The Dual Role of DNA Cross-linking Agents: From Genotoxicity to cGAS-STING-Mediated Immune Activation.

DNA cross-linking agents remain a cornerstone of cancer chemotherapy. While their efficacy was traditionally attributed to direct genotoxicity, blocking DNA replication and transcription to induce cell death, recent evidence suggests that their therapeutic activity also involves innate immune activation. Aberrant repair of DNA lesions generates cytosolic DNA fragments, which are sensed by cyclic GMP-AMP synthase (cGAS). This activates the cGAS-STING (stimulator of interferon genes) pathway, driving the production of type I interferons and proinflammatory cytokines, thereby eliciting antitumor …

Nano-immunomodulators co-activate immunogenic cell death and the cGAS-STING pathway for breast cancer immunotherapy.

Breast cancer generally does not respond to immunotherapy due to its inherently low immunogenicity and insufficient coordination between innate and adaptive immune responses. Here, we demonstrated that concurrent activation of immunogenic cell death (ICD) and the cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway synergistically amplifies antitumor immunity by promoting dendritic cell maturation, co-stimulatory signaling, and T cell priming. Building on these mechanistic insights, we designed a nanoparticle-based immunomodulator (nanoIM) that integrates a serum albumin…

PKCα-mediated nuclear translocation of cGAS stabilizes β-catenin and drives metastasis.

Although recent studies have shown that 2'3'-cyclic GMP-AMP synthase (cGAS) has both canonical cytoplasmic and non-canonical nuclear functions, the role of nuclear cGAS in cancer metastasis remains unclear. Here, we identify a STING-independent mechanism by which nuclear cGAS activates Wnt/β-catenin signaling to promote metastasis in triple-negative breast cancer (TNBC). Protein kinase C alpha (PKCα) phosphorylates cGAS at Ser120, facilitating its nuclear translocation. Once in the nucleus, cGAS disrupts the interaction between β-catenin and tripartite motif-containing protein 33 (TRIM33), pre…

AMnESTI reverses age-associated lung immune decline to drive heterosubtypic protection by inactivated influenza vaccines.

Existing influenza vaccines fail to elicit heterosubtypic immunity and have limited efficacy in the elderly. We developed AMnESTI (AEC-targeting MN Encapsulated STING agonist), an alveolar epithelial cell (AEC)-targeting manganese-complexed liposome encapsulating the stimulator of interferon genes (STING) agonist ADU-S100. Intranasal administration activated the cyclic GMP-AMP synthase-STING pathway in AECs, robustly expanding plasmacytoid dendritic cells, monocyte-derived dendritic cells, and inflammatory monocytes while reducing regulatory T cells (Treg cells) and immunosuppressive cDC2β+ ce…

Mechanistic insights and therapeutic potential of targeting the cGAS-STING pathway in neurodegenerative diseases.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway is a central cytosolic DNA-sensing module that links DNA damage and mitochondrial dysfunction to innate immune activation. Here, we focus on canonical cGAS-STING signaling in the central nervous system (CNS) and discuss non-canonical branches only when directly relevant to neurodegeneration. We summarize structural and activation-termination mechanisms and synthesize cell-type-biased outputs across microglia, astrocytes, neurons, and oligodendroglial lineage cells. We then integrate Alzheimer's disease, Parkinson's…

Cytotoxic lymphocytes counteract viral type I interferon immune evasion.

Viruses are recognized by host cell innate immunity through viral RNA/DNA sensing by cyclic GMP-AMP synthase (cGAS) stimulator of interferon genes (STING). However, many viruses evade cGAS-STING signaling and antiviral IFN-β response. Here, we show that natural killer (NK) cells counteract immune evasion of type I interferon response upon human cytomegalovirus (HCMV) infection. NK cells enhance IFN-β response in virus-infected cells more efficiently than perforin-knockout and GrM-knockout NK cells. Mechanistically, GrM cleaves viral pp71 into two fragments, the first, like full-length pp71, st…

Dumbbell-shape DNA structure activates cGAS-STING pathway for enhanced checkpoint immunotherapy in colorectal cancer.

