Protein-based biosurfactants remain underexplored compared to glycolipids and lipopeptides, despite their unique interfacial properties and self-assembly behavior. PAC3, a surface-active protein produced by the marine fungus Acremonium sclerotigenum, exhibits dual behavior as both a biosurfactant and bioemulsifier. For this reason, it can be seen as a high molecular weight proteinaceous compound, able to efficiently reduce surface tension. Here, we identify PAC3 as the first member of a previously unrecognized family of fungal protein biosurfactants. The complete amino acid sequence of PAC3 wa

BACKGROUND: Inflammatory bowel disease (IBD) is a type of persistent chronic inflammation in the colon and can cause malignant transformation of colonic mucosa through various mechanisms. The dysbiosis of gut microbiota is closely related to the development and maintenance of IBD. Studies have shown that a proper intake of dietary fiber serves to maintain intestinal homeostasis by modulating commensal microbiomes. The present study aims to reveal the effect of soluble dietary fiber from foxtail millet bran (FMB-SDF) on chronic colitis and gut fungal community.
RESULTS: C57BL/6J mice were oral

Chinese Distillers' grains (CDGs), a typical agro-industrial byproduct, contain a highly recalcitrant lignin that limits the utilization. In this study, a dual-fungal solid-state fermentation system consisting of Phanerochaete chrysosporium and Pleurotus eryngii was constructed for lignin conversion in CDGs. The lignin degradation ratio reached 58.66%, indicating that the system can effectively convert lignin. Laccase (Lac) and manganese peroxidase (MnP) activities peaked at 10 d, reaching 38.54 and 50.36 U/g, respectively, whereas lignin peroxidase (LiP) reached its maximum activity (51.67 U/

BACKGROUND: Mycobacterium tuberculosis (Mtb) remains a leading cause of global morbidity and mortality. Viral, bacterial, fungal, and parasitic co-infections substantially modify tuberculosis (TB) pathogenesis, and an increasingly recognised population of TB survivors develops post-TB lung disease (PTLD), which predisposes them to recurrent respiratory infections.
OBJECTIVES: To summarise current evidence on 1) Mtb pathophysiology and immunopathogenesis; 2) mechanisms by which major viral, bacterial, fungal, and parasitic co-infections alter TB progress; and 3) infectious complications associ

Fungal antagonism shapes microbial community structure and provides an ecological basis for biological control. Although mycoviruses have been implicated in fungal antagonism, their effects are generally attributed to indirect modulation of resident host physiology. Here, we show that a mycovirus can function as a mobile effector of antagonism through cross-taxonomic transmission, representing the first documentation of viral transfer from an entomopathogenic/endophytic fungus (Beauveria) to a phytopathogenic fungus (Sclerotinia) via a plant‑mediated route. We characterized Beauveria brongniar

For the first time, a comprehensive analytical approach is introduced that simultaneously quantifies a metabolic precursor (tryptophan), a quorum-sensing biomarker for fungal infections (tryptophol), and an antifungal drug (voriconazole) within a single platform using reversed-phase high-performance liquid chromatography coupled with diode array detection (HPLC-DAD). The method utilizes a Pursuit PFP column featuring a unique pentafluorinated structure, with a mobile phase of methanol: water (60:40, v/v), a flow rate of 1.0 mL/min, and a detection wavelength set at 254.0 nm. BACKGROUND/OBJECTIVES: Small Intestinal Bacterial Overgrowth (SIBO) and Small Intestinal Fungal Overgrowth (SIFO) are distinct yet often overlapping conditions characterized by an abnormal increase in microbial populations within the small intestine. SIBO results from an overgrowth of colonic bacteria, while SIFO is driven by fungal overgrowth, primarily involving Candida species. Both conditions present with nonspecific gastrointestinal (GI) symptoms such as bloating, abdominal pain, diarrhea, and malabsorption, making differentiation between SIBO and SIFO challenging. This review aims to elu

The global incidence of invasive fungal infections (IFIs) has increased over the past few decades, mainly in immunocompromised patients, and is associated with high mortality and morbidity. Aspergillus fumigatus is one of the most common and deadliest IFI pathogens. Major hurdles to treating fungal infections remain the lack of rapid and definitive diagnosis, including the frequent need for invasive procedures to provide microbiological confirmation, and the lack of specificity of structural imaging methods. To develop an Aspergillus-specific positron emission tomography (PET) imaging agent, w

