Homocysteine induces vascular inflammatory response via SMAD7 hypermethylation in human umbilical vein smooth muscle cells.

Homocysteine (Hcy) can induce atherosclerosis through the inflammatory response and DNA methylation disorder. Our recent study has reported a novel epigenetic modified gene related to atherosclerosis -SMAD7. To further understand the pathogenesis of atherosclerosis, the current study was designed to investigate an inflammatory role of Hcy in human umbilical vein smooth muscle cells (HUVSMCs) through interfering with SMAD7 methylation. Using MALDI-TOF MS, we found that Hcy increased DNA methylation levels of SMAD7 promoter in a dose and time-dependent manner in HUVSMCs. Meanwhile, both SMAD7 mR

The interaction of IGF-1/IGF-1R and hydrogen sulfide on the proliferation of mouse primary vascular smooth muscle cells.

Hydrogen sulfide (H2S) is mostly produced by cystathionine-gamma-lyase (CSE) in vascular system and it inhibits the proliferation of vascular smooth muscle cells (SMCs). Insulin-like growth factor-1 (IGF-1), via its receptor (IGF-1R), exerts multiple physiological and pathophysiological effects on the vasculature, including stimulating SMC proliferation and migration, and inhibiting SMC apoptosis. Since H2S and IGF-1/IGF-1R have opposite effects on SMC proliferation, it becomes imperative to better understand the interaction of these two signaling mechanisms on SMC proliferation. SMCs isolated

Smooth muscle cell sheet transplantation preserve cardiac function and minimize cardiac remodeling in a rat myocardial infarction model.

BACKGROUND: We examined whether a vascular smooth muscle cell (SMC) sheet is effective in the treatment of a rat myocardial infarction (MI) model. METHODS: We examined the effect of SMC sheet on the cardiac function and cardiac remodeling in a rat MI model in comparison with their effect of dermal fibroblast (DFB) sheet in vivo. Furthermore, we estimated the apoptosis and secretion of angiogenic factor of SMC under hypoxic condition in comparison with DFB. Seven days after MI, monolayer cell sheets were transplanted on the infarcted area (SMC transplantation group, SMC-Tx; DFB transplantation

Integrin-α5β1 is not required for mural cell functions during development of blood vessels but is required for lymphatic-blood vessel separation and lymphovenous valve formation.

Integrin α5β1 is essential for vascular development but it remains unclear precisely where and how it functions. Here, we report that deletion of the gene encoding the integrin-α5 subunit (Itga5) using the Pdgfrb-Cre transgenic mouse line, leads to oedema, haemorrhage and increased levels of embryonic lethality. Unexpectedly, these defects were not caused by loss of α5 from Pdgfrb-Cre expressing mural cells (pericytes and vascular smooth muscle cells), which wrap around the endothelium and stabilise blood vessels, nor by defects in the heart or great vessels, but were due to abnormal developme

Mitochondrial fission induced by platelet-derived growth factor regulates vascular smooth muscle cell bioenergetics and cell proliferation.

Vascular smooth muscle cells (VSMCs) develop a highly proliferative and synthetic phenotype in arterial diseases. Because such phenotypic changes are likely integrated with the energetic state of the cell, we hypothesized that changes in cellular metabolism regulate VSMC plasticity. VSMCs were exposed to platelet-derived growth factor-BB (PDGF) and changes in mitochondrial morphology, proliferation, contractile protein expression, and mitochondrial metabolism were examined. Exposure of VSMCs to PDGF resulted in mitochondrial fragmentation and a 50% decrease in the abundance of mitofusin 2. Syn

Aortic aneurysm generation in mice with targeted deletion of integrin-linked kinase in vascular smooth muscle cells.

RATIONALE: Integrin-linked kinase (ILK) is located at focal adhesions and links the extracellular matrix (ECM) to the actin cytoskeleton via β1- and β3-integrins. ILK plays a role in the activation of kinases including protein kinase B/Akt and glycogen synthase kinase 3β and regulates cell proliferation, motility, and survival. OBJECTIVE: To determine the function of ILK in vascular smooth muscle cells (SMCs) in vivo. METHODS AND RESULTS: SM22Cre(+)Ilk(Fl/Fl) conditional mutant mice were generated in which the Ilk gene was selectively ablated in SMCs. SM22Cre(+)Ilk(Fl/Fl) conditional mutant

Imipramine inhibition of TRPM-like plasmalemmal Mg2+ transport in vascular smooth muscle cells.

