Programmable engineered bacteria manipulate metabolism and remodel the TME in situ for enhancing adoptive cell therapy.

In solid tumors, the efficacy of adoptive T cell therapy (ACT) is limited by a metabolically constrained tumor environment that undermines T cell persistence, fitness, and infiltration. Here, we engineered a hypoxia-activated bacterial hybrid system (MM@TMV) to address these barriers. This hybrid system integrates metabolically engineered bacteria and tumor membrane vesicles (TMVs) to achieve tumor-restricted metabolic reprogramming and immune reinforcement within hypoxic tumors. Within hypoxic tumors, D-mannose is produced in situ to support stem-like phenotypes and limit exhaustion, while TM

Full-length SARS-CoV-2 envelope protein adopts an interfacial topology in lipid bilayers.

The envelope protein is essential for the replication of coronaviruses. Several structures of the envelope protein of the SARS-CoV-2 and the closely related SARS viruses are available. However, an experimental structure of the entire full-length protein in a native lipid environment has not been determined. In addition, significant structural differences exist between published models, and the conformation adopted by the C-terminal domain is only partially understood. CW-EPR power saturation and EPR lineshape analysis of 15 different labeled sites were carried out on the full-length envelope p

CERTOMICS: trusted single-cell multiomics pipeline for high-resolution profiling of adoptive cellular immunotherapies.

SUMMARY: Adoptive cellular immunontherapies, such as chimeric antigen receptor (CAR) T cell therapy, have transformed cancer treatment, yet challenges such as resistance, relapse, and high costs limit their efficacy and accessibility. A comprehensive understanding of cellular heterogeneity and molecular profiles is essential to improve these therapies. Advanced single-cell multiomics technologies have the power to analyze the complex interactions between CAR-engineered cells, immune cells, and tumor cells. However, standardized single-cell multiomics computational pipelines specifically tailor

Nanomedicine Meets Immunotherapy: Advancing Adoptive Cell Therapy with Nanoparticles in the Treatment of Cancer with Sustainability Perspectives.

Immunotherapy has achieved remarkable clinical success in certain cancers, particularly through adoptive cell therapy (ACT), where T cell engineering with chimeric antigen receptor (CAR) has driven major clinical breakthroughs in the treatment of hematologic malignancies. However, efficacy against solid tumors remains limited due to multiple barriers, including the scarcity of tumor-specific antigens, antigen heterogeneity, immunosuppressive tumor microenvironment, and physical obstruction of T cell infiltration by dense extracellular matrix. Nanoparticle (NP)-based approaches can overcome the

Adoptive MAIT cell therapy for liver cancer enhanced by multimodal modulation of immune receptors and checkpoints.

BACKGROUND: Mucosal-associated invariant T (MAIT) cells are innate-like T lymphocytes that recognize riboflavin-derived metabolites presented by the monomorphic major histocompatibility complex (MHC)-class I-related molecule MR1. MAIT cells are highly enriched in the human liver and are increasingly implicated in liver cancer immunity, yet their antitumor potential and therapeutic applicability remain incompletely defined. METHODS: Here, we profiled MAIT cells derived from peripheral blood of healthy donors and patients with hepatocellular carcinoma, revealing disease-associated phenotypic al

Thymus engraftment and developmental progression of adoptively transferred progenitor T cells are affected by host conditioning.

Hematopoietic stem cell transplantation as treatment for patients with leukemia relies on chemotherapy and/or irradiation-based conditioning regimens to prevent graft rejection and cancer relapse. Despite advances in radiotherapy, total body irradiation for myeloablative conditioning is associated with dose and temporal heterogeneity, and how these variables impact thymus reconstitution and production of new T cells has not been fully elucidated. Here, we used in vitro-generated mouse progenitor T (proT) cells to investigate the dynamics of donor-derived cells in host thymuses under different

In situ macrophage reprogramming via micropatch engineering for safe and effective antitumor adoptive cell therapy.

