Development and standardization of a rapid, PCR-based method for the detection of Wuchereria bancrofti in mosquitoes, for xenomonitoring the human prevalence of bancroftian filariasis.

PCR has recently been studied as a promising tool for monitoring the progress of efforts to eliminate lymphatic filariasis. PCR can be used to test concurrently at least 30 pools, with as many as 40 mosquitoes in each pool, for the presence of filarial larvae. The SspI PCR assay for the detection of Wuchereria bancrofti DNA in pools of mosquitoes has been used since 1994 in a variety of laboratories worldwide. During that time, the original assay has been modified in these different laboratories and no standardized assay currently exists. In an effort to standardize and improve the assay, a me

Schistosoma mansoni and Biomphalaria: past history and future trends.

Schistosoma mansoni is one of the most abundant infectious agents of humankind. Its widespread distribution is permitted by the broad geographic range of susceptible species of the freshwater snail genus Biomphalaria that serve as obligatory hosts for its larval stages. Molecular phylogenetic studies suggest that Schistosoma originated in Asia, and that a pulmonate-transmitted progenitor colonized Africa and gave rise to both terminal-spined and lateral-spined egg species groups, the latter containing S. mansoni. Schistosoma mansoni likely appeared only after the trans-Atlantic dispersal of Bi

Recombinant vaccinia viruses and gene gun vectors expressing the large subunit of Schistosoma mansoni calpain used in a murine immunization-challenge model.

Schistosomiasis is a parasitic disease affecting over 200 million people every year in tropical regions of the world. Drug treatment and other existing control measures are costly and have failed to eliminate the incidence of infection, morbidity and mortality due to Schistosoma mansoni infection. Vaccination of susceptible individuals using recombinant vaccines encoding key S. mansoni antigens may be the most effective and least expensive means of controlling schistosomiasis. A candidate vaccine antigen is p80, the large subunit of the S. mansoni protease, calpain. In our vaccine studies, we

Simple blood-spot sampling with nested polymerase chain reaction detection for epidemiology studies on Brugia malayi.

In the absence of a suitable Brugia malayi antigen detection assay, PCR remains one of the more sensitive alternatives to Giemsa-stained thick blood films for B. malayi detection. The need for refrigerated storage and transportation of blood has limited the use of PCR for large-scale epidemiology studies in remote endemic areas. Here we report simple finger-prick blood-spot collection, a one-tube DNA template extraction method and the development of a B. malayi-specific nested PCR assay. The assay was tested on 145 field samples and was positive for all 30 microscopy-positive samples and for a

A multiplex PCR-based sequencing method for the diagnosis and differentiation of echinococcosis using plasma samples, a proof-of-concept study.

Echinococcosis poses a significant public health threat, and its diagnosis relies on imaging. Non-invasive plasma detection of Echinococcus cell-free DNA (cfDNA) offers a promising diagnostic approach. We developed a low-cost multiplex PCR panel with 230 primer pairs targeting Echinococcus mitochondrial genomes and nuclear DNA repeat regions. We validated the panel using gradient dilutions of two pathogen species' DNA and 81 plasma samples (53 patients and 28 controls), differentiating species through sequencing analysis. Our method detected Echinococcus genomic DNA at an input as low as 1 fg

Redescription of two Allocreadium species and molecular dating of the family Allocreadiidae (Trematoda: Gorgoderoidea).

The family Allocreadiidae comprises diverse parasites of freshwater fishes, but their evolutionary timescale remains poorly understood. We provide a molecular dating analysis based on an expanded 28S rRNA dataset and redescriptions of two East Asian species, Allocreadium hasu and A. pseudaspii, with new morphological and genetic data. Allocreadium hasu from Lake Biwa (Japan) is genetically close to Far Eastern A. khankaiense and A. anastasii (0.09-0.97% divergence in 28S) but has not been recorded in the Russian Far East. Allocreadium pseudaspii, first recorded in the Bolshaya Ussurka River, p

First report of fatal baylisascariasis-induced acute pancreatitis in a giant panda.

