Polycaprolactone/poly L-lactic acid nanofibrous scaffold improves osteogenic differentiation of the amniotic fluid-derived stem cells.

Using stem cells is one of the most important determining factors in repairing lesions using regenerative medicine. Obtaining adult stem cells from patients is a perfect choice, but it is worth noting that their differentiation and proliferation potential decreases as the patient ages. For this reason, the use of amniotic fluid stem cells can be one of the excellent alternatives. This research aimed to investigate the osteogenic differentiation potential of the amniotic fluid stem cells while cultured on the polycaprolactone/poly L-lactic acid nanofibrous scaffold. Scaffolds were qualitatively

Stem cell biology and regenerative medicine for neonatal lung diseases.

Lung diseases remain one of the main causes of morbidity and mortality in neonates. Cell therapy and regenerative medicine have the potential to revolutionize the management of life-threatening and debilitating lung diseases that currently lack effective treatments. Over the past decade, the repair capabilities of stem/progenitor cells have been harnessed to prevent/rescue lung damage in experimental neonatal lung diseases. Mesenchymal stromal cells and amnion epithelial cells exert pleiotropic effects and represent ideal therapeutic cells for bronchopulmonary dysplasia, a multifactorial disea

Mass cytometry-based single-cell analysis of human stem cell reprogramming uncovers differential regulation of specific pluripotency markers.

Human induced pluripotent stem cells (hiPSCs) are reprogrammed from somatic cells and are regarded as promising sources for regenerative medicine and disease research. Recently, techniques for analyses of individual cells, such as single-cell RNA-Seq and mass cytometry, have been used to understand the stem cell reprogramming process in the mouse. However, the reprogramming process in hiPSCs remains poorly understood. Here we used mass cytometry to analyze the expression of pluripotency and cell cycle markers in the reprogramming of human stem cells. We confirmed that, during reprogramming, th

Machine learning-assisted prognosis of multiple myeloma side population cells via SRGs and OCLR stemness index.

BACKGROUND AND OBJECTIVE: Relapse in Multiple Myeloma, driven by therapy-resistant cancer stem cells, necessitates the development of more specific and accurate prognostic models. Existing stemness indices often lack specificity for the unique biology of Multiple Myeloma. This study aimed to develop, validate, and optimize a novel prognostic gene signature derived from Side Population cells, a well-defined cancer stem cell-enriched subpopulation in Multiple Myeloma. METHODS: A core stemness gene module was identified from Side Population cell transcriptomes (GSE109651) using Weighted Gene Co-

[Tolerance in transplantation: potential contribution of haematopoietic transplantation and cell therapy].

The ultimate objective of organ transplantation is to obtain a state of tolerance, i.e. long-term acceptance of the graft without immunosuppressive therapy in order to limit the complications of these treatments (viral infections, tumours, etc.). The various immunological mechanisms allowing a state of tolerance will be described in this review. Among these various experimental strategies, combined bone marrow (or haematopoietic stem cell) transplantation and organ transplantation, made possible by the development of non-myeloablative or less intensive conditioning, appears to be one of the mo

A Rho Kinase (ROCK) Inhibitor, Y-27632, Inhibits the Dissociation-Induced Cell Death of Salivary Gland Stem Cells.

Salivary gland stem cells (SGSCs) are potential cell sources for the treatment of salivary gland diseases. The control of cell survival is an essential factor for applying stem cells to regenerative medicine or stem cell-based research. The purpose of this study was to investigate the effects of the ROCK inhibitor Y-27632 on the survival of SGSCs and its underlying mechanisms. SGSCs were isolated from mouse submandibular glands and cultured in suspension. Treatment with Y-27632 restored the viability of SGSCs that was significantly decreased during isolation and the subsequent culture. Y-27632

Hair Follicle Stem Cells: Emerging Physiology and Therapeutic Applications.

Hair follicle stem cells (HFSCs) have been studied for their role in developmental biology, hair cycling and transplantation. However, recent research reframes the hair follicle as a heterogeneous regenerative ecosystem capable of responding to injury cues and contributing to tissue repair beyond trichology. Despite advances, our understanding of HFSC physiology and therapeutic applications remains fragmented. Many reviews fail to bridge the gap between mechanistic biology and translatable therapeutic modalities, often overlooking critical aspects such as stemness-preserving culture systems, b

A systematic review, umbrella review, and quality assessment on clinical translation of stem cell therapy for knee osteoarthritis: Are we there yet?

BACKGROUND: The success of stem cell therapy for knee osteoarthritis (KOA) in preclinical animal models has accelerated the pace of clinical translation. However, it remains uncertain whether the current scientific evidence supports the clinical application of stem cells in treating KOA. A comprehensive evaluation of the safety and efficacy of stem cell therapies and scientific evidence quality is necessary. METHODS: Using "stem cells" and "knee osteoarthritis" as the search terms, several databases, including PubMed, Web of Science, Cochrane, Embase, and Clinicaltrials.gov, were searched on

[The application of breast cancer organoids in drug screening and gene function research].

