Depressed superoxide radical generation by neutrophils from patients with rheumatoid arthritis and neutropenia: correlation with neutrophil reactive IgG.

Neutrophils of 31 patients with neutropenia and rheumatoid arthritis (RA) have been studied to assess their ability to generate superoxide radicals (O-2) on activation. Seventeen patients had classical Felty's syndrome and 14 presumed chrysotherapy related neutropenia. Results were compared with those from age and sex matched controls with uncomplicated RA and from normal subjects. Neutrophils from patients with Felty's syndrome had a significantly reduced ability to generate superoxide radicals when compared with the other three groups. In addition, serum levels of IgG polymorphonuclear leuco

Studies on the mechanism of toxicity of the mycotoxin sporidesmin. IV. Inhibition by copper-chelating agents of the generation of superoxide radical by sporidesmin.

Sporidesmin, the mycotoxin responsible for "facial eczema" in ruminants, has previously been shown to generate superoxide free radical, the latter being formed during autoxidation of the reduced form of the mycotoxin, a dithiol. The autoxidation of reduced sporidesmin is catalysed by iron and by copper, although cobalt, nickel, manganese, cerium, vanadium and molybdenum were found to be without effect. The catalytic activity of copper was some 900-fold that of iron. Cyanide and certain organic complexing agents inhibit superoxide production from reduced sporidesmin by virtue of their ability t

Selenium, zinc and copper in Down's syndrome (trisomy 21): blood levels and relations with glutathione peroxidase and superoxide dismutase.

Increased superoxide dismutase and glutathione peroxidase activities have been reported in erythrocytes of subjects with Down's syndrome. Since these enzymes contain specific trace-elements as essential components, we have determined copper, zinc and selenium levels in plasma and erythrocytes of 29 trisomy 21 patients compared with 32 age-matched controls and examined the relations with the enzymes' activities. In plasma, mean zinc and copper levels were normal, but selenium was found to be significantly decreased (p less than 0.001). In red cells, the increase of activity of the selenoenzyme

Oxidative stress and NO generation in the rat pancreatitis induced by pancreatic duct ligation.

The interaction between nitric oxide (NO) and superoxides is critical in the development of an acute pancreatitis. Previously, we reported that the expression of superoxides and of the NO-generating enzyme (NO synthase, NOS) was up-regulated in the human pancreatitis, especially within the exocrine compartment indicating an exceptional susceptibility of the exocrine parenchyma to oxidative stress. The aim of the present study was to compare the regulation of NO signalling pathways in the human pancreatitis and in an animal model of an acute pancreatitis induced by pancreatic duct ligation (PDL…

Neoagarotetraose from Marine Red Algae Attenuates Hypertensive Vascular Remodeling by Targeting FoxO4 to Inhibit Mitochondrial Superoxide Generation.

Oxidative stress is a key driver of hypertensive vascular remodeling. Neoagarotetraose (NA4), a marine-derived oligosaccharide, possesses potent antioxidant activity, but its effects on hypertension remain unexplored. Here, we investigated the role of NA4 in vascular smooth muscle cells (VSMCs) from spontaneously hypertensive rats (SHR) and in SHR in vivo. NA4 treatment significantly reduced mitochondrial superoxide, H2O2 levels, and NADPH oxidase activity in SHR-VSMCs and suppressed their hyperproliferation and migration. RNA-seq identified FoxO4 as a significantly downregulated target. FoxO4

Evaluation of two novel superoxide dismutases (SODs) from the freshwater diatom Fragilaria saxoplanctonica and their specific responses to metals and pesticides.

Superoxide dismutase (SOD) is a key antioxidant enzyme that functions as a primary defense system against cellular oxidative stress. Here, we determined two novel SOD genes, FsMnSOD and FsCuZnSOD, from the freshwater diatom Fragilaria saxoplanctonica, and examined their molecular features and transcriptional responses to environmental contaminants. FsMnSOD and FsCuZnSOD encoded 240 amino acids (aa) with conserved metal-binding residues and 160 aa with conserved metal-binding site residues in diatoms, respectively. The phylogenetic analysis revealed that the two FsSODs formed distinct branches,

Oxidative stress markers in bipolar disorder and first-degree relatives: differential associations of ischemia-modified albumin and superoxide dismutase.

