Spontaneous Abortion and Myocardial Infarction: A Mendelian Randomization Investigation and Transcriptomic Analysis.

BACKGROUND: A link has been found between spontaneous abortion (SA) and myocardial infarction (MI). However, there is still a lack of comprehensive knowledge regarding the genetic links and biological mechanisms between SA and MI. An investigation of the causal association between SA and MI, along with the associated signaling networks, was conducted using univariate Mendelian randomization (MR) and transcriptome analysis. METHODS: Data from genome-wide association studies (GWAS) for SA and MI were analyzed using the FinnGen consortium database. To assess the causality between SA and MI, vari

Causal Association Between Inflammatory Factors and Hypertrophic Scar: A Two-Sample Mendelian Randomization Study.

BACKGROUND: Hypertrophic scars result from abnormal healing following skin injuries. AIM: To delve deeper into the causal association between inflammatory factors and hypertrophic scars. METHODS: This study utilized genetic data from the FINN cohort and pertinent literature to scrutinize the nexus between a spectrum of inflammatory factors-encompassing IL-1β, interleukin 1 receptor-like 1, MCP1, RANTES/CCL5, TNFα, IL-8, IL-18, and CTACK/CCL27-and the risk of hypertrophic scarring. Our analytical strategy was based on the inverse variance weighted (IVW) approach, further bolstered by MR-Egger

Blunted cortisol as a biomarker of depression based on the attenuation hypothesis: A Mendelian randomization analysis using depression as exposure.

BACKGROUND: Both elevated and blunted cortisol responses have been associated with depression. Previous Mendelian randomization (MR) studies have largely ruled out cortisol as a cause of depression. Based on the attenuation hypothesis, this MR study used depression as exposure to assess whether cortisol might be a consequence and therefore a biomarker of depression. METHODS: Strong (P < 5 × 10-8) and independent (r2 < 0.001) single nucleotide polymorphisms (SNPs) associated with broadly defined depression (294,322 cases, 741,438 controls) were used as instruments. These were applied to geneti

Exploring the causal relationship between 25-hydroxyvitamin D and asthma: A bidirectional Mendelian randomization study.

The role of 25-hydroxyvitamin D (25(OH)D) in disease has attracted much attention. However current research findings on the relationship between 25(OH)D and asthma are not completely consistent. This study aims to explore the potential causal relationship between 25(OH)D levels and asthma using a bidirectional Mendelian randomization (MR) approach. Data from large genome-wide association studies related to the study phenotypes were utilized. Genetic correlation data for 25(OH)D were obtained from the European Bioinformatics Institute database, and genetic correlation data for asthma risk were

Causality of immune cells on primary sclerosing cholangitis: a bidirectional two-sample Mendelian randomization study.

BACKGROUND: Observational studies have indicated that immune dysregulation in primary sclerosing cholangitis (PSC) primarily involves intestinal-derived immune cells. However, the causal relationship between peripheral blood immune cells and PSC remains insufficiently understood. METHODS: A bidirectional two-sample Mendelian randomization (MR) analysis was implemented to determine the causal effect between PBC and 731 immune cells. All datasets were extracted from a publicly available genetic database. The standard inverse variance weighted (IVW) method was selected as the main method for the

Appraising the causal association among depression, anxiety and intracranial aneurysms: Evidence from genetic studies.

BACKGROUND: The risk of intracranial aneurysms (IAs) is increased in individuals with depression and anxiety. This indicates that depression and anxiety may contribute to the development of physical disorders. Herein, to investigate the association between genetic variants related to depression and anxiety and the risk of IA, two-sample Mendelian randomization was performed. METHODS: The genome-wide association study (GWAS) comprised genome-wide genotype data of 2248 clinically well-characterized patients with anxiety and 7992 ethnically matched controls from four European countries. Sex-spec

Causal association between circulating inflammatory markers and sciatica development: a Mendelian randomization study.

