Targeting the NLRP3 inflammasome alleviates the comorbidity of chronic pain and depression via enhancing the autophagy.

The comorbidity of chronic pain and depression exhibits the bidirectional relationship and forms a vicious cycle in disease development, diagnosis and therapy. NLRP3 inflammasome-mediated neuroinflammation is a crucial factor for the pathogenesis of pain and depression. The underlying mechanisms of NLRP3 in the comorbidity of chronic pain and depression is unclear. In this study the spared nerve injury (SNI) mouse model was constructed, NLRP3 level was pharmacologically (MCC950) and genetically (Nlrp3−/−) regulated. As a result, SNI surgery induced pain hypersensitivity, depression-like, motor

STK39 interacts with MAPK14 to facilitate cardiomyocyte pyroptosis in septic cardiomyopathy by inducing NLRP3 inflammasome.

BACKGROUND: Septic cardiomyopathy (SCM) is a severe manifestation of sepsis characterized by myocardial dysfunction and systemic inflammation. The NLRP3 inflammasome-driven pyroptosis plays a pivotal role in SCM pathogenesis. This study aimed to elucidate the upstream molecular regulators of pyroptosis in SCM and investigate the functional role of serine/threonine kinase 39 (STK39) in modulating NLRP3 inflammasome activation. METHODS: Peripheral blood samples were collected from 44 patients with sepsis (19 with SCM) and 30 healthy controls. A murine cecal ligation and puncture (CLP) model and

Mechanistic insights into natural product-driven modulation of NLRP3-inflammasome signalling in metabolic syndrome.

INTRODUCTION: Metabolic syndrome (MetS) comprises a cluster of interrelated metabolic abnormalities, including obesity, insulin resistance, dyslipidaemia and hypertension, mainly driven by chronic low-grade inflammation. Among innate immune pathways, the Nod-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome has emerged as a critical molecular link between metabolic stress and inflammatory signalling, promoting caspase-1 activation, interleukin (IL)-1β/IL-18 maturation and pyroptosis across metabolically active organs such as adipose tissue, liver and pancreas. Persistent NLRP

Paeonol mitigates age-related osteoporosis via mitophagy-mediated NLRP3 inflammasome inhibition.

Age-related osteoporosis, a progressive skeletal disorder inherent to aging, is pathologically defined by diminished bone mass and deteriorated bone microarchitecture, resulting in heightened skeletal fragility. Despite extensive research, effective therapeutic interventions for age-associated bone loss remain limited. Accumulating evidence indicates that aging-induced immune dysregulation contributes significantly to chronic low-grade inflammation, thereby exacerbating osteoporotic progression. In this study, we identify Paeonol (PAE) as a potent mitigator of senile osteoporosis, acting throu

Deciphering the NEK7-NLRP3 inflammasome assembly: from conformational activation to allosteric drug discovery.

The NLRP3 inflammasome, a pivotal component of innate immunity, orchestrates immune defense and inflammatory responses. NEK7, an essential upstream regulator, drives inflammasome assembly through direct interaction with NLRP3. This review systematically summarizes the molecular mechanisms, upstream regulatory networks, and therapeutic targeting of the NEK7-NLRP3 axis. Structurally, NEK7 binds to the leucine-rich repeat (LRR) domain of NLRP3 via its catalytic domain, inducing conformational rearrangement and oligomerization. This structural shift exposes NLRP3's PYRIN domain (PYD), enabling ASC

Loss of Progranulin Expression Decreases NLRP3 Inflammasome-Mediated Inflammation and Enhances Bone Anabolism.

Progranulin (PGRN), a glycosylated protein, is expressed in most tissues, including bones, and its level is elevated in the serum and joints of individuals with inflammatory bone loss disorders such as rheumatoid arthritis (RA). Previously, using global and macrophage-specific Grn deletion mice, we demonstrated that loss of PGRN protects against age-related bone loss selectively in females, suggesting a sex-dependent role for macrophage-derived PGRN in skeletal homeostasis. Here, we investigated the role of PGRN in the regulation of macrophage-mediated inflammation and bone formation. Immune-p

Betamethasone Dipropionate Inhibits NLRP3 Inflammasome Activation by Suppressing Pro-IL-1β Expression.

