TP53 mutations in workers exposed to occupational carcinogens.

In some cases, evidence exists that exogenous carcinogenic exposures contribute to the mutation spectrum of the TP53 gene (p53) in human cancers. Although the clearest examples come from dietary and environmental sources, only a restricted number of papers have concentrated specifically on TP53 mutations in tumors from workers exposed to occupational carcinogens. In populations exposed to dietary aflatoxin B1 with liver cancer (AFB1) and ultraviolet (UV)-radiation with skin cancer, a single specific-looking TP53 mutation has been described in some of the tumors. Whether these fingerprints in t

Chitosan-ε-polylysine‑sodium tripolyphosphate based Pickering emulsions for co-encapsulation of cinnamaldehyde and linalool: An effective sustained-release system against toxigenic fungi.

Chitosan-ε-polylysine‑sodium tripolyphosphate (CS-εPL-STPP) ternary solid particles were fabricated to stabilize Pickering emulsions for co-encapsulating cinnamaldehyde (CN) and linalool (LN). The incorporation of ε-PL into the CS-STPP system enhanced structural stability via hydrogen bonding and van der Waals-mediated crosslinking, as confirmed by FT-IR, XPS, and molecular docking, concurrently improving thermal stability. The optimized CN-LN-PE (oil fraction 4 %, CN: LN = 4:2) exhibited not only high encapsulation efficiency (86.67 ± 0.61 %) and temperature-responsive release properties but

Turn-On Near-Infrared Lateral Flow Immunoassay with Synergistic Energy Transfer for Direct and Ultrasensitive Detection in Complex Matrices.

The rapid, sensitive, and quantitative detection of small-molecule analytes in complex matrices remains fundamentally constrained by severe optical interference and limited signal transduction efficiency. Herein, we report a turn-on near-infrared (NIR) fluorescence lateral flow immunoassay (NFLFIA) that integrates indocyanine green-embedded nanoparticles and horseradish peroxidase-loaded gold nanorods as a spectrally matched donor-acceptor pair. Benefiting from the intrinsically low optical background of the NIR window and a cascade quenching mechanism governed by synergistic inner-filter effe

Ferroptosis in Mycotoxin-Induced Toxicity: Molecular Mechanisms, Intervention Implications, and Future Directions.

Chronic exposure to mycotoxin-contaminated food and feed induces systemic toxicity in humans and animals, with emerging evidence identifying ferroptosis as a key mechanism driving this pathology. Key mycotoxins, including deoxynivalenol, aflatoxin B1, T-2 toxin, fumonisin B1, zearalenone, ochratoxin A, and patulin, trigger ferroptosis through interconnected pathways: iron metabolism disruption, lipid peroxides accumulation, Glutathione Peroxidase 4 (GPX4) antioxidant system imbalance, Ferroptosis Suppressor Protein 1 (FSP1) pathway modulation, and mitochondrial homeostasis disruption. This rev

Curcumin attenuates AFB1-induced oxidative stress and pyroptosis-associated responses while improving autophagy-related signaling in HD11 macrophages.

Aflatoxin B1 (AFB1) induces oxidative stress, autophagy-related impairment, and pyroptosis-associated responses in immune cells. This study evaluated the protective effects of curcumin against AFB1-induced cytotoxicity in HD11 macrophages and examined the involvement of Nrf2 signaling, autophagy-related activity, and the NLRP3 inflammasome using ML385, 3-MA, and MCC950. AFB1 suppressed Nrf2-related antioxidant signaling, reduced autophagy-related markers, and enhanced NLRP3-associated pyroptosis-related responses. Curcumin increased Nrf2-related activity, improved autophagy-related signaling,

Characterization of a bZIP Transcription Factor ZipD in Aspergillus flavus.

Aspergillus flavus produces aflatoxin B1, a Group 1 carcinogen that causes severe economic and agricultural losses and can lead to invasive aspergillosis in humans. During its growth, A. flavus develops hyphae that form a continuous mycelium, which is associated with host colonization and toxin production. Transcription factors are considered regulators of fungal development and pathogenicity. Among these families, bZIP proteins are associated with growth, metabolism, and stress tolerance. In A. fumigatus, the bZIP transcription factor ZipD regulates calcium signaling pathways and cell wall in

An Unusual Outbreak of Ochratoxicosis Associated with Trigonella foenum-graecum Ingestion in Ruminants from Different Farms of Sicily.