Depression is associated with vascular disease, such as myocardial infarction and stroke. Pharmacological treatments may contribute to this association. On the other hand, Mg(2+) deficiency is also known to be a risk factor for the same category of diseases. In the present study, we examined the effect of imipramine on Mg(2+) homeostasis in vascular smooth muscle, especially via melastatin-type transient receptor potential (TRPM)-like Mg(2+) -permeable channels. The intracellular free Mg(2+) concentration ([Mg(2+) ](i) ) was measured using (31) P-nuclear magnetic resonance (NMR) in porcine car

An angiogenesis inhibitor TNP-470 (AGM-1470) suppresses vascular smooth muscle cell proliferation in experimental rat aortotomy models.

AIM: Vascular intimal hyperplasia is an important clinical concern in vascular diseases, such as anastomotic stricture as a possible complication of cardiovascular surgery. We recently suggested that a rat aortotomy model could be substituted for a vascular anastomotic stricture around a suture line. TNP-470 is known as an angiogenesis inhibitor and has demonstrated abilities to inhibit DNA synthesis of smooth muscle cells (SMCs) and SMCs proliferation. The aim of this study was to investigate the effect of TNP-470 on SMC proliferation using rat aortotomy models. METHODS: Longitudinal aortoto

Intracellular calcium transients are necessary for platelet-derived growth factor but not extracellular matrix protein-induced vascular smooth muscle cell migration.

PURPOSE: Vascular smooth muscle cell (SMC) migration is a critical component of the hyperplastic response that leads to recurrent stenosis after interventions to treat arterial occlusive disease. We investigated the relationship between intracellular calcium ([Ca(2+)](i)) and migration of vascular SMCs in response to platelet-derived growth factor (PDGF) and extracellular matrix (ECM) proteins. METHODS: Human saphenous vein SMCs were used for all experiments. SMC migration in response to agonists was measured with a microchemotaxis assay. A standard fluorimetric assay was used to assess chang

Vascular smooth muscle cells and calcification in atherosclerosis.

Vascular calcification is a prominent feature of atherosclerosis but the mechanisms underlying vascular calcification are still obscure. Since bone-associated proteins such as osteonectin, osteocalcin, and matrix Gla protein have been detected in calcified vascular tissues, calcification has been considered to be an organized, regulated process similar to mineralization in bone tissue. Vascular smooth muscle cells (VSMCs) are currently considered to be responsible for the formation of vascular calcifications. Apoptosis of VSMCs appears to be a key factor in this process, while other factors in

STF083010 Mitigates Thoracic Aortic Dissection by Modulating the NLRP3 Inflammasome-Dependent Pyroptosis.

Thoracic aortic dissection (TAD) is a life-threatening vascular pathology with limited therapeutic options. The role of the small molecule STF083010, a novel IRE1α RNase-specific inhibitor, in TAD has not yet been defined. We aimed to target the IRE1α-XBP1s axis pharmacologically using STF083010, and evaluate its therapeutic potential in the occurrence and progression of TAD. Using a β-aminopropionitrile monofumarate (BAPN)-induced murine TAD model combined with interleukin 1β (IL1β)-stimulated vascular smooth muscle cells (VSMCs), we systematically evaluated the effects of STF083010 on the de

Exercise Attenuates Aortic Dissection Via PDE5A-Mediated Inhibition of Vascular Smooth Muscle Cell Phenotypic Switch.

BACKGROUND: Aortic dissection (AD) is a life-threatening condition and involves pathological vascular smooth muscle cell (VSMC) phenotypic switching from a contractile to a synthetic state. Although exercise confers broad cardiovascular benefits, its role in AD pathogenesis remains unclear. This study aimed to determine whether exercise attenuates AD by modulating VSMC phenotype and to elucidate the underlying molecular mechanism. METHODS: Human aortic tissues from AD patients were analyzed for VSMC phenotypic markers and PDE5A (phosphodiesterase 5A) expression. A β-aminopropionitrile induced

VSMC-derived 3HAA activates AhR to drive abdominal aortic aneurysm formation via the p62-Nrf2 axis.

BACKGROUND: Abdominal aortic aneurysm (AAA) remains a life-threatening vascular disease without effective pharmacologic therapy. Although 3-hydroxyanthranilic acid (3HAA), a kynurenine-pathway metabolite with dual redox effects, has been implicated in vascular diseases. How 3HAA instigates AAA is poorly defined. METHODS: We combined human AAA tissues, genetically modified mice, pharmacologic kynurenine-pathway inhibition, and mechanistic vascular smooth muscle cell (VSMC) studies to define 3HAA-AhR signaling in aneurysm formation. RESULTS: In angiotensin (Ang) II-infused apoE-/- mice, plasma

CXCL16 promotes macrophage-driven inflammation and vascular smooth muscle cell phenotypic switching during carotid plaque destabilization.

