diverse procedures. Both acute and subchronic administration of S32212 reduced immobility time in a forced-swim test in rats. Acutely, it also suppresse

Plasma dehydroepiandrosterone sulphate and insulin-like growth factor I levels in obstructive sleep apnoea syndrome.

CONTEXT: We aimed to assess whether obstructive sleep apnoea syndrome (OSAS) affects plasma IGF-1 and dehydroepiandrosterone sulphate (DHEA-S) levels in men, factors implicated in the development of age-related metabolic disorders. DESIGN: We conducted a cross-sectional and longitudinal clinical study. PATIENTS AND SETTING: We measured plasma IGF-1 and DHEA-S levels in 191 non-drug-treated Japanese men (34 primary snorers (PS), 88 patients with mild-to-moderate OSAS and 69 patients severe OSAS ). RESULTS: Plasma IGF-1 and DHEA-S were negatively correlated with age. Plasma IGF-1 was also neg

Role of gabapentin enacarbil XR in restless legs syndrome.

Gabapentin enacarbil XR is a new extended-release formulation which attempts to overcome the reduced efficacy of shorter-acting gabapentin, with sustained delivery over a 24-hour period. It is a gabapentin prodrug which is efficiently and rapidly converted to gabapentin during active transport throughout the length of the intestine via high-capacity monocarboxylate type 1 nutrient transporters unlike its predecessor, which is absorbed via low-capacity transporters largely confined to the upper intestinal region. Its lack of saturable absorption allows for dose-proportional absorption and hence

EEG changes and seizure outcomes following anterior corpus callosotomy in adults with Lennox-Gastaut syndrome: A single-center experience.

INTRODUCTION: Lennox-Gastaut syndrome (LGS) is a drug-resistant epileptic encephalopathy that typically begins in infancy or childhood but often persists into adulthood, becoming a lifelong condition. Typical electroencephalographic (EEG) findings include generalized paroxysms of 2.5 Hz slow spike-wave and bursts of generalized paroxysmal fast activity and anterior predominant slow spike-wave during sleep. For adult patients with LGS, corpus callosotomy (CC) has proven to be an effective surgical intervention for seizure management, yet its effects on clinical and EEG outcomes in this populati

Quantitative Assessment of Cerebrospinal Fluid-Vascular Association in Alzheimer's Disease Using Phase-Contrast and 4D-Flow Magnetic Resonance Imaging.

From a neuropathological perspective, the most well-known pathological change in Alzheimer's disease (AD) to date is primarily tau protein hyperphosphorylation, which leads to the formation of intracellular neurofibrillary tangles (NFTs) and amyloid protein deposition-namely, the accumulation of amyloid protein (Aβ) and other protein aggregates outside cells, resulting in neurofibrillary tangles. The glymphatic system, driven by arterial pulsation, clears waste, including amyloid-β, via perivascular spaces (PVS). In AD, vascular dysfunction and slow-wave sleep speculation have been separately

Hypothalamic aging and hormones.

It is the heterogeneous changes of hypothalamic functions that determine the chronological sequence of aging in mammals. Recently, it was hypothesized by Cai the decrease in slow-wave sleep (SWS) resulting from skin aging as responsible for the degeneration of hypothalamic suprachiasmatic nucleus (SCN). It was soon hypothesized by the European people in television that the increase in body fat as responsible for the degeneration of male preoptic sexually dimorphic nucleus (SDN-POA), via the aromatase converting testosterone to estradiol as proposed by Cohen. It is the hypothalamic paraventricu

Electrical status epilepticus during sleep in patients with congenital Zika virus syndrome: An unprecedented clinical finding.

BACKGROUND: Brazil experienced a disproportionately higher rate of microcephaly cases in November 2015 with evidence of a causal link with Zika virus (ZIKV) infections during pregnancy. Epilepsy is a major neurological feature seen as part of congenital Zika virus syndrome (CZVS). Different seizure types and electroencephalographic (EEG) abnormalities have been described in association with this syndrome. However, clinical and neurophysiological features of epilepsy seen in children with CZVS are not fully understood. METHODS: We evaluated children with CZVS showing an EEG pattern of electric

Stress and Depression: a Crucial Role of the Mineralocorticoid Receptor.

Cortisol and corticosterone act on the appraisal process, which comprises the selection of an appropriate coping style and the encoding of the experience for storage in the memory. This action exerted by the stress hormones is mediated by mineralocorticoid receptors (MRs), which are expressed abundantly in the limbic circuitry, particularly in the hippocampus. Limbic MR is down-regulated by chronic stress and during depression but induced by antidepressants. Increased MR activity inhibits hypothalamic-pituitary-adrenal axis activity, promotes slow wave sleep, reduces anxiety and switches circu

Effect of aging on metabolic respiratory control in sleeping dogs.

