The left-right side-specific endocrine signaling in the effects of brain lesions: questioning of the neurological dogma.

Each cerebral hemisphere is functionally connected to the contralateral side of the body through the decussating neural tracts. The crossed neural pathways set a basis for contralateral effects of brain injury such hemiparesis and hemiplegia as it has been already noted by Hippocrates. Recent studies demonstrated that, in addition to neural mechanisms, the contralateral effects of brain lesions are mediated through the humoral pathway by neurohormones that produce either the left or right side-specific effects. The side-specific humoral signaling defines whether the left or right limbs are aff…

Identification of c-Kit receptor as a regulator of adult neural stem cells in the mammalian eye: interactions with Notch signaling.

Neural stem cells are present in specific regions of the adult central nervous system (CNS). Recent evidence suggests that the ciliary epithelium (CE), a CNS derivative, in the adult mammalian eye, harbors a quiescent population of neural stem cells. Here, we report the identification of c-Kit signaling as one of the regulators of adult CE neural stem cells in vitro. c-Kit receptors are expressed in proliferating adult CE neural stem cells and colocalized with neural progenitor markers. Perturbation of c-Kit signaling influences the self-renewal and differentiation of CE neural stem cells, thu

Electrical Microenvironment Reconstruction and the Application of Biomaterials in Spinal Cord Injury.

The key challenge in restoring neural function after spinal cord injury stems from a vicious cycle triggered by the collapse of the bioelectrical microenvironment at the injury site: an 'electrical silence-neuronal degeneration-glial proliferation' cascade that conventional therapies fail to reverse. This review systematically summarizes the pathological mechanisms of electrical microenvironment imbalance and its critical role in neural regeneration. Furthermore, current intervention strategies based on biomaterials are outlined: evolving from passive reconstruction of electrical pathways usin

The role of the precuneus and posterior cingulate cortex in the neural routes to action.

Источник пометил публикацию как отозванную

Neural substrates of action to the object or this specific direct route, however, remain unclear, especially for the connection from the visual pathway to the motor cortex. The study examined this issue by conducting an fMRI experiment, in which two action generation tasks involving pictures of real objects (PA) and the object's nouns (NA) were used, with pictures naming (PN) and covert noun reading (NR) being the control tasks. The result showed that the model predefined for the PCC and precuneus connecting IPL to the posterior-medial frontal cortex dominated over the others (with 0.45 probab

Transcriptomes of Dravet syndrome iPSC derived GABAergic cells reveal dysregulated pathways for chromatin remodeling and neurodevelopment.

Dravet syndrome (DS) is an early onset refractory epilepsy typically caused by de novo heterozygous variants in SCN1A encoding the α-subunit of the neuronal sodium channel Nav1.1. The syndrome is characterized by age-related progression of seizures, cognitive decline and movement disorders. We hypothesized that the distinct neurodevelopmental features in DS are caused by the disruption of molecular pathways in Nav1.1 haploinsufficient cells resulting in perturbed neural differentiation and maturation. Here, we established DS-patient and control induced pluripotent stem cell derived neural prog

Norepinephrinergic projection from locus coeruleus to parafascicular nucleus promotes pain and anxiety-like behaviors in mice.

Chronic neuropathic pain is frequently comorbid with anxiety disorders, yet the neural circuits underlying this interaction remain poorly defined. The parafascicular nucleus (PF) of the thalamus integrates nociceptive and affective signals, but its specific regulatory mechanisms in pain-anxiety comorbidity are not well known. Using spared nerve injury (SNI) model mice, we combined viral neural tracing, chemogenetics, pharmacology, and electrophysiology to dissect the locus coeruleus (LC)-PF neural pathway. Viral tracing revealed monosynaptic projections from norepinephrinergic (NEergic) neuron

Regulatory gene analysis in enteric neural crest cells stratified by GDNF activation status: An insight into Hirschsprung disease pathophysiology.