BACKGROUND: This research explores the causal association between circulating inflammatory markers and the development of sciatica, a common and debilitating condition. While previous studies have indicated that inflammation may be a factor in sciatica, but a thorough genetic investigation to determine a cause-and-effect relationship has not yet been carried out. Gaining insight into these interactions may uncover novel treatment targets. METHODS: We utilized data from the OpenGWAS database, incorporating a large European cohort of 484,598 individuals, including 4,549 sciatica patients. Our s

Genetic variation reveals the influence of steroid hormones on the risk of retinal neurodegenerative diseases.

It is difficult to get evidence from randomized trials of a causal relationship between steroid hormones produced by the adrenal gland and gonad and retinal neurodegenerative disorders (RND). In this study, genetic variations of aldosterone (Aldo), androstenedione (A4), progesterone (P4), hydroxyprogesterone (17-OHP), and testosterone/17β-estradiol (T/E2) were obtained from genome-wide association studies as instrumental variables. Mendelian randomization (MR) analysis was used to assess the impact on the risk of RND, including glaucoma (8,591 cases and 210,201 controls), diabetic retinopathy

Bidirectional association between hypothyroidism and myasthenia gravis: a Mendelian randomized study.

OBJECTIVES: Although observational studies have suggested a link between hypothyroidism and myasthenia gravis (MG), a causal relationship has not been established. We aimed to investigate the causal association using a two-sample Mendelian randomization (MR) study. METHODS: Using summary statistics from genome-wide association studies involving 494,577 and 38,243 individuals, single-nucleotide polymorphisms exhibiting no linkage disequilibrium (r2 ≤ 0.001) and displaying significant differences (p ≤ 5 × 10-8) were selected for hypothyroidism and MG. To assess the potential causality relations

Functional variants in the LRRK2 gene confer shared effects on risk for Crohn's disease and Parkinson's disease.

Crohn's disease (CD), a form of inflammatory bowel disease, has a higher prevalence in Ashkenazi Jewish than in non-Jewish European populations. To define the role of nonsynonymous mutations, we performed exome sequencing of Ashkenazi Jewish patients with CD, followed by array-based genotyping and association analysis in 2066 CD cases and 3633 healthy controls. We detected association signals in the LRRK2 gene that conferred risk for CD (N2081D variant, P = 9.5 × 10-10) or protection from CD (N551K variant, tagging R1398H-associated haplotype, P = 3.3 × 10-8). These variants affected CD age of

Investigating Causal Links Between Uterine Prolapse, Urinary Tract Infections, and Lower Urinary Tract Symptoms: A Two-Sample Mendelian Randomization Study.

BACKGROUND: Uterine prolapse (UP), a subtype of pelvic organ prolapse (POP), frequently coexists with urinary tract infections (UTIs) and lower urinary tract symptoms (LUTS), including urinary frequency (UF), urinary incontinence (UI), and urinary retention (UR), is a common pelvic floor dysfunction disorder in women. However, the causal relationship between these conditions remains controversial. This study aims to fill this gap by applying two-sample Mendelian randomization. METHODS: A two-sample Mendelian randomization (MR) study was conducted to genetically evaluate associations between U

How aging causes osteoarthritis: An evolutionary physiology perspective.

Late-life diseases result from the poorly understood process of senescence (aging), which is largely genetically determined. According to a recently proposed evolutionary physiology-based account, the multifactorial model, senescence is largely caused by evolved but non-adaptive programmatic mechanisms specified by the wild-type (i.e. normal) genome. These act together with disruptions to wild-type function (due e.g. to infectious pathogens, mechanical injury and malnutrition) in a variety of combinations to generate diverse late-life diseases. Here, I explore the utility of this model by test

Identifying the Genetic Association Between Sjogren's Syndrome and Autoimmune Thyroid Disease: A Bidirectional Two-Sample Mendelian Randomization Study.

