A Dual-Loop Task-Orthogonal Framework for Sequential Multitask Learning in Echo State Networks.

Sequential multitask learning is a major problem for Echo State Networks (ESNs) because of catastrophic forgetting and interference between the tasks. To solve this, a Dual-Loop Task-Orthogonal Framework of Echo State Networks (DLTOF-ESN) model is proposed. It has a well-scaling architecture based on orthogonal parameter updates and a new mechanism named Adaptive Stability Regulation (ASR). DLTOF-ESN is based on dual-loop learning. The inner loop uses orthogonal updates to minimize gradient overlap. The outer loop adapts regularization strength based on changes in the relationships of tasks. A

Do Hopfield networks dream of stored patterns? A statistical-mechanical theory of dreaming in multidirectional associative memories.

We introduce the Dreaming L-directional Associative Memory (DLAM), a multi-layer Hebbian architecture in which off-line dreaming and supervised heteroassociative coupling coexist within a single cost function, placing our approach within the framework of energy-based models (EBMs). The replica-symmetric free energy, derived via the Guerra interpolation scheme, yields self-consistency equations governing the order parameters across the control-parameter space. The effective local field decomposes into signal, intra-layer dreaming noise, and inter-layer noise. Dreaming improves retrieval by diff

The Adaptive Cellular State Model: A Conceptual Framework Linking Dietary Quality to Exercise-Induced Cellular Adaptation.

Exercise is a recurrent multidimensional perturbation that initiates biological adaptation, yet responses to comparable training programmes remain heterogeneous. This critical narrative Review introduces the Adaptive Cellular State (ACS) Model as a heuristic framework for examining how the biological context present before training may contribute to variation in the subsequent processing and consolidation of exercise-induced perturbations. ACS is defined as a pre-existing, dynamic and multidimensional biological configuration that may condition how exercise-induced perturbations are received,

Predictability of sleep slow oscillation emergence and spatial extent from pre-onset neural dynamics.

Slow oscillations (SOs; ~ 0.5-1.5 Hz) are a hallmark of non-rapid eye movement sleep and are known to support memory consolidation. Although their dynamics are well characterized, processes that precede SO initiation and predict their spatial extent remain largely unknown, despite their potential utility for closed-loop intervention. Using high-density EEG from 29 healthy adults, we examined neural activity in the 2-s interval preceding the SO trough. SOs were classified into Global and Frontal subtypes, which differ in propagation. We quantified instantaneous spectral power, time-frequency dy…

Phase-targeted auditory stimulation enhances slow-wave activity during sleep in Huntington's disease: A pilot crossover study.

BackgroundSleep disturbances, including fragmentation and reduced slow-wave sleep (SWS), are common in Huntington's disease (HD). SWS contributes to synaptic homeostasis, metabolic clearance, and memory consolidation. Phase-targeted auditory stimulation (PTAS) enhances slow-wave activity (SWA) in healthy individuals and Parkinson's disease, but its feasibility and physiological effects in HD remain unknown.ObjectiveTo assess the feasibility and physiological effects of PTAS in individuals with HD in a home-based setting.MethodsTen participants with genetically confirmed HD were enrolled in a r…

Learning how to experience the world: From circuits to cell types to genes.

Perception depends on the brain's ability to transform high-dimensional sensory inputs into low-dimensional internal models that support ada … dent manner. This could potentially enable active sensation during behavioral engagement as well as memory consolidation and model updating during behavioral quiescence. Together, these studies suggest that examining how gene expression programs equip specific cell types with relevant computational properties is a promising approach that can integrate these frameworks …

Targeting PARP1-dependent parthanatos in Alzheimer's disease: Mechanisms and therapeutic opportunities.

Alzheimer's disease (AD) is the predominant cause of dementia globally. This review clarifies the dual function of poly(ADP-ribose) polymerase 1 (PARP1) in AD pathogenesis, emphasizing its role in mediating parthanatos, a unique caspase-independent cell death mechanism. We analyze contemporary literature regarding PARP1 expression, parthanatos signaling, and pharmaceutical treatments in AD models. In addition, PARP1 exhibits context-dependent duality: its physiological nuclear expression in hippocampus neurons is essential for memory consolidation and decreases early in cognitive impairment, s…

Spatially Extensive LFP Correlations Identify Slow-Wave Sleep in Marmoset Sensorimotor Cortex.

