A postmortem study suggests a revision of the dual-hit hypothesis of Parkinson's disease.

The dual-hit hypothesis of Parkinson's disease (PD) originally postulated that a neurotropic pathogen leads to formation of α-synuclein pathology in the olfactory bulb (OB) and dorsal motor nucleus of the vagus (DMV) and then invades the brain from these two entry points. Little work has been conducted to validate an important underlying premise for the dual-hit hypothesis, namely that the initial Lewy pathology does arise simultaneously in the OB and the enteric nervous system (ENS) plexuses and DMV at the earliest disease stage. We conducted a focused re-analysis of two postmortem datasets, …

PROKR2 and PROK2 mutations cause isolated congenital anosmia without gonadotropic deficiency.

OBJECTIVE: Isolated congenital anosmia (ICA) is a rare phenotype defined as absent recall of any olfactory sensations since birth and the absence of any disease known to cause anosmia. Although most cases of ICA are sporadic, reports of familial cases suggest a genetic cause. ICA due to olfactory bulb agenesis and associated to hypogonadotropic hypogonadism defines Kallmann syndrome (KS), in which several gene defects have been described. In KS families, the phenotype may be restricted to ICA. We therefore hypothesized that mutations in KS genes cause ICA in patients, even in the absence of fa…

THE LIMITED NEUROTROPIC CHARACTER OF THE ENCEPHALITIS VIRUS (ST. LOUIS TYPE) IN SUSCEPTIBLE MICE.

1. St. Louis encephalitis virus injected intracerebrally into susceptible mice multiplies there to reach a titre of 10(9) intracerebral lethal doses. It is found also in the blood in small amounts immediately following injection and preceding death. 2. Injected intraperitoneally or subcutaneously the virus circulates in the blood for several hours and survives in the spleen for days. It does not multiply in the brain and cause encephalitis, however, unless overwhelming doses are injected or the brain is traumatized. 3. Virus dropped into the nares is demonstrable in the olfactory bulbs at 24 h…

Acute N-Acetylcysteine Administration Ameliorates Loss of Olfactory Neurons Following Experimental Injury In Vivo.

The olfactory epithelium (OE) is the peripheral organ for the sense of smell, housing primary sensory neurons that project axons from the no … n a reduction of olfactory neuronal loss from the OE of the nose following surgical ablation of the olfactory bulb. At 1 week postlesion, we identified 54 ± 8.1 mature neurons per 0.5 mm epithelium in NAC-treated animals vs. 28 ± 4.2 in vehicle-treated controls (P = 0.02). Furthermore, in an olfactory cell culture model, we have identified significant alteration …

Expression of L1 and N-CAM cell adhesion molecules during development of the mouse olfactory system.

The expression of the neural adhesion molecules L1 and N-CAM has been studied in the embryonic and early postnatal olfactory system of the m … y expressed at contact sites between olfactory axons and their target cells in the glomeruli of the olfactory bulb. L1 and N-CAM 180, the component of N-CAM that accumulates at cell contacts by interaction with the cytoskeleton are detectable as early as the first axons extend toward the primordial olfactory bulb. L1 remains prominent throughout development on a …

Regeneration in the olfactory system of vertebrates.

The olfactory sensory neurons, which are of placodal origin, maintain in adult vertebrates, including mammals, a neurogenetic matrix that allows for their replacement without impairment of function, both in normal circumstances (turnover) and after experimental intervention. The unique characteristics of these "disposable" neurons have recently led to a number of experiments whose results may have some importance in our understanding of the olfactory system and of the nervous system as a whole.

A molecular approach to the pathophysiology of the X chromosome-linked Kallmann's syndrome.

The human KAL gene is responsible for the X chromosome-linked Kallmann's syndrome, which consists of an association between hypogonadotropic hypogonadism and anosmia (or hyposmia). Additional symptoms are occasionally observed. The olfactory defect is associated with hypoplasia of the olfactory bulbs and tracts. The hypogonadism may be due to a defect in the embryonic migratory process of GnRH-synthesizing neurones from the olfactory pits up to the brain. The human and chicken KAL genes have been isolated. From the amino acid sequences deduced, it has been postulated that the KAL protein is an…

Localization of phosphotyrosine adaptor protein ShcD/SHC4 in the adult rat central nervous system.

