Sustainable Four-Component Annulation for the Synthesis of 2,3,4,6-Tetraarylpyridines.

A one-pot, four-component annulation of 2,3,4,6-tetraarylpyridines from aromatic aldehydes, methyl ketones, diaryl ethanones, and ammonium acetate is described. The reaction features high functional group compatibility in air under solvent-free conditions without any additive and only water as the nontoxic byproduct, providing a strategy for the facile, economical, and eco-friendly construction of multiaryl-substituted pyridines from simple and readily available reactants.

Direct Conversion of Hydrazones to Amines using Transaminases.

Transaminase enzymes (TAms) have been widely used for the amination of aldehydes and ketones, often resulting in optically pure products. In this work, transaminases were directly reacted with hydrazones in a novel approach to form amine products. Several substrates were investigated, including those with furan and phenyl moieties. It was determined that the amine yields increased when an additional electrophile was added to the reaction mixture, suggesting that they can sequester the hydrazine released in the reaction. Pyridoxal 5'-phosphate (PLP), a cofactor for transaminases, and polyethyle…

Glycine and serine markedly eliminate methylglyoxal in the presence of formaldehyde via the formation of imidazole salts.

l-glycine and l-serine are the building blocks of proteins and exhibit various biological activities. This work found that l-glycine and l-serine show low scavenging capacity for methylglyoxal at moderate conditions (pH 7.0, 37 °C). However, they efficiently eliminate methylglyoxal and formaldehyde when the two aldehydes co-exist, via generation of imidazole salt, a compound formed by one molecule of methylglyoxal and formaldehyde, and two molecules of amino acids. The imidazole salts were identified in biscuits and fried potato crisps. Moreover, the formation of imidazole salts greatly decrea…

Expansion of Substrate Scope for Nitroxyl Radical/Copper-Catalyzed Aerobic Oxidation of Primary Alcohols: A Guideline for Catalyst Selection.

Four distinctive sets of optimum nitroxyl radical/copper salt/additive catalyst combinations have been identified for accommodating the aerobic oxidation of various types of primary alcohols to their corresponding aldehydes. Interestingly, less nucleophilic catalysts exhibited higher catalytic activities for the oxidation of particular primary allylic and propargylic alcohols to give α,β-unsaturated aldehydes that function as competent Michael acceptors. The optimum conditions identified herein were successful in the oxidation of various types of primary alcohols, including unprotected amino a…

A comparative analysis of virucidal efficiency of biocide agents.

The main groups of biocide agents used for inactivation of bacteria and viruses were studied for their virucidal activity against enveloped (HIV, viral hepatitis C, influenza virus A) and non-enveloped viruses (poliovirus, adenovirus). Their efficiency was analyzed. Quarterly ammonium compounds (QAC) themselves are not able to properly inactivate non-enveloped viruses. However, they can be successfully applied in combination with other biocides (guanidines, aldehydes). Effective composition of QAC with amines and guanidines provided inactivation of viruses (4.0 lgTCID50) in concentrations of 0…

Cascade oxime formation, cyclization to a nitrone, and intermolecular dipolar cycloaddition.

Simple haloaldehydes, including enolisable aldehydes, were found to be suitable for the formation of cyclic products by cascade (domino) condensation, cyclisation, dipolar cycloaddition chemistry. This multi-component reaction approach to heterocyclic compounds was explored by using hydroxylamine, a selection of aldehydes, and a selection of activated dipolarophiles. Initial condensation gives intermediate oximes that undergo cyclisation with displacement of halide to give intermediate nitrones; these nitrones undergo in situ intermolecular dipolar cycloaddition reactions to give isoxazolidine…

Synthesis of 5-alkylated barbituric acids and 3-alkylated indoles via microwave-assisted three-component reactions in solvent-free conditions using Hantzsch 1,4-dihydropyridines as reducing agents.

Reaction of barbituric acids with aldehydes and dihydropyridines in one pot under microwave (MW) irradiation in the absence of solvent, affords 55–82% of the 5-benzylated barbituric acids. Use of alkyl nitriles or barbituric acids with indole-3-aldehyde and dihydropyridine (DHP) afforded 3-alkylated indoles in 57–76 % yield. In each case DHPs are converted to pyridines.

The Structure-Antioxidant Activity Relationship of Ferulates.

The antioxidant activity of ferulic acid (1), iso-ferulic acid (2), coniferyl aldehyde (3), methyl ferulate (4), and ethyl ferulate (5) were investigated using 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and ferric-reducing antioxidant power (FRAP) assays and autoxidation of triacylglycerols of commercially available sunflower oil (TGSO). The compounds tested for ability to scavenge ABTS radical cations was in the order of ferulic acid > coniferyl aldehyde ≈ iso-ferulic acid > ethyl ferulate ≈ methyl ferulate. The results of the FRAP assay for ferulic acid, iso-ferulic acid,

Green leaf volatiles enhance methyl jasmonate response in Arabidopsis.

Plants emit green leaf volatiles (GLVs) in response to insect or pathogen damage. GLVs consist of C6 and C9 aldehydes, alcohols, and their acetate esters, and play important roles in the plant defense response. One of the functions of GLVs in the defense response is priming. Plants pretreated by GLVs can induce a defense response more rapidly and effectively than unpretreated plants when they are damaged by pathogens or insects. In this study, we focused on the priming effects of GLVs on jasmonic acid response involved in the defense response. When Arabidopsis was pretreated with aldehyde GLVs…

Mechanistic studies on chloral toxicity: relationship to trichloroethylene carcinogenesis.

