Defects of leukocyte locomotion in multiple myeloma.

23 patients with multiple myeloma of IgG, IgA and light chain type, one patient with benign monoclonal IgE gammopathy and 23 healthy controls were investigated for their leukocyte locomotion (LL), for the ability of their sera to generate chemotactic factors upon activation with zymosan, and for inhibitory effects of their sera upon LL. While no significant impairment of the patients' LL was observed when casein was used as cytotaxin, LL was markedly reduced in myeloma patients with zymosan activated control serum as chemoattractant. The ability of serum to generate chemotactic factors upon ac

Unravelling the multi-scale structural organisation of in vivo ileal digesta from diets containing protein-seaweed polysaccharide blends.

Seaweed-derived polysaccharides such as agar, alginate, carrageenan, and cellulose are increasingly used as functional ingredients in food products, yet their influence on the gastrointestinal digestion process remains poorly understood. This study investigated how different fibres modulate the digestion and nanoassembly process of two model food proteins, casein and whey protein isolate, under physiologically relevant conditions using an in vivo pig model. Ileal digesta were analysed through compositional, rheological, microstructural, and nanostructural characterisation. The results demonstr

Inhibiting CK2 in breast cancer: From molecular targets to drug candidates.

Casein kinase 2 (CK2) is a constitutively active serine/threonine kinase that orchestrates multiple oncogenic signaling networks, including PI3K/Akt, NF-κB, JAK/STAT3, Wnt/β-catenin, and DNA damage response pathways. Its broad involvement in cell survival, proliferation, and therapy resistance has positioned CK2 as a compelling target in oncology, particularly in breast cancer where CK2α and CK2β are frequently overexpressed and correlate with poor prognosis. Over the past decades, a wide range of CK2 inhibitors has been developed ranging from classical ATP-competitive scaffolds (TBB, DMAT, CX

Development of Potent and Selective CK1α Molecular Glue Degraders.

Molecular glue degraders (MGDs) are small molecules that facilitate proximity between a target protein and an E3 ubiquitin ligase, thereby inducing target protein degradation. Glutarimide-containing compounds are MGDs that bind cereblon (CRBN) and recruit neosubstrates. Through explorative synthesis of a glutarimide-based library, we discovered a series of molecules that induce casein kinase 1 alpha (CK1α) degradation. By scaffold hopping and rational modification of the chemical scaffold, we identified an imidazo[1,2-a]pyrimidine compound that induces potent and selective CK1α degradation. A

Effect of dipotassium phosphate and heat on milk protein beverage viscosity and color.

Our objective was to determine the effect of addition of dipotassium phosphate (DKP) at 3 different thermal treatments on color, viscosity, and sensory properties of 7.5% milk protein-based beverages during 15 d of storage at 4°C. Micellar casein concentrate (MCC) and milk protein concentrate (MPC) containing about 7.5% protein were produced from pasteurized skim milk using a 3×, 3-stage ceramic microfiltration process and a 3×, 3-stage polymeric ultrafiltration membrane process, respectively. The MCC and MPC were each split into 6 batches, based on thermal process and addition of DKP. The 6 b

Glycomacropeptide Safety and Its Effect on Gut Microbiota in Patients with Phenylketonuria: A Pilot Study.

Glycomacropeptide (GMP) represents a good alternative protein source in Phenylketonuria (PKU). In a mouse model, it has been suggested to exert a prebiotic role on beneficial gut bacteria. In this study, we performed the 16S rRNA sequencing to evaluate the effect of 6 months of GMP supplementation on the gut microbiota of nine PKU patients, comparing their bacterial composition and clinical parameters before and after the intervention. GMP seems to be safe from both the microbiological and the clinical point of view. Indeed, we did not observe dramatic changes in the gut microbiota but a speci

A review of the materials and allergens in protective gloves.

The ingredients previously reported to cause protective glove allergy are presented and evaluated for strength of evidence. Allergens that have caused both delayed hypersensitivity and contact urticaria are considered for rubber, plastic, leather, and textile gloves. The current guidelines regarding glove manufacture are described. A list of materials confirmed by the industry to be used in glove production is presented together with a suggested series for investigating patients with delayed type hypersensitivity and contact urticaria secondary to glove use.

Neonatal jaundice and diet.