BACKGROUND: Oxidative stress has been implicated in the pathophysiology of bipolar disorder (BD), potentially contributing to neuroprogression and cellular dysfunction. However, whether specific oxidative stress markers reflect disease expression or familial vulnerability remains unclear. This study aimed to evaluate thiol-disulfide homeostasis (TDH), ischemia-modified albumin (IMA), ferroxidase, and superoxide dismutase (SOD) levels in patients with BD, their unaffected first-degree relatives (FDRs), and healthy controls (HCs). METHODS: In this cross-sectional study, 50 patients with BD, 40

Antagonistic effects of typical veterinary antibiotic and herbicide on Cu/Zn superoxide dismutase activity: Competitive binding at the dimer interface.

Veterinary antibiotics and herbicides could be simultaneously ingested by livestock (especially cattle) due to their co-occurrence in feed, disrupting redox homeostasis. However, limited information is available on the combined molecular-level effects of these agrochemicals on antioxidant enzymes. In this study, sulfadiazine (SD) and metamitron (MET) were respectively selected as the representatives of veterinary antibiotics and herbicides to explore their combined toxic effects on SOD and the underlying molecular mechanisms, using multi-spectroscopic technologies, X-ray photoelectron spectros

A Near-Infrared Fluorescent Probe for Dynamic Monitoring of Superoxide Anions for Early Diagnosis of Cuproptosis-Mediated Acute Kidney Injury.

Copper serves an essential trace element in the human body, playing an indispensable role in various physiological processes. Cuproptosis, a recently discovered form of copper dependent programmed cell death, has emerged as a significant factor in the pathogenesis of several diseases. Acute kidney injury (AKI), as a high mortality disease, has attracted widespread attention. However, the precise mechanisms underlying the interplay between cuproptosis and AKI remain largely elusive. Here, we report a superoxide anion (O2•-) activated near-infrared fluorescent probe for investigating the relatio

Iron Superoxide Dismutase (Fe-SODe) as a sensitive antigenic marker for serodiagnosis of Leishmania spp. in urban schoolchildren of Central Mexico.

Leishmaniasis remains a neglected tropical disease with significant diagnostic challenges due to serological cross-reactivity with other trypanosomatids. This study evaluated the diagnostic performance of Iron Superoxide Dismutase (Fe-SODe), an excreted enzyme from Leishmania spp., as a specific antigen in detecting anti-Leishmania antibodies in school-aged children from urban areas of Santiago de Querétaro, Mexico. A total of 131 blood samples were analyzed using ELISA and Western blot assays with Fe-SODe from L. mexicana, L. braziliensis, and L. infantum. ELISA-FeSODe revealed seroprevalence

Deleting Mitochondrial Superoxide Dismutase 2 in Salivary Gland Ductal Epithelial Cells Recapitulates Non-Sjögren's Sicca Syndrome.

Elevated oxidative stress can play a pivotal role in autoimmune diseases by exacerbating inflammatory responses and tissue damage. In Sjögren's disease (SjD), the contribution of oxidative stress in the disease pathogenesis remains unclear. To address this question, we created mice with a tamoxifen-inducible conditional knockout (KO) of a critical antioxidant enzyme, superoxide dismutase 2 (Sod2), in the salivary glands (i-sg-Sod2 KO mice). Following tamoxifen treatment, Sod2 deletion occurred primarily in the ductal epithelium, and the salivary glands showed a significant downregulation of So

Endurance exercise preconditioning alleviates ferroptosis induced by doxorubicin-induced cardiotoxicity through mitochondrial superoxide-dependent AMPKα2 activation.