BACKGROUND: Carotid plaque instability is a major determinant of ischemic stroke and is characterized by heightened inflammation and structural remodeling of the vessel wall. Although macrophages and vascular smooth muscle cells (VSMCs) are central to plaque vulnerability, the mechanisms coordinating immune activation with vascular remodeling remain incompletely understood. METHODS: Bulk transcriptomic data from multiple Gene Expression Omnibus (GEO) datasets were integrated to compare unstable and stable carotid plaques. Dual Lineage Tracing Identifies Cellular Mechanisms Underlying Radiation-Associated Changes in Atherosclerotic Lesion Composition-Brief Report.

BACKGROUND: Phenotypic plasticity of smooth muscle cells (SMCs) and endothelial cells (ECs) contributes to atherosclerotic plaque composition and stability, yet how shifts in one population influence the contribution and function of the other under conditions of vascular stress, such as irradiation, is poorly understood. A major limitation has been the inability to simultaneously fate-map both cell types within the same lesion, with most studies mapping one lineage while inferring the other using unreliable dynamically changing marker genes, risking false-positive and false-negative assignment

CD81 restricts decidual IGF2-mediated smooth muscle cell dedifferentiation during spiral artery remodeling in preeclampsia.

BACKGROUND: Preeclampsia (PE) is a pregnancy-specific complication characterized by defective spiral artery remodeling, notably due to the abnormal retention of differentiated vascular smooth muscle cells (VSMCs). Although elevated CD81 levels in the placenta and maternal circulation contribute to this impairment, the definite mechanisms remain elusive. This study aimed to investigate how CD81 overexpression in extravillous trophoblasts (EVTs) contributes to defective VSMC dedifferentiation within uterine spiral arteries, as well as the involved molecular mechanisms. METHODS: Placental basal

ATAD3A Limits Aortic Dissection via Mito-Lysosome Contacts and Lipoylation.

BACKGROUND: Aortic aneurysm and dissection (AAD) is a fatal vascular emergency with limited mechanism-based therapies. ATAD3A (ATPase family AAA [ATPases associated with diverse cellular activities] domain-containing protein 3A), a mitochondrial AAA+ ATPase enriched at organelle contact sites, has been implicated in mitochondrial signaling, but its role in AAD remains unclear. METHODS: AAD was induced in wild-type, ATAD3A knock-in, and vascular smooth muscle cell (VSMC)-specific knockdown mice generated using short hairpin RNA against ATAD3A (sh-ATAD3A) by 3-week β-aminopropionitrile monofuma

Cellular crosstalk between endothelial and vascular smooth muscle cells via the BACH1/COL3A1 axis promotes vascular calcification in chronic kidney disease.

BACKGROUND: High phosphate levels lead to significant pathological changes in vascular smooth muscle cells (VSMCs) and endothelial cells, driving vascular disease in chronic kidney disease (CKD). Endothelial cell senescence plays a major role in this process, promoting vascular calcification by fostering a pro-inflammatory and pro-osteogenic environment. Interaction between VSMCs and endothelial cells further accelerates calcification, although the precise mechanisms remain incompletely understood. METHODS: Exosomes were isolated and characterized, with RT-qPCR, western blot, immunohistochemi

Indoxyl Sulfate Promotes Vascular Calcification in Association with Oxidative Stress and Activation of ERK and Wnt/β-Catenin Signaling Pathways.

Vascular calcification (VC) is a major complication of chronic kidney disease (CKD) that is strongly associated with cardiovascular mortality. While indoxyl sulfate (IS), a protein-bound uremic toxin, has been implicated in the progression of VC, the underlying molecular mechanisms remain unclear. We investigated the procalcific effects of IS using a two-step nephrectomy-induced CKD mouse model and cultured vascular smooth muscle cells. In vivo, progressive renal impairment was associated with elevated circulating IS levels and enhanced VC. In vitro, IS dose- and time-dependently induced calci

Growth factors involved in vascular remodeling in pulmonary arterial hypertension.

Pulmonary arterial hypertension (PAH) is a rare and fatal cardiovascular disease characterized by pulmonary vascular remodeling, leading to a progressive increase in pulmonary vascular resistance and pulmonary arterial pressure (PAP). Elevated PAP induces right ventricular hypertrophy and eventually progresses to right heart failure. Pulmonary vascular remodeling is primarily caused by the excessive proliferation and migration of pulmonary arterial smooth muscle cells (PASMCs) in the medial layer. This is mediated by the binding of growth factors to their specific receptor tyrosine kinases. To