OBJECTIVE: To determine whether an earlier observation, that infants fed a casein-hydrolysate formula (Nutramigen) have lower neonatal jaundice levels than those fed standard formulas, would be repeated in a larger independent group of infants with more frequent measurements and more rigorous statistical analysis. DESIGN: Newborn infants were fed human milk, a standard whey-predominant formula (Enfamil), or Nutramigen (n = 20 for each group) during the first 3 weeks of life. Transcutaneous jaundice index was measured daily for the first week of life and every 2 to 3 days thereafter, using a n

Exploration of ovine milk whey proteome during postnatal development using an iTRAQ approach.

BACKGROUND: Ovine milk is a rich source of bioactive proteins that supports the early growth and development of the newborn lambs. A large number of researches had targeted to the identification of ovine milk fat globule membrane proteins (MFGMPs), caseins (CNs), mastitis milk proteins in past years, but the dynamic change tendency of milk whey proteins during postnatal development has received limited attention. This research aimed to investigate the dynamic changes of ovine milk whey proteins after delivery, and explore the functions of whey proteins on early development of the newborns. ME…

Stages of granulovacuolar degeneration: their relation to Alzheimer's disease and chronic stress response.

Granulovacuolar degeneration (GVD) is characterized by the presence of vacuolar cytoplasmic lesions in nerve cells of the medial temporal lobe. These changes occur in older non-diseased individuals as well as in patients with Alzheimer's disease (AD), progressive supranuclear palsy (PSP), Pick's, sporadic Parkinson's (PD), and Guam diseases. We stained representative paraffin sections from all parts of the brain with anti-pTDP43, anti-CK1δ or anti-CK1ε from 14 non-demented elderly, 19 AD, 17 non-AD tauopathy, 9 sporadic PD, and 5 TDP43-proteinopathy [amyotrophic lateral sclerosis (ALS) and fro

The Evaluation of Yeast Protein Quality as a New and Sustainable Source of Dietary Protein.

Facing the global shortage of protein resources, yeast protein (YP) has emerged as a sustainable alternative to dietary proteins. This study employed a weaned SD rat model to evaluate the quality of YP by comparing plant-based protein sources and animal-derived protein. Supplementation of YP with whey protein isolate (WPI) or soybean protein isolate (SPI) achieved body weight gains comparable to those of casein. The tryptophan (Trp) in YP was the amino acid with the highest content in true ileal digestible (TID), accounting for 99.92%. Significant enhancements (p < 0.05) in true digestibility

Yogurt fortified with various protein hydrolysates: Texture and functional properties.

This work evaluated the impact of incorporating 1% of commercial protein hydrolysates [rice protein hydrolysate (RPH), pea protein hydrolysate (PPH), and casein hydrolysate (CH)] on the functional, microstructure, and texture properties of set yogurt. Yogurt prepared with RPH exhibited the highest viability number of Streptococcus thermophilus. The addition of three hydrolysate types to yogurt revealed significant increases in the antioxidant and ACE-inhibitory activities, where the highest values were noted for the yogurt prepared with RPH. RPH exhibited no differences in texture properties (

Caecal digestibility as an approximation of ileal protein digestibility evaluation in rats.

The rat model can be used to assess ileal protein digestibility rapidly and in first intention, but no standardised method exists. Our objective was to compare methods to assess protein digestibility, depending on collection site (ileum/caecum) and use of a non-absorbable marker. A meal containing either casein, gluten or pea protein and chromium oxide as non-absorbable marker was given to male Wistar rats and the entire digestive content was collected 6 h later. Total chromium recovery was incomplete and variable, depending on protein source. We observed no significant difference in digestibi

Physiological functions of CKIP-1: From molecular mechanisms to therapy implications.

The casein kinase 2 interacting protein-1 (CKIP-1, also known as PLEKHO1) is initially identified as a specific CK2α subunit-interacting protein. Subsequently, various proteins, including CPα, PAK1, Arp2/3, HDAC1, c-Jun, ATM, Smurf1, Rpt6, Akt, IFP35, TRAF6, REGγ and CARMA1, were reported to interact with CKIP-1. Owing to the great diversity of interacted proteins, CKIP-1 exhibits multiple biologic functions in cell morphology, cell differentiation and cell apoptosis. Besides, these functions are subcellular localization, cell type, and regulatory signaling dependent. CKIP-1 is involved in bio
