Injectable polysaccharide-based hydrogel delivering Poria cocos extracellular vesicles promotes hair regeneration in androgenetic alopecia via Wnt/β-catenin signaling.

Androgenetic alopecia (AGA) is characterized by progressive hair follicle miniaturization driven by premature follicle regression. While extracellular vesicles (EVs) are potent bioactive therapeutics, their clinical translation is hindered by the low yields and poor stability of mammalian-derived sources. Herein, we developed an injectable biomacromolecular hydrogel composed of sodium carboxymethylcellulose (CMCNa), sodium alginate (SA), and hyaluronic acid (HA) to deliver plant-derived Poria cocos extracellular vesicles (PCEVs) for the topical treatment of AGA. This polysaccharide matrix was

Topical Application of Manuka Honey for the Treatment of Non-Healing Venous Leg Ulcers.

Background/Objectives: Issues related to the chronic venous leg ulcer (VLU) treatment and prevention of recurrences remain the subject of research, but so too do common clinical problems in daily medical practice. Due to its medicinal properties, Manuka honey is increasingly used in the treatment of wounds of various origins. The aim of the study was to investigate the effectiveness of Manuka honey for the topical treatment of non-healing, chronic, venous leg ulcers. Methods: Eighty patients with chronic VLU participated in the study and were randomized into two equinumerous groups. In group 1

The efficacy of topical cetirizine using microneedling in androgenetic alopecia male patients.

BACKGROUND: Androgenetic alopecia (AGA) affects both sexes equally and lasts long. AGA is the most common kind of hair loss in men. Gradual, pattern-based hair thinning on the scalp is a hallmark of the disease. The front, back, center, and sides of a man's head are the most common areas where hair thinning and loss occur. AIM: To evaluate the efficacy of microneedling-administered topical cetirizine in treating androgenetic alopecia in men. METHODS: A total of 80 AGA male patients were randomly assigned to this single-blind, parallel placebo-controlled study conducted at Beni-Suef Universit

In Vitro Proof-of-Concept Study: Lidocaine and Epinephrine Co-Loaded in a Mucoadhesive Liquid Crystal Precursor System for Topical Oral Anesthesia.

Background: Local anesthesia is essential for most dental procedures, but its parenteral administration is often painful. Topical anesthetics are commonly used to minimize local anesthesia pain; however, commercial formulations fail to fully prevent the discomfort of local anesthetic injection. Methods: We developed and characterized a novel lidocaine and epinephrine co-loaded liquid crystalline precursor system (LCPS) for topical anesthesia. The formulation was structurally characterized using polarized light microscopy (PLM) and small-angle X-ray scattering (SAXS). Rheological behavior was a

Minocycline and Diacetyl Minocycline Eye Drops Reduce Ocular Neovascularization in Mice.

PURPOSE: To evaluate the efficacy of minocycline and a novel, modified minocycline analogue that lacks antimicrobial action, diacetyl minocycline (DAM), on choroidal neovascularization (CNV) in mice of both sexes. METHODS: CNV was induced via laser injury in female and male C57BL/6J mice. Minocycline, DAM, or saline was administered via topical eye drops twice a day for 2 weeks starting the day after laser injury. CNV volume was measured using immunohistochemistry labeling and confocal microscopy. RESULTS: Minocycline reduced lesion volume by 79% (P ≤ 0.0004) in female and male mice. DAM red

Fluoxetine Protects Retinal Ischemic Damage in Mice.

BACKGROUND: To evaluate the neuroprotective effect of the topical ocular administration of fluoxetine (FLX) in a mouse model of acute retinal damage. METHODS: Ocular ischemia/reperfusion (I/R) injury in C57BL/6J mice was used to elicit retinal damage. Mice were divided into three groups: control group, I/R group, and I/R group treated with topical FLX. A pattern electroretinogram (PERG) was used as a sensitive measure of retinal ganglion cell (RGC) function. Finally, we analyzed the retinal mRNA expression of inflammatory markers (IL-6, TNF-α, Iba-1, IL-1β, and S100β) through Digital Droplet

The effect of rosmarinic acid on the prevention of myringosclerosis.

OBJECTIVES: Myringosclerosis commonly occurs as a long-term complication of ventilation during the treatment of otitis media. We aimed to determine the effects of rosmarinic acid as an antioxidant on experimentally induced myringosclerosis. METHODS: Twenty-four male Sprague-Dawley rats, weighing 250-300 g, were unilaterally myringotomized and randomly separated into three groups. Group 1 received no treatment (control group) (n = 8); Group 2 received topical rosmarinic acid (n = 8); Group 3 received oral rosmarinic acid (n = 8). On the twenty-first day, the right ears were examined by otomicr

Topical treatment with nanoliposomal Amphotericin B reduces early lesion growth but fails to induce cure in an experimental model of cutaneous leishmaniasis caused by Leishmania mexicana.

Leishmania mexicana infection causes localized skin lesions that can lead to tissue damage and permanent disfigurement if not resolved. Currently, recommended treatments include intravenous administration of Amphotericin B, which is undesirable due to the associated cost and patient burden related to receiving regular injections. In this study, we evaluated the effect of topical treatment with a nanoliposomal formulation of Amphotericin B that is penetrable to the skin (SinaAmphoLeish 0.4%) in mice infected with L. mexicana by using ulcerated (BALB/c) and non-ulcerated (129SVE) models. First-line treatment of pemphigus vulgaris with a combination of rituximab and high-potency topical corticosteroids.

