Insights from paleomicrobiology into the indigenous peoples of pre-colonial America - a review.

This review investigates ancient infectious diseases in the Americas dated to the pre-colonial period and considers what these findings can tell us about the history of the indigenous peoples of the Americas. It gives an overview, but focuses on four microbial pathogens from this period: Helicobacter pylori, Mycobacterium tuberculosis, Trypanosoma cruzi and Coccidioides immitis, which cause stomach ulceration and gastric cancer, tuberculosis, Chagas disease and valley fever, respectively. These pathogens were selected as H. pylori can give insight into ancient human migrations into the America…

C-type lectin receptors differentially induce th17 cells and vaccine immunity to the endemic mycosis of North America.

Vaccine immunity to the endemic mycoses of North America requires Th17 cells, but the pattern recognition receptors and signaling pathways that drive these protective responses have not been defined. We show that C-type lectin receptors exert divergent contributions to the development of antifungal Th17 cells and vaccine resistance against Blastomyces dermatitidis, Histoplasma capsulatum, and Coccidioides posadasii. Acquired immunity to B. dermatitidis requires Dectin-2, whereas vaccination against H. capsulatum and C. posadasii infection depends on innate sensing by Dectin-1 and Dectin-2, but…

Travel and transplantation: travel-related diseases in transplant recipients.

PURPOSE OF REVIEW: Travel-related diseases may be seen in transplant recipients after travel, after transplant tourism, and via transmission from blood and organ donors, augmented by recent increases in travel, migration, and globalization. Such infections include tuberculosis, Plasmodium (malaria), Babesia, Trypanosoma cruzi (Chagas disease), Strongyloides, Coccidioides, Histoplasma, Leishmania, Brucella, HTLV, dengue, among numerous others. RECENT FINDINGS: Review of cohorts of transplant recipients show that they tend to have minimal or suboptimal preparation prior to travel, with limited …

[First report of coccidioidomycosis associated with Sweet syndrome].

BACKGROUND: We describe the first reported case associating coccidioidomycosis and Sweet syndrome, two uncommon diseases in Europe. CASE RE … n showed an F-precipitin on agar (IgM) and an IgG-titre of 1:8, leading to the diagnosis of primary Coccidioides immitis infection with a probable lung localization. DISCUSSION: Search for an associated disease should be made in patients with Sweet syndrome. The Sweet syndrome coccidioidomycosis association could be related to TH-1 lymphocyte proliferation.

Coccidioidomycosis among persons with AIDS in the United States.

Adults and adolescents diagnosed with AIDS from 1987 through 1992 residing in counties endemic (selected counties in California, Arizona, Texas, New Mexico, Nevada, Utah) and not endemic for Coccidioides immitis were assessed to determine the frequency of and risk factors for disseminated coccidioidomycosis. Of 602 AIDS patients reported with disseminated coccidioidomycosis, 323 (1.1% of AIDS patients) resided in C. immitis-endemic counties and 279 (0.1% of AIDS patients) resided in C. immitis-nonendemic counties in 35 states. In multivariate analysis, patients with disseminated coccidioidomyc…

Community-acquired fungal pneumonias.

Histoplasma capsulatum, Coccidioides immitis, Blastomyces dermatitidis, and Cryptococcus neoformans can cause acute community-acquired pneumonia. All are soil-dwelling fungi and disturbance of contaminated soil is necessary for infection in man. Each has particular epidemiologic considerations that may offer a clue to diagnosis, but the clinical presentation is not sufficiently characteristic to be of diagnostic help. Infection may produce variable symptoms, ranging from asymptomatic skin test conversion to fulminant respiratory failure. Chest roentgenograms are likewise nonspecific. Rapid mic…

Fungal autofluorescence with ultraviolet illumination.

Fungal autofluorescence of hematoxylin and eosin (H & E)-stained tissue sections viewed under ultraviolet illumination was evaluated for diagnostic utilization. Cases examined included coccidioidomycosis, candidiasis, aspergillosis, mucormycosis, and histoplasmosis. The method was most useful in identifying Coccidioides immites, Candida spp., and Aspergillus spp. in sections from solid parenchymatous organs, loose connective tissue, granulomata, or necrotic tissue debris. The architectural detail of spherules, yeast forms, and hyphae was very well delineated. Histoplasma capsulatum was not aut…

Delayed hypersensitivity responses of experimental animals to histoplasmin from the yeast and mycelial phases of Histoplasma capsulatum.

