Search for Molecular Markers of Myocardial Injury in a Mouse Model of Obstructive Nephropathy Using Nestin Promoter-Driven GFP Expression.

The expression of cardiac dysfunction markers and the number of nestin-positive cells in the myocardium were analyzed in a mouse model of obstructive nephropathy using animals expressing GFP under the nestin promoter. Unilateral ureteral obstruction (UUO) led to an increase in the number of nestin-positive cells in cardiac tissue, accompanied by elevated expression of B-type natriuretic peptide, which positively correlated with GFP levels. A trend toward reduced expression of β-myosin heavy chain and the gap junction protein connexin-43 was observed, whereas the expression of the progenitor ce

Polystyrene microplastics impact on cardiac and pulmonary physiology and microenvironment in a mouse model: Role of taurine supplementation and molecular docking insights.

Polystyrene microplastics (PS-MPs) have recently gained attention as widespread environmental contaminants posing risks to both human and animal health. In this study, we investigated the potential protective effect of taurine (200 mg/kg b.wt) against cardiopulmonary toxicity induced by PS-MPs (10 mg/kg b.wt) in male Swiss mice following a 60-day oral exposure. Molecular docking investigation for both proteins and mRNA targets was carried out utilizing a global, flexible docking strategy that allowed for full ligand conformational freedom and binding surface exploration. We designed an experim

Hydration and interfacial behavior of 7-ketocholesterol monolayers: A comparative molecular dynamics study with cholesterol using the OPC water model.

We investigate how oxidation of cholesterol at the C7 position alters sterol hydration and interfacial organization by combining Langmuir monolayer experiments with all-atom molecular dynamics simulations employing the OPC water model. While cholesterol forms tightly packed and weakly hydrated monolayers, 7-ketocholesterol exhibits increased molecular area, enhanced compressibility, and pronounced orientational disorder. These differences originate from altered hydration: the hydroxyl group of 7-ketocholesterol at C3 position retains cholesterol-like hydration, whereas the additional carbonyl

The Triangular Model of Psychological Stress, Sleep Disorders and Food Addiction in T2DM: An Integrative Review Based on Shared Molecular Mechanisms.

The prevalence of type 2 diabetes mellitus (T2DM) continues to rise, and traditional models fail to fully explain its pathogenesis, particularly the frequent co-occurrence of T2DM with mental health disorders. Based on a systematic integration of epidemiological and molecular biological studies, this review organises existing evidence into a "psychological stress-sleep disturbance-food addiction" triangular framework, drawing together observations that have mostly been discussed in isolation. In this model, the three factors form a self-perpetuating vicious cycle through bidirectional interact

Fluorescence from dual molecular rotors allows sensitive detection of widespread brain pathology in a mouse model of Alzheimer's disease.

Amyloid fibrils formed by the misfolding and aggregation of proteins are a pathological hallmark of many neurodegenerative conditions including Alzheimer's disease (AD). Although recent studies have shown that pre-fibrillar species including low molecular-weight oligomers are more toxic in vitro than mature fibrils, and correlate better with cognitive decline in AD patients, techniques to study this subtle pathology remain limited. Here, we describe the use of the dye pair 9-(dicyanovinyl)julolidine (DCVJ) and crystal violet (CV), two fluorescent molecular rotors, to detect widespread patholog

Investigation on the Molecular Mechanism of ORF7a Suppressing BST-2 Antiviral Activity through Structural Modeling of Their Protein-Protein Interface.

In the present work, a structural model for the protein-protein interface of bone marrow stromal cell antigen 2 (BST-2) and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) open reading frame 7a (ORF7a) is predicted using multiscale molecular dynamic simulations. Coarse-grained simulations enabled broad conformational sampling, and all-atom simulations refined the resulting structures. A machine learning-based clustering analysis was applied to the simulation ensemble to categorize the dominant heterodimeric conformations that captured the principal interaction geometries. NAStructuralDB : structural database to facilitate computational studies of molecular modeling and recognition of proteins with special focus on antibody-antigen interactions.

Studying the interactions between antibodies and antigens is fundamental to the development of novel therapeutic biologics. Predictions of such interactions start with data collection. Though there exist reliable resources to identify antibody structures in the Protein Data Bank (PDB), such data still requires substantial processing to be usable in predictive tasks. Redundancy in sequences needs to be removed to avoid data leakages between train, test, and validation sets. Descriptors such as surface accessibility, secondary structure, and antibody region information need to be additionally an

Molecular classification and construction of prognostic risk model via machine learning based on metabolic-related genes in hepatocellular carcinoma.

BACKGROUND: Despite recent advances in therapeutic strategies for hepatocellular carcinoma (HCC), patient prognosis remains unsatisfactory. Accumulating evidence indicated that dysregulation of metabolism-related pathways and genes plays a pivotal role in HCC progression. Accordingly, metabolic-associated genes hold promise as prognostic biomarkers and potential predictors of therapeutic response. This study aimed to identify distinct metabolic subtypes of HCC and compare their clinicopathological and genomic features. Prognosis-associated metabolic genes were screened using an integrated mach

Proteomic analysis of tissue-derived extracellular vesicles shows region-specific molecular changes in a rat model of takotsubo syndrome.