Organoids refer to cell clusters with certain spatial structures formed by human adult stem cells or pluripotent stem cells through three-dimensional culture in vitro, which can be stably passaged. Organoids can simulate real organs in both structure and function, and to the greatest extent reproduce the structure and function of living organs or tissues. They have three-dimensional structural features, retain cellular heterogeneity, and can self-renew and self-organize. In the field of breast cancer research, breast cancer organoid models can faithfully reproduce the heterogeneity, tissue str

Pathophysiology of Fibrosis in the Vocal Fold: Current Research, Future Treatment Strategies, and Obstacles to Restoring Vocal Fold Pliability.

Communication by voice depends on symmetrical vibrations within the vocal folds (VFs) and is indispensable for various occupations. VF scarring is one of the main reasons for permanent dysphonia and results from injury to the unique layered structure of the VFs. The increased collagen and decreased hyaluronic acid within VF scars lead to a loss of pliability of the VFs and significantly decreases their capacity to vibrate. As there is currently no definitive treatment for VF scarring, regenerative medicine and tissue engineering have become increasingly important research areas within otolaryn

Functional maturation of immature β cells: A roadblock for stem cell therapy for type 1 diabetes.

Type 1 diabetes mellitus (T1DM) is a chronic autoimmune disease caused by the specific destruction of pancreatic islet β cells and is characterized as the absolute insufficiency of insulin secretion. Current insulin replacement therapy supplies insulin in a non-physiological way and is associated with devastating complications. Experimental islet transplantation therapy has been proven to restore glucose homeostasis in people with severe T1DM. However, it is restricted by many factors such as severe shortage of donor sources, progressive loss of donor cells, high cost, etc. As pluripotent stem

Targeting ferroptosis: opportunities and challenges of mesenchymal stem cell therapy for type 1 diabetes mellitus.

Type 1 diabetes mellitus (T1DM) is characterized by progressive β-cell death, leading to β-cell loss and insufficient insulin secretion. Mesenchymal stem cells (MSCs) transplantation is currently one of the most promising methods for β-cell replacement therapy. However, recent studies have shown that ferroptosis is not only one of the key mechanisms of β-cell death, but also one of the reasons for extensive cell death within a short period of time after MSCs transplantation. Ferroptosis is a new type of regulated cell death (RCD) characterized by iron-dependent accumulation of lipid peroxides.

Emerging therapies for brain recovery after IVH in neonates: Cord blood derived Mesenchymal Stem Cells (MSC) and Unrestricted Somatic Stem Cells (USSC).

In this report, we summarize evidence on mechanisms of injury after intraventricular hemorrhage resulting in post-hemorrhagic white matter injury and hydrocephalus and correlate that with the possibility of cellular therapy. We describe how two stem cell lines (MSC & USSC) acting in a paracrine fashion offer promise for attenuating the magnitude of injury in animal models and for improved functional recovery by: lowering the magnitude of apoptosis and neuronal cell death, reducing inflammation, and thus, mitigating white matter injury that culminates in improved motor and neurocognitive outcom

MiR-378 promoted cell proliferation and inhibited apoptosis by enhanced stem cell properties in chronic myeloid leukemia K562 cells.

Dysregulation of miR-378 has been found in diverse types of tumors as well as in leukemia. The role of miR-378 in chronic myeloid leukemia (CML) remains unclear. The aim of the study was to reveal the potential effects of miR-378 in the pathological process and progress in CML. Our results showed general level of miR-378 was significant higher in CML patients compared to controls. Overexpression of miR-378 dramatically promoted cell proliferation and drug-resistance. Additionally, apoptosis was inhibited in cells transfected with miR-378. More and bigger stem cell sphere formation was observed

Harnessing mechanobiology of human pluripotent stem cells for regenerative medicine.

Recent advances in human pluripotent stem cells (hPSCs) open new doors for therapeutics of motor neuron (MN)-associated neurodegenerative diseases. However, the MN differentiation process is not yet completely understood. In this Viewpoint, we stress the concept of designing synthetic cell culture surfaces with precisely controlled mechanical properties (such as rigidity) to improve the efficiency of MN differentiation from hPSCs. Emerging evidence strongly supports the potent role of mechanobiology in controlling stem cell fate. Leveraging the intrinsic mechanosensitive properties of hPSCs in

The role of quantum dots in enhancing the therapeutic targeting of cancer stem cells.

In recent years, cancer stem cells have emerged as an interesting field in oncology due to their metastatic and resistance potential to chemotherapy and radiation therapy, thus resulting in the resurfacing of cancer even after multiple treatment attempts. The interest in these cells aims to address the key challenges associated with cancer treatments and to offer insights that may aid in better understanding the biology of cancer, with the possibility of introducing advanced or novel treatment methods. Conventional treatments often fail to eradicate the cancer stem cells, which then results in
