A comparison between the effects of edrophonium and choline in the skeletal muscles of the cat.

The effects of edrophonium and choline have been compared with those of the depolarizing substances acetylcholine, decamethonium, and suxamethonium, in both innervated and chronically denervated tibialis anterior muscles of cats under chloralose anaesthesia. Both edrophonium and choline were more potent antagonists to paralysis by tubocurarine than could be accounted for by their ability to stimulate the motor end-plates directly. It appeared likely that direct depolarization of the end-plate played no part in the anti-curare action of edrophonium and only some part in the anti-curare action o

Can Resistance Training-Induced Skeletal Muscle Growth Improve Local Muscular Endurance?

Skeletal muscle growth occurs in response to training with a wide range of loads, despite traditionally being linked to high-load resistance training and maximal strength development. A body of literature suggests that with lower loads, training near fatigue/task failure is an important stimulus for muscle growth. We consider that training-induced adaptations often improve performance in the specific activity that stimulated them (e.g., increased cardiac output and endurance exercise). Accordingly, we explore skeletal muscle growth as a compensatory adaptation to intense fatiguing exercise, po

High-Resolution Skeletal Muscle Fiber Typing Using Four-Color Immunofluorescence Microscopy.

Skeletal muscle is composed of heterogeneous fiber types defined by distinct myosin heavy chain (MYH) isoforms. Accurate fiber type identification is essential for studying skeletal muscle development, plasticity, disease, and intervention responses. Traditional fiber type classification techniques are limited by either lack of specificity or the inability to simultaneously detect all fiber types in a single section. Here, we describe a novel four-channel immunofluorescent staining protocol that enables simultaneous visualization and quantification of type I, IIA, IIX, and IIB fibers within a

Inhibiting Glutaminase Reverses Senescence-Associated Mitochondrial Dysfunction and Skeletal Muscle Decline.

Age-associated loss of skeletal muscle mass and function, also known as sarcopenia, is closely linked to mitochondrial dysfunction. In this study, we report that senescent skeletal muscle myoblasts exhibit elevated activity and expression of the enzyme glutaminase (GLS1), which is mediated by p38 MAPK signaling and leads to intracellular urea accumulation that impairs mitochondrial function. Pharmacological inhibition of GLS1 with CB-839 reduced urea levels, restored the expression of electron transport chain (ETC) complexes, and improved mitochondrial function. Consistent with our in vitro fi

Aberrant skeletal muscle morphogenesis and myofiber differentiation characterize equine myotonic dystrophy.

Equine myotonic dystrophy (eMD) is a rare neuromuscular disorder of undetermined origin marked by muscle hypertrophy and stiffness, dystrophic muscle histopathology, and myotonic discharges. In humans, myotonic dystrophy (DM) arises from trinucleotide repeat expansions in dystrophia myotonica protein kinase (DMPK) (DM1) or tetranucleotide expansions in cellular nucleic acid-binding protein (CNBP) (DM2), which disrupt mRNA processing and induce embryonic splicing patterns across multiple genes. In 6 eMD Quarter Horse types, (2-36 months-of-age) and 8 control Quarter Horses we determined: (1) fi

E-cigarette exposure impairs skeletal muscle mitochondrial function in male mice.

While conventional tobacco-cigarette smoking continues to decline, e-cigarette (E-cig) use is rising, yet its physiological consequences remain poorly characterized. Chronic activation of the aryl hydrocarbon receptor (AHR) by tobacco smoke impairs skeletal muscle mitochondrial function. Here, we evaluated whether E-cig vapor elicits AHR activation and mitochondrial dysfunction in skeletal muscle. C2C12 mouse myoblasts were exposed to 1% dimethyl sulfoxide (vehicle), 0.02% tobacco-smoke condensate (TSC), or vape condensate (VC) at 0.006%, 0.06%, and 0.3%. Cell viability, AHR-pathway gene expre

Increasing skeletal muscle mass and strength during incretin-based weight loss.