IMPORTANCE: The main component of the first-line treatment of pemphigus vulgaris is high doses of systemic corticosteroids, but adverse effects of these drugs are frequent and sometimes severe. Rituximab has shown effectiveness as a corticosteroid-sparing agent or in case of relapse. To our knowledge, the effectiveness of rituximab as a first-line treatment without systemic corticosteroids has not been evaluated. OBSERVATIONS: Five women in their 50s, 60s, or 70s with pemphigus vulgaris (Pemphigus Disease Area Index score, 15-84 at diagnosis) and contraindications to systemic corticosteroid t

Nicotine facilitates reinnervation of phenol-injured perivascular adrenergic nerves in the rat mesenteric resistance artery.

Nicotine has been shown to have neuroprotective and neurotrophic actions in the central nervous system. To elucidate the peripheral neurotrophic effects of nicotine, we determined whether nicotine affected the reinnervation of mesenteric perivascular nerves following a topical phenol treatment. A topical phenol treatment was applied to the superior mesenteric artery proximal to the abdominal aorta in Wistar rats. We examined the immunohistochemistry of the distal small arteries 7 days after the treatment. The topical phenol treatment markedly reduced the density of tyrosine hydroxylase (TH)-LI

Nanoparticles as multifunctional devices for the topical treatment of cutaneous leishmaniasis.

INTRODUCTION: Cutaneous and mucocutaneous leishmaniasis are major tropical skin diseases. Topical treatment is currently limited to the least severe forms of cutaneous leishmaniasis (CL) without risk of dissemination. It is also recommended in combination with systemic therapy for more severe forms. Progresses in this modality of treatment are hindered by the heterogeneity of the disease and shortcomings in the clinical trials. AREAS COVERED: This review overlooks three major modalities of topical therapies in use or under investigation against CL: chemotherapy, photodynamic therapy and immun

Force requirements in topical medicine use--the squeezability factor.

INTRODUCTION: Patient compliance is essential to ensure efficacious treatment. The ease of topical drop delivery is of paramount importance. At least 50% of patients report difficulty in self-administration of topical ocular medicine. The two most frequently reported causes of difficulty include aiming the bottle and squeezing the bottle. PURPOSE: The aims of this study were to identify (I) the force required to deliver a single drop from a bottle (the squeezability factor), (II) are some bottle designs easier to use than others? (III) Do compliance aids reduce the finger strength required to

Survey on the use of ototopical medications by New Zealand otolaryngologist/head and neck surgeons.

AIM: There is increasing awareness of the potential for ototoxicity with the administration of aminoglycoside eardrops in the presence of a non-intact tympanic membrane, whether that is due to a perforation or due to the presence of a patent ventilating tube. Expert panels in the USA, Canada, UK, and Australia have all advocated the preferential use of non-ototoxic topical antibiotics for the discharging middle ear and/or mastoid cavity. The advent of non-ototoxic fluoroquinolone eardrops provides the first real opportunity to prescribe topical therapy in accordance with such guidelines. METH

Effects of topical anandamide-transport inhibitors, AM404 and olvanil, on intraocular pressure in normotensive rabbits.

PURPOSE: To evaluate the effects of topically applied anandamide transport inhibitors, AM404 and olvanil, on the intraocular pressure (IOP) of normotensive rabbits. To determine if the ocular hypotension induced by topical anandamide (AEA) can be potentiated by co-administered AM404. METHODS: Test compounds, in either hydroxypropyl-beta-cyclodextrin (HP-beta-CD) or propylene glycol, were administered unilaterally onto rabbit eyes. To determine if AM404 affects the IOP-profile of AEA, AM404 was administered ocularly 15 minutes before topical AEA. Phenylmethylsulfonyl fluoride (PMSF) (24 mg/kg,

Injuries to the iris and corneal endothelium in intraocular surgeries.

The changes in the corneal endothelium at the centre of the cornea were followed after intracapsular cataract extraction and penetrating keratoplasty, by means of specular microscopy and morphometry using a computerized image analyzer. The corneal endothelium undergoes progressive transformation in its size over a period of more than 1 year. In penetrating keratoplasty, progressive increase in the cell size appears to occur over a longer postoperative period. The injury to the iris leaves permanent tissue defects and this is due to insufficient fibroblastic activity of the iris. Iris injury pr

Ionic liquid-mediated delivery of a BCL-2 inhibitor for topical treatment of skin melanoma.

Skin melanoma is one of the most common cancer types in the United States and worldwide, and its incidence continues to grow. Primary skin melanoma can be removed surgically when feasible and if detected at an early stage. Anti-cancer drugs can be applied topically to treat skin cancer lesions and used as an adjunct to surgery to prevent the recurrence of tumor growth. We developed a topical formulation composed of Navitoclax (NAVI), a BCL-2 inhibitor that results in apoptosis, and an ionic liquid of choline octanoate (COA) to treat early-stage melanoma. NAVI is a small hydrophobic molecule th
