Developing a GC-EI-MS/MS method for quantifying warfarin and five hydroxylated metabolites generated by the Fenton reaction.

Since the 1950s, Warfarin has been used globally as both a prescription drug and a rodenticide. Research has shown that warfarin and other rodenticides are present in the environment and food chain. However, emerging contaminants are subject to degradation by biotic and abiotic processes and advanced oxidation processes. In some cases, detecting the parent compound may not be possible due to the formation of structurally changed species. This approach aims to identify hydroxylated transformation products of warfarin in a laboratory setting, even after the parent compound has undergone degradat

Synthesis and characterization of branched multiblock copolymers of polyhydroxybutyrate (PHB) autoxidized fatty acid and polymethylmethacrylate (PMMA).

The branched multiblock copolymer based on fatty acid, methyl methacrylate and PHB was first time synthesized. Autoxidation is the reaction of unsaturated fatty acids with air oxygen leading to macroperoxide initiators. Poly (3-hydroxy butyrate), PHB, is a natural bio polyester accumulated in the bacterium cell of some bacteria. Tri hydroxyl derivatives of PHB were obtained by the modification reactions of PHB with di-ethanol amine. Hydroxyl ends of the modified PHB were capped with the autoxidized unsaturated fatty acid oligomeric peroxides in order to the polyoleic acid, polylinoleic acid an

Aqueous Formation and Evolution of Phenolic Secondary Organic Aerosol: Excited by NO2- and Organic Triplet Excited State.

Liquid-phase photochemical aging critically affects the composition of secondary organic aerosols (SOAs), their optical properties, and heal … mechanisms: 3C* promotes triplet-state hydrogen abstraction, whereas NO2- degradation involves ∼46% hydroxyl radicals and the remainder mainly involves reactive nitrogen species. Product analysis indicates that NO2- photochemistry preferentially forms nitrophenols, whereas 3C*-mediated oxidation favors hydroxylated products and oligomerization. These distinct reac …

Particulate methane monooxygenase and cytochrome P450-induced reactive oxygen species facilitate 17β-estradiol biodegradation in a methane-fed biofilm.

Methane-fed biosystems have shown great potential for degrading various organic micropollutants, yet underlying molecular degradation mechan … particulate methane monooxygenase (pMMO) and cytochrome P450 monooxygenase (CYP450) could generate hydroxyl radicals (•OH), which are the primary ROS involved in E2 degradation. Molecular dynamics simulations suggest that E2 can enter the active catalytic site of pMMO through electrostatic attraction. Four amino acid residues are found to form stable hydrogen bon …

DFT Mechanistic Investigation into Ni(II)-Catalyzed Hydroxylation of Benzene to Phenol by H2O2.

Introduction of oxygen into aromatic C-H bonds is intriguing from both fundamental and practical perspectives. Although the 3d metal-catalyzed hydroxylation of arenes by H2O2 has been developed by several prominent researchers, a definitive mechanism for these crucial transformations remains elusive. Herein, density functional theory calculations were used to shed light on the mechanism of the established hydroxylation reaction of benzene with H2O2, catalyzed by [NiII(tepa)]2+ (tepa = tris[2-(pyridin-2-yl)ethyl]amine). Dinickel(III) bis(μ-oxo) species have been proposed as the key intermediate

A comprehensive review of structure-activity relationships and effect mechanisms of polyphenols on heterocyclic aromatic amines formation in thermal-processed food.

Heterocyclic aromatic amines (HAAs) are potent carcinogenic substances mainly generated in thermal-processed food. Natural polyphenols have been widely used for inhibiting the formation of HAAs, whereas the effect of natural polyphenols on HAAs formation is complex and the mechanisms are far from being clearly elucidated. In order to clarify the comprehensive effect of polyphenols on HAAs, this review focused on the structure-activity relationships and effect mechanisms of polyphenols on the formation of HAAs. In addition, the effects of polyphenols on HAAs toxicity were also first reviewed fr

Confined Cu Single Sites in ZSM-5 for Photocatalytic Hydroxylation of Benzene to Phenol.