BACKGROUND: Incretin-based therapies produce robust weight loss in obesity and type 2 diabetes mellitus (T2DM), but some weight lost is lean body mass (LBM), raising concern about sarcopenia and functional decline. Men with obesity or type 2 diabetes (T2DM) and testosterone deficiency may lose skeletal muscle mass (SMM) during weight reduction. Whether a program incorporating testosterone therapy and exercise can preserve SMM during incretin-mediated weight loss remains uncertain. METHODS: This retrospective case series included 93 men with obesity, testosterone deficiency symptoms and mornin

Mitochondrial Adaptations in Skeletal Muscle Following Incretin-Based Therapies: In Vitro.

BACKGROUND: Incretin-based therapies such as glucagon-like peptide-1 receptor agonists (GLP-1Ras), dual GLP-1/GIP agonists and amylin analogues have demonstrated significant weight loss benefits. However, their impact on skeletal muscle mitochondrial function, particularly under metabolic stress, remains unclear. This study aimed to investigate the effects of semaglutide (GLP-1RA), tirzepatide (dual GLP-1/GIP agonist) and cagrilintide (amylin analogue) on mitochondrial function in C2C12 skeletal muscle myotubes under both healthy and lipotoxic (palmitic acid-treated) conditions. Sustained Activation of CaMKII Promotes Skeletal Muscle Contractile Dysfunction in Aging.

Sarcopenia, the age-related loss of muscle strength and mass, contributes to adverse health outcomes in older adults. Exercise engages calcium (Ca2+)- and redox-dependent signaling pathways that enhance muscle performance and adaptation, whereas aging disrupts Ca2+ and redox homeostasis. CaMKII is a key transducer of both signals, raising the possibility that sustained CaMKII signaling becomes maladaptive with aging. Here, we show that CaMKII protein abundance is increased in aged mouse skeletal muscle and that sustained CaMKII activation in young muscle is sufficient to impair contractile fun

Canine Adipose MSC-Derived Exosomes Ameliorate Skeletal Muscle Injury in Mice.

Severe skeletal muscle injury in dogs can result in muscle atrophy, fibrotic remodeling, and fat accumulation, leading to skeletal muscle dysfunction and impaired quality of life. However, there is currently no effective treatment available. This study aims to investigate the potential of canine adipose mesenchymal stem cell-derived exosomes (cADMSC-Exos) as a novel acellular therapy for the repair of muscle atrophy and injury. cADMSCs and their derived exosomes were isolated and characterized. A dexamethasone-induced C2C12 myotube atrophy model was established to evaluate the effects of cADMS

RNA Therapeutics Targeting Skeletal Muscle: Emerging Antisense and Gene-Modifying Strategies.

RNA-based therapeutics are reshaping the treatment landscape for skeletal muscle disorders by enabling modulation of RNA processing or direct correction of disease-causing alleles. In Duchenne muscular dystrophy (DMD), four antisense oligonucleotides-eteplirsen, golodirsen, viltolarsen, and casimersen-have received FDA approval; these phosphorodiamidate morpholino oligomers (PMOs) induce exon skipping to restore the reading frame and enable expression of internally truncated dystrophin. Beyond splice switching, RNA therapeutics include RNase H-active gapmers and steric-blocking antisense oligo

Effects of low-dose γ-radiation on skeletal muscle stem cells.

Introduction: Exposure to low dose ionizing radiation (LDIR) has been associated with aging related health effects. The aging related decline in the regenerative capacity of muscle stem cells in elderly individuals, as well as other myopathy conditions, represents a major health issue. Effects of LDIR exposures, such as those from routine medical CT scans on functional status of muscle stem cells, are not known. Methods: We investigated how a single acute 60Co γ-irradiation (10 and 100 mGy) affected myogenesis into mature muscle fibers in cultures of mouse C2C12 myoblasts and biopsy-derived hum

Sarcolemma resilience and skeletal muscle health require O-mannosylation of dystroglycan.

