Tuning fluorescence of fexofenadine by switching OFF intramolecular photoinduced electron transfer: Application to development of one-step green and high throughput microwell spectrofluorimetric assay for analysis of tablets and plasma.

Fexofenadine (FEX) is a non-sedating antihistamine commonly used for the treatment of allergic conditions such as seasonal rhinitis and chronic idiopathic urticaria. This study describes the tuning "ON" the intrinsic fluorescence of FEX by switching "OFF" its intramolecular photoinduced electron transfer (PET) through the protonation of the piperidinyl nitrogen atom using sulfuric acid. The resulting fluorescence was utilized as a basis for the development of a highly sensitive microwell spectrofluorimetric assay (MW-SFA) for the one-step determination of FEX in pharmaceutical tablets and plas

A fluorescence nanoprobe of N-Acetyl-L-Cysteine capped CdTe QDs for sensitive detection of nitrofurazone.

A method was established for detecting the content of nitrofurazone (NFZ) by fluorescence quenching of N-Acetyl-L-Cysteine (NAC) coated cadmium telluride quantum dots (CdTe QDs). By means of transmission electron microscopy (TEM) and multispectral methods such as fluorescence and ultraviolet visible spectra (UV-vis), the synthesized CdTe QDs were characterized. The quantum yield (φ) of CdTe QDs was measured as 0.33 by reference method. The CdTe QDs had a better stability, the RSD of fluorescence intensity was 1.51% in three months. NFZ quenching the emission light of CdTe QDs was observed. The

Fluorine-Nitrogen-Codoped Carbon Dots as Fluorescent Switch Probes for Selective Fe(III) and Ascorbic Acid Sensing in Living Cells.

High-quality fluorescent probes based on carbon dots (CDs) have promising applications in many fields owing to their good stability, low toxicity, high quantum yield, and low raw material price. The fluorine- and nitrogen-doped fluorescent CDs (NFCDs) with blue fluorescence was successfully synthesized using 3-aminophenol and 2,4-difluorobenzoic acid as the raw material by the hydrothermal method. The NFCDs as probe can be used to directly and indirectly detect Fe3+ (detection range: 0.1-150 μM and detection limit: 0.14 μM) and ascorbic acid (AA) (detection range: 10-80 μM and detection limit:

Green synthesis of fluorescent carbon nanospheres from chrysanthemum as a multifunctional sensor for permanganate, Hg(II), and captopril.

A simple and green method for the synthesis of fluorescent carbon nanospheres (CNs) was proposed using chrysanthemum as a natural precursor and ethylenediamine as the co-reagent. The prepared CNs show strong blue fluorescence in water with quantum yield of 13.7 %, and distinguished fluorescent stability against photobleaching and ion strength. Meanwhile, the fluorescence signal of CNs is reversible and sensitive to temperature in the range of 20-80 °C, which makes CNs useful as a temperature sensor. More importantly, the CNs can serve as excellent fluorescent sensors for detecting MnO4- and Hg

A Novel Fluorescent Probe for the Detection of Cyanide Ions in Solutions and Studies on Its Biophysical Interactions with ctDNA and Proteases.

A new cationic indolium based styryl dye (Ci) as a fluorescent probe was synthesized and its anions selectivity/sensitivity properties/molecular interactions with protease enzymes (pepsin/trypsin) and ctDNA has been studied by spectroscopic and computational methods. The fluorescence measurements at different temperatures indicated that quenching mechanism of enzymes by Ci was static. ΔH and ΔS data pointed out electrostatic/hydrophobic interactions with pepsin, and also hydrogen bonds/van der Waals forces with trypsin of Ci. According to Förster's non-radiative energy transfer, binding distan

Luminescence turn-on response of naphthalene diimide based chemosensor with Formaldehyde: A novel stratagem for estimation of formaldehyde in storage fish samples.

A new strategy has been developed for selective estimation of toxic Formaldehyde (FA) in storage fish samples by a simple chemosensor (BNDI) based on naphthalene diimide core in aqueous medium at neutral pH. The rapid "lightning-up" fluorescence feature of BNDI has been implied to detect and estimate aqueous FA selectively at very low concentration. The chemosensing properties of BNDI with aqueous FA have been established through a unique interaction pattern which is proven by different spectroscopic and theoretical analysis.

Sulfonation of 3,3'-Diformyl-BINOL for Enantioselective Fluorescent Recognition of Amino Acids in Water.