Chloral (trichloroacetaldehyde), the major metabolite of trichloroethylene (TCE), was investigated for its potential to form DNA-protein cross-links (DPX), a lesion produced by other aldehydes. Chloral did not form DPX in rat liver nuclei at concentrations up to 250 mM for 30 min at 37 degrees C, while chloroacetaldehyde (47 mM) and acetaldehyde (200 mM) did form cross-links. Experiments with the aldehyde-trapping reagents thiosemicarbazide and semicarbazide showed that chloral did not react, in contrast with aldehydes that form DPX. This indicates a very strong hydration of chloral. Mice give…

Nutritional copper deficiency and penicillamine administration: some effects on bone collagen and arterial elastin crosslinking.

Nutritional copper deficiency effects marked changes in the crosslinking of collagen and elastin, presumably in relationship to copper's role as a cofactor for lysyl oxidase. Lysyl oxidase controls one of the initial steps in the crosslinking of elastin and collagen, i.e., the conversion of peptidyl lysine or hydroxylysine residues to peptidyl alpha-aminoadipic-delta-semialdehyde derivatives. Once lysine-derived aldehydic functions in collagen and elastin are formed, crosslinks occur via aldol and Schiff-base type condensations. A decrease in the degree of crosslinking results in changes in th

Wine-red lignin revisited, dirigent proteins, and native lignin macromolecular configuration.

The wine-red coloration of xylem in antisense cinnamyl alcohol dehydrogenase (CAD) down-regulated tobacco (Nicotiana tabacum) has long been … (MWD) of the lignin from the antisense CAD line. No evidence was obtained that poly-hydroxycinnamyl aldehydes contributed to bulk lignification; the slightly lower MWD may result from hydroxycinnamyl aldehydes functioning as terminators of polymerization during lignification. More importantly, these findings are placed in context of what is now known about …

Genome-wide identification of the CAD gene family and functional analysis of putative bona fide CAD genes in tobacco (Nicotiana tabacum L.).

Cinnamyl alcohol dehydrogenase (CAD) plays a crucial role in lignin biosynthesis, and the gene family encoding various CAD isozymes has been cloned and characterized in numerous plant species. However, limited information regarding the CAD gene family in tobacco is currently available. In this study, we identified 10 CAD genes in Nicotiana tabacum, four in N. tomentosiformis, and six in N. sylvestris. The nucleotide and amino acid sequences of these tobacco CADs demonstrate high levels of similarity, whereas the putative protein sequences conservatively possessed two Zn2+ binding motifs and an

Amplification of anti-diastereoselectivity via Curtin-Hammett effects in ruthenium-catalyzed hydrohydroxyalkylation of 1,1-disubstituted allenes: diastereoselective formation of all-carbon quaternary centers.

Under the conditions of ruthenium-catalyzed transfer hydrogenation, 1,1-disubstituted allenes 1a-c and alcohols 2a-g engage in redox-triggered generation of allylruthenium-aldehyde pairs to form products of hydrohydroxyalkylation 3a-g, 4a-g, and 5a-g with complete branched regioselectivity. By exploiting Curtin-Hammett effects, good to excellent levels of anti-diastereoselectivity (4:1 to >20:1) are obtained. Thus, all carbon quaternary centers are formed in a diastereoselective fashion upon carbonyl addition from the alcohol oxidation level in the absence of premetalated nucleophiles or stoic

Role of ALDH2 in myocardial ischemia-reperfusion injury: using ALDH2 as a potential therapeutic target.

Myocardial ischemia-reperfusion (I/R) injury remains a major barrier to the full benefit of timely coronary reperfusion. This review summarizes the role of mitochondrial aldehyde dehydrogenase 2 (ALDH2) as both an endogenous cardioprotective enzyme and a potential therapeutic target in myocardial I/R injury. During reperfusion, reactive oxygen species promote lipid peroxidation and the accumulation of cytotoxic aldehydes, particularly 4-hydroxy-2-nonenal (4-HNE) and malondialdehyde. ALDH2 detoxifies these aldehydes and thereby modulates several injury pathways, including mitochondrial dysfunct…

Effects of silylene ligands on the performance of carbonyl hydrosilylation catalyzed by cobalt phosphine complexes.

In order to study the effects of silylene ligands on the catalytic activity of carbonyl hydrosilylation catalyzed by cobalt phosphine complexes, readily available model catalysts are required. In this contribution, a comparative study of the hydrosilylation of aldehydes and ketones catalyzed by tris(trimethylphosphine) cobalt chloride, CoCl(PMe3)3 (1), and bis(silylene) cobalt chloride, Co(LSi:)2(PMe3)2Cl (2, LSi: = {PhC(NtBu)2}SiCl), is presented. It was found that both complexes 1 and 2 are good catalysts for the hydrosilylation of aldehydes and ketones under mild conditions. This catalytic …

Colonial seabird's paralytic perfume slows lice down: an opportunity for parasite-mediated selection?

Selection for chemical signals in birds could be influenced by parasitism as has been previously suggested for visual and acoustic displays. Crested auklets (Aethia cristatella), colonial seabirds from Alaska and Siberia, offer an example of how this might occur. Crested auklets secrete lipids in plumage, possibly as an indicator of status and attractiveness. Prominent among these secretions are aldehydes, which are noticeable as a pungent citrus-like odour. Octanal and hexanal, the most abundant aldehydes in the plumage of crested auklets, are potent invertebrate repellents, reported from the…