BACKGROUND: Maintaining the connection between skeletal muscle fibers and the surrounding basement membrane is essential for muscle function. Dystroglycan (DG) serves as a basement membrane extracellular matrix (ECM) receptor in many cells, and is also expressed in the outward-facing membrane, or sarcolemma, of skeletal muscle fibers. DG is a transmembrane protein comprised of two subunits: alpha-DG (α-DG), which resides in the peripheral membrane, and beta-DG (β-DG), which spans the membrane to intracellular regions. Extensive post-translational processing and O-mannosylation are required for

MicroRNA-33 inhibition ameliorates muscular dystrophy by enhancing skeletal muscle regeneration.

Muscular dystrophy is a group of diseases characterized by progressive weakness and degeneration of skeletal muscles, for which there is currently no cure. Here, we show that microRNA (miR)-33a/b play a crucial role in muscle regeneration. miR-33a was upregulated during myoblast differentiation and in skeletal muscles of mdx mice, a genetic model of Duchenne muscular dystrophy (DMD). miR-33a deficiency enhanced muscle regeneration response to cardiotoxin injury and attenuated muscle degeneration and fibrosis in mdx mice. Conversely, a humanized mouse model expressing miR-33a and miR-33b showed

Integrative multi-omics uncovers skeletal muscle enhancer programming of cardiorespiratory fitness.

Cardiorespiratory fitness (CRF) is a heritable trait associated with improved metabolic health and reduced mortality. To identify regulatory mechanisms associated with CRF variation, we generated and integrated 546 transcriptomic and epigenomic (chromatin accessibility and histone marks) profiles from 128 genetically heterogeneous rats selectively bred for high and low running capacity, a model that mirrors CRF-associated traits in humans. We found that selection for high CRF led to genetic convergence in skeletal muscle regulatory regions linked to genes involved in lipid metabolism and angio

Congenital Cutaneous Hamartomas With Skeletal Muscle Differentiation Associated With LUMBAR Syndrome.

BACKGROUND/OBJECTIVES: Congenital skin anomalies have been observed in LUMBAR syndrome, but their clinicopathologic significance remains unclear. This study aimed to investigate the congenital, nonvascular skin anomalies in LUMBAR syndrome (lower body infantile hemangiomas, urogenital anomalies and hemangioma ulceration, spinal cord malformations, bony deformities, anorectal/arterial anomalies, and renal anomalies). We hypothesized that an association exists between complex skin hamartomas and LUMBAR syndrome, similar to recent findings in PHACE syndrome (posterior fossa anomalies, hemangioma,

Underexplored terrain: effects of high priority environmental toxicants on skeletal muscle.

In recent decades, evidence has continuously mounted regarding the myriad adverse health effects that environmental exposures exert on human health, yet little attention has been given to skeletal muscle-related outcomes. With its important metabolic, hormonal, and functional properties, skeletal muscle exerts a critical effect on human health and quality of life. The aim of this review was to survey the literature regarding potential impacts of environmental exposures on skeletal muscle health. The focus was on 10 substances atop the Agency for Toxic Substances and Disease Registry's Substanc

TGFBI Facilitates Myogenesis and Limits Fibrosis in Mouse Skeletal Muscle Regeneration.

Skeletal muscles are essential for movement and support but are vulnerable to injury. Muscle regeneration relies on the extracellular matrix (ECM), which regulates key cellular processes. Transforming growth factor β-induced (TGFBI), an ECM component involved in cell adhesion, migration, and tissue development, has not been investigated in skeletal muscle regeneration. Here, we examined the role of TGFBI using Tgfbi knockout (KO) mice and C2C12 myoblasts. In vitro, C2C12 cells were treated with recombinant TGFBI following snake venom (SV)-induced injury, and myogenic differentiation and fusion
