Baicalin-loaded peony seed polypeptide nanoparticles via a pH-driven method: Characterization, release kinetics, and in vitro anti-inflammatory effects.

Baicalin (BA), a flavonoid with diverse bioactivities, faces clinical application challenges due to its poor solubility, low bioavailability, and limited stability. This study reports on the development of BA-loaded peony seed polypeptide nanoparticles (BA-PSP NPs) using a pH-driven method to overcome these limitations. These nanoparticles (NPs) were spherical, 88.20 ± 2.19 nm in size, with a zeta potential of -35 mV, demonstrating excellent stability under thermal, pH, salt ion, and storage conditions. Physicochemical assays confirmed that BA was encapsulated in an amorphous state within the

Development of Receptor-Binding Domain (RBD)-Loaded PEG-PCL Nanoparticle Formulations Against SARS-CoV-2.

The COVID-19 pandemic caused by the novel coronavirus SARS-CoV-2 has highlighted the critical need for safe and effective vaccines. In this study, subunit nanovaccine formulations were developed using the receptor-binding domain (RBD) of the SARS-CoV-2 spike (S) protein encapsulated in polymeric nanoparticles composed of poly(ethylene glycol)-block-poly(ε-caprolactone) (PEG-PCL). Two surfactants, poly(vinyl alcohol) (PVA) and sodium cholate (SC), were evaluated during formulation via a modified water-in-oil-in-water (w1/o/w2) emulsion-solvent evaporation method. The resulting nanoparticles wer

β-Glucan-Modified Liposomes for Microfold Cell-Targeted Oral Delivery of Food-Derived Angiotensin I-Converting Enzyme Inhibitory Peptides Using Microfluidics.

This study presents a novel oral delivery system for food-derived angiotensin I-converting enzyme (ACE) inhibitory peptides by integrating β-glucan-functionalized liposomes with microfluidic encapsulation and receptor-mediated targeting. Two peptides, RADHPFL and YAEERYPIL derived from egg white proteins, were encapsulated into liposomes (composed of lecithin, cholesterol, and DSPE-β-glucan) via a controlled microfluidic self-assembly process. Encapsulation significantly improved the gastrointestinal stability of the peptides and preserved their ACE inhibitory activity compared to free peptide

A general controllable release amplification strategy of liposomes for single-particle collision electrochemical biosensing.

As an important component of the COVID-19 mRNA vaccines, liposomes play a key role in the efficient protection and delivery of mRNA to cells. Herein, due to the controllable release amplification strategy of liposomes, a reliable and robust single-particle collision electrochemical (SPCE) biosensor was constructed for H9N2 avian influenza virus (H9N2 AIV) detection by combining liposome encapsulation-release strategy with immunomagnetic separation. The liposomes modified with biotin and loaded with platinum nanoparticles (Pt NPs) were used as signal probes for the first time. Biotin facilitate

Effect of redox-responsive DTSSP crosslinking on poly(l-lysine)-grafted-poly(ethylene glycol) nanoparticles for delivery of proteins.

Therapeutic proteins are utilized in a variety of clinical applications, but side effects and rapid in vivo clearance still present hurdles. An approach that addresses both drawbacks is protein encapsulation within in a polymeric nanoparticle, which is effective but introduces the additional challenge of destabilizing the nanoparticle shell in clinically relevant locations. This study examined the effects of crosslinking self-assembled poly(l-lysine)-grafted-poly(ethylene glycol) nanoparticles with redox-responsive 3,3'-dithiobis(sulfosuccinimidyl propionate) (DTSSP) to achieve nanoparticle de

Thioridazine in PLGA nanoparticles reduces toxicity and improves rifampicin therapy against mycobacterial infection in zebrafish.

Encapsulating antibiotics such as rifampicin in biodegradable nanoparticles provides several advantages compared to free drug administration, including reduced dosing due to localized targeting and sustained release. Consequently, these characteristics reduce systemic drug toxicity. However, new nanoformulations need to be tested in complex biological systems to fully characterize their potential for improved drug therapy. Tuberculosis, caused by infection with the bacterium Mycobacterium tuberculosis, requires lengthy and expensive treatment, and incomplete therapy contributes to an increasin

Intravenous administration of brain-targeted stable nucleic acid lipid particles alleviates Machado-Joseph disease neurological phenotype.