Trigonella foenum-graecum is a widely cultivated legume in Mediterranean regions, and it is used for human and animal consumption, as well as for medical purposes. High temperatures and abundant rainfall during the spring season in Sicily favor the formation of an environment suitable for the growth and proliferation of fungi with the production of mycotoxins. In this study, ochratoxin A, aflatoxin, deoxynivalenol, zearalenone, fumonisin, and T-2 toxin concentrations in Trigonella foenum-graecum were determined in feed administered to ruminants and also in blood samples from cattle and sheep i

The Mechanism of APOBEC3B in Hepatitis B Virus Infection and HBV Related Hepatocellular Carcinoma Progression, Therapeutic and Prognostic Potential.

Hepatocellular carcinoma (HCC) is one of the most prevalent malignant tumors globally. Prominent factors include chronic hepatitis B (CHB) and chronic hepatitis C (CHC) virus infections, exposure to aflatoxin, alcohol abuse, diabetes, and obesity. The prevalence of hepatitis B (HBV) is substantial, and the significant proportion of asymptomatic carriers heightens the challenge in diagnosing and treating hepatocellular carcinoma (HCC), necessitating further and more comprehensive research. Apolipoprotein B mRNA editing catalytic polypeptide (APOBEC) family members are single-stranded DNA cytidi

Human multi-organ chip co-culture of bronchial lung culture and liver spheroids for substance exposure studies.

Extrapolation of cell culture-based test results to in vivo effects is limited, as cell cultures fail to emulate organ complexity and multi-tissue crosstalk. Biology-inspired microphysiological systems provide preclinical insights into absorption, distribution, metabolism, excretion, and toxicity of substances in vitro by using human three-dimensional organotypic cultures. We co-cultured a human lung equivalent from the commercially available bronchial MucilAir culture and human liver spheroids from HepaRG cells to assess the potential toxicity of inhaled substances under conditions that permi

Data-driven methods for imputing national-level incidence in global burden of disease studies.

OBJECTIVE: To develop transparent and reproducible methods for imputing missing data on disease incidence at national-level for the year 2005. METHODS: We compared several models for imputing missing country-level incidence rates for two foodborne diseases - congenital toxoplasmosis and aflatoxin-related hepatocellular carcinoma. Missing values were assumed to be missing at random. Predictor variables were selected using least absolute shrinkage and selection operator regression. We compared the predictive performance of naive extrapolation approaches and Bayesian random and mixed-effects reg

The etiology of hepatocellular carcinoma and consequences for treatment.

Most patients with hepatocellular carcinoma (HCC) have liver cirrhosis, which develops following long periods of chronic liver disease. Cirrhosis is characterized by a decrease in hepatocyte proliferation, indicating an exhaustion of the regenerative capacity of the liver, and results in an increase in fibrous tissue and a destruction of liver cells, which may ultimately lead to the development of cancerous nodules. Half of all cases of HCC are associated with hepatitis B virus infection, with a further 25% associated with hepatitis C virus. Other risk factors for developing HCC include alcoho

Protection by dietary compounds against mutation in a transgenic rodent.

One of the most relevant biomarkers of genotoxicity and, potentially, carcinogenesis is the occurrence of mutations. Data indicate that carcinogens are highly specific with regard to their target tissue in inducing both tumors and mutations. This specificity may reflect the dependence on tissue-specific metabolic activation, the organ-specific environment or both. Ideally, therefore, mutation should be determined in a real animal rather than in a cell culture system. The lacI transgenic rodent model provides such a system. We have used this model to investigate tissue, species and sex specific

Modulation of Inflammation-Driven Hepatocarcinogenesis: The Hepatic Immune Microenvironment, Gut-Liver Axis, and Neuroregulation.

Hepatocellular carcinoma (HCC) is a highly malignant cancer closely related to the chronic inflammation induced by persistent liver damage. Various risk factors, including chronic hepatitis B/C virus infections, alcoholic liver disease, metabolic dysfunction-associated steatotic liver disease, aflatoxins exposure, and metabolic disorders, contribute to genetic mutations in hepatocytes, leading to sustained cellular damage and apoptosis. These processes foster a chronic inflammatory microenvironment that activates hepatic stellate cells, promotes extracellular matrix deposition, and triggers ab…
