Genetic dissection of lymphopenia from autoimmunity by introgression of mutated Ian5 gene onto the F344 rat.

Peripheral T cell lymphopenia (lyp) in the BioBreeding (BB) rat is linked to a frameshift mutation in Ian5, a member of the Immune Associated Nucleotide (Ian) gene family on rat chromosome 4. This lymphopenia leads to type 1 (insulin-dependent) diabetes mellitus (T1DM) at rates up to 100% when combined with the BB rat MHC RT1 u/u genotype. In order, to better study the lymphopenia phenotype without possible confounding effects of diabetes or other autoimmune disease, we generated congenic F344.lyp rats by introgression of lyp on diabetes-resistant MHC RT1 lv1/lv1 F344 rats. Analysis of thymic

An arginine residue (Arg101), which is conserved in many GroEL homologues, is required for interactions between the two heptameric rings.

Homologous recombination was used to construct a series of hybrid chaperonin genes, containing various lengths of Escherichia coli groEL replaced by the equivalent region from the homologous cpn60-1 gene of Rhizobium leguminosarum. Analysis of proteins produced by these hybrids showed that many of them formed structures with properties consistent with their being single heptameric rings under some conditions, as opposed to the double ring form in which both the GroEL and the Cpn60-1 proteins are found. By determining precise cross-over points, two regions in Cpn60-1 were defined which appeared

Thyroid nodularity and cancer among Chernobyl cleanup workers from Estonia.

Thyroid examinations, including palpation, ultrasound and, selectively, fine-needle aspiration biopsy, were conducted on nearly 2,000 Chernobyl cleanup workers from Estonia to evaluate the occurrence of thyroid cancer and nodular thyroid disease among men with protracted exposure to ionizing radiation. The examinations were conducted in four cities in Estonia during March-April 1995, 9 years after the reactor accident. The study population was selected from a predefined cohort of 4,833 cleanup workers from Estonia under surveillance for cancer incidence. These men had been sent to Chernobyl be

Epidemiological analysis of biofilm-forming methicillin-resistant Staphylococcus aureus clinical isolates.

INTRODUCTION: Methicillin-resistant Staphylococcus aureus (MRSA) remains a significant global concern in healthcare and community environments, posing serious risks to patients due to its ability to form biofilm. Monitoring and spread control of epidemic MRSA clones require robust epidemiological typing methods. METHODS: In this study, 30 MRSA isolates associated with significant morbidity were recovered from King Abdulaziz Specialist Hospital, Taif, Saudi Arabia. The strains were identified using the Vitek 2 automated system. The ability of MRSA to form biofilm on a polystyrene surface was e

Cytotoxicity and genotoxicity of cadmium oxide nanoparticles evaluated using in vitro assays.

Cadmium oxide nanoparticles (CdO NPs) are among some of the most studied and industrially used metal oxide NPs. They have been widely used for industrial application, such as paint pigments and electronic devices, and medical therapeutics. With increasing use of CdO NPs and concerns for their potential adverse effects on the environment and public health, evaluation of the cytotoxicity and genotoxicity of CdO NPs becomes very important. To date, there is a limited understanding of the potential hazard brought by CdO NPs and a lack of information and research, particularly on the genotoxicity a

Genetic mapping of fruit texture in American cranberry reveals multi-trait QTL and a complex genetic architecture.

Fruit texture is a key quality trait in cranberry (Vaccinium macrocarpon Ait.), with a direct impact on processing efficiency and overall profitability for the industry. To elucidate the genetic basis of cranberry texture, we conducted the first QTL mapping study in this crop implementing a fruit compression method with 10 mechanical texture traits in two biparental populations, CNJ02 (n=168) and CNJ04 (n=67), over two years. These 10 traits, associated with flesh hardness/stiffness and elasticity, showed similar phenotypic patterns of transgressive segregation and generally strong positive co

Combining computer-aided enzyme design and chromosomal integration for plasmid-free biosynthesis of 1,5-pentanediol in Escherichia coli.

BACKGROUND: 1,5-Pentanediol (1,5-PDO) is a high-value chemical with broad uses in polymer, cosmetic, and pharmaceutical industries. Although diverse biosynthetic pathways have been constructed, current recombinant strains typically rely on plasmid-based overexpression, which necessitates antibiotics and hinders industrial-scale production. RESULTS: We developed a robust, plasmid-free Escherichia coli platform for de novo 1,5-PDO synthesis by integrating pathway genes (davB, davA, gabT, yahK, car, sfp and yqhD) into the chromosome of a lysine-hyperproducing strain via CRISPR/Cas9. Genomic Imprinting, Epigenetic Dysregulation, and Neuropsychiatric Mechanisms in Prader-Willi Syndrome: A Multi-Level Integrative Review.

Prader-Willi syndrome (PWS) is a rare imprinting-related neurodevelopmental disorder caused by loss of paternally expressed genes within the chromosome 15q11-q13 region, including SNORD116, MAGEL2, and NDN. It provides a natural model for examining how genomic imprinting disruptions shape neural development and psychiatric vulnerability. This review synthesizes current evidence to clarify the mechanistic pathways linking imprinting defects and epigenetic dysregulation to neuropsychiatric outcomes in PWS. Published studies-including patient-derived induced pluripotent stem cell (iPSC) models, a

Leave-One-Chromosome-Out Association Testing Decodes Uncapping Behaviour in Honeybees During Varroa destructor Infestation.

BACKGROUND: Uncapping behaviour of Varroa‑infested brood cells is a critical social‑immunity mechanism in honeybees, enabling colonies to detect and remove parasitized brood. This trait reduces mite reproduction and pathogen transmission and is increasingly recognized as a valuable phenotypic marker for breeding Varroa‑resistant colonies. OBJECTIVES: This study aimed to identify robust genomic markers associated with uncapping behaviour by applying a statistically rigorous GWAS framework that overcomes the potential inflated associations and limited resolution of earlier fixed‑model approache

TP53 Mutations in Acute Leukemias and Myelodysplastic Syndromes: Insights and Treatment Updates.

TP53 mutations are found in 5%-10% of de novo myelodysplastic syndrome (MDS) and AML cases. By contrast, in therapy related MDS and AML, mutations in TP53 are found in up to 30%-40% of patients. The majority of inactivating mutations observed in MDS and AML are missense mutations localized in a few prevalent hotspots. TP53 missense mutations together with truncating mutations or chromosomal loss of TP53 determine a loss-of-function effect on normal p53 function. Clonal expansion of TP53-mutant clones is observed under the selection pressure of chemotherapy or MDM2 inhibitor therapy. TP53-mutan
