Molecular identification, antifungal resistance and virulence of Cryptococcus neoformans and Cryptococcus deneoformans isolated in Seville, Spain.

Cryptococcal meningitis is one of the leading causes of death in HIV/AIDS patients. Our aim was to in order to characterise the epidemiology, antifungal susceptibility pattern and virulence of 28 Cyptococcus sp. strains recovered from 12 AIDS patients during two years in a Spanish single institution. Antifungal susceptibility testing was performed according to the CLSI protocols. Clinical strains were molecularly characterised by serotyping, mating type, PCR fingerprinting (M13 and GACA4 microsatellites) and analysis of two rDNA regions (IGS1 and ITS). Sequencing of the ERG11 gene was used to

Characterization of environmentally persistent Escherichia coli isolates leached from an Irish soil.

Soils are typically considered to be suboptimal environments for enteric organisms, but there is increasing evidence that Escherichia coli populations can become resident in soil under favorable conditions. Previous work reported the growth of autochthonous E. coli in a maritime temperate Luvic Stagnosol soil, and this study aimed to characterize, by molecular and physiological means, the genetic diversity and physiology of environmentally persistent E. coli isolates leached from the soil. Molecular analysis (16S rRNA sequencing, enterobacterial repetitive intergenic consensus PCR, pulsed-fiel

Label-Free Raman Spectroscopy Reveals Metabolic Signatures Associated with MGMT Promoter Methylation Status in Glioblastoma.

Glioblastoma (GBM) response to Temozolomide (TMZ) critically depends on O6-methylguanine-DNA methyltransferase (MGMT) promoter methylation. Clinically, the unmethylated MGMT subtype─representing the predominant GBM population─universally develops TMZ resistance, driving an urgent need for novel therapeutics. While metabolic reprogramming represents a potential therapeutic target, current methylation detection methods cannot provide targetable metabolic information. To elucidate methylation-associated metabolic signatures while mitigating complex biological background noise, we established a st

Chromatin landscape and epigenetic heterogeneity of acute myeloid leukaemia.

Acute myeloid leukaemia (AML) is an aggressive blood cancer characterized by the unregulated proliferation of immature myeloblasts. Gene mutations have been shown to have a large effect on pathogenesis, inter-tumour heterogeneity and clinical outcomes in AML1-8; however, the role of epigenetic alterations in these respects has been investigated less extensively. Here we use ATAC-seq (assay for transposase-accessible chromatin with sequencing) in a cohort of 1,563 individuals with a recent diagnosis of AML (the 'eCHROMA' cohort) to show that AML can be classified into 16 subgroups on the basis

A novel colorimetric assay for early differentiation of mucocutaneous and cutaneous leishmaniasis via species-specific identification.

Mucocutaneous leishmaniasis (MCL) is a severe and debilitating progression of cutaneous leishmaniasis (CL) that occurs when the disease spreads to involve mucosal tissues. Contrasting CL, which can often be treated with local therapies, MCL requires aggressive systemic treatment, strict adherence to a 30-day regimen and regular monitoring to prevent recurrence. These requirements highlight the critical need for accurate and rapid early diagnosis to guide effective treatment strategies. However, differentiating between the Leishmania species responsible for MCL and CL remains a significant chal

Multitargeted docking approach reveals droxidopa against DNA replication and repair-related protein of cervical cancer.

Cervical cancer begins in the cells lining the cervix and is caused by persistent infection with certain types of human papillomavirus (HPV). Initially, it has no symptoms, and later it causes pelvic pain, abnormal vaginal bleeding, and pain during intercourse. It is the fourth-ranked cancer among women, and many women die from cervical cancer every year, particularly in low-income countries and the majority could be prevented with early detection and treatment. In this study, we have taken Cervical Cancer DNA Replication and Repair-Related Protein with the PDBID- 3H15, 5VBN, and 6NT9 and perf

Water-accommodated fractions of heavy and light oils impact DNA integrity, embryonic development, and immune system of Japanese medaka at early life stages.

Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous contaminants generally found in complex mixtures. PAHs are known to cause pleiotropic effects on living organisms, including developmental defects, mutagenicity, carcinogenicity and immunotoxicity, and endocrine disruptions. The main goal of this study is to evaluate the toxicity of water-accommodated fractions (WAFs) of oils in two life stages of the Japanese medaka, larvae and juveniles. The deleterious effects of an acute exposure of 48 h to two WAFs from Arabian light crude oil (LO) and refined oil from Erika (HO) were analyzed in both

A novel histological staging of hippocampal sclerosis that is evident in gray matter loss in vivo.

INTRODUCTION: Hippocampal sclerosis of aging (HS) is defined by end-stage histological findings, strongly associated with limbic-predominant age-related TAR DNA-binding protein 43 (TDP-43) encephalopathy (LATE). We aimed to characterize features of early HS to refine the understanding of its role within combined pathology. METHODS: We studied 159 brain donations from the multimodal Vallecas Alzheimer's Center Study. A staging system (0 to IV) was developed to account for HS progression and analyzed in relation to pre-mortem cognitive and magnetic resonance imaging (MRI) data. RESULTS: Our HS

Discovery of a potent TDP1 inhibitor through machine learning-driven predictive modeling combined with structure-based virtual screening and experimental validation.

Tyrosyl-DNA phosphodiesterase I (TDP1) repairs topoisomerase I (TOP1)-mediated DNA damage and is a promising anticancer target, particularly in combination with TOP1 inhibitors. However, the discovery of potent and drug-like TDP1 inhibitors remains challenging due to the limited structural diversity of known active compounds. Here, we developed an integrated computational framework combining machine learning (ML), deep learning (DL), and structure-based docking with experimental validation. A curated dataset of 2040 compounds (857 active, 1183 inactive) was assembled and analyzed by scaffold c

Beyond antibiotics: Essential oils as novel biofilm inhibitors.

