Breaking the Mold: Partnering with the National Institutes of Health Intramural Research Program to Accelerate PhD Training.

Immunology is an increasingly interdisciplinary field. Here we describe a new model for interinstitutional graduate training as partnerships between complementary laboratories. This collaborative model reduces time to graduation without compromising productivity or alumni outcomes. We offer our experience with one such program and thoughts on the ingredients for their success. Despite tremendous recent advances in technology, communications, and the translation of basic scientific discoveries into new diagnostics and therapies for human diseases, graduate training in immunology and other areas

When the science alone is not enough: embracing our responsibility as science communicators.

The US biomedical research enterprise has led the world in driving medicine forward but now faces unprecedented threats. Systematic efforts to defund and dismantle the academic research infrastructure, compounded by misinformation and questionable public support for science, have already impeded innovation and discouraged students from pursuing careers in research and healthcare, which ultimately will harm patients. This moment calls on us, as scientists and physicians, to effectively communicate the relevance of our research to the public and elected officials and ensure that the value of bas

Liquid Metal Molecular Medicine.

Biomolecules are central to disease onset, diagnosis, and treatment, yet materials capable of actively interacting with biological targets remain limited. Gallium-based liquid metals and their derivative composites possess a metallic liquid core, a dynamic oxide interface, and releasable metal species, which support diverse molecular interactions and regulatory processes. However, current advances remain scattered across different biomedical applications and are often discussed mainly from a materials perspective. Here, we propose liquid metal molecular medicine (LMMM) as an interdisciplinary

Summit proceedings: Biomedical countermeasure development for emerging vector-borne viral diseases.

Emerging and re-emerging infectious diseases are an expanding global threat to public health, security, and economies. Increasing populations, urbanization, deforestation, climate change, anti-vaccination movements, war, and international travel are some of the contributing factors to this trend. The recent Ebola, MERS-CoV, and Zika outbreaks demonstrated we are insufficiently prepared to respond with proven safe and effective countermeasures (i.e., vaccines and therapeutics). The State University of New York Upstate Medical University and the Trudeau Institute convened a summit of key opinion

Supermole-rat to the rescue: Does the naked mole-rat offer a panacea for all that ails us?

Over the previous several decades, many non-traditional research models have offered new avenues of exploration for biomedical research. The promise of these animals is primarily derived from adaptations to unique or challenging environments that share key factors with a disease or pathology of interest (e.g., hypoxemia or hypercarbia are clinically relevant and are also in vivo consequences of environmental hypoxia and hypercapnia, respectively). Animals adapted to such environments allow us to ask the question: how has nature solved a particular problem and what can we learn to inform novel

Biomarkers of acute lung injury: worth their salt?

The validation of biomarkers has become a key goal of translational biomedical research. The purpose of this article is to discuss the role of biomarkers in the management of acute lung injury (ALI) and related research. Biomarkers should be sensitive and specific indicators of clinically important processes and should change in a relevant timeframe to affect recruitment to trials or clinical management. We do not believe that they necessarily need to reflect pathogenic processes. We critically examined current strategies used to identify biomarkers and which, owing to expedience, have been do

Emerging Technologies in Cardiovascular Disease Modeling: Bridging Basic Science and Clinical Translation.

Cardiovascular diseases (CVDs) remain the leading cause of death worldwide, yet therapeutic development is limited by experimental models that incompletely recapitulate human physiology, disease heterogeneity, and treatment responses. Emerging human-relevant models, including patient-derived induced pluripotent stem cells, cardiac organoids, engineered tissues, organ-on-chip systems, genome editing, multi-omics, and computational approaches, are improving the ability to investigate cardiovascular disease mechanisms and therapeutic responses. In this review, we critically compare these technolo

Fibroblast Growth Factors and Their Receptors in Major Depressive Disorder: A Scoping Review of Translational Research.

