Detection of Permethrin Resistance and Fipronil Tolerance in Rhipicephalus sanguineus (Acari: Ixodidae) in the United States.

Permethrin is a commonly used acaricide for tick control on domestic animals and in residential environments, while fipronil use is restricted to on-animal treatment. Following widespread reports of permethrin and fipronil application failures to control indoor infestations of Rhipicephalus sanguineus (Latreille), the brown dog tick, 31 tick populations were obtained from Florida and Texas for acaricide resistance screening. These field-collected ticks from kennels and residential facilities were challenged with technical grade permethrin and fipronil to create dose response curves that were c

Thrombin-induced transfer of arachidonic acid in human platelets is not inhibited by trifluoperazine.

Human platelets have been shown to contain a Ca++- and CoA-independent transacylase enzyme that catalyzes the transfer of arachidonic acid from phosphatidylcholine (PC) to lysoplasmenylethanolamine. It has been suggested that this route may represent a major source for released arachidonic acid in stimulated platelets. In this study, we have shown using arachidonic-labelled human platelets that the thrombin-induced activation of a transacylase reaction was not affected by concentrations of trifluoperazine (TFP) (15 micrograms/2 X 10(9) cells) which abolished the accumulation of free [3H]arachi

Investigation of the Mechanism of Lorlatinib-Induced Hepatotoxicity: Reactive Metabolites of Lorlatinib Activate Inflammasomes.

Lorlatinib, a third-generation anaplastic lymphoma kinase (ALK) tyrosine kinase inhibitor, is widely used to treat ALK-positive nonsmall cell lung cancer. However, lorlatinib has occasionally been associated with severe liver injury. Immune-mediated adverse events are frequently provoked by certain pharmacological agents or their reactive metabolites. However, the specific contribution of immune pathways to hepatotoxicity caused by lorlatinib is yet to be fully elucidated. In the present study, the potential for direct inflammasome stimulation by lorlatinib was examined in differentiated THP-1

Halogen-Bond Dimensionality Controls π-π Packing to Unlock Ultrahigh Second-Harmonic Generation and Giant Birefringence.

Compact all-solid-state lasers require nonlinear optical crystals that combine strong second-harmonic generation (SHG) with large birefringence, yet planar π-conjugated molecules often pack antiparallel, canceling their macroscopic second-order response. Here, we break this trade-off through dimensional competition between halogen bonding and π-π stacking in two nitroazole-based organic frameworks. The parent (IClPzNO2)2·H2O forms a centrosymmetric two-dimensional framework with coplanar chromophores and a birefringence of 0.705 at 546 nm. Combined skeletal and halogen substitutional editing,

Targeting DNA gyrase B: medicinal chemistry strategies to combat antimicrobial resistance.

Antimicrobial resistance (AMR) represents one of the most pressing global healthcare challenges, necessitating the discovery of novel antibacterial agents with distinct mechanisms of action. DNA gyrase, an essential bacterial type II topoisomerase absent in humans, has emerged as an attractive antibacterial target. In particular, the ATPase domain of the GyrB subunit is highly conserved across ESKAPE pathogens, providing a strong rationale for the development of broad-spectrum antibacterial agents. Several natural product-derived antibiotics, including novobiocin, clorobiocin, and coumermycin

HyDRA-II: spectroscopic results and BEsT guesses for the mono- and dihydrate blind challenge.

HyDRA blind challenges aim at unbiased forecasts of water vibrations in vacuum-isolated hydrate complexes of organic molecules by different computational protocols. In this work, directly after the completion of the HyDRA II challenge and ignorant of its computational results, the experimental findings for the 9 new solvates with a total of 12 hydrogen-bonded water OH stretching wavenumbers are disclosed and discussed. We further summarise and analyse the 70 entries of the training database, which were made available at the start of the challenge. Monohydrates of 4 other previously uncharacter

Unlocking catalytic capacity of (S)-selective ω-transaminases toward aryl methyl ketones via pocket engineering and droplet screening.

ω-Transaminases (ω-TAs) exhibit remarkable efficiency and stereoselectivity in catalyzing the asymmetric synthesis of chiral amines, rendering them prime biocatalysts for pharmaceutical chiral intermediate production. Nevertheless, developing a generalizable strategy to significantly enhance the acceptance of bulky substrates across different ω-TAs remains a major challenge, as most enzyme engineering efforts are case-specific and lack of high-efficiency screening approaches. Herein, we presented an efficient ω-TAs engineering strategy, established via the direct conversion of 1-(6-(4-fluoro-1

Synthesis and photoluminescence of iridium(III) arylacetylide complexes with acetylide-localized emissive excited states.

Herein, we report a series of three bis-cyclometalated iridium(III) complexes each bearing an arylacetylide and an N-heterocyclic carbene (NHC) ligand. The cyclometalating ligand is 2-(2,4-difluorophenyl)pyrazole (F2ppz), chosen to ensure the 5d(Ir) → π*(F2ppz) 3MLCT excited-state is higher in energy than the 3(π → π*) state of the acetylide, allowing luminescence to originate from the latter. The strong-field NHC is included to alleviate photoluminescence (PL) quenching caused by the thermal population of the triplet metal-centered (3MC) states, but nevertheless, the three compounds are weakl

The developmental and inflammatory disease target protein ADAM17 is vulnerable to off-target interaction by the drug eltrombopag: Insights from molecular modeling.

The loss of proteolytic activity of ADAM17 causes birth defects and embryonic lethality. Conversely, inhibiting ADAM17 activity represents a potential strategy for treating inflammatory and autoimmune diseases. ADAM17 has an active site cleft with a divalent Zn ion and hydrophobic S1'/S3' subsites interconnected to form an L shaped cavity. However, it is currently unknown whether the active site of ADAM17 is susceptible to off-target inhibition by the small molecule drug eltrombopag, which contains metal-binding moieties and is classified as pregnancy category C by the FDA. The in-depth molecu

An Effective Reaction-Based Virtual Screening Method to Discover New CDK8 Ligands.

Cyclin Dependent Kinase 8 (CDK8) is a valuable drug target for cancer suppression. Through an effective reaction-based virtual screening method consisting of pharmacophore modeling, Scifinder database searches, and energy evaluations, a number of new type II CDK8 ligands were discovered with comparable or better binding free energies than the ones reported in literature. In this method, a pharmacophore model, derived from seven crystal structures of CDK8 and type II ligands, was able to catch the key interactions for ligands binding at the ATP binding site of CDK8. This model then was used to