Controlled yeast lysate (CYL) and controlled mycelial lysate (CML) histoplasmins were produced from Histoplasma capsulatum grown in a nutritionally lean, chemically defined medium. The lysates were assayed for skin-test activity in guinea pigs sensitized by infection with the homologous organism. In some studies, nonliving vaccine preparations were employed also. Inter-lot biological variation was minimal, and 20 lots of the CYL reagent elicited strong dermal reactions with high specificity. Further, CYL reagents were nonreactive in guinea pigs infected with Coccidioides immitis, whereas the c…

Delayed hypersensitivity to fungal antigens in mice. I. Use of the intradermal skin and footpad swelling tests as assays of active and passive sensitization.

Mice were sensitized to Coccidioides immitis and Candida albicans antigens and tested for sensitivity by the intradermal and footpad swelling methods. In mice actively sensitized with killed antigen, antigen-specific intradermal and footpad induration responses occurred 24 and 48 hr after sensitization. Antigen-specific intradermal and footpad responses were transferred to normal mice with spleen cells from immune animals. Such responses were also transferred with normal spleen cells that had been incubated in vitro with immune RNA preparations. Histologic studies of intradermal reactions show…

Surveillance for Coccidioidomycosis - United States, 2011-2017.

PROBLEM/CONDITION: Coccidioidomycosis (Valley fever) is an infection caused by the environmental fungus Coccidioides spp., which typically causes respiratory illness but also can lead to disseminated disease. This fungus typically lives in soils in warm, arid regions, including the southwestern United States. REPORTING PERIOD: 2011-2017. DESCRIPTION OF SYSTEM: Coccidioidomycosis has been nationally notifiable since 1995 and is reportable in 26 states and the District of Columbia (DC), where laboratories and physicians notify local and state public health departments about possible coccidioid…

Harnessing calcineurin-FK506-FKBP12 crystal structures from invasive fungal pathogens to develop antifungal agents.

Calcineurin is important for fungal virulence and a potential antifungal target, but compounds targeting calcineurin, such as FK506, are immunosuppressive. Here we report the crystal structures of calcineurin catalytic (CnA) and regulatory (CnB) subunits complexed with FK506 and the FK506-binding protein (FKBP12) from human fungal pathogens (Aspergillus fumigatus, Candida albicans, Cryptococcus neoformans and Coccidioides immitis). Fungal calcineurin complexes are similar to the mammalian complex, but comparison of fungal and human FKBP12 (hFKBP12) reveals conformational differences in the 40s…

Coccidioidomycosis: Surgical Issues and Implications.

BACKGROUND: Coccidioidomycosis, commonly called "valley fever," "San Joaquin fever," "desert fever," or "desert rheumatism," is a multi-system illness caused by infection with Coccidioides fungi (C. immitis or C. posadasii). This organism is endemic to the desert Southwest regions of the United States and Mexico and to parts of South America. The manifestations of infection occur along a spectrum from asymptomatic to mild self-limited fever to severe disseminated disease. METHODS: Review of the English-language literature. RESULTS: There are five broad indications for surgical intervention i…

[Case of cavitary coccidioidomycosis with fungus balls in the apices of both lungs].

Pulmonary cavitary coccidioidomycosis with a fungus ball was observed in a immunocompetent case. A 32-year-old Japanese man visited Arizona … osis was made. It is very rare for fungus ball formation and coexistence of spherules and hyphae of Coccidioides immitis to be seen. Fluconazole was temporarily effective, causing cavities to shrink and eosinophilia to decrease, however Amphotericin B needed to be used later. Eosinophilia was closely related to the severity of the disease gravity.

Epidemiology of coccidioidomycosis.

Coccidioides immitis naturally occurs in the soil and air of certain areas of the New World. These are generally arid to semiarid areas that have relatively modest rainfall, mild winters, and prolonged hot seasons. Coccidioidomycosis is usually a disease of human and nonhuman residents of these areas; but visitors may develop the disease after entering these areas and returning home long distances from the endemic areas. Inhalation (rarely percutaneous introduction) of arthroconidia of C. immitis leads to usually benign but occasionally severe and even fatal infection. Recovery from or asympto…

Coccidioidin and merthiolate in previously sensitized animals.

The effect of merthiolate, which is used as a preservative in skin test materials, on skin test reactions was determined in guinea pigs. In four groups of animals, merthiolate in basal medium produced skin tests at 24 and 48 h characterized by erythema and/or induration in an intermediate region, i.e., 5 plus or minus 2.2 mm. One of the four groups of animals was a nonsensitized control group. The other three groups were subcutaneously sensitized with (i) merthiolate and saline, (ii) killed Coccidioides immitis arthrospores, and (iii) merthiolate with killed C. immitis arthrospores.