Zeolites with band-like charge transport properties have exhibited their potential activities in sensing, optics, and electronics. Herein, a precisely designed Cu@ZSM-5 catalyst is presented with an ultra-wide bandgap of 4.27 eV, showing excellent photocatalytic activity in hydroxylation of benzene with benzene conversion 27.9% and phenol selectivity 97.6%. The SXRD and Rietveld refinement results illustrate that Cu@ZSM-5 has an average of 0.8 Cu atoms per unit cell and the single Cu atoms located in the cross-section of the sinusoidal and straight channels. XANES and EXAFS further demonstrate

Visible-light-driven amino acids production from biomass-based feedstocks over ultrathin CdS nanosheets.

Chemical synthesis of amino acids from renewable sources is an alternative route to the current processes based on fermentation. Here, we report visible-light-driven amination of biomass-derived α-hydroxyl acids and glucose into amino acids using NH3 at 50 °C. Ultrathin CdS nanosheets are identified as an efficient and stable catalyst, exhibiting an order of magnitude higher activity towards alanine production from lactic acid compared to commercial CdS as well as CdS nanoobjects bearing other morphologies. Its unique catalytic property is attributed mainly to the preferential formation of oxy

Asynchronous Radical Addition-Coupled Two-Electron Transfer in P450-Catalyzed Arene Oxidation.

The P450-catalyzed arene oxidation reaction (e.g., arene hydroxylation) is considered to have an arene oxide (or epoxide) intermediate. The two-electron transfer in the formation of arene oxide has two pathways. The electrophilic addition mechanism is initiated by substrate-to-porphyrin electron transfer, followed by electrophilic addition led by an FeIV-oxo complex on the arene carbon atom, resulting in an FeIV-alkoxyl complex and a substrate in the radical state. On the other hand, the radical addition by an FeIV-oxo complex transfers one electron from the substrate to the FeIV center, resul

Elucidating the catalytic degradation of enrofloxacin by copper oxide nanoparticles through the identification of the reactive oxygen species.

Copper oxide nanoparticles (CuO-NPs) have been suggested as effective catalysts to degrade many persistent organic contaminants. In parallel … radical species indicated production of a high percentage of superoxide (O2-.) radicals as well as hydroxyl radicals; this production is similar to the "radical production" activity of the superoxide dismutase (SOD) enzyme in the presence of hydrogen peroxide. This activity was also tested in the opposite direction, to examine if CuO-NPs show reactivity that pote …

Our data suggest that the method can be used for the quantification of ROS in individual living cells based on the measurement of fluorescence lifetime of those probes.

ESR studies on reaction of saccharide with the free radicals generated from the xanthine oxidase/hypoxanthine system containing iron.

The free radicals generated from the iron containing system of xanthine oxidase and hypoxanthine (Fe-XO/HX) were directly detected by using spin trapping. It was found that not only superoxide anion (O(2)*-) and hydroxyl radical (OH*), but also alkyl or alkoxyl radicals (R*) were formed when saccharides such as glucose, fructose and sucrose were added into the Fe-XO/HX system. The generated amount of R* was dependent on the kind and concentration of saccharides added into the Fe-XO/HX system and no R* were detected in the absence of saccharides, indicating that there is an interaction between …

Copper-zinc superoxide dismutase activity in normal and inflamed human dental pulp tissue.

Information regarding the presence of the free radical scavenging (inactivating, dismutating) enzyme superoxide dismutase in human dental pulp was sought. Free radicals, such as the superoxide anion radical (O2-) and the hydroxyl anion radical (OH.), are powerful biological oxidants produced by phagocytes during the normal tissue response to injury and infection. Also produced is hydrogen peroxide (H2O2), an aggressive oxygen species formed by the reaction of superoxide with itself, i.e., a dismutation in which one molecule of O2- is oxidized by the other. These three reactive oxygen intermedi

α/β Conflicting Aromaticity Under the Microscope: Study of Pro-Aromatic Radicals.

