Electrophysiological interactions of enkephalins with neuronal circuitry in the rat hippocampus. I. Effects on pyramidal cell activity.

Effects of enkephalins on hippocampal pyramidal cell activity were studied in situ and in the in vitro hippocampal slice. Active enkephalin derivatives produced a dose-dependent naloxone-reversible excitation in both preparations whereas inactive enkephalin derivatives had no effect. Several different types of experiments, carried out in the slice, strongly suggest that this excitation is due to blockade of inhibitory pathways. First, when the pyramidal cell population spike is increased during enkephalin administration, no change is seen in the simultaneously recorded EPSP. Second, the magnit

Maternal and fetal effects of epidural analgesia during labour.

We can state that an effective pain relief during labour, like epidural analgesia, is indicated and it prevents or modifies the pain-induced harmful effects on mother and the fetus. Maternal and fetal ACTH or BE response in labour are not altered, but the increase of maternal cortisol secretion is reduced, thus probably suggesting a lesser maternal stress, to which also point the changes of PRL. Epidural analgesia prevents harmful maternal metabolic changes like hyperventilation, hypocapnia, metabolic acidosis and lactic acid accumulation. These beneficial effects are also reflected in a bette

Influence of a Combined Gluten-Free and Casein-Free Diet on Behavior Disorders in Children and Adolescents Diagnosed with Autism Spectrum Disorder: A 12-Month Follow-Up Clinical Trial.

The use of alternative interventions, such as gluten-free and casein-free (GFCF) diets, is frequent due to limited therapies for Autism Spectrum Disorder (ASD). Our aims were to determine the influence of a GFCF diet on behavior disorders in children and adolescents diagnosed with ASD and the potential association with urinary beta-casomorphin concentrations. Thirty-seven patients were recruited for this crossover trial. Each patient consumed a normal diet (including gluten and casein) for 6 months and a GFCF diet for another 6 months. The order of the intervention (beginning with normal diet

Effects of morphiceptin in the medial preoptic area on male sexual behavior.

Morphiceptin, a selective mu opioid agonist, injected into the medial preoptic area (MPOA), delayed the onset of copulation in male rats, but did not affect genital reflexes, sexual motivation or general motor activity. In a dose-dependent manner, morphiceptin (100 ng and 1000 ng) injected into the MPOA increased mount and intromission latencies. Similar injections of morphiceptin into the ventromedial hypothalamus had no effect on any parameter of copulation. The increase in copulatory latencies following the injection of the highest dose of morphiceptin was blocked by pretreatment with the o

Psychosocial stress elevates blood pressure via an opioid dependent mechanism in normotensive rats.

Stress is an important risk factor in cardiovascular diseases, including hypertension. Endogenous opioids are known to be elevated in stress and in various models of hypertension with differing etiologies. Blockade of endogenous opioids with naloxone has been demonstrated to attenuate or reverse the elevation in blood pressure in both renovascular and spontaneous hypertension. In the current study, increased blood pressure was induced using a model of psychosocial stress. During the first week of stress, systolic blood pressure rose rapidly to reach a level that was sustained throughout the re

The effect of endogenous opioids on blood pressure during stress.

1. A series of experiments were conducted to investigate the effect of endogenous opioids on blood pressure of laboratory rats during stress. 2. Rats subjected to 120 min immobilization showed a significant drop in systolic pressure which could be prevented by pretreatment injections of naloxone. 3. Adrenalectomized rats subjected to the same kind of stress showed a drop in systolic pressure equivalent to only 30% of the systolic pressure drop in the intact animals. This decrease in systolic pressure could also be prevented by pretreatment injections of naloxone. 4. It was concluded that the d

Alterations in regional concentrations of endogenous opioids following traumatic brain injury in the cat.

Delayed injury following trauma to the central nervous system (CNS) may be due to the release or activation of endogenous factors. Endogenous opioid peptides have been proposed as one such class of injury factors, based on pharmacological studies demonstrating a therapeutic effect of naloxone and other opiate receptor antagonists following CNS injury. However, changes in brain opioid concentrations following injury have not been evaluated. In the present study, we measured regional alterations in dynorphin (ir-Dyn), leucine-enkephalin (ir-Enk) and beta-endorphin immunoreactivity (ir-End) follo

Ovarian steroid involvement in endogenous opioid modulation of LH secretion in seasonally anoestrous mature ewes.

In June, 16 mature ewes were ovariectomized and allocated to four groups: 1, saline; 2, naloxone; 3, progesterone implant plus naloxone; 4, oestrogen implant plus naloxone. Steroids were implanted at the time of ovariectomy. At 5 days after ovariectomy, the animals were intravenously infused with saline for 8 h and naloxone (50 mg/h) in saline for 8 h the following day. Three intact ewes were given naloxone in a similar way. During infusions and for 8 h on the day after naloxone, jugular venous blood samples were taken every 15 min and assayed for LH. Naloxone resulted in significant increases

Neurotransmitters and biphasic respiratory response to hypoxia.

