Three affected siblings with the hermaphrodism are described. The propositi showed the following characteristics: male phenotype and gender role, hypospadias, bilateral scrotal ovotestes with palpable nodules, and absence of müllerian structures. The X chromatin was positive and the Y chromatin was negative in the 3 affected subjects. Their karyotype in peripheral blood lymphocytes and in gonadal fibroblasts was 46,XX and no Y chromosome fluorescence was observed. Plasma FSH was elevated in the 2 older patients and plasma LH was elevated only in the oldest. Plasma testosterone was low and plas…

Cell culture studies were performed on members of a family in which two sisters, ages 9 and 12, have a similar disorder characterized clinically by severe scoliosis, joint laxity and recurrent dislocations, hyperextensible skin, and thin scars. The skin collagen from the sisters was markedly deficient in hydroxylysine, but other amino acids were present in normal amounts. Hydroxylysine in collagen from fascia and bone was reduced to a lesser extent. Since the most likely explanation for the hydroxylysine deficiency was a reduction in enzymatic hydroxylation of lysine residues in protocollagen,

Hypertensive disorders of pregnancy (HDP) are associated with increased maternal cardiovascular disease (CVD) mortality, with risks varying by HDP subtypes and subsequent pregnancy outcomes. The contribution of shared familial factors given this heterogeneity is unclear. We conducted a population-based study using Norwegian registries (1967-2020) including 1 106 658 women with complete pregnancy histories, of whom 628 345 had at least one full sibling. Women with HDP were classified into low-risk (gestational hypertension or term pre-eclampsia followed by no HDPs) and high-risk (all other patt

AIMS/HYPOTHESIS: First-degree relatives of Type II (non-insulin-dependent) diabetic patients in cross-sectional studies have increased insul … gen. Fasting lipid profiles, blood pressure and HOMA insulin sensitivity (%S) were also measured in siblings and in matched subjects without family history of diabetes.
RESULTS: Hyperglycaemic and normoglycaemic siblings (7 female, 6 male) were aged, mean (SD) 56.8 (8.7) and 55.8 (8.4) years. Hyperglycaemic siblings had increased PAI-1 antigen, geometric …

OBJECTIVE: As the prevalence of autism rises, attention to the well-being of siblings of autistic individuals is increasingly important. The present study aimed to identify predictors of clinically significant anxiety in non-autistic siblings of youth with autism.
METHOD: Data were derived from the Simons Simplex Collection. Participants included 53 siblings with clinically significant anxiety (anxious group) and 53 siblings without elevated anxiety (comparison group), who were matched to the anxious group on various demographic characteristics. Logistic regression was used to assess factors …

UNLABELLED: Adult-onset Alexander disease (AOAD) is a genetic neurodegenerative disease due to mutations in GFAP, often causing medullary degeneration.
OBJECTIVES: (i) In a kindred of three siblings with AOAD to compare and correlate clinical progression with MRI quantified medullary atrophic changes. (ii) To demonstrate how AOAD risks under-diagnosis in an older patient population.
METHODS: We retrospectively compared three siblings in their fifth and sixth decade from a family of Irish heritage with AOAD with respect to clinical phenotype, radiological measurements and delay to diagnosis.

OBJECTIVES: To evaluate the extent of clinical concordance and phenotypic variability among pediatric FMF sibling pairs carrying identical biallelic exon 10 MEFV variants.
METHODS: This cross-sectional study included 194 pediatric FMF patients from 97 families, all harboring identical biallelic pathogenic exon 10 variants. After excluding four monozygotic twin patients from two families, 190 non-twin siblings from 95 families were analyzed. Demographic information, clinical manifestations, disease severity scores (Pras, ISSF), colchicine response rates and genetic data were collected retrospe…

Glycogen storage disease 1b (GSD1b) typically presents in early infancy with poor fasting tolerance, hepatomegaly, and neutropenia. We report two siblings who were diagnosed with GSD1b in adulthood. Both had a normal fasting tolerance throughout childhood and, as adults, were able to fast for at least 16 h without developing hypoglycaemia. The older sibling developed nodular cirrhosis during adolescence. The younger sibling exhibited a more pronounced metabolic phenotype, including hyperuricaemia leading to recurrent gout and nephrolithiasis. He experienced occasional episodes of mild neutrope…

Lipoid proteinosis (LP), or Urbach-Wiethe disease, is an ultra-rare autosomal-recessive disorder caused by loss-of-function variants in the ECM1 gene. It is characterized by the progressive deposition of hyaline-like material in the skin, mucosae, and central nervous system. This series describes three siblings (aged 16, 24, and 25) from Gujranwala, Pakistan, born to non-consanguineous parents. All patients presented with infant-onset hoarseness of voice and recurrent vesiculobullous eruptions that evolved into waxy papules and hyperkeratotic plaques. Physical examination revealed the pathogno…

This mini review synthesizes current evidence on the psychosocial adaptation of non-autistic siblings of autistic children, adolescents, and adults. We searched PubMed, PsycINFO, and Web of Science (January 2021-March 2026) and synthesized the retrieved literature narratively along three organizing axes: ecological level, developmental stage, and the risk-resilience continuum. Existing research shows a heterogeneous profile in which vulnerability and resilience coexist: siblings may experience loneliness, anxiety, depressive symptoms, and role strain, but may also report empathy, maturity, and…