The aromaticity of a representative sample of pro-aromatic radicals and its nitro, amino, hydroxyl and imine substituted derivatives has been analysed by means of multicenter delocalization indices (MCI) and nuclear-independent chemical shifts (NICS). Because of their radical character, these compounds may exhibit conflicting α/β aromaticity, so that the contribution of α and β electrons to the MCI and NICS has been analysed separately and their values qualitatively interpreted in terms of the 2n+1/2n rule. All the monocyclic radicals investigated show conflicting α/β aromaticity. Thus, signif

[Mechanistic Analysis of Oxidative Stress-related Diseases Using Nitroxyl Radicals and Electron Spin Resonance].

Excessive production of reactive oxygen species (ROS) causes oxidative stress and is involved in the development and progression of a wide variety of diseases. Therefore, techniques for measuring oxidative stress are indispensable for analysis of the mechanisms of various diseases. The method involving ESR and the durable nitroxyl radical (ESR/spin probe method) is useful for this purpose, because the ESR signal intensity of the spin probe changes on reacting with ROS and other unstable radicals. In this review, the author's research applying the ESR/spin probe method to clarify disease mechan

Involvement of free radicals in excitotoxicity in vivo.

Recent evidence has linked excitotoxicity with the generation of free radicals. We examined whether free radical spin traps can attenuate excitotoxic lesions in vivo. Pretreatment with N-tert-butyl-alpha-(2-sulfophenyl)-nitrone (S-PBN) significantly attenuated striatal excitotoxic lesions in rats produced by N-methyl-D-aspartate (NMDA), kainic acid, and alpha-amino-3-hydroxy-5-methyl-isoxazole-4-propionic acid (AMPA). In a similar manner, striatal lesions produced by 1-methyl-4-phenylpyridinium (MPP+), malonate, and 3-acetylpyridine were significantly attenuated by either S-PBN or alpha-phenyl

Copper-catalysed dehydrogenation or lactonization of C(sp3)-H bonds.

Cytochrome P450 enzymes are known to catalyse bimodal oxidation of aliphatic acids via radical intermediates, which partition between pathways of hydroxylation and desaturation1,2. Developing analogous catalytic systems for remote C-H functionalization remains a significant challenge3-5. Here, we report the development of Cu(I)-catalysed bimodal dehydrogenation/lactonization reactions of synthetically common N-methoxyamides through radical abstractions of the γ-aliphatic C-H bonds. The feasibility of switching from dehydrogenation to lactonization is also demonstrated by altering reaction cond

Facile synthesis of pyrite FeS2 on carbon spheres for high-efficiency Fenton-like reaction.

Designing iron-based catalysts for Fenton-like reactions with peroxymonosulfate (PMS) as oxidants have attracted growing attentions. Herein, pyrite FeS2 supported on carbon spheres (FeS2@C) is synthesized by a facile low-temperature method. The FeS2@C/PMS system can degrade carbamazepine (CBZ) effectively in a wide pH range. Sulfate radicals (SO4·-), hydroxyl radicals (·OH), superoxide radical (O2·-), and singlet oxygen (1O2) are the responsible reactive oxygen species (ROSs) for CBZ degradation. Moreover, in the simulated fixed-bed reactor, the FeS2@C/PMS system can maintain a high CBZ remova…

Selective oxidation of tetracyclines by peroxymonosulfate in livestock wastewater: Kinetics and non-radical mechanism.

Tetracyclines (TCs) discharged from livestock wastewater have received worldwide concerns owing to their potential threats to the ecosystem and human health. Advanced oxidation processes always exhibit low efficiency to remove TCs in livestock wastewater due to the radical scavenging by water matrices. Herein, we report selective elimination of TCs by peroxymonosulfate (PMS) in livestock wastewater. A kinetic model was developed to describe the rapid degradation of TCs by PMS in the real livestock wastewater. The radical scavenging study and electron paramagnetic resonance (EPR) technique excl