A wild adult male giant panda that was rescued from a nature reserve in Sichuan Province, China, has died. The panda had been in poor physical condition: it was wheezing and had increased serum amylase. A pathological examination was performed in order to determine the cause of death. Gross examination revealed 1380 mL of yellowish fluid in the abdominal cavity, 356 nematodes in the digestive tract and one filling the pancreatic duct, contractions and variably-sized dark purple areas in the spleen, a collapsed right lung and consolidation of the left lung. Acute pancreatitis was confirmed hist

Morphological and Molecular Characteristics of the Gastro-Intestinal Nematode Parasite Ascaridia columbae Infecting the Domestic Pigeon Columba livia domestica in Saudi Arabia.

BACKGROUND: Parasitism is a complex problem that is often ignored in companion animals, including birds, unless it develops into a severe clinical disorder. The present study was, therefore, aimed to investigate the presence of the gastrointestinal nematode infecting the domestic pigeon and provide a complete morphological description and clarify its taxonomic position through phylogenetic analysis of the ITS1-5.8s-ITS2 rDNA gene region. MATERIALS AND METHODS: During the current study, a total of twenty-six domestic pigeons, Columba livia domestica, were collected and internal organs examined

Label-free nanostructured biosensor for Schistosoma mansoni detection in complex biological fluids.

Schistosomiasis is a neglected tropical disease with a worldwide prevalence. Neuroschistosomiasis is the most severe presentation of the disease and affects the central nervous system. In this work, Schistosoma mansoni detection was based on self-assembled layers of 3-mercaptopropyltrimethoxysilane (MPTS) and electrosynthesized gold nanoparticles (AuNPs). The DNA probe was chemisorbed onto AuNPs. The biosensor was characterized by electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). The impedimetric response of the MPTS-AuNPs-DNAprobe system indicates an effective modifica

O-5S quantitative real-time PCR: a new diagnostic tool for laboratory confirmation of human onchocerciasis.

BACKGROUND: Onchocerciasis is a parasitic disease caused by the filarial nematode Onchocerca volvulus. In endemic areas, the diagnosis is commonly confirmed by microscopic examination of skin snip samples, though this technique is considered to have low sensitivity. The available melting-curve based quantitative real-time PCR (qPCR) using degenerated primers targeting the O-150 repeat of O. volvulus was considered insufficient for confirming the individual diagnosis, especially in elimination studies. This study aimed to improve detection of O. volvulus DNA in clinical samples through the deve

Elucidation of the first definitively identified life cycle for a marine turtle blood fluke (Trematoda: Spirorchiidae) enables informed control.

Blood flukes of the family Spirorchiidae are significant pathogens of both free-ranging and captive marine turtles. Despite a significant proportion of marine turtle mortality being attributable to spirorchiid infections, details of their life cycles remain almost entirely unknown. Here we report on the molecular elucidation of the complete life cycle of a marine spirorchiid, identified as Amphiorchis sp., infecting vermetid gastropods and captive hatched neonate Caretta caretta in the Oceanogràfic Aquarium, in Valencia, Spain. Specimens of a vermetid gastropod, Thylaeodus cf. rugulosus (Monte

Identity of rumen fluke in deer.

As evidence is growing that in many temperate areas paramphistome infections are becoming more common and widespread, this study was undertaken to determine the role of deer as reservoirs for rumen fluke infections in livestock. A total of 144 deer faecal samples (88 from fallow deer, 32 from red deer and 24 samples from sika, sika/red deer hybrids) were screened for the presence of fluke eggs. Based on the ITS-2 rDNA locus plus flanking 5.8S and 28S sequences (ITS-2+), fluke eggs were identified to species level. Our results indicate that, of the 3 deer species, fallow deer had the highest fl

Alloglossidium floridense n. sp. (Digenea: Macroderoididae) from a spring run in North Central Florida.