BACKGROUND: Incomplete migration of enteric neural crest cells (ENCCs) into the colon underlies the pathogenesis of Hirschsprung disease, resulting in aganglionosis and disordered intestinal motility. Although activation of the glial cell line-derived neurotrophic factor (GDNF) pathway has been implicated in neurosphere differentiation, specific regulatory genes governing ENCC differentiation and proliferation remain poorly characterized. This study seeks to elucidate regulatory genes and pathways involved in neural crest cell differentiation, furthering our understanding of GDNF's role in ent

The glutamatergic NTS-PAG/PVN pathway underlies myocardial injury and anxiety-like behavior induced by myocardial ischemia-reperfusion.

For patients with myocardial ischemia-reperfusion injury (MIRI), cardiac dysfunction accompanied by anxiety is an extremely common comorbidity. Although the association between these two conditions has received some attention, the underlying neural mechanisms remain unclear. Here, we functionally characterized two distinct glutamatergic (Glu) neural pathways in the nucleus tractus solitarius (NTS) of male mice, a pathway from the NTS to the periaqueductal gray (PAG) that mediates anxiety-like behaviors in MIRI mice, and a pathway from the NTS to the paraventricular nucleus of the hypothalamus …

A dual path graph neural network framework for dementia diagnosis.

Dementia typically results from damage to neural pathways and the consequent degeneration of neuronal connections. Graph neural networks (GNNs) have been widely employed to model complex brain networks. However, leveraging the complementary temporal, spatial, and spectral features for diagnosing neurocognitive disorders remains challenging. To address this issue, we propose a Bi-path Multi-scale Graph Neural Network (Bi-MCGNN), which integrates two paths : one focusing on time and spatial relationships, and the other on spatial and frequency patterns. By unifying these pathways, Bi-MCGNN integ…

Neural Tube Defects and Folate Deficiency: Is DNA Repair Defective?

Neural tube defects (NTDs) are complex congenital malformations resulting from failure of neural tube closure during embryogenesis, which is affected by the interaction of genetic and environmental factors. It is well known that folate deficiency increases the incidence of NTDs; however, the underlying mechanism remains unclear. Folate deficiency not only causes DNA hypomethylation, but also blocks the synthesis of 2'-deoxythymidine-5'-monophosphate (dTMP) and increases uracil misincorporation, resulting in genomic instabilities such as base mismatch, DNA breakage, and even chromosome aberrati

Common and distinct neural underpinnings of the association between childhood maltreatment and depression and aggressive behavior.

BACKGROUND: Although childhood maltreatment (CM) is widely recognized as a transdiagnostic risk factor for various internalizing and externalizing psychological disorders, the neural basis underlying this association remain unclear. The potential reasons for the inconsistent findings may be attributed to the involvement of both common and specific neural pathways that mediate the influence of childhood maltreatment on the emergence of psychopathological conditions. METHODS: This study aimed to delineate both the common and distinct neural pathways linking childhood maltreatment to depression …

Impaired connectivity between the thalamus and the visual pathway in schizophrenia: a multimodal magnetic resonance imaging study.

BACKGROUND: Schizophrenia is a severe psychiatric disorder characterized by cognitive deficits as well as positive and negative symptoms. It is considered a disorder of widespread network dysconnectivity, including aberrant connectivity between the thalamus and the visual pathway. However, the relationships between the thalamus and various regions of the dorsal and ventral visual pathways in schizophrenia, and how the thalamus affects interactions among these visual regions, remain unclear. METHODS: Resting-state functional magnetic resonance imaging, task-state functional magnetic resonance

The attentional switching framework: A dual process account of how the brain switches when attention switches.

The capacity to flexibly shift attention between external and internal events is critical for everyday tasks, yet research on how this shifting operates at the cognitive and neural level is still emerging. Here, we examined the question: How does the brain switch when attention switches between external and internal states? We propose the Attentional Switching Framework, which postulates that switching between external and internal attention states involves two seemingly opposing mechanisms subserved by distinct neural bases. First, based on our Resource Dependent Account, the regulation of at

Signals controlling neural crest contributions to the heart.

Cardiac neural crest cells represent a unique subpopulation of cranial neural crest cells that are specified, delaminate and migrate from the developing neural tube to the caudal pharynx where they support aortic arch artery development. From the caudal pharynx, a subset of these cells migrates into the cardiac outflow tract where they are needed for outflow septation. Many signaling factors are known to be involved in specifying and triggering the migration of neural crest cells. These factors have not been specifically studied in cardiac crest but are assumed to be the same as for the other
