Psoriasis is a common immune-mediated inflammatory disease that often worsens during cold weather, yet the mechanisms linking cold exposure to enhanced psoriatic inflammation remain poorly understood. Shuai and colleagues identify a distinct macrophage population induced by cold stress in the dermal adipose layer, characterized by heightened lipid metabolism and IL-1β production. These macrophages promote IL-17A-producing γδT-cell responses in a mouse model of psoriasis, uncovering a potential cellular mechanism through which cold environments exacerbate psoriatic inflammation via immune-metab

Psoriasis is a chronic disease with elusive pathogenesis linked to genetic, and immune factors. Studies have shown most patients have experienced high levels of oxidative stress, which can lead to inflammation or damage. Withaferin A is a natural product with multiple pharmacological activities, including anti-tumor and anti-inflammatory effects. We investigated the effects of WA in treating Imiquimod (IMQ)-induced psoriasis mice. The epidermal pathology of mice was significantly improved after treatment. WA inhibited inflammation by decreasing the production of IL-1β, IL-6, and IFN-γ, which w

Sézary syndrome (SS) is a rare and aggressive type of cutaneous T-cell lymphoma, with an abnormal inflammatory response in affected skin. The cytokines IL-1B and IL-18, as key signaling molecules in the immune system, are produced in an inactive form and cleave to the active form by inflammasomes. In this study, we assessed the skin, serum, peripheral mononuclear blood cell (PBMC) and lymph-node samples of SS patients and control groups (healthy donors (HDs) and idiopathic erythroderma (IE) nodes) to investigate the inflammatory markers IL-1B and IL-18 at the protein and transcript expression

Alopecia areata (AA) and vitiligo are both common skin diseases of autoimmune origin. Both alopecia areata and vitiligo have shown to be affected by oxidative stress. The present work is aimed at evaluating and comparing the serum proinflammatory cytokine levels in AA and nonsegmental vitiligo (NSV). A cross-sectional study was conducted of 33 patients with AA, 30 patients with NSV, and 30 healthy controls. Serum levels of interferon γ (IFN-γ), interleukin- (IL-) 1β, and IL-6 were determined quantitatively by ELISA method. Our analysis identified a signature of oxidative stress associated with

Radiotherapy is known to trigger immunogenic cell death and activate local anti-tumor immune responses. However, its systemic immunomodulatory effects remain poorly understood. Here, we discovered that irradiated tumor cell-derived microparticles (RT-MPs) are released into the circulation and subsequently taken up by neutrophils in the spleen. The mitochondrial DNA contained within RT-MPs promotes the hyperactivation of neutrophils, leading to the secretion of interleukin-1beta (IL-1β) via the STING/NLRP3/GSDMD axis. IL-1β, in turn, enhances the antigen-presenting capacity of dendritic cells (…

Mevalonate kinase deficiency is a rare autosomal recessive inborn error of metabolism with an autoinflammatory phenotype. In this review we discuss its pathogenesis, clinical presentation and treatment. Mutations in both copies of the MVK-gene lead to a block in the mevalonate pathway. Interleukin-1beta mediates the inflammatory phenotype. Shortage of a non-sterol isoprenoid product of the mevalonate pathway, Geranylgeranylpyrophosphate leads to aberrant activation of the small GTPase Rac1, and inflammasome activation. The clinical phenotype ranges widely, depending on the severity of the enzy…

This study presents the first case of internal root resorption in monozygotic twins. Genetic factors have been implicated in the development of root resorption lesions. The link between interleukin (IL)-1 gene polymorphism and root resorption had been reported. However, these studies have involved external root resorption. Twin healthy 17 year old Caucasian sisters were diagnosed with internal root resorption in their upper left central incisor. Neither had a history of orofacial trauma and neither had received orthodontic treatment. The presence of IL-1beta+C3953 allele is the sign of predisp

BACKGROUND: Selenoprotein S (SelS) protects the functional integrity of the endoplasmic reticulum against the deleterious effects of metabolic stress. SEPS1/SelS polymorphisms have been involved in the increased release of pro-inflammatory cytokines interleukin (IL)-1beta, tumor necrosis factor (TNF)-alpha and IL-6 in macrophages. We aimed at investigating the role of the SEPS1 variants previously associated with higher plasma levels of these cytokines and of the SEPS1 haplotypes in the susceptibility to develop immune-mediated diseases characterized by an inflammatory component.

Both CCL27 and CCL28 are ligands for CCR10 and attract CCR10(+) lymphocytes. We previously demonstrated that CCL27 and CCL28 were strongly expressed in sera and lesional keratinocytes of patients with atopic dermatitis and psoriasis vulgaris. However, the regulation of CCL27 and CCL28 production in keratinocytes has not been well documented. In this study, we showed that CCL27 and CCL28 expression and production by a human keratinocyte cell line, HaCaT cells, were strongly induced by inflammatory cytokines tumor necrosis factor-alpha and interleukin-1beta. CCL27 production was downregulated by…

Several autoimmune diseases are believed to be mediated, in part, by interleukin (IL)-18. Many are those with associated elevated interferon-gamma levels, such as systemic lupus erythematosus, macrophage activation syndrome, rheumatoid arthritis, Crohn's disease, psoriasis, as well as graft-versus-host disease. Clinical and animal studies also support the concept that IL-18 is a key player in atherosclerosis, acute renal ischemia and hepatitis. IL-18 is a member of the IL-1 family; IL-1beta and IL-18 are closely related, and both require the intracellular cysteine protease caspase-1 for biolog

Inflammatory mediators are multifunctional cytokines that play important roles both in normal central nervous system (CNS) development and in the response of the brain to diverse forms of injury. Interleukin (IL)-1beta, tumor necrosis factor-alpha and IL-6 are among the best-characterized early-response cytokines. Recent data suggest that they may be synthesized and secreted by several CNS cell types, including microglia, astrocytes and neurons. Biological effects of these cytokines that could influence the progression of injury in the brain include stimulating the synthesis of other cytokines

In this study dengue virus (DV) was found to infect primary endothelial cells derived from human umbilical cord veins (HUVEC) and alter their cytokine production. Dengue virus infection of HUVEC was confirmed by an increase in plaque-forming units in the culture supernatant and by immunofluorescence assay. HUVEC produced large amounts of interleukin (IL)-6 and IL-8 but not IL-1beta after DV infection. Both the replication of DV and the production of IL-6 and IL-8 by HUVEC after DV infection were inhibited by ribavirin, an antiviral synthetic guanosine analogue. Additionally, increased serum le