A new species of Alloglossidium is described from the intestines of 2 madtom species (Noturus leptacanthus and Noturus gyrinus) that were collected from the run of a small, unnamed spring system that drains into the Santa Fe River, Florida. Alloglossidium floridense n. sp. is morphologically very similar to other nonprecocious Alloglossidium spp. that use ictalurids as definitive hosts, but can be distinguished by a combination of its smaller overall size (length and width), large eggs in relation to its small body size, position of the vitellaria, ovary shape, and position of the ovary in rel

Mitochondrial phylogeny of the genus Echinococcus (Cestoda: Taeniidae) with emphasis on relationships among Echinococcus canadensis genotypes.

The mitochondrial genomes of the genus Echinococcus have already been sequenced for most species and genotypes to reconstruct their phylogeny. However, two important taxa, E. felidis and E. canadensis G10 genotype (Fennoscandian cervid strain), were lacking in the published phylogeny. In this study, the phylogeny based on mitochondrial genome sequences was completed with these taxa. The present phylogeny highly supports the previous one, with an additional topology showing sister relationships between E. felidis and E. granulosus sensu stricto and between E. canadensis G10 and E. canadensis G6

Prevalence survey and first molecular characterization of Echinococcus granulosus in France.

Human cystic echinococcosis (hydatid disease) caused by the Echinococcus granulosus tapeworm continues to be a substantial cause of morbidity and mortality in many parts of the world. France is still considered as endemic area, but the current infestation by E. granulosus of intermediate hosts in France remains currently unknown due to the absence of official data reporting for the last 20 years. A 1-year prevalence survey was conducted in the 24 slaughterhouses of ten departments of the South of France. We demonstrate that the E. granulosus parasite is still currently present at low prevalenc

Ascariasis in people and pigs: new inferences from DNA analysis of worm populations.

Ascaris is a large parasitic roundworm (nematode) of the small intestine of humans and pigs. These roundworms cause the socioeconomically important disease, ascariasis. For the past 20 years, molecular markers have been used in studies on Ascaris and ascariasis, and added valuable information to the understanding of these roundworms. Here, we provide a review of these studies on human and pig roundworms. We begin with a summary of studies using molecular phenotypic markers to compare Ascaris from humans and pigs, followed by a synopsis of comparisons using genetic markers. We then draw forth i

The potential of molecular diagnosis of cutaneous ectopic schistosomiasis.

A 28-year-old woman presented with extensive erythematous lesions on her back after visiting Malawi. Skin biopsies showed ova, which could belong to Schistosoma spp. Sequencing of the Schistosoma 28S rRNA gene, extracted and amplified from paraffin biopsies, identified DNA of Schistosoma haematobium. Cutaneous ectopic schistosomiasis can present with extensive lesions and should be considered in the differential diagnosis of skin lesions in returning travelers. Microscopy and serology are the classical methods to obtain a diagnosis. Alternatively, molecular methods can be a valuable new tool f

[DNA diagnosis of mixed invasions of Opisthorchis felineus and Metorchis bilis by polymerase chain reaction].

Opisthorchiasis is one of the significant naturofocal diseases in Russia. The diagnosis of opisthorchiasis may mask some diseases, such as opisthorchiasis, metorchiasis, and clonorchiasis - biohelminthoses that are induced by various representatives of the family Opisthorchiidae-Opisthorchis felineus/O.viverrini, Metorchis bilis, and Clonorchis sinensis, respectively. Coproovoscopy and serologic methods fail to accurately define the species-specific affiliation; in this connection the identification of opisthorchids, by using DNA diagnostic techniques, becomes urgent. The present paper gives t

Taenia crassiceps cysticercosis: variations in its parasite growth permissiveness that encounter with local immune features in BALB/c substrains.

This study describes the first days of Taenia crassiceps infection in BALB/c substrains, BALB/cAnN and BALB/cJ, using two stocks of the same strains which were kept in different animal facilities, conventional and pathogen-free conditions, respectively. This study shows that parasite growth restriction shown by conventional BALB/cJ mice changed to parasite growth permissiveness when pathogen-free BALB/cJ mice were used. In addition, the higher number of macrophages, NK cells and intraperitoneal level of IFN-gamma found in the conventional restrictive BALB/cJ substrain vanished when the permiss
