Cauda equina-derived extracellular matrix for fabrication of nanostructured hybrid scaffolds applied to neural tissue engineering.

Extracellular matrix (ECM) components have become important candidate materials for use as neural scaffolds for neural tissue engineering. In the current study, we prepared cauda equina-derived ECM materials for the production of scaffolds. Natural porcine cauda equina was decellularized using Triton X-100 and sodium deoxycholate, shattered physically, and made into a suspension by differential centrifugation. The decellularization procedure resulted in the removal of >94% of the nuclear material and preserved the extracellular collagen and sulfated glycosaminoglycan. Immunofluorescent stainin…

Cryo-chemical decellularization of the whole liver for mesenchymal stem cells-based functional hepatic tissue engineering.

Liver transplantation is the ultimate treatment for severe hepatic failure to date. However, the limited supply of donor organs has severely hampered this treatment. So far, great potentials of using mesenchymal stem cells (MSCs) to replenish the hepatic cell population have been shown; nevertheless, there still is a lack of an optimal three-dimensional scaffold for generation of well-transplantable hepatic tissues. In this study, we utilized a cryo-chemical decellularization method which combines physical and chemical approach to generate acellular liver scaffolds (ALS) from the whole liver.

Phenotypic changes in cultured smooth muscle cells: limitation or opportunity for tissue engineering of hollow organs?

Smooth muscle cells (SMCs) are typically used as a cell source for the reconstruction of hollow organs by conventional tissue engineering techniques. However, the necessity for and advantage of the use of tissue-specific SMCs are unknown. The present study investigated the phenotypic changes that occur following isolation and in vitro expansion of rat SMC populations isolated from three different tissues: the aorta, oesophagus and urinary bladder. rSMCs were isolated by enzymatic dispersion and expanded by conventional cell culture techniques, yielding microscopically homogeneous populations. …

Tissue engineering of a small hand phalanx with a porously casted polylactic acid-polyglycolic acid copolymer.

In this study, we evaluate a porous polylactic acid-polyglycolic acid (PLGA) polymer in tissue engineering a small hand phalanx. The PLGA polymer was processed with a unique solvent cast/salt-leaching method in the shape of a little finger distal phalanx. An 85% polylactic acid (PLA)/15% polyglycolic acid (PGA) copolymer was determined to be the most favorable mixture for creating a small digital phalanx in precise shape and size. The pore sizes averaged 250-425 microm. Bovine periosteum was wrapped around each phalangeal polymer. Chondrocytes were injected into the articular end. Six construc…

Polylactic acid/hydroxyapatite composite filaments: Balancing printability, mechanical integrity, and osteogenic performance for bone tissue engineering.

Biomedical devices for bone tissue repair require materials with a balanced combination of printability, structural integrity, and biological performance. Their fabrication is increasingly shifting toward additive manufacturing, in which thermoplastic biodegradable polymers, particularly poly(lactic acid) (PLA), serve as matrices. In this study, PLA composite filaments with varying hydroxyapatite (HAp) contents (0, 5, 10, 20, and 30 wt%) were fabricated via a solvent-assisted non-solvent-induced precipitation method and evaluated for extrusion-based bone tissue engineering applications. HAp co…
