Rethinking the trade-off hypothesis in evolutionary epidemiology.

The trade-off hypothesis plays a foundational role in the theoretical framework of modern evolutionary epidemiology, proposing that pathogen fitness is maximized at intermediate levels of virulence. Despite its influence on the study of host-pathogen interactions, the hypothesis has been criticized for its limited empirical support and the conceptual ambiguities surrounding the definition of virulence. This paper reassesses the scientific status of the trade-off hypothesis by interpreting the dependencies among epidemiological parameters as constitutive principles. Therefore, the results of th…

Fungal MicroRNA-Like RNAs Facilitate Convergent Attack on a Host Immune Signalling Network.

Cross-kingdom RNA interference has emerged as an important mechanism in plant-pathogen interactions, yet how fungal small RNAs coordinate host immune suppression remains unclear. Here, we demonstrate that Alternaria alternata f. sp. mali, the causal agent of apple leaf spot, suggests microRNA-like RNAs (milRNAs) into host tissues in association with extracellular vesicles (EVs). Small RNA sequencing of fungal EVs identified multiple milRNAs, among which AamilR251, AamilR292 and AamilR004 were the three most abundant species. These AamilRNAs target three apple (Malus domestica) immune-related k

Potato purple top phytoplasma infection induces autophagy-associated lipid dynamics that support pathogen proliferation.

Phytoplasmas are unculturable, phloem-restricted bacterial pathogens responsible for devastating diseases in crops and ornamentals worldwide. Their mechanism for nutrient acquisition from host plants remains largely unknown. This study demonstrated that infection with potato purple top phytoplasma induced extensive remodeling of lipid metabolism in tomato plants, closely linked to autophagy activation. Western blot and confocal analyses revealed increased ATG8 lipidation and autophagosome formation at endoplasmic reticulum stress sites, alongside the redistribution of lipid droplets toward phy

Tomato Spotted Wilt Virus Reprogrammes Host Glycolysis to Facilitate Proliferation by a Phase-Separated Co-Aggregate of Nucleocapsid Protein and Phosphoglycerate Kinase.

Efficient viral proliferation within the host is a critical step in pathogenicity and requires adenosine triphosphate (ATP). The replication, movement and immune evasion of many plant viruses within their hosts are associated with phase separation (PS)-derived aggregates formed by viral components. However, the host factors that drive the formation of these condensates remain largely unknown. This study provides evidence that the nucleocapsid protein (N) of tomato spotted wilt virus (TSWV) recruits the host factor phosphoglycerate kinase (NbPGK) from Nicotiana benthamiana to form phase-separat

Insect immune systems: same same but different but still same.

Insects are the most diverse group of animals in nature, occupying nearly every ecological niche and playing central roles as pollinators, pests, and disease vectors. Despite this vast diversity, insects rely on a set of conserved yet evolutionarily adaptable immune pathways to defend against pathogens. Early studies in insect immunity have laid the foundation for human immunology, and recent advances in genomic and transgenic technologies have renewed interest in understanding how immune responses vary across insect orders. Insects are highly diverse in their immune systems; each species has

Exploring the roles of fungal extracellular vesicles in fungal pathogenesis and symbiosis: Current understandings and future perspectives.

Fungal extracellular vesicles (FEVs) have emerged as sophisticated emissaries in cross-kingdom communication, redefining our understanding of fungal secretome in pathogens like Cryptococcus neoformans and Candida species. Rather than mere cellular debris, these bilayered structures are actively exported across the fungal cell wall via complex biogenesis mechanisms, carrying a potent cargo of virulence factors, genetic material, proteins, lipids, and polysaccharides. In human hosts, FEVs facilitate myco-pathogenesis by interacting directly with immune cells to modulate the immune response, eith

Mpox virus immunology: exploring links to autoimmune diseases.

The reemergence of the mpox virus (MPXV) as a global health threat, with >100 000 cases reported across 115 countries since the WHO's 2022 alert, has intensified research into its pathophysiology. MPXV exists in two main clades with distinct geographic and clinical profiles. Despite progress, the impact of MPXV infection in patients with autoimmune diseases remains underexplored. Emerging data highlight immunomodulatory MPXV proteins that suppress innate immunity and dysregulate adaptive responses, through inhibition of type I interferons, nuclear factor kappa-B, and tumor necrosis factor-α. I

Adaptive Evolution of Dengue Virus: Interplay Between Viral Genetics and Host Immunity.

Dengue virus (DENV), a major arboviral pathogen of global concern, evolves rapidly through a combination of intrinsic molecular mechanisms, vector adaptation, and host immune pressures. Despite extensive research, major gap remains in understanding how these forces, collectively generate, shape, and fix adaptive mutations that drive DENV evolution. In this review, we explore the molecular mechanisms underlying DENV evolution, with emphasising the error-prone RNA-dependent RNA polymerase, template switching, recombination, and the modulatory effects of viral RNA secondary structures. We also hi

Unraveling dengue virus-host interactions through transcriptomics: Insights into disease progression and severity signatures.

Dengue virus (DENV) remains a major global health concern, characterized by complex virus-host interactions that are not yet fully understood. Advances in transcriptomic technologies have become crucial for uncovering the molecular mechanisms underlying DENV infection. This review summarizes recent transcriptomic studies, spanning microarrays, bulk RNA sequencing (RNA-seq), single-cell RNA sequencing (scRNA-seq), virus-inclusive single-cell RNA-seq (ViscRNA-seq), and spatial transcriptomics. that have deepened our understanding of how DENV modulates host gene expression. These approaches have

HAdV55 reprograms host 3D genome architecture and mitochondrial metabolism to drive pathogenesis.

