Corneal blindness has been a significant, in most cases, reversible cause of visual impairment worldwide due to donor deficiency and donor graft failure, which has encouraged the consideration of donor-independent techniques for regeneration. This review aims to discuss the advances in corneal organoid-based tissue engineering and its potential application in the translation to vision restoration. This review was conducted through an analysis of publications related to corneal organoids, biomaterials, bioprinting, preclinical models, and early human studies, published between 2005-2025 in Scop…

BACKGROUND: In vitro intestinal epithelial models are being developed for studying gut physiology, barrier function, and drug transport. There are two major in vitro models which are currently used: cell line-derived monolayers (such as Caco-2/HT29 cocultures) and primary cell monolayers derived from human intestinal organoids.
AIM: To conduct a side by side comparison of organoid-derived and cell line-derived intestinal epithelial monolayers, assessing their morphological features, barrier performance, and secretome under similar culture conditions.
METHODS: Human duodenum-derived organoids…

Male reproductive health has declined markedly over the past decades, with environmental chemicals, pharmaceuticals, and lifestyle factors i … nsion, alongside matrix choices and microfluidic culture platforms. It then examines how testicular organoids have been deployed to assess endocrine-disrupting chemicals, chemotherapeutics, and other pharmacological agents, where several studies report the detection of effects at concentrations appreciably below those required in monolayer systems, particul …

Human brain organoids have evolved from early neurodevelopmental models into platforms for therapeutic discovery. Here, we highlight two cases in which organoid-derived findings enabled FDA-approved clinical trials. Patient-derived organoids modeling Pitt-Hopkins syndrome revealed human-specific, TCF4-dependent abnormalities and supported the development of a regulated AAV gene therapy. In parallel, Rett syndrome organoids cultured aboard the International Space Station uncovered space-induced neural senescence, characterized by retroelement-associated neuroinflammation, prompting evaluation o…

Cervical cancer, a human papillomavirus (HPV) -driven malignancy with inherent viral antigenicity, serves as a key model in immuno-oncology. Yet, the variable efficacy of immune checkpoint inhibitors highlights a critical gap: the lack of preclinical models that accurately mirror the patient-specific tumor microenvironment (TME). Patient-derived organoids (PDOs) offer a transformative solution, preserving autologous tumor and immune components for personalized therapeutic testing. This study provides the first bibliometric analysis of this evolving field, mapping its knowledge structure from m…

Neurological diseases are a leading cause of disability, morbidity, and mortality, affecting 43% of the world's population. The detailed study of neurological diseases, testing of drugs, and repair of site-specific defects require physiologically relevant models that recapitulate key events and dynamic neurodevelopmental processes in a highly organized fashion. As an evolving technology, self-organizing and self-assembling brain organoids offer the advantage of modeling different stages of brain development in a 3D microenvironment. Herein, we review the utility, advantages, and limitations of…

Small cell carcinoma of the ovary, hypercalcemic type (SCCOHT) is a rare and lethal tumor in adolescent and young adult patients. Now, there is no standard-of-care treatment for these patients. Reliable models that represent this disease and can be used for translational research are scarce. To model SCCOHTs, we have established eight patient-derived tumoroid lines from tumor lesions of three patients with SCCOHT. The tumoroids recapitulate genomic and transcriptomic characteristics of the corresponding patient tumors and capture intrapatient tumor heterogeneity. Organoid drug profiling using

The tongue is essential for swallowing, taste perception, and mechanosensation. The anterior and posterior parts of the tongue have region-specific developmental origins and are maintained by adult epithelial stem/progenitor cells. In vitro models that can be used to investigate anterior tongue biology have been lacking. Here, a protocol is developed to generate a long-term expanding organoid model from the adult mouse dorsal anterior tongue. Anterior tongue organoids consist of Lgr6+ cells, Sox2+ stem/progenitor cells, and Hoxc13+ filiform papillae progenitor cells. Furthermore, anterior tong…

Intestinal organoids have become an important model system in basic and translational research, but their culture typically relies on an ill-defined laminin-rich extracellular matrix (ECM). Using tunable and chemically defined 3D hydrogels, we systematically explored the role of the ECM during murine and human intestinal organoid development. We discovered that without exogenous laminin, stem cells developed into intestinal epithelia with a large proportion of regenerative cells. This population secreted a laminin-rich basement membrane that functioned as a de novo stem cell niche, promoting o…

The biliary system is vital to hepatobiliary function, yet diseases like primary sclerosing cholangitis, biliary atresia, cystic fibrosis, and cholangiocarcinoma remain poorly understood due to the limitations of traditional two-dimensional (2D) cell cultures and animal models, which fail to replicate complex human biliary physiology. Biliary organoids, an innovative three-dimensional (3D) in vitro model, have emerged to bridge this gap, closely mimicking tissue structure and function. This review systematically summarizes the construction methods of biliary organoids-matrix-independent method…

Brain organoid technology has revolutionized in vitro modeling of human neurodevelopment and disease, providing unprecedented insights into cortical patterning, neural circuit assembly, and pathogenic mechanisms of neurological disorders. Critically, human brain organoids uniquely recapitulate human-specific developmental processes-such as the expansion of outer radial glia and neuromelanin-that are absent in rodent models, making them indispensable for studying human brain evolution and dysfunction. However, a major bottleneck persists: Extended culture periods (≥6 months) are empirically req…

BACKGROUND: The advent of organoid culture systems has revolutionized our ability to model and study complex tissues in vitro. The placenta is one of the last human organs to have a functional organoid model developed: trophoblast organoids. These 3-dimensional structures, derived from placental tissue, offer researchers a valuable tool for studying previously inaccessible processes that occur within the womb and play a significant role in determining the health of the offspring. While primarily used for research, trophoblast organoids hold promise for clinical applications, including prenatal…

Trophoblast organoids can provide crucial insights into mechanisms of placentation, however their potential is limited by highly variable extracellular matrices unable to reflect in vivo tissues. Here, we present a bioprinted placental organoid model, generated using the first trimester trophoblast cell line, ACH-3P, and a synthetic polyethylene glycol (PEG) matrix. Bioprinted or Matrigel-embedded organoids differentiate spontaneously from cytotrophoblasts into two major subtypes: extravillous trophoblasts (EVTs) and syncytiotrophoblasts (STBs). Bioprinted organoids are driven towards EVT diff…

The drug discovery and development process faces significant challenges, including high attrition rates and substantial financial investment, in part due to the limitations of traditional 2-dimensional (2D) cell culture systems and animal models to predict human drug metabolism, efficacy, and toxicity. This review highlights the emergence of novel in vitro human cell culture and organoid systems, such as 3-dimensional (3D) cultures, self-assembling organoids, induced pluripotent stem cell-derived models, and microphysiological system or organ-on-a-chip systems, as transformative solutions to t…

OBJECTIVE: Prenatal exposure to environmental endocrine-disrupting chemicals (EDCs) has been increasingly linked to neurodevelopmental impairment. Bisphenol S (BPS) and perfluoro-octane sulfonate (PFOS), two widely distributed EDCs detected in maternal and fetal tissues, raise concern due to their potential to interfere with brain development even at low environmental doses.
METHODS: a phenotypic screening on human iPSC-derived cerebral organoids was performed to explore whether chronic exposure to BPS and PFOS could affect key neurodevelopmental processes.
RESULTS: Both compounds affected k…