Takotsubo syndrome (TS) is characterized by transient regional wall motion abnormalities (RWMA) of the heart following stress. Extracellular vesicles (EVs) play a significant role in cellular communication and disease pathophysiology, but remain unexplored in TS. Using a high-fidelity rat model of TS induced by isoprenaline infusion (n = 16), we isolated EVs from the tissue of affected apical and unaffected basal segments of the left ventricle at 24 h post-induction. The TS phenotype and cardiac function were assessed using high-resolution echocardiography. EVs were characterized by electron m

Design, synthesis, molecular modelling and biological evaluation of oxiconazole-based 1,2,3-triazoles as dually acting antifungal and antitumor agents.

The rise of azole-resistant and the elevated prevalence of fungal infections in immuno-compromised cancer patients emphasize the necessity for strategies that can tackle both fungal resistance and tumor expansion. We postulated that the appropriate alteration of the azole pharmacophore through the inclusion of a 1,2,3-triazole ring might result in dual-acting drugs that preserve antifungal efficacy while exhibiting anticancer properties. In accordance with the aforementioned reasoning, a targeted library of oxiconazole-derived 1,2,3-triazoles was synthesized and assessed for antifungal potency

Artificial intelligence-guided discovery of marine scalaranes as selective COX-2 inhibitors through machine learning and molecular modelling.

Anti-inflammatory agents are critical for managing chronic conditions, yet traditional non-selective inhibitors often lead to severe gastrointestinal or cardiovascular events. This study presents an integrated artificial intelligence (AI) and molecular modeling pipeline to discover safer alternatives from the Red Sea sponge Hyrtios erectus. We screened a comprehensive library of 8415 COX2-related records using four machine learning algorithms, identifying a random forest model (93% accuracy, 86% MCC) as the most robust predictor. Using this model to screen H. erectus-derived scalarane sesterte

Characterization of central venous stenosis induced by indwelling catheter and balloon injury in a rabbit model: angiographic, histological, and molecular evaluations.

BACKGROUND: Central venous stenosis (CVS) is a significant complication in patients undergoing hemodialysis. Although mechanical injury and foreign body reactions are known triggers, the differential pathophysiological mechanisms underlying CVS induced by transient balloon injury and chronic indwelling catheters remain unclear. This study compared stenotic phenotypes and molecular pathways in rabbit models of balloon-induced (Ap) and catheter-related (Ac) CVS. METHODS: Rabbit models were established using balloon dilation (Ap group, n = 9) or indwelling catheter implantation (Ac group, n = 10

Hydrogen-bond network reorganization and reference-state-dependent barriers in the microhydrated CH2OO + H2S reaction: A molecular modeling study.

Explicit water molecules reorganize proton-transfer networks, but cluster recruitment may offset intrinsic catalysis. We model direct (A0), one-water (A1), and two-water (A2) pathways for CH2OO + H2S using a common omegaB97M-V/def2-TZVPP/SMD(water)//omegaB97X-D/def2-TZVP/SMD(water) cluster-continuum protocol with quasi-RRHO thermochemistry. Intrinsic free-energy barriers decrease from 5.52 kcal mol-1 (A0) to 3.86 (A1) and 3.30 (A2), whereas formal 1 M overall barriers increase from 9.88 to 12.18 and 15.20 kcal mol-1 because larger clusters are costlier to assemble. Ethnicity-specific molecular subtypes and a machine-learning risk model in Asian patients with non-muscle-invasive bladder cancer.

Non-muscle-invasive bladder cancer (NMIBC) has high rates of recurrence and progression, yet its molecular landscape across different ethnicities remains poorly defined. We assembled a meta-cohort of 202 Asian NMIBC transcriptomes and identified three novel molecular subtypes (AC0, AC1, AC2) using non-negative matrix factorization clustering. These subtypes, each associated with distinct biological pathways and clinical outcomes, were externally validated across 22 datasets. Subtype-specific biomarkers (RABL6, MYBL2, RAD54L, and FAM64A) were confirmed by immunohistochemistry in an independent

Behavioral and molecular characterization of various muscle-based surgical strategies for neuroma treatment in a peroneal nerve transection rat model.

Chronic pain resulting from neuroma formation is a prevalent and challenging problem to treat. Clinical studies suggest (1) surgically intervening at the time of nerve injury rather than in a delayed fashion and (2) providing denervated muscle targets, such as vascularized denervated muscle targets (VDMTs), leads to greater pain resolution than innervated muscle targets (bury-in-muscle [BIM]), although the mechanisms underlying these benefits are unclear. We compared behavioral, histological, and transcriptional signatures associated with nerve transection, immediate VDMT (I-VDMT), BIM, and de

Multiscale molecular modeling-directed ROS-responsive nanotherapy for dual-axis regulation of fibrotic and inflammatory signaling in alport nephropathy.

BACKGROUND: In Alport nephropathy, tightly interconnected fibrotic, inflammatory, and oxidative cascades are activated, elevating reactive oxygen species (ROS) that intensify renal injury. The broad activation of stress-responsive and profibrotic pathways further induces disease progression and limits the efficacy of monotherapies. RESULTS: We performed structure-based docking and long-timescale molecular dynamics simulations to identify mechanistically complementary agents, enabling the assessment of ligand stability, specificity, and suitability for selecting an effective drug combination.
