Prediction of femoral head collapse in osteonecrosis using 18F-fluoride positron emission tomography.

OBJECTIVE: We investigated the usefulness of (18)F-fluoride PET as a predictor of femoral head collapse in patients with osteonecrosis (ON) before radiographic changes occur. PATIENTS AND METHODS: Sixty-six hips of 42 patients diagnosed with ON of the femoral head were included in this study. We evaluated the relationship between the maximum standardized uptake value (SUV(max)) in (18)F-fluoride PET and the Ficat classification. We evaluated the relationship between SUV(max) and the presence of femoral head collapse on plain radiography at 12 months after (18)F-fluoride PET in 23 hips of 18 p

Effects of life-long fluoride intake on bone measures of adolescents: a prospective cohort study.

Controversy persists concerning the impact of community water fluoridation on bone health in adults, and few studies have assessed relationships with bone at younger ages. Ecological studies of fluoride's effects showed some increase in bone mineral density of adolescents and young adults in areas with fluoridated water compared with non-fluoridated areas. However, none had individual fluoride exposure measures. To avoid ecological fallacy and reduce bias, we assessed associations of average daily fluoride intake from birth to age 15 yr for Iowa Bone Development Study cohort members with age 1…

Integration of fluoridated implants in onlay autogenous bone grafts - an experimental study in the rabbit tibia.

INTRODUCTION: Bone augmentation before treatment with endosseous implants is a common procedure for rehabilitation of the edentulous jaw. Both machined and surface modified implants have been used in one-stage and two-stage surgery protocols with varying results and survival rates. The influence of surface modification on the integration of implants has been documented in both non-grafted and grafted bone. The aim of this study was to compare the integration and stability of surface modified fluoridated vs. machined implants when placed simultaneously with an onlay bone graft. MATERIAL AND ME

Broad-spectrum antimalarial activity of peptido sulfonyl fluorides, a new class of proteasome inhibitors.

Despite declining numbers of cases and deaths, malaria remains a major public health problem in many parts of the world. Today, case management relies heavily on a single class of antimalarial compounds: artemisinins. Hence, development of resistance against artemisinins may destroy current malaria control strategies. Beyond malaria control are elimination and eradication programs that will require drugs with good activity against acute infection but also with preventive and transmission-blocking properties. Consequently, new antimalarials are needed not only to ensure malaria control but also

Biochemical alterations induced by oral subchronic exposure to fipronil, fluoride and their combination in buffalo calves.

The effects of various pesticides and minerals on biochemical parameters have been explored in different species, but hardly any data exist regarding the combined toxicological effect of pesticides and minerals on these parameters in animals. The present study investigated the effects of fipronil and fluoride co-exposure on biochemical parameters in buffalo calves. Twenty-four healthy male buffalo calves divided into four groups were treated for 98 consecutive days. Group I, receiving no treatment served as the control. Animals of groups II and III were orally administered with fipronil @ 0.5m

Atlas of sodium fluoride PET bone scans: atlas of NaF PET bone scans.

Sodium fluoride (NaF) is a bone-seeking positron-emitting tracer with high sensitivity and specificity for detection of osseous lesions, particularly osteolytic lesions. We believe that NaF positron emission tomography (PET)/computed tomography (CT) scans can provide a more thorough and conclusive evaluation of bone diseases than conventional Technetium-99m-methylene diphosphonate bone scans. Understanding both normal and pathologic patterns is important for the evaluation and interpretation of these studies. Thus, an atlas of NaF positron emission tomography/computed tomography bone scans dem

The effect of sodium fluoride on the stability of cyanide in postmortem blood samples from fire victims.

Assigning a level of significance to cyanide concentrations found in the blood of fire victims is often hampered by the fact that cyanide is inherently unstable in cadavers and in stored blood samples. A few researchers have proposed that sodium fluoride can be used to minimize the instability of cyanide in blood samples; however, controlled studies have not been performed to support validation of this hypothesis. To test the sodium fluoride hypothesis, both treated and control blood samples from 14 autopsied fire victims were tested over a 25-30 day period. A 2% concentration of sodium fluori

Enduring fluoride health hazard for the Vesuvius area population: the case of AD 79 Herculaneum.

BACKGROUND: The study of ancient skeletal pathologies can be adopted as a key tool in assessing and tracing several diseases from past to present times. Skeletal fluorosis, a chronic metabolic bone and joint disease causing excessive ossification and joint ankylosis, has been only rarely considered in differential diagnoses of palaeopathological lesions. Even today its early stages are misdiagnosed in endemic areas. METHODOLOGY/PRINCIPAL FINDINGS: Endemic fluorosis induced by high concentrations of fluoride in water and soils is a major health problem in several countries, particularly in vol

Influence of a magnesium-fluoride coating of magnesium-based implants (MgCa0.8) on degradation in a rabbit model.

