Relative to non-human primates, in humans the cerebellum, and prefrontal cortex are brain regions which have undergone major evolutionary changes. In recent decades, progress in molecular biology and advances in the development of functional neuroimaging analysis have shown that the evolution of the human cerebellum was accompanied by the acquisition of more functions than were previously deduced from human post-mortem studies and animal experimentation. These new cerebellar functions included the control of attention and other cognitive functions, emotions and mood, and social behavior, which

Three cases of Rift Valley fever in human individuals are reported. The virus was recovered from the respiratory tract of the patients and was transmitted to ferrets by the intranasal route. The experimental disease so produced in ferrets is characterized by fever, marked pulmonary lesions, and hemorrhagic phenomena. The results indicate that the virus of Rift Valley fever belongs to the group of filterable viruses which may gain entrance to the human body through the respiratory tract. The differential diagnosis of Rift Valley fever and influenza is discussed. While, clinically, this is a dif

It is now apparent that HIV infection leads to a gradual collapse of a complex system of lymphoid tissue. This collapse tends to be associated with a change in virus tropism from macrophages to T lymphocytes, and a change in phenotype from nonsyncytium inducing (NSI) to syncytium inducing (SI). An experimental system is required to study the role of this change in HIV pathogenesis in lymphoid tissue. Here we describe such a system. Histocultures of human lymphoid tissue preserve their general cytoarchitecture, including a network of follicular dendritic cells (FDCs). Histocultures of tonsils,

Praziquantel is the current drug of choice for human schistosomiasis. Recent reports from laboratory and field studies concerning reduced praziquantel efficacy against Schistosoma mansoni have generated some controversy. The prevailing question is whether the emergence of strains of schistosome resistant to praziquantel is a fact, or an artifact resulting from erroneous field or laboratory experimentation. In this article, Padraic Fallon, Liang-feng Tao, Magdi Ismail and James Bennett examine the available evidence for schistosome resistance to praziquantel. Contributory factors to the schisto

Direct-fed probiotics in poultry can improve animal nutrition and health while also competing with human foodborne pathogens for colonization of the gastrointestinal tract. The objective of this study was to perform whole-genome sequencing on lactic acid bacteria isolated from commercial chicken litter to characterize genetic markers associated with probiotic potential. Three Ligilactobacillus salivarius strains were selected for whole-genome sequencing using Oxford Nanopore Technology. The genomes contained genes associated with sialic acid utilization, bacteriocin production, and antimicrobi

No useful purpose is served by developing therapeutic interventions that are applicable only in nonexistent patient populations. The history of laboratory hemorrhagic shock research may be a case in point because although many interventions have been proposed on the basis of animal experimentation, few if any have found a place in the treatment of human beings. For a laboratory shock model to have clinical relevance, it must replicate important aspects of shock as seen in human beings during or following massive blood loss. The difficulty in developing an animal model that incorporates these h

Animal models have long been the foundation of biomedical research and experimentation. However, the utility of animal models in both research and preclinical testing of drugs has been challenged by the insufficiency and cost of such methods, including the fact that over 90 % of preclinically successful compounds ultimately fail in human trials, as well as concerns about irreproducibility, scalability, and ethics in animal studies; their limitations are increasingly recognized. Advances in artificial intelligence (AI) now offer a promising complementary approach through the creation of "virtua

BACKGROUND: Surfactants elicit alterations in the stratum corneum. Predictive tests that avoid animal experimentation are needed.
OBJECTIVE: This study compares three methods of rating and predicting shampoo-induced irritation.
METHODS: Corneosurfametry entails collection of stratum corneum followed by brief contact with diluted surfactants and measurement of variations in staining of samples.
RESULTS: Corneosurfametry appears to correlate well with in vivo testing in volunteers with sensitive skin. However, corneosurfametry presents less interindividual variability than in vivo testing and

Both genetic and environmental factors contribute to autoimmune disease development. Previously, we evaluated genetic factors in a humanized mouse model of Hashimoto's thyroiditis (HT) by immunizing human leucocyte antigen DR3 (HLA-DR3) and HLA-DQ8 transgenic class II-knock-out non-obese diabetic (NOD) mice. DR3+ mice were susceptible to experimental autoimmune thyroiditis (EAT) induction by both mouse thyroglobulin (mTg) and human (h) Tg, while DQ8+ mice were weakly susceptible only to hTg. As one environmental factor associated with HT and tested in non-transgenic models is increased sodium

Differentiation of human neural progenitors into neuronal and glial cell types offers a model to study and compare molecular regulation of neural cell lineage development. In vitro expansion of neural progenitors from fetal CNS tissue has been well characterized. Despite the identification and isolation of glial progenitors from adult human sub-cortical white matter and development of various culture conditions to direct differentiation of fetal neural progenitors into myelin producing oligodendrocytes, acquiring sufficient human oligodendrocytes for in vitro experimentation remains difficult.

