Anti-P antibodies that impair memory perturb hippocampal glutamatergic receptor trafficking, synapse structure and microglia.

BACKGROUND: Anti-ribosomal P protein autoantibodies (anti-P) are associated with psychosis and cognitive dysfunction in patients with systemic lupus erythematosus (SLE), yet the underlying mechanisms remain undefined, hindering targeted therapies. Anti-P cross-react with a neuronal surface protein (NSPA), alter glutamatergic synaptic transmission and plasticity in hippocampal slices, and impair spatial memory in a short-term passive transfer mouse model. NSPA knockout mice display spatial memory deficit linked to reduced NMDAR activity and postsynaptic density (PSD) levels, along with an incre

The evolving biology of T cell memory: A dynamic continuum of function and persistence.

Immunological memory underpins long-term protection and vaccination. Although early work established the durability and recall capacity of memory T cells, understanding of CD8 T cell memory generation, maintenance, and plasticity has advanced substantially over recent decades. Initial models viewed memory as an outcome of peak effector responses and were defined mainly by persistence. Successive waves of technological innovation, from MHC tetramers and genetic lineage-tracing approaches to single-cell and epigenomic profiling and modern high-throughput gene-perturbation screens, have reshaped

A Critical Time Window of Neocortex Modulation for Remote Fear Memory in Rats and Humans.

Persistent fear memories, central to trauma-related psychopathology, rely on long-term consolidation. While initial encoding involves subcortical circuits, the precise timing and necessity of neocortical engagement for remote fear memory expression remain elusive. In the present translational study from rats to humans, we identified a late consolidation phase in the medial prefrontal cortex (mPFC) that occurs around 18 h after fear conditioning, a phase essential for remote memory. Neuronal ensembles in the mPFC activated during this phase mediate the expression of remote fear memory in rats.

Direct electrical stimulation of the human amygdala enhances recognition memory for objects but not scenes.

Research from studies using invasive neuroanatomy-inspired direct manipulations suggests the basolateral amygdala (BLA) mediates a generalized modulation of many different types of memory. In contrast, noninvasive indirect correlations suggest that specificity exists in how the BLA prioritizes experiences in memory. We used direct electrical stimulation of the BLA to investigate the specificity of the memory enhancement in the human brain. Patients undergoing intracranial monitoring via depth electrodes viewed object and scene images, half of which were followed by BLA stimulation. Stimulation

Astrocytic PI3Kα controls synaptic plasticity and cognitive function via serine metabolism.

Astrocytes modulate neuronal activity by gliotransmission and through metabolic regulation, yet the relationship between these functions remains poorly understood. Here we show that phosphatidylinositol 3-kinase (PI3K) signaling in astrocytes controls synaptic plasticity and memory by regulating cellular metabolism. Deletion of the p110α isoform of PI3K in hippocampal astrocytes impairs NMDA receptor activation during the induction of hippocampal long-term potentiation (LTP). This defect is rescued by either D-serine (co-agonist of synaptic NMDA receptors) or its precursor L-serine, indicating

Sex-Dependent Effects of Maternal Caloric Restriction, but Not Intermittent Fasting, on Offspring Hippocampal Memory and Antioxidant Activity.

During the lactation period, maternal malnutrition can lead the offspring to deficiencies in brain development and impact behavior throughout life. This study evaluated the effects of maternal dietary restrictions, caloric restriction (CR), and intermittent fasting (IF) during lactation on the memory and hippocampal redox state of the adolescent offspring (males and females). The litters (dam and 8 pups) received the diets: control-received the lab chow (Nuvilab CR-1) ad libitum, CR-received 50% of the chow consumed by the control group, and IF-received lab chow ad libitum for the first 24 h,

Rapid identification of antigen-specific monoclonal antibodies from HIV-1 elite controllers using the Beacon platform.

Elite controllers (ECs) are rare human immunodeficiency virus (HIV)-infected individuals who manage viral replication without antiretroviral therapy. While their cellular immunity is well characterized, humoral responses remain limited, which provides an uncommon window into antibody responses to HIV-1. Using single-cell functional screening, this study aimed to isolate and characterize HIV-1 envelope (Env)-specific monoclonal antibodies (mAbs) from ECs. The Beacon platform was used for enrichment, activation, and screening of cluster of differentiation 27+ (CD27+) memory B cells. Among ~ 12,0

Targeted NMDA receptor knockdown in recall-activated neuronal ensembles impairs remote fear extinction.

Fear extinction training in rodents decreases fear responses, providing a model for the development of post-traumatic stress disorder therapeutics. Fear memory recall reactivates the consolidated fear memory trace across multiple brain regions, and several studies have suggested that these recall-activated neurons are re-engaged during extinction. However, the molecular mechanisms linking this reactivation to extinction remain largely elusive. Here, we investigated the role of N-Methyl-D-Aspartate receptors (NMDARs) in remote memory recall-activated neurons within the basolateral amygdala and

Inhibition of inflammation is not enough for recovery of cognitive impairment in hepatic encephalopathy: Effects of minocycline and ibuprofen.

There is evidence that hyperammonia and inflammation play crucial roles in hepatic encephalopathy. This study intends to determine neuroprotective effects of minocycline (MINO) and ibuprofen (IBU), and also set out to assess whether inhibition of inflammation is enough to achieve optimal improvement of hepatic encephalopathy symptoms. The hepatic encephalopathy was induced by bile-duct ligation (BDL), and the animals received first dose of MINO and/or IBU 15 days later and then every day until the 28 day. The rats were divided into the 6 groups of control, sham, BDL + V and BDL + IBU, BDL + MI

The Effects of Mitral Valve Repair on Memory Performance, Executive Function, and Psychological Measures in Patients With Heart Failure.