MgCa0.8 cylinders (2.5 x 25 mm(2)) were coated with a magnesium-fluoride layer and implanted in the marrow cavities of both tibiae of 10 New Zealand White rabbits. The implantation duration was 3 and 6 months. The implants were clinically well tolerated. Micro-computed tomography revealed a new bone formation at the edges of the implants as well as an endosteal and periosteal remodeling. Using EDX-analysis, a calcium and phosphorus rich degradation layer could be found on the implant surface. It was covered by an incomplete layer containing fluoride. The analysis by weight before implantation

Activation of PERK signaling through fluoride-mediated endoplasmic reticulum stress in OS732 cells.

Our proteomical analysis of osteoblasts exposed to fluoride revealed a distinctive upregulation of proteins in osteoblast. These upregulated proteins play key roles in the protein folding. The PRK-like ER kinase (PERK) signaling, one branch of unfolded protein response (UPR) to combat ER stress, is a transcription factor needed for osteoblast proliferation and differentiation. The mechanism of skeletal fluorosis by which fluoride regulates osteoblast is not fully defined. Here we studied the effect of fluoride on PERK signaling genes and x-box binding protein 1 (xbp-1) in OS7232 cells (human o

Effect of fluoride on the morphology of calcium phosphate crystals grown on acid-etched human enamel.

The aim of this study was to examine the effect of fluoride ion concentration on the morphology of calcium phosphate crystals grown on acid-etched enamel as a model for tooth enamel erosion. Samples were immersed in calcification solution for 16 h and changes in crystal morphology were monitored by field emission scanning electron microscopy. Without fluoride, plate-like octacalcium phosphate crystals (20 nm thick, 2-10 microm wide) were formed. With 1-10 mg/l fluoride, arrays of denser needle-like nanocrystals (20-30 nm wide, >500 nm in length) were formed. We conclude that there is a minimal

Developmental defects of enamel in children of Davangere District and their relationship to fluoride levels in drinking water.

To assess the prevalence and severity of developmental defects and their relationship to fluoride levels in drinking water. 739 (406 male and 333 female) schoolchildren aged 13 years formed the study population. We used the ion selective electrode method to estimate the fluoride concentration in drinking water. A modified developmental defects of enamel index was used for recording the developmental defects. The Karl Pearson coefficient for correlation and the chi-square test were used for statistical estimation. The fluoride concentration ranged from 0.64 to 2.64 ppm. The prevalence of develo

Fourier transform microwave spectroscopy and molecular structure of the 1,1-difluoroethylene-hydrogen fluoride complex.

Fourier transform microwave rotation spectra in the 7-21 GHz region are obtained for the complex formed between 1,1-difluoroethylene and hydrogen fluoride, including the normal isotopomer and two singly substituted (13)C species obtained in natural abundance. Spectra are also obtained for the analogous three species formed using deuterium fluoride. Analysis of the spectra provides rotational and hyperfine constants that are used, in combination with information from the analogous complex, 1,1-difluoroethylene-acetylene, to determine a structure for CH(2)CF(2)-HF. This structure is similar to t

Ultrastructural and immunohistochemical study of the influence of fluoride excess on the development of rat incisor tooth buds.

Little information is available on the pathogenesis of fluorosis during the fetal and initial postnatal period. In the present study, female rats received 0 (control), 7 or 100 ppm of sodium fluoride in drinking water, one week before breeding and throughout gestation and nursing periods. The hemimandibles of the offspring were collected at 0, 7 and 14 days of postnatal life (n = 5) and processed for morphological analyses by light and electron microscopy, immunohistochemical analysis for amelogenin and morphometric study of enamel matrix and ameloblasts of incisors. The results showed a decre

Effects of water fluoride exposure at crown completion and maturation on caries of permanent first molars.

Pre-eruptive fluoride exposure has been shown to be important for caries prevention. This paper aimed to determine the relative effects of water fluoride exposure during crown completion (CC) and maturation on caries experience in first permanent molars. Parental questionnaires covering residential history were linked to oral examinations of 19,885 6- to 15-year-old Australian children conducted in 1992 by the School Dental Services of South Australia and Queensland. The percentage of lifetime exposed to optimally fluoridated water at CC, maturation (MAT) and post-eruption (POST) was calculate

Durability of FTLA treatment as a medicament for dentin hypersensitivity--abrasion resistance and profiles of fluoride release.

The purpose of this study was to evaluate the durability of tubules occluded with FTLA treatment by toothbrush abrasion test on the applied surface and by measuring fluoride release from the FTLA components. Dentin specimens with simulated hypersensitive surfaces were treated with APF containing tannic acid. After which, the specimens received lanthanum-chloride-with-powdered-fluoroapatite-glass-ceramics treatment. The specimens were subjected to toothbrush abrasion test up to 6,000 strokes. SEM observation revealed that dentinal tubules of the FTLA treated specimens were completely occluded w