Colorectal cancer (CRC), a prevalent malignancy of the digestive tract with poor responsiveness to existing immunotherapies, presents an urgent need for strategies to overcome therapeutic resistance. The cyclic GMP-AMP synthase (cGAS)- stimulator of interferon genes (STING) pathway, a key mediator of innate antitumor immunity through DNA sensing and type I interferon (IFN-I) signaling, offers a promising target for intervention. However, conventional STING agonists are limited by insufficient tumor targeting and systemic adverse effects. To address these challenges, this study developed a tumo…

Context-dependent actions of STING pathway in colitis and associated colon cancer.

Inflammatory bowel disease (IBD), a prevalent chronic inflammatory disorder with unsatisfactory therapeutic outcomes, significantly increases the risk of colorectal cancer. The cyclic GMP-AMP synthase (cGAS) and stimulator of interferon gene (STING), highly expressed in human IBD, are potential anti-inflammatory and anti-tumor immunotherapeutic targets. However, conflicting evidence regarding the dual roles of the STING pathway has significantly hindered its development as a therapeutic target for innovative treatments. Previous studies have predominantly suggested that hyperactivation of the …

Dual-targeted molybdenum nanomedicine treats acute pancreatitis by blocking mitochondrial DNA-triggered cGAS-STING signaling via ROS modulation.

Acute pancreatitis (AP), a potentially fatal disorder driven by macrophage-associated inflammation, involves mitochondrial reactive oxygen species (ROS) overproduction with pancreatic acinar cells (PACs). This ROS surge damages mitochondria, causing mitochondrial DNA (mt-DNA) leakage and PACs apoptosis. Released mt-DNA then activates the pro-inflammatory cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway in macrophages, exacerbating disease progression. Critically, while scavenging mitochondrial ROS can halt this cycle, existing mitochondria-targeted drugs fail to pene…

VDAC1 oligomerization-mediated mtDNA release under sublethal oxidative stress: A novel inflammatory mechanism in vitiligo.

Oxidative stress is a critical initiating factor in vitiligo, yet the early molecular events linking redox imbalance to melanocyte immune activation remain unclear. Here, we demonstrate that sub-lethal hydrogen peroxide (H2O2, 0.1 mM) exposure in human epidermal melanocytes induces a robust pro-inflammatory response independent of apoptosis or pyroptosis. This response is driven by the selective cytosolic release of mitochondrial DNA (mtDNA), which activates the cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway. Mechanistically, voltage-dependent anion channel 1 (VDAC…

ATP-Responsive Manganese-Based Bacterial Materials Synergistically Activate the cGAS-STING Pathway for Tumor Immunotherapy.

Stimulating the cyclic guanosine monophophate(GMP)-adenosine monophosphate (AMP) synthase (cGAS)-stimulator of interferon genes (STING) pathway is a crucial strategy by which bacteria activate the tumor immune system. However, the limited stimulation capability poses significant challenges in advancing bacterial immunotherapy. Here, an adenosine 5'-triphosphate (ATP)-responsive manganese (Mn)-based bacterial material (E. coli@PDMC-PEG (polyethylene glycol)) is engineered successfully, which exhibits an exceptional ability to synergistically activate the cGAS-STING pathway. Molecular docking, dynamics and in vitro analysis of multi-target inhibitors for Clostridioides difficile.

The opportunistic pathogen, Clostridioides difficile owes its extreme pathogenicity for its ability to develop antibiotic resistance and recurrent infections. The current antibiotics used for the treatment are showing declining sensitivity and rising antibiotic resistance. Therefore, it is of interest to develop the anti-clostridial drugs to overcome these issues. Hence, we have explored ZINC library to find the suitable lead compounds against five target proteins of C. difficile. Multistep virtual screening is performed to find the suitable compounds that are checked for their stability using

DNA Damage-driven Inflammatory Cytokines: Reprogramming of Tumor Immune Microenvironment and Application of Oncotherapy.