BACKGROUND: The NOD-like receptor Family, pyrin domain-containing 3 protein (NLRP3) inflammasome is a macromolecular complex critical for inflammatory responses. Its excessive activation or improper regulation is intimately associated with the development of various inflammatory diseases. However, currently available drugs directly targeting the NLRP3 inflammasome are limited. In preliminary analyses, detecting the level of secreted interleukin-1 beta (IL-1β) revealed that betamethasone-17,21-dipropionate (also known as betamethasone dipropionate, BD), a clinically used glucocorticoid, potenti

THBS1+ Macrophages Exacerbate Modic Changes via SDC4-Dependent Activation of NLRP3 Inflammasome.

Modic changes (MCs) in the lumbar vertebral endplates are a common imaging finding in patients with low back pain. Their pathogenesis and progression are closely linked to the local immune microenvironment, underscoring the need to investigate the immunomodulatory mechanisms involved in MCs. Here, we identified the THBS1+ macrophages displaying a strongly pro-inflammatory phenotype. The THBS1+ macrophages were enriched at lesion sites in both human lumbar MCs specimens and the C. acnes-induced mouse model of MC. In vitro, C. acnes and its key metabolites promoted THBS1 expression in macrophage

Potential mechanisms of natural products in improving gouty arthritis: Focusing on NLRP3 inflammasome regulation.

Gouty arthritis (GA) is a metabolic inflammatory disorder characterized by the deposition of monosodium urate crystals. Although conventional treatments like colchicine can relieve symptoms, they come with several limitations, such as gastrointestinal side effects, hepatorenal toxicity, and poor target specificity. In contrast, natural products, known for their multi-pathway and multi-target properties, present distinct advantages and significant potential for intervening in GA. The primary pathological mechanism of GA involves the overactivation of the NOD-like receptor family pyrin domain-co

Inhibition of ERK5 activity ameliorates osteoarthritis by suppressing NLRP3 inflammasome-mediated chondrocyte pyroptosis.

Osteoarthritis (OA) is a widespread degenerative joint disorder manifesting as chronic pain, functional impairment, and progressive disability, imposing a substantial burden on global public health. Despite extensive research, the precise molecular pathways underlying OA pathogenesis remain incompletely elucidated. Our data indicated that human OA cartilage and cartilaginous tissues from aged mice exhibited markedly elevated levels of extracellular signal-related kinase 5 (ERK5) enzyme activity. Using Col2a1-CreERT2/Erk5flox/flox mice, we found that Erk5 deletion in cartilage inhibited the pro

Vilazodone inhibits NLRP3 inflammasome assembly by targeting the NACHT domain and alleviates inflammatory disease.

The NLRP3 inflammasome plays a central role in innate immunity and inflammatory diseases, yet effective inhibitors remain limited. Here, we identified vilazodone, an FDA-approved antidepressant, as a potent inhibitor of NLRP3 inflammasome activation by small-molecule screening. Vilazodone significantly suppressed IL-1β secretion in multiple macrophage models in response to diverse NLRP3 stimuli. Mechanistically, vilazodone did not affect the priming step but selectively inhibited inflammasome activation. It disrupted NLRP3 interactions with NEK7 and ASC, thereby blocking inflammasome assembly,

A sesterterpenoid derivative alleviates ulcerative colitis through targeting ProRS to suppress the NLRP3 inflammasome.

Ulcerative colitis (UC) is a prevalent inflammatory bowel disease that affects millions of people worldwide, for which no definitive cure currently exists. Herein, we report the discovery of a novel sesterterpenoid derivative, ColD14, with a complicated cage-like structure and contiguous stereogenic centers, which exhibits significant anti-NLRP3 inflammasome activity. Proteomic and biochemical studies identified glutamyl-prolyl tRNA synthetase (EPRS) as a direct target of ColD14. Both EPRS knockdown and pharmacological inhibition by ColD14 reduced NLRP3 inflammasome activation, underscoring th

Processing of lysophosphatidylcholines by ExoU secreted by P. aeruginosa leads to inflammasome activation and cell death.

