Gestational Diabetes and Hypothyroidism: Do Other Factors Play a Role? A Retrospective Case-Control Study.

BACKGROUND: During pregnancy, extensive hormonal and metabolic changes can occur. Gestational diabetes can cause maternal and neonatal complications. Follow-ups have shown that women who first develop metabolic diseases during pregnancy have an increased risk of developing other metabolic diseases, including hypothyroidism. There are contradictory results. The aim of this study is to provide an overview of the relationship between gestational diabetes and gestational thyroid dysfunctions. METHODS: This study is a retrospective case-control study conducted in 2025 on 353 pregnant women in Iran

The blue light receptor gene PoWC-1 plays a role in the formation of Pleurotus ostreatus primordia.

Blue light is a critical environmental signal that triggers fruiting body initiation in mushroom-forming fungi. Pleurotus ostreatus is a widely cultivated model mushroom, yet the molecular mechanisms linking light perception to primordia formation remain poorly understood. Here, we functionally characterized the blue light receptor gene PoWC-1, a homolog of Neurospora crassa white collar 1, during early morphogenesis. Antisense-mediated silencing of PoWC-1 caused a marked delay in primordia formation under light/dark cycles and complete arrest at the undifferentiated mycelial stage under conti

Sorting endosomes play key roles in presentation of Mycobacterium tuberculosis-derived ligands to MAIT cells.

The immune system has developed specialized mechanisms to recognize intracellular pathogens such as Mycobacterium tuberculosis (Mtb). Major Histocompatibility Complex Class I-Related protein 1 (MR1) is a conserved nonclassical antigen presenting molecule that presents ligands derived from microbial riboflavin synthesis to Mucosal Associated Invariant T (MAIT) cells. While endosomal trafficking facilitates MR1 antigen presentation during Mtb infection, the exact mechanisms by which MR1 loading of Mtb-derived ligands occurs are not known. We found that trafficking through sorting endosomes media

RING1 and BMI1 catalytic activities play distinct roles in plant PcG-mediated gene regulation.

PcG proteins play a crucial role in the regulation of eukaryotic development. Two major complexes, PRC1/2, are responsible for H2Aub and H3K27me3, respectively. While the core composition of PRC2 is conserved across species and its activity is required for gene repression, PRC1 components are diverse, and the role of H2Aub is not yet fully understood. In Arabidopsis (Arabidopsis thaliana), both B-cell-specific Moloney murine leukemia virus integration site 1 (BMI1A/B/C) and Really Interesting New Gene 1 (RING1A/B) display H2Aub activity, but whether any link or difference exists between RING1

Arylacetamide deacetylase plays a protective role against amodiaquine-induced liver injury by suppressing ferroptosis.

Amodiaquine (AQ), an antimalarial drug, is metabolized by cytochrome P450 3A4 (CYP3A4) to quinoneimine forms, which are proposed to cause liver injury; however, the underlying mechanism remains unclear. Ferroptosis, a type of regulated cell death driven by iron-dependent lipid peroxidation, has been implicated in certain cases of drug-induced liver injury. Arylacetamide deacetylase (AADAC), a drug-metabolizing hydrolase, regulates hepatic iron homeostasis through non-enzymatic mechanisms and may protect against ferroptosis-associated drug-induced liver injury. This study aimed to elucidate the

Zoonotic cutaneous leishmaniasis caused by Leishmania major: Do humans play a role in amplifying transmission?

Zoonotic cutaneous leishmaniasis (ZCL) caused by Leishmania (L.) major is the most widespread form of CL worldwide, with hundreds of thousands of cases annually. It is associated with significant morbidity and economic burden, particularly in North Africa and the Middle East. For over a century, the zoonotic transmission of ZCL, involving wild gerbils (Muridae: Gerbillinae) as the reservoir and Phlebotomus (Ph.) papatasi as the vector, has been considered an established fact, with no contradictory debate. However, the pronounced endophilic and anthropophilic behaviors of Ph. papatasi, the expo

What Roles Does EBV Play in Systemic Lupus Erythematosus? A Synthesis of Its Action Mechanisms.

Systemic lupus erythematosus (SLE) is a heterogeneous chronic autoimmune disease. Exogenous pathogen infections act as key environmental triggers for autoimmune activation and the development of SLE, with Epstein-Barr virus (EBV) being a particularly prominent one. EBV, a gamma herpesvirus, has been associated with the development of various autoimmune diseases, including SLE, rheumatoid arthritis, Sjögren's syndrome, and multiple sclerosis. Current studies provide sufficient evidence supporting an association between EBV and SLE; however, the specific underlying mechanisms remain unclear. In

Do apelin and ghrelin play a role in multiple sclerosis? The analysis of patients treated with immunomodulatory therapies.