Human adenovirus type 55 (HAdV55), an emerging recombinant pathogen linked to severe respiratory outbreaks, exhibits heightened virulence and replication efficiency, yet its mechanisms of host interaction remain poorly resolved. Here, we integrate transcriptomic and 3D genomic analyses to define the spatiotemporal interplay between HAdV55 and A549 cells. Temporal profiling revealed triphasic host transcriptional reprogramming, with distinct clusters of genes governing cell survival, apoptosis, and innate immunity dynamically modulated during infection. Crucially, HAdV55 reshapes the 3D chromat

Infected host competence overshadows heterogeneity in susceptibility in shaping experimental epizootics.

The accelerated rate of disease emergence in recent decades underscores the need to understand conditions that promote or dampen epidemics. Theoretical models consistently show that epidemics are smaller in populations with higher among-individual heterogeneity in susceptibility. Experimental tests of these predictions are rare but critical for understanding how heterogeneity in susceptibility shapes epidemics in natural systems. We directly link data-parameterized models from previous dose response experiments in the house finch and Mycoplasma gallisepticum system to experimental epidemics in

A genome-wide CRISPR screen defines host determinants of early Brucella infection in human macrophage-like cells.

Brucella spp. are widespread intracellular animal pathogens that cause brucellosis, a significant zoonosis. Despite the global impact of brucellosis on animal and human health, the host genes that support Brucella infection remain incompletely defined. To address this knowledge gap, we developed a flow cytometry-based infection assay with fluorescent Brucella and performed a genome-wide CRISPR-Cas9 loss-of-function screen in human macrophage-like cells. Disruption of >150 host genes significantly reduced intracellular B. abortus signal at 3 hours post-infection. In addition to recovering known

Immuno-phage synergy driven by fitness costs: turning bacterial phage resistance into therapeutic gain through evolutionary traps.

The host immune system typically recognizes administered bacteriophages as foreign entities, potentially acting as a rate-limiting factor in phage therapy via neutralizing antibody production and immune-mediated clearance. Furthermore, the emergence of bacterial phage resistance during treatment remains a major hurdle for its clinical application. Although employing phage cocktails is the standard strategy to suppress phage resistance, clinical trials occasionally report the emergence of phage-resistant variants despite cocktail treatments. Therefore, a more proactive strategic approach that g

Beneath the Bark: Molecular Determinants of Adaptation and Infection in Bacterial Pathogens of Woody Hosts.

Bacterial pathogens of woody plants represent a pervasive yet underexplored threat to global agriculture and forestry, causing devastating diseases in fruit trees, shrubs, forest trees and ornamental species. Unlike pathogens of herbaceous plants, the molecular strategies enabling bacteria to colonise and damage woody tissues remain poorly understood, owing to the experimental challenges associated with woody host systems and the distinctive biology of these plants. Moreover, persistence within long-lived woody tissues requires prolonged modulation of host immune signalling, enabling pathogens

Variable Staphylothrombin Activity in Staphylococcus aureus Bloodstream Infections.

BACKGROUND: Although manipulation of host hemostasis is a central mechanism in Staphylococcus aureus virulence, clinically relevant phenotypes of S aureus coagulase activity and the variability thereof in the context of bloodstream infections are unknown. METHODS: We created a high-throughput staphylothrombin activity and growth fitness assay to characterize fibrin formation and S aureus replication by adding bacterial culture supernatant to human blood plasma. Supernatant is rich in bacterial proteins, including coagulases secreted during growth, but it contains relatively few viable cells.

The host immune response to Mycobacterium tuberculosis determining protection or disease progression.

Tuberculosis (TB) remains a global health issue and leading cause of death. Understanding the immune response to Mycobacterium tuberculosis infection is critical to advance TB control. Although immune factors involved in protection against TB have long been identified, these cannot predict the efficacy of TB vaccines, nor which individuals will control the infection or progress to active TB disease. This is especially challenging given the complexity of M. tuberculosis infection states and the breadth of TB disease severities, which is increasingly apparent from basic, clinical and translation

The ppsA-ppsE gene cluster in Mycobacterium bovis: structure, function, and role in virulence.

Phthiocerol dimycocerosates (PDIMs) are hallmark virulence-associated lipids within the Mycobacterium tuberculosis complex, including M. bovis. The ability to produce PDIMs depends on the presence of the ppsA-ppsE gene cluster, which harbors a series of modular type I PKS that coordinate the phthiocerol backbone of PDIMs. In this review, the genomic organization, biochemical function, and pathogenic relevance of the ppsA-ppsE cluster in M. bovis have been studied, integrating evidence from comparative genomics, lipid biochemistry, and host-pathogen interaction. In the M. tuberculosis complex,

Complement Resistance Assays.

Like many other pathogens of medical importance, pathogenic Leptospira employ diverse strategies to circumvent Complement System activation. Under physiological conditions, this central humoral arm of innate immunity is tightly controlled by negative Complement regulatory proteins. However, upon infection, pathogenic microorganisms interfere with normal Complement host defense mechanisms by recruiting or mimicking Complement regulators, and by secreting endogenous proteases or acquiring host's proteases that inactivate key Complement components. In this chapter, we describe in detail some of t

Oncoviruses at the Crossroads of Ageing and Cancer: Molecular Mechanisms and Key Signalling Pathways.

Oncoviruses are responsible for a substantial fraction of human cancers and have emerged as critical modulators of host cellular pathways that also govern ageing. The convergence of viral oncogenesis, ageing, and cancer reflects shared molecular mechanisms, including genomic instability, epigenetic alterations, telomere attrition, and chronic inflammation. Increasing evidence indicates that oncoviruses such as human papillomavirus, Epstein-Barr virus, hepatitis B and C viruses, and Kaposi's sarcoma-associated herpesvirus actively reprogramme host signalling networks to promote persistent infec