BACKGROUND: Mammalian Shc (Src homology and collagen) proteins comprise a family of four phosphotyrosine adaptor molecules which exhibit var … d distribution in the adult brain and spinal cord, with prominent levels of staining throughout the olfactory bulb, as well as in sub-structures of the cerebellum and hippocampus, including the subgranular zone. Co-localization studies confirm the expression of ShcD in mature neurons and progenitor cells. ShcD immunoreactivity is primarily localized to axons and …

Neurophysiologic detector-a selective and sensitive tool in high-performance liquid chromatography.

In the present study neurons from the olfactory system of the fish crucian carp, Carassius carassius L. were used as components in an in-line neurophysiologic detector (NPD) to measure physiological activities following the separation of substances by high-performance liquid chromatography (HPLC). The skin of crucian carp, C. carassius L. contains pheromones that induce an alarm reaction in conspecifics. Extra-cellular recordings were made from neurons situated in the posterior part of the medial region of the olfactory bulb known to mediate this alarm reaction. The nervous activity of these s…

Two cases of subfrontal schwannoma, including a rare case located between the endosteal and meningeal layers of the dura.

Subfrontal schwannomas arising from the olfactory groove are rare and their origin remains uncertain because olfactory bulbs do not possess Schwann cells. We present two cases of subfrontal schwannomas treated with surgical resection. In one case, the tumor was located between the endosteal and meningeal layers of the dura mater. This rare case suggests that subfrontal schwannomas may originate from the fila olfactoria.

Structure of the principal olfactory tract.

Although the purpose and importance of the sense of smell in human beings has not been totally clarified, it is one of the principal informa … ctory tract components. Our study of the connections between the olfactory mucosa and the principal olfactory bulb deserves special mention. The distribution of second neuron connections of the olfactory tract with the central nervous system is quite complex and diffuse. This indicates an interrelation between the sense of smell and a multitude of functions. The …

Olfactory but not vomeronasal mediation of scent marking by male golden hamsters.

Although olfactory and vomeronasal projections to the olfactory bulb and to the rest of the brain are quite distinct, it is not clear how the functions of these two chemosensory systems differ. In these experiments we attempted to determine the roles of the main olfactory and vomeronasal sensory systems in mediating scent marking by male golden hamsters, a behavior known to be stimulated by odors of other hamsters. Lesions of the main olfactory mucosa by irrigation of the nasal cavity with a solution of zinc sulfate dramatically reduced the frequency of marking. The degree of reduction in mark…

Dopaminergic modulation at the olfactory nerve synapse.

Dopamine can change the membrane potential, regulate cyclic nucleotides, and modulate transmitter release in central neurons. In the olfactory bulb (OB), the dopamine synthetic enzyme, tyrosine hydroxylase, is largely confined to neurons in the glomerular layer. After demonstrating dopamine D2 receptors in the glomerular and olfactory nerve (ON) layers, Nickell et al. [W.T. Nickell, A.B. Norman, L.M. Wyatt, M.T. Shipley, Olfactory bulb DA receptors may be located on terminals of the olfactory nerve, NeuroReport, 2 (1991) 9-12.] proposed that these receptors may reduce transmitter release due t…

Associations between olfactory dysfunction and structural MRI findings in Parkinson's disease: a systematic review.

Olfactory dysfunction is a common and early non-motor symptom of Parkinson's disease (PD). Structural Magnetic Resonance Imaging (MRI) studies have reported changes in olfactory-related brain regions, but findings remain inconsistent. This review synthesizes MRI evidence to clarify neuroanatomical correlates of olfactory dysfunction in PD. We performed a systematic review to find the associations between olfactory dysfunction and structural MRI findings in people with PD. A systematic search in PubMed, Scopus, and Web of Science yielded 20 eligible studies. The eligible studies involved 1,092