Recent work from this laboratory (J. Appl. Physiol.: Respirat. Environ. Exercise Physiol. 55:483-488, 1983) has shown that the biphasic respiratory response to hypoxia in piglets is due to changing central neural respiratory output. To test the hypothesis that either adenosine or opiatelike neurotransmitters mediate the failure to sustain hyperpnea in response to hypoxia, 12 piglets were studied at a mean age of 2.9 +/- 0.4 days (range 2-6 days). Animals were anesthetized, paralyzed, and ventilated using a servo-controlled system that maintained end-tidal CO2 constant. Electrical activity of t

Opioid antagonists and spinal reflexes in the anaesthetized cat.

In barbiturate-anaesthetized cats, intravenous naloxone (0.025-0.10 mg/kg) increased the amplitude of monosynaptic reflexes produced by electrical stimulation of the nerves to the biceps-semitendinosus and gastrocnemius muscles and the complex reflexes to electrical stimulation of myelinated afferents of the sural and tibial nerves and reflexes to electrical stimulation of unmyelinated primary afferents of the tibial nerve. Increases in reflexes were also produced by the (-)- but not the (+)-isomer of the opiate antagonist N- furylmethylnormetazocine (both isomers being given in the dose range

Neurohypophysial hormones and brain function: the neurophysiological effects of oxytocin and vasopressin.

An increasing body of evidence suggests that the neurohypophysial hormones, in addition to their classical actions, may also act as neurotransmitters. They have a widespread but discontinuous distribution in the CNS; apart from their presence in the magnocellular nuclei they may be found in the hippocampus, amygdala, septum, substantia nigra, brainstem and spinal cord. They exert profound effects on behavior, particularly on memory, a function frequently ascribed to the hippocampus, amygdala and septum; on memory consolidation, internal reward and self stimulation functions frequently ascribed

Enkephalins co-exist with oxytocin and vasopressin in nerve terminals of rat neurohypophysis.

Evidence is accumulating that opiates inhibit the release of oxytocin and vasopressin by acting on nerve terminals in the neurohypophysis. Extracts of neurohypophysis have been shown to contain substantial amounts of Met- and Leu-enkephalin, and Leu-enkephalin-immunoreactive (IR) nerve fibres originating in the magnocellular hypothalamic nuclei have been described in the neural lobe, where the two hormones are secreted. We have compared the distribution of oxytocin, vasopressin and enkephalin immunoreactivity (IR) in the neurohypophysis of the rat, and report here that Met-enkephalin-IR is inv

Characterisation of the pro opiocortin family of peptides in human cerebrospinal fluid.

Chromatography under acid dissociating conditions in conjunction with radioimmunoassay has been employed to investigate the nature of peptides related to opiocortin in human cerebrospinal fluid. Samples of cerebrospinal fluid (CSF) were collected for chromatography from 15 patients prior to air encephalography. 2 patients had pituitary dependent Cushing's disease, 3 non-endocrine neurological disease and 10 non-ACTH related pituitary disease. The column fractions were assayed for N- and C-terminal beta-lipotropin, N-terminal ACTH and gamma-MSH immunoreactivity. Elution profiles obtained from c

Opiate agonists inhibit neurogenic plasma extravasation in the rat.

Neurogenic plasma extravasation (NPE) evoked by antidromic stimulation of the saphenous nerve was inhibited by more than 50% by i.p. administration of the synthetic enkephalin analogue (D-Met2, Pro5)-enkephalinamide, 0.3-10 mg/kg and, to a lesser extent, by morphine. The effect of enkephalinamide (10 mg/kg) was reversed by naloxone (1 mg/kg, s.c.). Evidence is presented that the effect of enkephalinamide was not secondary to haemodynamic changes or alteration of mast cell function. It is suggested that opiate agonists inhibit NPE through specific opiate receptors, located peripherally on senso

[Changes in the gonadotrophins, in the prolactin and in the sexual steroid levels throughout the normal menstrual cycle (author's transl)].

The authors describe the hormone changes that occur in a normal woman throughout the menstrual cycle. These is no circadian rhythm in the release of gonadotrophins after puberty. LH is liberated in a pulsatile way throughout the day and the periodicity and amplitude of the episodes vary throughout the cycle with a notable peak of this hormone before ovulation. The level of FSH is greater early in the follicular phase and in mid-cycle, where there is also a cycle together with that of the LH. In spite of these variations a single estimation of gonadotrophins does represents their secretion over

Enkephalin-, VIP- and substance P-like immunoreactivity in the carotid body.

The carotid body type I cell contains amines and has features, both morphological and cytochemical, which indicate that it may also produce a peptide. Many regulatory peptides are now known to be present in both central and peripheral tissues. In the periphery these neuropeptides occur in both classical endocrine (APUD) cells and the neurones of the autonomic nervous system. We have now investigated the possible presence of neuropeptides in the cat carotid body using both immunocytochemistry and radioimmunoassay. Met- and Leu-enkephalin-like material occurred in considerable quantities in caro