BACKGROUND: Multidrug and extensively antibiotic-resistant microorganisms, also known as "superbugs," are now becoming a global threat to mankind. Bacteria constitute a protective layer of extracellular polymeric substances (EPS) around their colonies, which produces persistent bacterial infection. Bacteria coordinate in aggregated biofilm with each other through a mechanism named quorum sensing. OBJECTIVE: This study aimed to explore the potential of essential oils as novel agents for inhibiting bacterial biofilm formation, specifically through their quorum sensing inhibitory properties. ME

Drug-Induced Epigenetic Alterations: A Set of Forensic Toxicological Fingerprints?-A Systematic Review.

BACKGROUND/OBJECTIVES: Epigenetics refers to heritable modifications in gene expression that do not involve changes to the DNA sequence. Among these, DNA methylation, histone modifications, and non-coding RNAs play a key role in regulating gene activity and are influenced by environmental factors, including exposure to psychoactive substances. In recent years, it has been hypothesized that such alterations may serve as molecular markers with forensic relevance. This systematic review aims to evaluate whether current evidence supports the use of drug-induced epigenetic changes as potential toxi

Arcanobacterium bialowiezense sp. nov. and Arcanobacterium bonasi sp. nov., isolated from the prepuce of European bison bulls (Bison bonasus) suffering from balanoposthitis, and emended description of the genus Arcanobacterium Collins et al. 1983.

A taxonomic study was performed on 13 bacterial strains isolated from preputial swabs of European bison (Bison bonasus) bulls suffering from balanoposthitis. The isolates were Gram-positive, non-motile, facultatively anaerobic, diphtheroid-shaped cells. Based on biochemical profiles and BOX-PCR-generated genomic fingerprints, the isolates were grouped into two clusters represented by four and nine strains, respectively. Strains 1(W3/01)T and 2(W106/04)T, selected as representatives of the two clusters, shared 97.2 % 16S rRNA gene sequence similarity. The highest gene sequence similarities foun

Neither gastric topological distribution nor principle virulence genes of Helicobacter pylori contributes to clinical outcomes.

AIM: Studies on Helicobacter pylori (H pylori) and gastroduodenal diseases have focused mainly on the distal sites of the stomach, but relationship with the gastric cardia is lacking. The aim of this study is to determine if the gastric topology and genotypic distribution of H pylori were associated with different upper gastrointestinal pathologies in a multiethnic Asian population. METHODS: Gastric biopsies from the cardia, body/corpus and antrum were endoscoped from a total of 155 patients with dyspepsia and/or reflux symptoms, with informed consent. H pylori isolates obtained were tested f

The ultraviolet fingerprint dominates the mutational spectrum of the p53 and Ha-ras genes in psoralen + ultraviolet A keratoses from psoriasis patients.

Psoriasis patients exposed to high cumulative doses of psoralen + ultraviolet A frequently exhibit so-called "psoralen + ultraviolet A keratoses" (i.e., hyperkeratotic lesions with varying degrees of histologic atypia). The exact causes and molecular mechanisms of psoralen + ultraviolet A keratoses however, are not clear. We therefore performed DNA mutational analysis of the tumor suppressor gene p53 (exons in psoralen + ultraviolet A keratoses from 10 long-term psoralen + ultraviolet A-treated psoriasis patients. We detected 39 p53 mutations in 16 of 28 psoralen + ultraviolet A keratoses (57%

Use of a repetitive DNA probe to type clinical and environmental isolates of Aspergillus flavus from a cluster of cutaneous infections in a neonatal intensive care unit.

Aspergillus flavus is second to A. fumigatus as a cause of invasive aspergillosis, but no standard method exists for molecular typing of strains from human sources. A repetitive DNA sequence cloned from A. flavus and subcloned into a pUC19 vector, pAF28, was used to type 18 isolates from diverse clinical, environmental, and geographic sources. The restriction fragment length polymorphisms generated with EcoRI- or PstI-digested genomic DNA and probed with digoxigenin-labeled pAF28 revealed complete concordance between patterns. Eighteen distinct fingerprints were observed. The probe was used to

Transformative Potential of Laser Induced Breakdown Spectroscopy (LIBS) for the Analysis of Crime Scene Evidences: Application in Forensic Science.

The strengthening and growth of forensic science is vital for a Nation's law enforcement and criminal justice system. Forensic agencies serve the criminal justice system by analyzing crime scene evidence, such as DNA, ballistic, gunshot residue, fingerprints, and questioned documents. With the constraint of minimal quantity of evidence recovered from crime scenes, there is a growing need for nondestructive analysis techniques that can preserve evidence for further testing or legal proceedings. The advent of Laser-Induced Breakdown Spectroscopy (LIBS) in the late twentieth century has revolutio

A Crosstalk Analysis of high-risk human papillomavirus, microbiota and vaginal metabolome in cervicovaginal microenvironment.

The microbial community has a profound effect on the host microenvironment by altering metabolites. Persistent high-risk human papillomavirus (HRHPV) infection has been implicated as contributors to the initiation and progression of cervical cancer, but the involved mechanisms are unknown. Assessing the metabolic profile of the cervicovaginal microenvironment has the potential to reveal the functional interactions among the host, metabolites and microbes in HRHPV persistence infection and progression to cancer. The vaginal swabs of women were collected and divided into three groups according t