Identifying neural signatures of slow-wave sleep (SWS) is important for a number of reasons including diagnosing potential sleep disorders and examining its role in memory consolidation ( Diekelmann and Born, 2010; Klinzing et al., 2019; Brodt et al., 2023). Studies of sleep in the common marmoset (Callithrix jacchus) have revealed similarities to humans and other nonhuman primates, including distinct sleep stages ( Crofts et al., 2001) and diurnal sleep patterns ( Hoffmann et al., 2012). Advances in applying wireless technology for recording neural activity during natural, unrestrained behavi…

Theta rhythm as a real-time quantitative marker for non-invasive analysis of adult neurogenesis in the intact brain.

BACKGROUND: Adult neurogenesis is a regenerative mechanism of the brain that contributes to neuroplasticity and memory consolidation. Aberrant neurogenesis is considered a key pathogenic hallmark of a wide array of neurocognitive disorders. While the functional significance of adult neurogenesis is well established in most experimental and wild animals, its occurrence in the aging human brain remains uncertain. OBJECTIVE: Most studies on adult neurogenesis in humans rely on post-mortem analysis, as there is currently no method to accurately evaluate the neurogenic process in the intact brain.…

Transformed Visual Working Memory Representations in Human Occipitotemporal and Posterior Parietal Cortices.

Recent fMRI studies reported transformed representations between perception and visual working memory (VWM) in the human early visual cortex (EVC). This is inconsistent with the still widely cited original proposal of the sensory account of VWM, which argues for a shared perception-VWM representation based on successful cross-decoding of the two representations. Although cross-decoding was usually lower than within-VWM decoding and consistent with transformed VWM representations, this has been attributed to experimental differences between perceptual and VWM tasks: once they are equated, the s

Regulatory T cells suppress myeloma-specific immunity during autologous stem cell mobilization and transplantation.

Autologous stem cell transplantation (ASCT) is the standard of care consolidation therapy for eligible patients with myeloma but most patients eventually progress, an event associated with features of immune escape. Novel approaches to enhance antimyeloma immunity after ASCT represent a major unmet need. Here, we demonstrate that patient-mobilized stem cell grafts contain high numbers of effector CD8 T cells and immunosuppressive regulatory T cells (Tregs). We showed that bone marrow (BM)-residing T cells are efficiently mobilized during stem cell mobilization (SCM) and hypothesized that mobil

A mental workload and biomechanical assessment during split-belt locomotor adaptation with and without optic flow.

Adaptive human performance relies on the central nervous system to regulate the engagement of cognitive-motor resources as task demands vary. Despite numerous studies which employed a split-belt induced perturbation to examine biomechanical outcomes during locomotor adaptation, none concurrently examined the cerebral cortical dynamics to assess changes in mental workload. Additionally, while prior work suggests that optic flow provides critical information for walking regulation, a few studies have manipulated visual inputs during adaption to split-belt walking. This study aimed to examine the

Facial nerve axotomy induces morphological changes in hippocampal pyramidal neurons.

Facial nerve injury in rats have been widely used to study functional and structural changes that occur in the injured motoneurons and other central nervous system structures related with sensorimotor processing. A decrease in long-term potentiation of hippocampal CA3-to-CA1 commissural synapse has recently been reported related to this peripheral injury. Additionally, it has been found increased corticosterone plasmatic levels, impairment in spatial memory consolidation, and hippocampal microglial activation in animals with facial nerve axotomy. In this work, we analyzed the neuronal morpholo…

Tau-Induced Elevation of the Activity-Regulated Cytoskeleton Associated Protein Arc1 Causally Mediates Neurodegeneration in the Adult Drosophila Brain.

Alzheimer's disease and other tauopathies are neurodegenerative disorders pathologically defined by aggregated forms of tau protein in the brain. While synaptic degradation is a well-established feature of tau-induced neurotoxicity, the underlying mechanisms of how pathogenic forms of tau drive synaptic dysfunction are incompletely understood. Synaptic function and subsequent memory consolidation are dependent upon synaptic plasticity, the ability of synapses to adjust their structure and strength in response to changes in activity. The activity regulated cytoskeleton associated protein ARC ac…

Couple Relationships Are Associated With Increased REM Sleep-A Proof-of-Concept Analysis of a Large Dataset Using Ambulatory Polysomnography.

Background/Objectives: Rapid Eye Movement (REM) sleep is associated with memory consolidation and several health effects including stress response, mental health, and longevity. Recently, it has been shown that regularly co-sleeping couples have increased and stabilized REM sleep when co-sleeping as compared to sleeping individually. However, it remained unclear whether this is due to a specific effect of altering the usual sleeping environment by partner deprivation or due to a generalizable REM-sleep promoting effect of couple relationships. The present study aims to clarify this ambiguity. …