Current vaccine development efforts encompass diverse and innovative approaches; however, studies with basic vaccines (e.g., whole cell inactivated) continue to inform these efforts. Perhaps the world's most effective and durable human vaccine, the query (Q) fever vaccine known as Q-VAX, can trigger a severe post-vaccination hypersensitivity reaction, precluding its widespread deployment. The history of Q fever vaccine development serves as a rich example of the power of scientific collaboration, elegant experimentation, and the study of adverse post-vaccination events. Here, we review the his

Since 1978, in the first decades of in vitro fertilization (IVF), the use of ovarian hyperstimulation allowed for the development and transfer of multiple embryos. As IVF technology improved, the number of multiple pregnancies increased, which led to gradual reduction in the number of embryos that were transferred. Embryo freezing (vitrification) was recommended to allow subsequent transfer if the fresh cycle was unsuccessful. However, experimentation has continued to improve pregnancy outcomes. We discuss here the significance of frozen embryo transfer cycle and the impact of uterine and peri

For a variety of reasons--both medical and medicolegal--a more aggressive approach is taken in canine than in human rheumatoid arthritis. While cytotoxic agents are probably the drugs of choice in the dog, they are normally the last resort of the rheumatologist. On the other hand, the nonsteroidal anti-inflammatory drugs, which are the cornerstone of treatment in man, are of questionable safety--if not absolutely contraindicated--in the dog. Canine rheumatoid arthritis is an uncommon diagnosis, and it is not the intent of this discussion to encourage either the use of cytotoxic drugs or the ex

Through the analysis of case studies of chimpanzees (Pan troglodytes troglodytes) in residence at a sanctuary, who previously sustained prolonged captivity and biomedical experimentation, we illustrate how human psychological models of diagnosis and treatment might be approached in great apes. This study reflects growing attention to ethical, scientific, and practical problems associated with psychological well-being of animals. The analysis concludes that a diagnosis of Complex PTSD in chimpanzees is consistent with descriptions of trauma-induced symptoms as described by the DSM-IV and human

We report and investigated a case of inadvertent contamination of 125 mice (housed in two germ-free positive-pressurized isolators) with emerging human and coral pathogen Aspergillus sydowii. The infected mice correspond to genetic line SAMP1/YitFc, which have 100% immune predisposition to develop Crohn's disease-like spontaneous pathologies, namely, inflammatory bowel disease (IBD). Pathogen update based on a scoping review of the literature and our clinical observations and experimentation are discussed. The unwanted infection of germ-free mice (immunologically prone to suffer chronic inflam

Although James Blundell is rightly acknowledged as the father of modern transfusion therapy, a review of the events surrounding the initial human-to-human transfusions in the first decades of the 19th century reveals substantial contributions by Blundell's collaborators. Bundell's uncle John Haighton provided substantial support for animal experimentation in the growing field of physiology studies. John Leacock of Barbados provided the essential original experiments that focused the path of investigation on within-species transfusions. London instrument makers such as Laundy and Lloyd provided

Human anxiety and vestibular disorders have long been known to co-occur. Paralleling human clinical and non-clinical data, mounting genetic, pharmacological and behavioral evidence confirms that animal anxiety interplays and co-exists with vestibular/balance deficits. However, relatively few animal models have addressed the nature of this relationship. This paper examines side-by-side human psychiatric and otovestibular phenotypes with animal experimentation data, and outlines future directions of translational research in this field. Discussed here are recently developed specific animal model

UNLABELLED: New approach methodologies, such as high-complexity in vitro systems, are increasingly prioritized in biomedical research as potential alternatives to animal experimentation. We show that cerebral organoids derived from human induced pluripotent stem cells can be leveraged to (i) investigate isolate-specific replication dynamics of Nipah virus and (ii) model key histopathological lesions found in the brain tissue of infected human patients. Furthermore, we discuss the importance of organoid models for the study of Risk Group 4 viruses.
IMPORTANCE: Advanced development of medical c

As influenza A viruses (IAV) continue to cross species barriers and cause human infection, the establishment of risk assessment rubrics has improved pandemic preparedness efforts. In vivo pathogenicity and transmissibility evaluations in the ferret model represent a critical component of this work. As the relative contribution of in vitro experimentation to these rubrics has not been closely examined, we sought to evaluate to what extent viral titer measurements over the course of in vitro infections are predictive or correlates of nasal wash and tissue measurements for IAV infections in vivo.