Reaction of (R)-3,3'-diformyl-1,1'-bi-2-naphthol with concentrated sulfuric acid gives the corresponding 6,6'-disulfonated compound (R)-2 selectively. This provides a simple and efficient method to convert a water-insoluble compound to a water-soluble fluorescent probe. It is found that (R)-2 in combination with Zn2+ shows a highly enantioselective fluorescent response toward various amino acids in the aqueous HEPES buffer solution at pH 7.4. For example, an enantioselective fluorescence enhancement ratio [ef=ΔID /ΔIL ] up to 35.8 is observed for the recognition of asparagine. NMR and mass spe

Highly selective and sensitive detection of hydrogen sulfide in aqueous medium and live cells using peptide-based bioprobe to mimic the binding sites of the ceruloplasmin for Cu(II) ions.

Hydrogen sulfide (H2S) plays a vital role in biological systems, so it is important to develop new methods for monitoring H2S with high selectively and sensitivity. In this study, we report the efficient solid phase synthesis of a new peptide-based bioprobe L that mimics the Cu(II) binding sites of ceruloplasmin. L exhibits highly sensitive colorimetric and fluorescent "turn off" responses to Cu2+ with high selectively among various metal ions over a broad pH range in 100% aqueous solutions. Interestingly, as a new promising analytical bioprobe, the L-Cu ensemble can monitor H2S by both colori

Endogenous Fluorescence Dissimilarity Assessment of Four Potential Biomarkers of Early Liver Fibrosis by Preservation Media Effect.

Build-up of extracellular matrix in liver fibrosis results in changes on endogenous molecules expression that may be studied through the fluorescence characterization of ex vivo liver samples. To the best of our knowledge, no investigations have provided in-depth evidence and discussion on the changes of the endogenous fluorescence in ex vivo tissue due to the effects of the preservation media. In this work, we contrast and analyze the endogenous fluorescence from tryptophan, vitamin A, hydroxyproline and elastin cross-links potential biomarkers of the liver fibrosis, in in vivo measurements a

A new benzimidazole-based selective and sensitive 'on-off' fluorescence chemosensor for Cu2+ ions and application in cellular bioimaging.

Two new twinborn benzimidazole derivates (L and A), which bonded pyridine via the ester space on the opposite and adjacent positions of the benzene ring of benzimidazole respectively, were designed and synthesized. Compound L displayed fluorescence quenching response only towards copper(II) ions (Cu2+ ) in acetonitrile solution with high selectivity and sensitivity. However, compound A presented 'on-off' fluorescence response towards a wide range of metal ions to different degrees and did not have selectivity. Furthermore, compound L formed a 1:1 complex with Cu2+ and the binding constant betw

Facile reflux synthesis of polyethyleneimine-capped fluorescent carbon dots for sequential bioassays toward Cu2+ /H2 S and its application for a logic system.

In this work, we present a novel facile strategy for green synthesis of polyethyleneimine (PEI)-capped carbon dots (PEI-CDs), in which citric acid and PEI were chosen as reactants and highly fluorescent PEI-CDs could be readily obtained via a simple one-pot refluxing under 120 °C within 2 H. Fluorescence studies indicate that the as-prepared PEI-CDs exhibit strong fluorescence emission at 446 nm with excitation at 365 nm. Upon the sequential addition of Cu2+ and H2 S, PEI-CDs result in an interesting "ON-OFF-ON" three-state emission responses, promising a bifunctional sensory platform. Moreove

Supramolecular Detection of a Nerve Agent Simulant by Fluorescent Zn-Salen Oligomer Receptors.

We report on new Zn-Salen oligomer receptors able to recognize a nerve agent simulant, namely dimethyl methylphosphonate (DMMP), by a supramolecular approach. In particular, three Zn-Salen oligomers (Zn-Oligo-A, -B, and -C), differing by the length distribution, were obtained and characterized by NMR, Gel Permeation Chromatography (GPC), UV-Vis, and fluorescence spectroscopy. Furthermore, we investigated their recognition properties towards DMMP by using fluorescence measurements. We found that the recognition ability depends on the length of the oligomeric chain, and the Zn-Oligo-C shows a bi

Novel strategy to improve the sensing performances of split ATP aptamer based fluorescent indicator displacement assay through enhanced molecular recognition.