Others and we showed that RNA interference holds great promise for the treatment of dominantly inherited neurodegenerative disorders such as Machado-Joseph disease (MJD), for which there is no available treatment. However, successful experiments involved intracranial administration of viral vectors and there is a need for a safer and less invasive procedure. In this work, we successfully generated stable nucleic acid lipid particles (SNALPs), incorporating a short peptide derived from rabies virus glycoprotein (RVG-9r) and encapsulating small interfering RNAs (siRNAs), which can target mutant

Immunopotentiating nano-chitosan as potent vaccine carter for efficacious prophylaxis of filarial antigens.

Lymphatic filariasis (LF), a morbid vector-borne parasitic infection affects millions in tropical areas. Complete eradication can only be achieved by the development of a potent vaccine. Among the various filarial antigens that have been characterized, antigens Brugia malayi thioredoxin (TRX) and abundant larval transcript (ALT) have produced recognizable level of protection in Jirds, thereby evidenced to be good vaccine candidates. In this study an attempt was made to enhance their immunoprophylactic activity by encapsulating them in natural polysaccharide chitosan forming nanospheres (CN). H

Treponema pallidum in gel microdroplets: a novel strategy for investigation of treponemal molecular architecture.

Controversy exists regarding the constituents and antigenic properties of the Treponema pallidum outer membrane; a major point of contention concerns the cellular location(s) of the spirochaete's lipoprotein immunogens. To address these issues and circumvent problems associated with prior efforts to localize treponemal surface antigens, we developed a novel strategy for investigating T. pallidum molecular architecture. Virulent treponemes were encapsulated in porous agarose beads (gel microdroplets) and then probed in the presence or absence of Triton X-100. Intact, encapsulated treponemes wer

A wireless, battery-free device enables oxygen generation and immune protection of therapeutic xenotransplants in vivo.

The immune isolation of cells within devices has the potential to enable long-term protein replacement and functional cures for a range of diseases, without requiring immune suppressive therapy. However, a lack of vasculature and the formation of fibrotic capsules around cell immune-isolating devices limits oxygen availability, leading to hypoxia and cell death in vivo. This is particularly problematic for pancreatic islet cells that have high O2 requirements. Here, we combine bioelectronics with encapsulated cell therapies to develop the first wireless, battery-free oxygen-generating immune-i

Preventing hypoxia-induced cell death in beta cells and islets via hydrolytically activated, oxygen-generating biomaterials.

A major hindrance in engineering tissues containing highly metabolically active cells is the insufficient oxygenation of these implants, which results in dying or dysfunctional cells in portions of the graft. The development of methods to increase oxygen availability within tissue-engineered implants, particularly during the early engraftment period, would serve to allay hypoxia-induced cell death. Herein, we designed and developed a hydrolytically activated oxygen-generating biomaterial in the form of polydimethylsiloxane (PDMS)-encapsulated solid calcium peroxide, PDMS-CaO(2). Encapsulation

Microencapsulated Immunoassays for Detection of Cytokines in Human Blood.

Cytokines are produced by leukocytes in blood and may be used as indicators of malignancies or infections. The objective of this study was to develop a strategy for immunosensing cytokines in whole, unprocessed human blood. Microfluidic droplet generation was employed to fabricate ∼400 μm diameter microcapsules with a hydrogel shell and an aqueous core containing sensing microbeads. The hydrogel shell was composed of poly(ethylene glycol) forming a thin (∼10 μm) immunoisolation layer protecting antibody-modified microbeads inside the capsule from immune cells on the outside. The microbeads wer

Noncovalent Macrocyclic Encapsulation via Terminal "Dynamic Locking" Enabling High-Performance Nonfused Ring Electron Acceptors.