Adoptive cell therapy with macrophages holds great promise for treating solid tumors, but its clinical impact has been limited. The problem is twofold: the immunosuppressive tumor microenvironment quickly silences the anti-tumor activity of transferred cells, and systemic activation strategies often provoke off-target toxicity. Here, we describe a simple yet effective approach to overcome these barriers. We engineered primary macrophages with anti-CD11b-functionalized zinc-aluminum layered double hydroxide micropatches (LDHMP)-a surface-anchoring strategy that leaves the cells otherwise unalte

The implications of adopting the race-neutral spirometry equations in the detection of airway obstruction in Chilean children with asthma.

OBJECTIVES: This study compared the performance of the GLI-2012 and GLI-2022 equations for detecting airway obstruction in Chilean asthmatic children. METHODS: This cross-sectional analytical study compared the predicted values and z-scores for forced expiratory volume in 1 s (FEV1), forced vital capacity (FVC), and the FEV1/FVC ratio, calculated using the GLI-2012 Caucasian and race-neutral GLI-2022 equations, in 885 Chilean children with asthma aged 6-16 years. Airway obstruction was defined as an FEV1/FVC z-score of less than -1.64. Multivariable generalised estimating equations (GEE) logi

Stunting, underweight and thinness in internationally adopted children: prevalence and associated factors in a large cohort study.

Internationally adopted children (IAC) are at high risk of growth impairment due to early-life deprivation, institutional care, and prenatal exposures. Data on determinants of stunting and other forms of malnutrition in this population remain limited. To evaluate the prevalence of stunting, underweight, and thinness, and to identify factors associated with these conditions in a large cohort of internationally adopted children (IAC), we conducted a large cohort study including 1,955 IAC referred to a tertiary pediatric center in Italy between 2015 and 2025. The cohort mainly included children a

Clinical determinants of survival outcomes and acute toxicity after adoptive cellular therapy for solid tumors.

BACKGROUND: The efficacy of cellular therapies has been demonstrated in hematologic malignancies, and their use is expanding to solid tumors. Predictors of outcomes and toxicities remain limited in patients undergoing cellular therapies for solid tumors. METHODS: Data on patients with solid tumors who received cellular therapies between January 2016 and July 2025 in the Department of Investigational Cancer Therapeutics (Phase I Clinical Trials Program) at The University of Texas MD Anderson Cancer Center were reviewed retrospectively using the institutional CHIMERA database platform. Modeling Adoptive Cell Therapy in Bladder Cancer from Sparse Biological Data using PINNs.

Physics-informed neural networks (PINNs) are neural networks that embed the laws of dynamical systems modeled by differential equations into their loss function as constraints. In this work, we present a PINN framework applied to oncology. Here, we seek to learn time-varying interactions due to a combination therapy in a tumor microenvironment. In oncology, experimental data are often sparse and composed of a few time points of tumor volume. By embedding inductive biases derived from prior information about a dynamical system, we extend the physics-informed neural networks (PINN) and incorpora

Enforced E-selectin ligand installation enhances homing and efficacy of adoptively transferred T cells.

Adoptive T-cell transfer has revolutionized the treatment of hematological malignancies. However, this approach has had very limited success in treating solid tumors, largely due to inadequate infiltration of vascularly administered T cells at tumor sites. The shear-resistant interaction between endothelial E-selectin and its cognate ligand expressed on leukocytes, sialyl Lewis X (sLeX), is an essential prerequisite for extravasation of circulating leukocytes. Here, we report that enforced E-selectin ligand expression (enforced sLeX display) on antigen-specific T cells can be achieved by fucos

EZH1/EZH2 inhibition enhances adoptive T cell immunotherapy against multiple cancer models.

Tumor resistance to chimeric antigen receptor T cell (CAR-T) and, in general, to adoptive cell immunotherapies (ACTs) is a major challenge in the clinic. We hypothesized that inhibiting the tumor drivers' methyltransferases EZH2 and EZH1 could enhance ACT by rewiring cancer cells to a more immunogenic state. In human B cell lymphoma, EZH2 inhibition (tazemetostat) improved the efficacy of anti-CD19 CAR-T by enhancing activation, expansion, and tumor infiltration. Mechanistically, tazemetostat-treated tumors showed upregulation of genes related to adhesion, B cell activation, and inflammatory r

The role of cultural values and norms in the adoption and implementation of foreign innovations in health service delivery in China.

