Mesenchymal stem cell 1 (MSC1)-based therapy attenuates tumor growth whereas MSC2-treatment promotes tumor growth and metastasis.

BACKGROUND: Currently, there are many promising clinical trials using mesenchymal stem cells (MSCs) in cell-based therapies of numerous diseases. Increasingly, however, there is a concern over the use of MSCs because they home to tumors and can support tumor growth and metastasis. For instance, we established that MSCs in the ovarian tumor microenvironment promoted tumor growth and favored angiogenesis. In parallel studies, we also developed a new approach to induce the conventional mixed pool of MSCs into two uniform but distinct phenotypes we termed MSC1 and MSC2. METHODOLOGY/PRINCIPAL FIND

Characterization of Novel Mycobacteriophages of Subcluster A3: Evolutionary Plasticity of the Lysogeny Module and Lytic Potential.

The increasing prevalence of drug-resistant Mycobacteriumtuberculosis (Mtb) and nontuberculous mycobacteria (NTM) underscores the need for novel strategies. Mycobacteriophages are attracting growing interest for therapeutic and diagnostic applications. Here, we describe four new subcluster A3 phages (Cat, Goose1D, Goose1X, and Shep) and investigate their genomic diversity and lytic activity. The phages share >98% nucleotide identity with known A3 members but carry distinct deletions within the lysogeny module: Cat and Goose1X have different repressor-loss variants, and Goose1D lacks the integr

Synergistic inhibition of Streptococcus mutans biofilms by fluoride and epigallocatechin gallate: insights from multi-omics analysis.

INTRODUCTION: Streptococcus mutans biofilms are central to the development of dental caries, and strategies that effectively attenuate biofilm formation remain essential for disease control. This study examined the combined antibiofilm effects of sodium fluoride (NaF) and epigallocatechin gallate (EGCG), a green tea-derived polyphenol, and delineated the underlying mechanisms using an integrative multi-omics framework. MATERIALS AND METHODS: Synergistic interactions between NaF and EGCG were first assessed by crystal violet staining and checkerboard microdilution analysis. Biofilm biomass, vi

Controllable Engineered Probiotic Hydrogel Coatings Prevent Drainage Tube-Associated Infections by Disrupting Pathogen Oxidative Stress Responses.

Drainage tubes (DTs) are essential medical devices in clinical practice; however, preventing DT-related infections remains a significant challenge. Probiotics are considered a promising alternative therapy, but their cellular activity is susceptible to external stress. Here, we developed a strategy that integrates genetic engineering with a bilayer-structured encapsulation system to create a living probiotic coating for DTs. To enhance the safety and controllability of Saccharomyces boulardii, dual nutrient-deficient strains were constructed. ARO10 was overexpressed to improve the antagonistic

Effects of 275 nm Ultraviolet Light-Emitting Diode Irradiation on Oral Bacteria In Vitro and Toothbrush Sanitization.

The oral cavity harbors a complex microbial community where pathogens implicated in dental caries and periodontitis can heavily colonize toothbrushes, transforming them into persistent sources of contamination that threaten both oral and systemic health. Consequently, this study evaluated the bactericidal efficacy of 275 nm ultraviolet light-emitting diode (UV-LED) irradiation against common oral bacteria in vitro and its practical utility for extraoral toothbrush sanitization. Suspensions of Streptococcus mutans, Streptococcus sanguinis, Porphyromonas gingivalis, and Fusobacterium nucleatum w

Design and Evaluation of Gellan/Boron-dipyrromethene as a Photoactive Hydrogel and Efficiency as Antiyeast Activity.

The development of fluorescent sugar hydrogels has addressed robust challenges in health safety, low cost, versatile functionality, and bio-inspired utilization for application purposes. Here, we introduce a fluorescent sugar hydrogel prepared using crosslinked, low-priced gellan (GN) as a sugar resource and a synthesized boron-dipyrromethene derivative (BODIPY) that GN@BODIPY serves as a yeast inhibitor and singlet oxygen photosensitizer. The GN@BODIPY hydrogel showed maximum emission at ∼590 nm under 490 nm excitation, with a quantum yield (Φ) of 0.403, indicating efficient photoemission sui

Eco-Friendly Activation of Silicone Surfaces and Antimicrobial Coating with Chitosan Biopolymer.

Silicone is widely used in medical devices due to its mechanical properties and biocompatibility; however, microbial contamination of silicone surfaces, which can lead to nosocomial infections, remains a significant concern. This can be countered by surface modification using techniques commonly involving oxidative plasma activation or ozone treatments, followed by treatment with silanization agents. Here, we report an alternative surface modification procedure involving treatment with non-toxic organic hydroxyl amines or diamine dissolved in eco-friendly solvents, thus avoiding using reactive

Long-term antifungal drug-delivery denture materials with plasma-initiated surface functionalization.