Nonfused ring electron acceptors (NFREAs) have garnered significant attention recently; however, designing high-performance NFREAs remains challenging, where achieving an efficient charge transport channel via precisely controlling molecular packing and aggregation compactness is crucial. Herein, we proposed an innovative noncovalent macrocyclic encapsulation molecular strategy for high-performance NFREAs, and 3TT-Ph1 was designed and synthesized by introducing terminal phenyl lateral chains surrounding the trithieno[3,2-b]thiophene (3TT) core with an inward orientation. These terminal phenyl

Expanding the Therapeutic Potential of NUIG4: A Multifunctional Metal-Organic Framework for Dual Anticancer and Antibacterial Drug Delivery.

Metal-organic frameworks (MOFs) have emerged as highly tunable materials for targeted and controlled drug delivery. In this study, NUIG4 is presented as a multifunctional MOF-based carrier capable of encapsulating both anticancer and antimicrobial therapeutics. NUIG4 exhibits high-surface area, water stability, and suitable porosity, supporting the efficient loading of a range of drug molecules. Its dual anticancer drug delivery performance is assessed using doxorubicin (DOX) and 5-fluorouracil (5-FU). pH-Sensitive Multiliposomal Containers for Encapsulation and Rapid Release of Bioactive Substances.

A new method of the design of stimuli-sensitive multiliposomal containers for encapsulation and controlled drug release is described. Despite quite a wide choice of pH-sensitive containers, there is still a considerable challenge to synthesize those that respond quickly to small variations in pH and release most of the encapsulated drug in a short time. The suggested AMS-containing multiliposomal complexes demonstrated an excellent rate of encapsulated substance release under altering the pH of the outer solution. To improve the efficiency of the delivery of bioactive compounds to target cells

Sustained delivery of CpG oligodeoxynucleotide by acetalated dextran microparticles augments effector response to Computationally Optimized Broadly Reactive Antigen (COBRA) influenza hemagglutinin.

A subunit or protein-based influenza vaccine can be a safer alternative to live attenuated vaccine (Flumist) and require fewer boosts than an inactivated vaccine (e.g. Fluzone). However, to form an effective subunit vaccine, an adjuvant is often needed. In this work we used electrospray to encapsulate the hydrophilic adjuvant CpG into microparticles made from the hydrophobic biodegradable polymer acetalated dextran. To understand the rate of particle degradation on CpG release, polymer that was slow (21 h at phagosomal pH 5) and fast (0.25 h at pH 5) degrading was used to encapsulate the adjuv

Using crystallography tools to improve vaccine formulations.

This article summarizes developments attained in oral vaccine formulations based on the encapsulation of antigen proteins inside porous silica matrices. These vaccine vehicles show great efficacy in protecting the proteins from the harsh acidic stomach medium, allowing the Peyer's patches in the small intestine to be reached and consequently enhancing immunity. Focusing on the pioneering research conducted at the Butantan Institute in Brazil, the optimization of the antigen encapsulation yield is reported, as well as their distribution inside the meso- and macroporous network of the porous sil

Electrostatic Self-Assembled Chitosan-Pectin Nano- and Microparticles for Insulin Delivery.

A polyelectrolyte complex system of chitosan-pectin nano- and microparticles was developed to encapsulate the hormone insulin. The aim of this work was to obtain small particles for oral insulin delivery without chemical crosslinkers based on natural and biodegradable polysaccharides. The nano- and microparticles were developed using chitosans (with different degrees of acetylation: 15.0% and 28.8%) and pectin solutions at various charge ratios (n⁺/n- given by the chitosan/pectin mass ratio) and total charge. Nano- and microparticles were characterized regarding particle size, zeta potential,

Comparison of liposome based antigen delivery systems for protection against Leishmania donovani.

Liposomes have been widely exploited as antigen delivery systems for a variety of diseases including leishmaniasis. These vesicles can be prepared in various ways which may affect the immunogenicity of the encapsulated antigens. In this study we compared the vaccine potentiality of three cationic formulations with Leishmania donovani promastigote membrane antigens (LAg) and the best vesicle was evaluated for long-term protection against experimental visceral leishmaniasis. We immunized mice with LAg encapsulated in multilamellar vesicles (MLV), dehydration-rehydration vesicles (DRV) and revers