Natural polymers have recently been investigated for various applications, such as 3D printing and healthcare, including treating infections. Among microbial infections, fungal diseases remain overlooked, with limited therapeutic options and high recurrence. Cutaneous cryptococcosis is an opportunistic fungal infection triggered by mechanical inoculation or hematogenous dissemination of the yeast that causes cryptococcal pneumonia and meningitis. Every year, Cryptococcus neoformans endanger the lives of immunosuppressed hosts, resulting in 180,000 deaths per year. Nonetheless, healthy individu

Arbuscular mycorrhizal (AM) fungi play an important role in organic matter (OM) stabilization in Fe ore tailings for eco-engineered soil formation. However, little has been understood about the AM fungi-derived organic signature and organo-mineral interactions in situ at the submicron scale. In this study, a compartmentalized cultivation system was used to investigate the role of AM fungi in OM formation and stabilization in tailings. Particularly, microspectroscopic analyses including synchrotron-based transmission Fourier transform infrared (FTIR) and scanning transmission X-ray microspectro

Infections caused by the fungal pathogen Aspergillus fumigatus are increasingly resistant to first-line azole antifungal drugs. However, despite its clinical importance, little is known about how susceptible patients acquire infection from drug-resistant genotypes in the environment. Here, we present a population genomic analysis of 218 A. fumigatus isolates from across the UK and Ireland (comprising 153 clinical isolates from 143 patients and 65 environmental isolates). First, phylogenomic analysis shows strong genetic structuring into two clades (A and B) with little interclade recombination

Fungi from the Candida genus are widespread commensals and, at the same time, are the leading cause of fungal infections worldwide. For instance, vulvovaginal candidiasis (VVC) affects approximately 75% of women at least once in their lifetime, remaining the second most common gynecological infection. On the contrary, hospital-acquired fungal bloodstream infections (BSIs), although less frequent, are characterized by a high mortality rate. Undoubtedly, the main reason for this situation are virulence factors that these yeast-like fungi can produce, and the ability to form a biofilm is one of t

Oxygen is fundamental to the life of aerobic organisms and is not always available to Paracoccidioides cells. During the life cycle stages, reduced oxygen levels directly affect general metabolic processes and oxygen adaptation mechanisms may play a fundamental role on fungal ability to survive under such condition. Heme proteins can bind to oxygen and participate in important biological processes. Several fungi, including Paracoccidioides, express a heme-binding globin (fungoglobin - FglA) presumable to regulate fungal adaptation to hypoxia. However, the characterization of fungoglobin in Par

Sexual mating of compatible sporida is essential for Sporisorium scitamineum to form dikaryotic mycelia and then cause infection on sugarcane. Our previous work identified a Pseudomonas sp. ST4 from a soil sample, which showed a promising biocontrol potential by inhibiting the mating of S. scitamineum sporida and hyphal growth. In this study, we set to isolate the active compounds from Pseudomonas sp. ST4 through solid fermentation. High-performance liquid chromatography (HPLC) separation coupling with bioassay showed that Pseudomonas sp. ST4 produced a range of antimicrobial compounds. Two of

Cryptococcus neoformans, the encapsulated yeast acquired through inhalation, remains localized in lungs, but harbours the CNS in immunocompromised individuals. Several treatment regimes have failed combating this disease totally, but long-term usage of drugs leads to organ damage. As T11-target structure (T11TS) has documented profound immune potentiation, we aimed to investigate the role of microglia, pivotal immune cells of brain in ameliorating cryptococcosis, with T11TS immunotherapy. Murine model with C neoformans infection was prepared by intraperitoneal injection and the brains of rats

Candida dubliniensis is an important human fungal pathogen that causes oral infections in patients with AIDS and diabetes mellitus. However, C. Dubliniensis has been frequently reported in bloodstream infections in clinical settings. Like its phylogenetically related virulent species C. albicans, C. Dubliniensis is able to grow and switch between yeast form and filamentous form (hyphae) and develops biofilms on both abiotic and biotic surfaces. Biofilms are recalcitrant to antifungal therapies and C. Dubliniensis readily turns drug resistant upon repeated exposure. More than 80% of infections

We describe bilateral Encephalitozoon hellem keratoconjunctivitis in a 39-year-old immunocompetent woman with two years of daily close contact with a pet parrot. She presented with a three-week history of bilateral ocular redness, itching, foreign-body sensation, and blurred vision that had not improved with topical fluorometholone, lubricants, and levofloxacin prescribed for presumed dry eye disease. Slit-lamp examination showed bilateral conjunctival inflammation and diffuse superficial punctate corneal infiltrates. Giemsa staining of a left-eye corneal scraping demonstrated oval spore-like