INTRODUCTION: Multiple sclerosis (MS) is an autoimmune disorder with a complex and multifactorial etiology. Increasing evidence suggests that peptide hormones may significantly influence the pathogenesis of MS by affecting inflammatory processes and metabolic dysregulation. The aim of the study was to evaluate the levels of apelin and ghrelin, which are two peptide hormones, in patients with relapsing-remitting MS (RRMS) receiving natalizumab and fingolimod compared to healthy controls. METHODS: We enrolled forty-nine patients with RRMS and thirty-eight healthy individuals. The participants w

RAGE Cytosolic Partner Diaph1 Does Not Play an Essential Role in Diabetic Peripheral Neuropathy Progression.

Receptor for advanced glycation end-products (RAGE) activation by hyperglycemia-induced AGE (advanced glycation end-products) accumulation is likely to play a crucial role in the development of complications such as diabetic peripheral neuropathy (DPN). RAGE signaling is mediated via its cytosolic tail. Through its cytosolic tail, RAGE recruits diaphanous-related formin 1 (Diaph1), a protein involved in actin filament organization. Disruption of RAGE-Diaph1 interactions using small molecules alleviates diabetic complications in mice; however, the role of Diaph1 in DPN progression has not been

Overexpression of the signaling coordinator GAB2 can play an important role in acute myeloid leukemia progression.

Mutations that initiate acute myeloid leukemia (AML) can cause clonal expansion without transformation (clonal hematopoiesis). Cooperating mutations, usually in signaling genes, are needed to cause overt disease, but these may require a specific fitness state to be tolerated. Here, we show that nearly all AMLs arising in a mouse model expressing 2 common AML-initiating mutations (Dnmt3aR878H and Npm1cA) acquired a single copy amplification of chromosome 7 (chr7), followed by activating mutations in signaling genes. We show that overexpression of a single gene on chr7 (Gab2, which coordinates s

Intestinal Form of Human Coronavirus 229E Plays No Role in Peritoneal Sclerosis Pathology in Dialysis Patients.

Infectious peritonitis is found to be a leading factor in the development of viral peritonitis (VP) and encapsulating peritoneal sclerosis (EPS) in patients undergoing peritoneal dialysis (PD) treatment. Bacterial and fungal infections are a major cause of peritonitis in PD patients. Viral infections have rarely been reported in association with peritonitis in PD patients; about 20% of cases are fungal and bacterial culture-negative (so-called sterile peritonitis). Several possible viral causes are reported to cause peritonitis, Coronaviruses are an important virus group that has been found to

Estrogen Receptor May Play Important Role in Gender Differences of Early-Onset Colorectal Cancer.

BACKGROUND: The incidence of early-onset colorectal cancer (EOCRC) is increasing year by year. However, it is still unclear and controversial whether there is a significant gender difference. Our aims to investigate the clinicopathological characteristics of EOCRC, especially gender differences, and their correlation with the expression of estrogen receptor (ER), so as to provide a scientific basis for optimizing the early screening and treatment strategy of EOCRC. METHODS: All surgical specimens of CRC were collected from January 2015 to July 2024. Then, the clinicopathological characteristi

Infiltrating macrophages replace Kupffer cells and play diverse roles in severe alcohol-associated hepatitis.

Patients with alcohol-associated cirrhosis (AC) may develop severe alcohol-associated hepatitis (sAH), a disease with high short-term mortality. Our previous studies demonstrated that sAH, but not AC livers, are infiltrated with a high number of self-sustaining IL-8+ neutrophils that likely drive the transition from AC to sAH. Monocyte-derived macrophages (MoMFs) also infiltrate the liver in sAH, but their roles remain largely obscure. In the present study, we characterized liver macrophages in human liver explants from sAH and AC patients. Our data revealed a marked reduction in Kupffer cells

Caffeine plays a prevention role in stress-induced depression by modulating gut-brain axis function.

BACKGROUND: Caffeine intake is inversely associated with depression in epidemiological studies and can impact gut microbiota. Considering the close relationship between depression and gut microbiota, we conducted this study to investigate whether prophylactic caffeine use could influence the development of depression by affecting gut-brain axis. METHODS: Male C57BL/6J mice were randomly divided into three groups: a control group, one group receiving chronic unpredictable stress (CUS) as CUS group, and one group receiving CUS after intraperitoneal injection with caffeine (CAF) as CAF group.