Pseudomonas aeruginosa (P. aeruginosa) thrives in the lungs of patients with cystic fibrosis (CF) which results in pulmonary inflammation and decline in lung function. Inflammasome signaling promotes host cell death and processing of IL-1 which results in bacterial control. Since the type 3 secretion system (T3SS) of P. aeruginosa induces inflammasome signaling, we evaluated the role of ExoU, a phospholipase which is secreted through the T3SS and promotes host susceptibility. Our results indicate that the genes associated with T3SS-function display increased polymorphism and are poorly express

Targeting the NLRP3 Inflammasome with Colchicine in COVID-19: Therapeutic Evidence and Future Implications for Influenza.

Coronavirus disease 2019 (COVID-19) and influenza share key pathogenic mechanisms, including viral invasion, dysregulated innate immune activation, and the development of severe systemic complications. A central mediator of disease progression in both infections is the hyperactivation of the NLRP3 inflammasome, which drives excessive production of pro-inflammatory cytokines, immunothrombosis, multiorgan injury, and increased mortality. Colchicine possesses a unique pharmacokinetic property of preferential accumulation within myeloid cells, where sufficiently high intracellular concentrations i

NLRP3 inflammasome-mediated platelet hyperreactivity in sickle cell mice is targetable by BTK inhibition.

The platelet nucleotide-binding domain leucine-rich repeat-containing protein 3 (NLRP3) inflammasome is upregulated in sickle cell disease (SCD) and promotes platelet aggregation. We previously identified Bruton tyrosine kinase (BTK) as a critical regulator of the platelet NLRP3 inflammasome. However, whether NLRP3 contributes to platelet function beyond aggregation in SCD and whether these effects can be modulated through BTK inhibition, has been incompletely understood. Here, we show that platelet secretion, platelet spreading, platelet aggregation, and in vitro thrombus formation in respons

TLR2-Driven NLRP3 Inflammasome Activation Mediates EV71-Induced Neuropathogenesis in Neonatal Mice.

EV71 is a leading cause of HFMD-associated neurological complications, yet its CNS pathogenic mechanisms remain unclear. Using neonatal BALB/c mice, we identified 10 days post-infection (dpi) as the disease peak, characterized by neuronal necrosis and robust NLRP3 inflammasome activation. We demonstrate that EV71 upregulates TLR2, triggering the NF-κB pathway to prime and activate the NLRP3 inflammasome within microglia. This process drives pro-inflammatory cytokine release and neuroinflammatory damage. Pharmacological blockade of TLR2 with C29 suppressed NF-κB phosphorylation and NLRP3 assemb

Serum Amyloid A-Dependent Inflammasome Activation and Acute Injury in a Mouse Model of Experimental Stroke.

Serum amyloid A (SAA) proteins increase significantly in the blood following inflammation. Recently, SAAs were increased in humans following stroke and in ischemic animal models. However, the impact of SAAs on whether this signal is critical in the ischemic brain remains unknown. Therefore, we investigated the role of SAA and SAA signaling in the ischemic brain. Wild-type and SAA-deficient mice were exposed to middle cerebral artery occlusion and reperfusion and examined to determine the impact of infarct volumes, behavioral changes, inflammatory markers, TUNEL staining, and BBB changes. Decoupling musculoskeletal damage from antiviral control: the NLRP3 inflammasome in arthritogenic alphavirus infection.

Mosquito-borne arthritogenic alphaviruses, including chikungunya virus (CHIKV), Ross River virus (RRV), and Mayaro virus (MAYV), cause acute febrile polyarthralgia and can leave a substantial proportion of patients with persistent or relapsing musculoskeletal disease. In many cohorts, chronic arthritis continues after systemic viremia has declined, suggesting that late tissue pathology is sustained largely by host inflammatory programs rather than by ongoing productive viral replication. This Mini Review examines the NLRP3 inflammasome as a pathogenic node linking viral danger sensing to chron