Doxorubicin-induced cardiotoxicity (DIC) adversely impacts patients' long-term health and quality of life. Its underlying mechanism is complex, involving regulatory cell death mechanisms, such as ferroptosis and autophagy. Moreover, it is a challenge faced by patients undergoing cardiac rehabilitation. Endurance exercise (E-Exe) preconditioning effectively counters DIC injury, potentially through the adenosine monophosphate-activated protein kinase (AMPK) pathway. However, detailed studies on this process's mechanisms are scarce. Here, E-Exe preconditioning and DIC models were established usin

Sirtuin 3-mediated deacetylation of superoxide dismutase 2 ameliorates sodium fluoride-induced mitochondrial dysfunction in porcine oocytes.

Fluoride exerts detrimental effects on germ cells and increases the infertility rate in women. Nevertheless, the precise mechanisms behind the developmental abnormalities caused by fluoride in oocytes remain poorly comprehended. The current study, we established mitochondrial damage model in oocytes via 50 μg/mL sodium fluoride (NaF) supplementation. We then examined the effects of honokiol in preventing mitochondrial deficits caused by NaF and investigated the mechanisms through which honokiol protects oocytes. The findings investigated that NaF increased levels of mitochondrial reactive oxyg

Formalin-Fixed Paraffin-Embedded Proteomics of Malignant Mesothelioma and New Candidate Biomarkers Thioredoxin and Superoxide Dismutase 2 for Immunohistochemistry.

The pathogenesis of malignant mesothelioma (MM) has been extensively investigated, focusing on stress derived from reactive oxygen species. We aimed to identify diagnostic biomarkers of MM by analyzing proteins in formalin-fixed paraffin-embedded specimens using liquid chromatography-mass spectrometry. We extracted proteins from formalin-fixed paraffin-embedded sections of MM tissues (n = 7) and compared their profiles with those of benign mesothelial tissues (n = 4) and alveolar tissue (n = 1). Proteomic data were statistically assessed and profiled using principal component analysis. We were

P2X7 receptor activation mediates superoxide dismutase 1 (SOD1) release from murine NSC-34 motor neurons.

Mutant superoxide dismutase 1 (SOD1) can be constitutively released from motor neurons and transmitted to naïve motor neurons to promote the progression of amyotrophic lateral sclerosis (ALS). However, the biological impacts of this process and the precise mechanisms of SOD1 release remain to be fully resolved. Using biochemical and fluorescent techniques, this study aimed to determine if P2X7 receptor activation could induce mutant SOD1 release from motor neurons and whether this released SOD1 could be transmitted to motor neurons or microglia to mediate effects associated with neurodegenerat

Exogenous ketone salts inhibit superoxide production in the rat caudal solitary complex during exposure to normobaric and hyperbaric hyperoxia.

The use of hyperbaric oxygen (HBO2) in hyperbaric and undersea medicine is limited by the risk of seizures [i.e., central nervous system (CNS) oxygen toxicity, CNS-OT] resulting from increased production of reactive oxygen species (ROS) in the CNS. Importantly, ketone supplementation has been shown to delay onset of CNS-OT in rats by ∼600% in comparison with control groups (D'Agostino DP, Pilla R, Held HE, Landon CS, Puchowicz M, Brunengraber H, Ari C, Arnold P, Dean JB. Am J Physiol Regu Integr Comp Physiol 304: R829-R836, 2013). We have tested the hypothesis that ketone body supplementation

An easy-to-implement combinatorial approach involving an activity-based assay for the discovery of a peptidyl copper complex mimicking superoxide dismutase.

A combinatorial approach using a one-bead-one-compound method and a screening based on a SOD-activity assay was set up for the discovery of an efficient peptidyl copper complex. The complex exhibited good stability constants, suitable redox potentials and excellent intrinsic activity. This complex was further assayed in cells for its antioxidant properties and showed beneficial effects when cells were subjected to oxidative stress.