OBJECTIVES: Heart failure (HF) is a prevalent disease that remains costly and associated with a high mortality rate. HF is also associated with poor neurocognitive functioning. For the treatment for HF patients with severe mitral regurgitation, the MitraClip device has emerged as a promising interventional tool that reduces the mitral valve leakage and thus increases cardiac output. Currently, there is only limited knowledge on changes in cognitive and psychosocial functioning before and after the MitraClip intervention. METHODS: Cognitive function (memory and executive function) and psychoso

Impairment of cognitive function by chemotherapy: association with the disruption of phase-locking and synchronization in anterior cingulate cortex.

BACKGROUND: Patients following prolonged cancer chemotherapy are at high risk of emotional and cognitive deficits. Research indicates that the brain neuronal temporal coding and synaptic long-term potentiation (LTP) are critical in memory and perception. We studied the effects of cisplatin on induction of LTP in the basolateral amygdala (BLA)-anterior cingulate cortex (ACC) pathway, characterized the coordination of spike timing with local theta oscillation, and identified synchrony in the BLA-ACC network integrity. RESULTS: In the study presented, the impacts of cisplatin on emotional and co

Exposure to social defeat stress in adolescence improves the working memory and anxiety-like behavior of adult female rats with intrauterine growth restriction, independently of hippocampal neurogenesis.

Intrauterine growth restriction (IUGR) is a risk factor for memory impairment and emotional disturbance during growth and adulthood. However, this risk might be modulated by environmental factors during development. Here we examined whether exposing adolescent male and female rats with thromboxane A2-induced IUGR to social defeat stress (SDS) affected their working memory and anxiety-like behavior in adulthood. We also used BrdU staining to investigate hippocampal cellular proliferation and BrdU and NeuN double staining to investigate neural differentiation in female IUGR rats. In the absence

Acquisition and long-term retention of spatial learning in the human immunodeficiency virus-1 transgenic rat: effects of repeated nicotine treatment.

The HIV-1 transgenic (HIV-1Tg) rat shows a deficit in learning to locate a submerged platform in a multiple-trial water maze task compared to transgenic littermate and F344 control rats (Vigorito et al., J.Neuroimmune Pharmacol 2:319-328, 2007; Lashomb et al., J.Neurovirol 15:14-24, 2009). Nicotine is known to have neuroprotective effects possibly by minimizing cytotoxic effects of glutamate or by modulating a cholinergic anti-inflammatory pathway. Nicotine also improves performance in a variety of learning tasks by enhancing attention and short-term memory (STM). The purpose of this study was

Chronic intranasal URB597 treatment reverts short-term memory deficits in a rat model of metabolic syndrome.

AIMS: Metabolic syndrome (MetS) is characterized by several metabolic alterations that may increase the risk of memory alterations. While metabolic consequences of MetS are well documented, its memory impact remains underexplored. This study aimed at investigating the progression of MetS-associated impairments and evaluating the therapeutic potential of URB597, a fatty-acid amide hydrolase inhibitor, that increases the N-acylethanolamine levels. MATERIALS AND METHODS: In Experiment 1, male Sprague-Dawley rats were fed a control (CTRL), high-fat (HF), high-carbohydrate (HC), or combined HF/HC

Simultaneous STING and lymphotoxin-β receptor activation induces B cell responses in tertiary lymphoid structures to potentiate antitumor immunity.

B cell-rich tertiary lymphoid structures (TLS) are associated with favorable prognosis and positive response to immunotherapy in cancer. Here we show that simultaneous activation of innate immune effectors, STING and lymphotoxin-β receptor (LTβR), results in CD8+ T cell-dependent tumor suppression while inducing high endothelial venule development and germinal center-like B cell responses in tumors to generate functional TLS in a T cell-dependent manner. In a neoadjuvant setting, activation of STING and LTβR by their agonists effectively immunized mice against tumor recurrence, leading to long

Adverse performance effects of acute lorazepam administration in elderly long-term users: pharmacokinetic and clinical predictors.

BACKGROUND: The benzodiazepine lorazepam is widely utilized in the treatment of elderly individuals with anxiety disorders and related conditions. Negative effects of acute lorazepam administration on cognitive performance, especially memory, have been reported in both previously untreated elderly and in individuals who have received short term (up to three weeks) treatment with therapeutic doses. However, it remains unclear if these adverse cognitive effects also persist after long-term use, which is frequently found in clinical practice. METHODS: Cognitively intact elderly individuals (n=37

The nBAF complex subunit CREST/SS18L1 regulates hippocampal memory processes via tyrosine 397 and histone acetyltransferase CBP.

Chromatin-modifying and -remodeling machineries are important for learning-induced transcriptional activity, yet it remains unclear how they coordinate to drive de novo gene expression for memory formation. Here, we examine the transcription factor known as calcium-responsive transactivator (CREST) in memory formation, synaptic plasticity, and learning-induced gene expression. CREST is known to bind major chromatin-modifying and -remodeling machineries via interaction with CREB-binding protein (CBP) and brahma-related gene 1 (BRG1), respectively. In silico modeling of CREST identified tyrosine