BACKGROUND: Although observational studies have suggested a correlation between Sjogren's syndrome (SS) and autoimmune thyroid disease (AITD), a conclusive evidence supporting a causal relationship is still lacking. This study aims to explore the potential causal relationship between SS and AITD. METHODS: Using genome-wide association studies, we performed a bidirectional two-sample Mendelian randomization (MR) analysis. In our analysis, the random-effects inverse variance weighted (IVW) method was predominantly employed, followed by several sensitivity analyses, which include heterogeneity,

Changes in iron load in specific brain areas lead to neurodegenerative diseases of the central nervous system.

The causes of neurodegenerative diseases remain largely elusive, increasing their personal and societal impacts. To reveal the causal effects of iron load on Parkinson's disease (PD), Alzheimer's disease (AD), amyotrophic lateral sclerosis and multiple sclerosis, we used Mendelian randomisation and brain imaging data from a UK Biobank genome-wide association study of 39,691 brain imaging samples (predominantly of European origin). Using susceptibility-weighted images, which reflect iron load, we analysed genetically significant brain regions. Inverse variance weighting was used as the main est

Fifty years of recognizable patterns of human malformation: Insights and opportunities.

Now in its 7th edition, Smith's Recognizable Patterns of Human Malformation was first published in 1970. This 1st edition comprised 135 "dysmorphic syndromes of multiple primary defects" and 12 "single syndromic malformations resulting in secondary defects." Of the former, other than a few chromosomal and environmental disorders, most were heritable conditions of then unknown etiology. In 2021, the majority of these conditions are now "solved," a notable exception is Hallermann-Streiff syndrome. The "solved" conditions were typically clinically delineated decades prior to understanding the und

Cumulative Incidence in Monogenic Alzheimer's Disease and Frontotemporal Dementia: Gene-Gene Interaction Effect.

Monogenic forms of Alzheimer's Disease (AD) and Frontotemporal Dementia (FTD) represent the two principal neurodegenerative disorders leading to early-onset dementia, primarily linked to mutations in key AD- and FTD-associated genes. The marked heterogeneity in age at onset and penetrance among carriers of pathogenic mutations suggests that monogenic variants act within a broader polygenic background. The combined impact of AD- and FTD-related genetic variation on disease incidence in monogenic forms remains largely unexplored. Herein, we investigate gene-gene interaction patterns in monogenic

Genetic correlations and causal associations between BMI, HDL-C, and postoperative infections: a two-sample Mendelian randomization study.

Infections are serious postoperative complications, and strongly affects the mortality and prognosis of patients. Body mass index (BMI) and lipids are factors in postoperative infection, but a causal relationship has not been know. In this Mendelian randomization (MR) study, we utilized genome-wide association study (GWAS) data from the Global Lipids Genetics Consortium and FinnGen database, treating lipids and BMI as exposures. Postoperative infection GWAS data from the UK Biobank served as the outcome. We utilized linkage disequilibrium score regression (LDSC) analysis to evaluate the geneti

Associations between lipid-lowering drugs and urate and gout outcomes: a Mendelian randomization study.

OBJECTIVE: Gout is a common inflammatory arthritis and lipid metabolism plays a crucial role in urate and gout. Potential associations between urate and gout and lipid-lowering drugs have been revealed in observational studies. However, the effects of lipid-lowering drugs on urate and gout remain controversial. The aim of this study was to investigate the genetic association between lipid-lowering drugs and urate and gout. METHODS: In this study, two genetic proxies were employed to approximate lipid-lowering drug exposure: expression quantitative trait loci (eQTL) associated with drug-target

Characterization of genetic and phenotypic heterogeneity of obstructive sleep apnea using electronic health records.

BACKGROUND: Obstructive sleep apnea (OSA) is defined by frequent episodes of reduced or complete cessation of airflow during sleep and is linked to negative health outcomes. Understanding the genetic factors influencing expression of OSA may lead to new treatment strategies. Electronic health records (EHRs) can be leveraged to both validate previously reported OSA-associated genomic variation and detect novel relationships between these variants and comorbidities. METHODS: We identified candidate single nucleotide polymorphisms (SNPs) via systematic literature review of existing research. Usi