Major depressive disorder (MDD) is a complex psychiatric condition whose pathophysiology extends beyond the classic monoamine hypothesis, involving impaired neuroplasticity, neuroinflammation, HPA axis dysregulation, and altered neurotrophic signaling. The fibroblast growth factor (FGF) system, comprising ligands and their receptors (FGFRs), has emerged as a key candidate regulator of these processes. This scoping review qualitatively synthesizes current clinical, postmortem, and preclinical evidence on the role of the FGF system in MDD and antidepressant response. A structured literature sear

Targeting the Tumor Microenvironment Network to Overcome Immunotherapy Resistance in Non-Small Cell Lung Cancer: Mechanisms to Clinical Translation.

BACKGROUND: Immunotherapy has transformed the treatment landscape for advanced non-small cell lung cancer (NSCLC), yet resistance mediated by the tumor microenvironment (TME) limits its efficacy in most patients. This review aims to synthesize the current understanding of the TME as an integrated network driving immunotherapy resistance and to evaluate emerging therapeutic strategies aimed at overcoming it. METHODS: We conducted a comprehensive narrative review of key literature in the field, sourced from PubMed and major conference proceedings. The focus was on mechanistic studies elucidatin

Cancer-associated fibroblasts in therapeutic resistance of hepatocellular carcinoma: evidence, mechanisms, and translational implications.

Hepatocellular carcinoma (HCC) remains difficult to treat because responses to tyrosine kinase inhibitors (TKIs) and immune checkpoint inhibitors (ICIs) are limited and heterogeneous. Cancer-associated fibroblasts (CAFs) can influence therapeutic responses through paracrine signaling, immune regulation, and extracellular matrix remodeling, but CAF-driven tumor progression should not be equated with therapeutic resistance. This review critically evaluates HCC-specific evidence according to defined drug exposure, treatment-related outcomes, and functional rescue. The most mechanistically resolve

Induced pluripotent stem cell-derived macrophages (iMacs) in translational medicine: Disease models, drug testing, and therapeutic applications.

Macrophages play key roles in innate immunity, inflammation, and homeostasis. However, conventional macrophage models, particularly primary macrophages derived from donor monocytes (MDMs), are limited by donor-to-donor variability, limited proliferative capacity and non-renewable nature, low scalability, and poor genetic tractability. Induced pluripotent stem cell-derived macrophages (iMacs) have emerged as an alternative that offers a renewable source of engineered human macrophages. This review summarizes the approaches used to generate and characterize iMacs, highlighting their functional p

Clarity and inclusivity as precursors to translating yoga health evidence: a diverse cross-sectional study on barriers, facilitators, and associations of yoga.

BACKGROUND: Translating evidence-based interventions (EBIs) to advance health behaviors and well-being takes 15-17 years. This translational gap is lengthened for specific EBI and populations, such as yoga for people of color, with higher weight, or who live in rural areas. Yoga principles have positively impacted myriad populations and outcomes and align with scientific rigor, but misinformation persists. Applying a translational lens to input from under-represented yoga end-users may speed the gap. PURPOSE: To examine how underrepresented yoga end-users perceive yoga with consideration to y

Metabolic crosstalk in breast cancer in patients with Type 2 diabetes mellitus: pathophysiology and nanotherapeutic opportunities for clinical translation.

The intersection of Type 2 Diabetes Mellitus (T2DM) and breast cancer defines a complex onco-metabolic axis where chronic hyperglycaemia, hyperinsulinemia, and oxidative stress synergistically drive malignant transformation, metabolic reprogramming, and chemoresistance. Despite epidemiological evidence linking these conditions, clinical management typically addresses each disease individually according to established protocols; however, the presence of T2DM significantly alters tumour biology, therapeutic response, and outcomes, necessitating greater consideration of metabolic context in breas

Superoxide Dismutase-Centered Modulation by Curcumin in Cardiovascular Diseases: Mechanistic Insights and Translational Implications.

Cardiovascular diseases (CVD) remain the leading global cause of morbidity and mortality, driven in part by dysregulated redox homeostasis and chronic inflammation. Superoxide dismutase (SOD), a key enzymatic defence against reactive oxygen species (ROS), plays a central role in maintaining cardiovascular integrity through regulation of oxidative stress across cytosolic (SOD1), mitochondrial (SOD2) and extracellular (SOD3) compartments. Impairment of SOD function contributes directly to endothelial dysfunction, myocardial injury and vascular remodelling. Curcumin (Cur), a pleiotropic polypheno

From FLOW to translational positioning in chronic kidney disease: mechanistic complementarity of SGLT2 inhibitors and GLP-1 receptor agonists.