BACKGROUND: In pursuit of the ambitious and large scale 2009 national health reform objectives, China has turned to the introduction and implementation of foreign best practices in health service delivery. While it is well known that cultural differences may significantly impact the adoption and implementation of innovations, there is little specific knowledge on how these impact foreign health service innovations adopted in China. Our aim is therefore to identify which cultural norms and values affect the adoption and implementation of foreign innovations in health service delivery in the Chi…

Recent advances in feeder-free NK cell expansion as a future direction of adoptive cell therapy.

Adoptive cell therapy has progressed rapidly following the success of CAR-T therapy, with NK cells emerging as strong candidates for next-generation treatments. NK cells naturally recognize abnormal cells without genetic modification and do not cause graft-versus-host disease, making them suitable for mass production from donor blood. Traditional NK cell expansion often relies on cancer-derived feeder cells, posing safety and quality concerns. This review examines both feeder cell-based and feeder-free NK cell culture methods. We analyze the limitations of feeder-based approaches and categoriz

Adoptive Transfer of Lepr+ Bone Marrow Cells Attenuates the Osteopetrotic Phenotype of db/db Mice.

Leptin-deficient (ob/ob) and leptin receptor (Lepr)-deficient db/db mice develop a mild form of osteoclast-rich osteopetrosis, most evident in long bone epiphyses, implying leptin is important for normal replacement of cartilage during skeletal maturation. However, it is unclear whether leptin acts as a permissive or regulatory factor and whether its actions are mediated via peripheral pathways. Here we show the osteopetrotic phenotype is not evident in ob/+ or db/+ mice, suggesting that leptin acts as a critical but permissive factor for skeletal maturation. The importance of leptin is furthe

Assessment of Feasibility of the M2 Macrophage-Based Adoptive Gene Transfer Strategy for Osteoarthritis with a Mouse Model.

Current osteoarthritis (OA) therapies fail to yield long-term clinical benefits, due in part to the lack of a mechanism for the targeted and confined delivery of therapeutics to OA joints. This study evaluates if M2 macrophages are effective cell vehicles for the targeted and confined delivery of therapeutic genes to OA joints. CT bioluminescence in vivo cell tracing and fluorescent microscopy reveal that intraarticularly injected M2 macrophages were recruited to and retained at inflamed synovia. The feasibility of an M2 macrophage-based adoptive gene transfer strategy for OA was assessed usin

A Review of CAR T Cells and Adoptive T-Cell Therapies in Lymphoid and Solid Organ Malignancies.

Chimeric antigen receptor (CAR) T cells are genetically engineered T lymphocytes that express a synthetic receptor that recognizes a tumor cell surface antigen, which causes the T lymphocyte to kill the tumor cell. As of December 2024, the US Food and Drug Administration (FDA) approved six CAR T-cell therapies, with ten CAR T-cell therapies commercially available globally, which target the CD19 and B-cell maturation antigen (BCMA) molecules and with approved indications that include B-cell acute lymphoblastic leukemia (ALL), large B-cell lymphoma (LBCL), follicular lymphoma, mantle cell lympho

Adopting telehealth service for lymphedema care: Insights from a Filariasis Management Clinic, Puducherry, India.

Lymphatic filariasis (LF) is a neglected tropical disease affecting communities in tropical and subtropical regions marked by poor socio-economic conditions. Regular hospital-based follow-up and home-based Morbidity Management and Disability Prevention (MMDP) are below par among LF patients due to reasons ranging from travelling costs to stigma. Telehealth interventions are suggested as a promising adjunct tool in such scenarios, integrating medical expertise and accessibility, but rarely used for LF. This article reflects on the insights from an LF clinic on the potential of telehealth interv