We examined the effects of aging on the metabolic respiratory control system by measuring changes with time in steady-state minute volume of ventilation (VI), alveolar carbon dioxide pressure (PACO2), and ventilatory and arousal responses to hypercapnia and hypoxia during slow-wave sleep (SWS). Studies were performed longitudinally in six healthy dogs over a span of 3 to 7 yr, corresponding biologically to 12 to 24 human yr. In each of the dogs aging was associated with a decrease in steady state VI during SWS, from 6.53 +/- 1.08 (mean +/- SEM) to 5.56 +/- 0.90 L/min (p < 0.01), and with an in

Binding and neuropharmacological profile of zaleplon, a novel nonbenzodiazepine sedative/hypnotic.

The binding properties of CL284,846 (zaleplon), a novel nonbenzodiazepine sedative/hypnotic, at benzodiazepine receptor subtypes were evaluated. Zaleplon was 14.3 times more potent at inhibiting [3H]flunitrazepam binding to membrane preparations of the cerebellum than to membrane preparations of the spinal cord. The gamma-aminobutyric acid (GABA) ratio of zaleplon was 2.07. Zaleplon produced significant increases in muscimol binding similar to those of diazepam, and it was antagonized by flumazenil. Furthermore, zaleplon showed little affinity for other receptors. Spectral analysis of the elec

Subthreshold membrane potential dynamics of posterior parietal cortical neurons coupled with hippocampal ripples.

During behavioral states of immobility, sleep, and anesthesia, the hippocampus generates high-frequency oscillations called ripples. Ripples occur simultaneously with synchronous neuronal activity in the neocortex, known as slow waves, and contribute to memory consolidation. During these ripples, various neocortical regions exhibit modulations in spike rates and local field activity irrespective of whether they receive direct synaptic inputs from the hippocampus. However, little is known about the subthreshold dynamics of the membrane potentials of neocortical neurons during ripples. We patch-

[A case of Kallmann syndrome with bilateral calcifications at the basal ganglia].

A twenty-year-old male with hematoemesis was transported to our department in an unconscious state. His past history was epilepsy in infancy and the evacuation of an undescended testis for cryptochidism at three years of age. His familial history included duodenal ulcers and amyotrophic lateral screlosis among his relatives on his father's side of the family. On arrival, he demonstrated a sleeping tendency regardless of his stabilized vital signs. He had experienced a delayed puberty. A head CT examination revealed bilateral calcification at both the globus pallidus, caudate nucleus and perive

EEG in Creutzfeldt-Jakob disease.

Electroecenphalography (EEG) is an integral part of the diagnostic process in patients with Creutzfeldt-Jakob disease (CJD). The EEG has therefore been included in the World Health Organisation diagnostic classification criteria of CJD. In sporadic CJD (sCJD), the EEG exhibits characteristic changes depending on the stage of the disease, ranging from nonspecific findings such as diffuse slowing and frontal rhythmic delta activity (FIRDA) in early stages to disease-typical periodic sharp wave complexes (PSWC) in middle and late stages to areactive coma traces or even alpha coma in preterminal E

[Recent progress in development of psychotropic drugs (5)--Hypnotics].

Regarding the treatment of insomnia, it is said that a considerable degree of advancement has been made to date in terms of controlling toxicity, preventing the development of tolerance and avoiding addiction, which were often observed when administering barbiturates. Nevertheless, the majority of hypnotics used today, which are benzodiazepines, still retain certain side effects such as hangover effects, development of dependence, rebound insomnia as a withdrawal symptom, muscular atonia resulting in falling down and causing bone fracture, inhibition of respiratory system, negative interaction

Feasibility of Using a Wearable Biosensor Device in Patients at Risk for Alzheimer's Disease Dementia.

BACKGROUND: Alzheimer's disease (AD) is the most common and most costly chronic neurodegenerative disease globally. AD develops over an extended period prior to cognitive symptoms, leaving a "window of opportunity" for targeted risk-reduction interventions. Further, this pre-dementia phase includes early physiological changes in sleep and autonomic regulation, for which wearable biosensor devices may offer a convenient and cost-effective method to assess AD-risk. METHODS: Patients with a family history of AD and no or minimal cognitive complaints were recruited from the Alzheimer's Prevention

[Biological markers in schizophrenia].

Based on a review of the literature, the article deals with the major biological markers of schizophrenia. Recent developments in molecular biology have shown a possible association between schizophrenia and various HLA markers (A1, A2, A9, A10, A28, B27, BW16), and a linkage--in several families--between the disease and some polymorphisms of chromosome 5. On the other hand, chromosome X might also be involved. Neuropathological abnormalities have often been found in the brains of schizophrenics, such as cellular alterations in the basal ganglia and the limbic structures. Investigations by mea