Chronic kidney disease is a central node of the cardio-kidney-metabolic continuum and carries substantial residual cardiovascular and kidney risk. Sodium-glucose cotransporter 2 inhibitors (SGLT2i) have reproducibly reduced kidney disease progression and heart failure risk in CKD outcomes trials, whereas GLP-1RAs have established cardiovascular benefit across cardiometabolic populations. The FLOW trial (Evaluate Renal Function With Semaglutide Once Weekly) established hard kidney-outcome evidence for semaglutide in type 2 diabetes with CKD, although whether this renal benefit represents a broa

From molecular networks to translational intervention: current progress in the mechanisms of gemcitabine resistance in bladder cancer.

Bladder cancer remains one of the most common malignancies of the urinary tract. Although the treatment landscape has expanded rapidly in recent years, gemcitabine still occupies a central position in intravesical treatment for non-muscle-invasive bladder cancer, in perioperative systemic therapy for muscle-invasive disease, and in platinum-based first-line regimens for advanced urothelial carcinoma. Yet the long-term benefit of gemcitabine is frequently curtailed by primary non-response or acquired resistance. In practice, this problem is often recognized only after radiographic progression o

Mesenchymal stem cell-derived secretome in traumatic brain injury: Stage-specific paracrine mechanisms and translational challenges.

Traumatic brain injury (TBI) is a stage-dependent disorder that evolves from acute neuroinflammation, blood-brain barrier (BBB) disruption, oxidative stress, excitotoxicity, and apoptosis to subacute neurovascular remodeling and chronic impairment of neuroregeneration and circuit repair. Current clinical management improves survival by stabilizing physiological status and limiting secondary injury, but it rarely rebuilds damaged neural networks or restores long-term neurological function. Mesenchymal stem cells (MSCs) have therefore attracted attention as regenerative candidates, with increasi

Growth factor applications and clinical translation: advances and challenges.

BACKGROUND: Growth factors (GFs), as key molecules regulating cellular processes, demonstrate therapeutic potential in fields such as regenerative medicine, oncology, and metabolic regulation. However, inherent pharmacokinetic limitations, off-target effects, and safety concerns significantly hinder clinical translation. Although in-depth studies on mechanisms of action, smart delivery systems, and regulatory networks have improved GFs' stability, targeting, and safety - leading to clinical approvals of some related drugs - high costs, complex large-scale production processes, and the intricac

Single-cell and spatial transcriptomics analysis of osteoarthritis: pathway regulation, cell interaction networks, and therapeutic translation.

BACKGROUND: Osteoarthritis (OA) transcends the outdated paradigm of mere "wear and tear" cartilage loss. It is now recognized as a complex, whole-organ disease driven by multifaceted interactions among diverse cell populations across synovium, cartilage, subchondral bone, and infrapatellar fat pad. The profound heterogeneity within these tissues and the spatiotemporal dynamics of pathological processes have, until recently, remained a "black box," limiting our understanding of disease initiation and progression. The convergence of single-cell RNA sequencing (scRNA-seq) and spatial transcriptom

Overcoming Immunological Barriers in MSC-Derived Insulin-Producing Cells through CRISPR-Based Hypoimmunogenic Engineering and Translational Perspectives for Type 1 Diabetes.

Mesenchymal stromal cell (MSC)-derived insulin-producing cells (IPCs) represent an emerging strategy for β-cell replacement in type 1 diabetes mellitus (T1DM) owing to their differentiation potential, intrinsic immunomodulatory properties, and lower tumorigenic risk compared with pluripotent stem cell-derived platforms. However, accumulating evidence indicates that differentiation-associated immunogenicity, context-dependent immune recognition, and recurrent autoimmune responses may substantially limit long-term graft survival and therapeutic durability following transplantation.