Split aptamer strategy was often used to improve the sensitivity of aptasensor. However, traditional split aptamer strategy can not be directly used to improve the label-free aptamer based Thioflavin T (ThT) displacement assay for ATP because the split ATP aptamer display much lower enhancement effects on the fluorescence of ThT than intact aptamer. In order to address this issue, this is the first report using G-rich DNA sequence to enhance the affinity of the two split ATP aptamer halves to ThT and offer lower limit of detection (LOD), wider linear range and higher selectivity through the en

Kinetic and Conformational Insights into Islet Amyloid Polypeptide Self-Assembly Using a Biarsenical Fluorogenic Probe.

Amyloid fibril formation and tissue deposition are associated with many diseases. Studies have shown that prefibrillar intermediates, such as oligomers, are the most toxic proteospecies of the amyloidogenic cascade. Thus, understanding the mechanisms of formation and the conformational ensemble of prefibrillar species is critical. Due to their transient and heterogeneous nature, detection and characterization of prefibrillar species remain challenging. The fluorogenic probe fluorescein arsenical hairpin (FlAsH), which recognizes a tetracysteine motif, has been recently used to detect the oligo

High-performance liquid chromatographic determination of 2-aminoethylphosphonic acid and 2-amino-3-phosphonopropionic acid in seawater matrix using precolumn fluorescence derivatization with o-phthalaldehyde-ethanethiol.

2-Aminoethylphosphonic acid (2-AEP) and 2-amino-3-phosphonopropionic acid (2-AP3) are two types of abundant and ubiquitous naturally occurring phosphonates used as sources of phosphorus by many prokaryotic lineages. The potential utilization mechanism of 2-AEP and 2-AP3 in eukaryotic phytoplankton is currently under investigation. However, the lack of suitable analytical methods in saline samples are the limitation of such researches. Herein, a high-performance liquid chromatography (HPLC) method for monitoring 2-AEP and 2-AP3 using precolumn fluorescence derivatization with o-phthalaldehyde-e

Small molecular probe as selective tritopic sensor of Al3+, F- and TNP: Fabrication of portable prototype for onsite detection of explosive TNP.

Schiff base organic compound (SOC) has been prepared as a tritopic chemosensor for selective sensing by fluorescence signalling towards ions like Al3+, F- and explosive molecule like TNP. In general, fluorescence like photophysical property has been used for selective detection of analyte where Al3+ and F- show turn-on fluorescence signal at different wavelengths (nm) however, quenching was found with TNP. As a consequence, the chemosensor has become a selective sensor for Al3+, F- and TNP. Reversibility of fluorescence responses for Al3+ and F- are observed in presence of ammonium nitrate and

Application of Fluorescence Emission for Characterization of Albendazole and Ricobendazole Micellar Systems: Elucidation of the Molecular Mechanism of Drug Solubilization Process.

Albendazole (ABZ) and ricobendazole (RBZ) are referred to as class II compounds in the Biopharmaceutical Classification System. These drugs exhibit poor solubility, which profoundly affects their oral bioavailability. Micellar systems are excellent pharmaceutical tools to enhance solubilization and absorption of poorly soluble compounds. Polysorbate 80 (P80), poloxamer 407 (P407), sodium cholate (Na-C), and sodium deoxycholate (Na-DC) have been selected as surfactants to study the solubilization process of these drugs. Fluorescence emission was applied in order to obtain surfactant/fluorophore

Fluorescent Probe Encapsulated in SNAP-Tag Protein Cavity To Eliminate Nonspecific Fluorescence and Increase Detection Sensitivity.

Despite the promising improvements made recently on fluorescence probes for the detection of enzymes and reactive small molecules, two fundamental problems remain: weaker fluorescence of many dyes in aqueous buffers and strong nonspecific signals in samples containing high protein levels. In this paper, we introduce a novel fluorescent probe encapsulated in protein cavity (FPEPC) concept as demonstrated by SNAP-tag protein and three environment-sensitive fluorescence probes to overcome these two problems. The probes were constructed by following the current probe design for enzymes and reactiv

Facile synthesis of red emitting 3-aminophenylboronic acid functionalized copper nanoclusters for rapid, selective and highly sensitive detection of glycoproteins.

As an emerging class of fluorescent probes, copper nanoclusters (Cu NCs) have been considered as an intriguing candidate for detecting biomoleculars due to their outstanding fluorescent properties, excellent biocompatibility and low cost. Herein, we fabricated bovine serum albumin (BSA) protected Cu NCs (BSA-Cu NCs) and further functionalized them with 3-aminophenylboronic acid (APBA) for selectively discerning glycoproteins. In aqueous solution, Cu(2+) ions were directly reduced into BSA-Cu NCs by hydrazine hydrate (N2H4·H2O) at room-temperature using BSA as the capping agent. The synthetic p