Conventional polymethylmethacrylate (PMMA)-based denture materials are susceptible to colonization and biofilm-formation by Candida species, leading to Candida-associated denture stomatitis (CADS), a recurring fungal infection that affects a large patient population. In this report, we developed a two-step, plasma-initiated surface functionalization approach to covalently bond poly (methacrylic acid) (PMAA) onto denture resins. Various reaction conditions were investigated and the effects of PMAA grafting yields on the physical properties of the resulting denture materials were determined. The

Mechanism-Based Pharmacokinetic/Pharmacodynamic Modeling for Iron-Regulated Hematopoietic Stem and Progenitor Cells' Commitment toward Erythroid and Megakaryocytic Lineages.

Iron replenishment is a cornerstone therapy for anemia in diverse diseases. While its role in erythrocyte hemoglobinization is well-established, the broader impact of iron on other aspects of hematopoiesis, such as thrombopoiesis, remains poorly understood. In this study, we demonstrate that iron plays a regulatory role in the commitment of hematopoietic stem and progenitor cells (HSPCs) toward erythroid and megakaryocytic lineages. Using colony-forming unit assays and flow cytometry, we observed that iron increases the proportion of erythroid cells while reducing the proportion of megakaryocy

In Vitro Inhibitory Effect of Silver Diamine Fluoride Combined with Potassium Iodide against Mixed-Species Biofilm Formation on Human Root Dentin.

Applying a saturated potassium iodide (KI) solution immediately after silver diamine fluoride (SDF) application may affect the inhibitory effects of SDF on biofilm formation. This study compared the efficacy of 38% SDF with and without KI on preventing mixed-species biofilm formation on human root dentin surfaces and assessed ion incorporation into root dentin. The biofilms, composed of Streptococcus mutans, Lactobacillus rhamnosus, and Actinomyces naeslundii, were grown on specimen surfaces treated with either SDF or SDF + KI. After 24 h, the biofilms were evaluated using scanning electron mi

Enhanced medullary and extramedullary granulopoiesis sustain the inflammatory response in lupus nephritis.

OBJECTIVES: In SLE, deregulation of haematopoiesis is characterised by inflammatory priming and myeloid skewing of haematopoietic stem and progenitor cells (HSPCs). We sought to investigate the role of extramedullary haematopoiesis (EMH) as a key player for tissue injury in systemic autoimmune disorders. METHODS: Transcriptomic analysis of bone marrow (BM)-derived HSPCs from patients with SLE and NZBW/F1 lupus-prone mice was performed in combination with DNA methylation profile. Trained immunity (TI) was induced through β-glucan administration to the NZBW/F1 lupus-prone model. Disease activit

Bone marrow hematopoietic dysfunction in untreated chronic lymphocytic leukemia patients.

The consequences of immune dysfunction in B-chronic lymphocytic leukemia (CLL) likely relate to the incidence of serious recurrent infections and second malignancies that plague CLL patients. The well-described immune abnormalities are not able to consistently explain these complications. Here, we report bone marrow (BM) hematopoietic dysfunction in early and late stage untreated CLL patients. Numbers of CD34+ BM hematopoietic progenitors responsive in standard colony-forming unit (CFU) assays, including CFU-GM/GEMM and CFU-E, were significantly reduced. Flow cytometry revealed corresponding r

Wnt10b deficiency results in age-dependent loss of bone mass and progressive reduction of mesenchymal progenitor cells.

Wnt10b is a canonical Wnt ligand expressed in developing bone and has been linked to mesenchymal progenitor functions in mice and humans. Because Wnt signaling has been shown to play an important role in progenitor maintenance in a variety of adult tissues, we examined bone deposition and growth rates throughout postnatal development in Wnt10b-null mice. Using bone histomorphometry and micro-computed tomographic (µCT) studies, we demonstrate that trabecular bone deposition is slightly enhanced in Wnt10b-null mice at 1 month of age, followed by progressive loss with age. Importantly, we find th

Adhesion of Porphyromonas gingivalis strains to cultured epithelial cells from patients with a history of chronic adult periodontitis or from patients less susceptible to periodontitis.

BACKGROUND: The present study aimed to explain the interindividual variation in periodontitis susceptibility by differences in the initial adhesion rate of Porphyromonas gingivalis to the pocket epithelium of these individuals, and/or by inter-P. gingivalis strain differences in association capacity (adhesion and internalization). METHODS: Adhesion assays were performed on epithelial monolayers (cultured in vitro from pocket epithelium belonging to patients who were less or more susceptible to chronic adult periodontitis) using 11 genetically different clinical strains of P. gingivalis.
