Microbial functions in skin inflammation: a Mendelian randomization study.

Inflammatory skin diseases (ISDs) represent a significant global health burden, yet the role of the human microbiome in their pathogenesis remains unclear. This study employed a comprehensive two-sample Mendelian randomization (MR) framework to investigate potential causal associations between multi-site microbiota (3117 oral, 412 gut, and 150 skin microbial features) and five major ISDs: psoriasis, atopic dermatitis, acne, seborrheic dermatitis, and urticaria. Genetic instruments were selected from large-scale genome-wide association studies (GWAS) of microbiota composition and ISD outcomes.

Face mask-induced skin changes: a new common phenomenon during the coronavirus disease 2019 pandemic.

Coronavirus disease 2019 is a severe acute respiratory syndrome, and wearing a mask is an important preventive measure. However, long-term use can cause maskne, or mask-induced acne. This study evaluated the prevalence, clinical characteristics, and severity of mask-induced acne and its effect on pre-existing dermatosis using an online questionnaire. The questionnaire comprised 15 questions on 6 primary topics: demographic data, type of face mask, dermatological symptoms after using a mask, preexisting dermatological conditions, the duration of use, and the effects of mask use on existing derm

Most common pediatric skin conditions managed in outpatient dermatology clinics in the United States stratified by race and ethnicity.

A better understanding of what skin conditions are most commonly diagnosed in different pediatric racial and ethnic groups in outpatient dermatology clinics could help guide the development of pediatric dermatology educational initiatives for primary care providers and general dermatologists who have limited access to pediatric dermatologists. Using a nationally representative dataset, we evaluated the most common diagnoses in patients younger than 15 years of age (children) and 15-24 years of age (youth) who present to outpatient dermatology clinics, stratified by race and ethnicity. While ac

Pimecrolimus in dermatology: atopic dermatitis and beyond.

Pimecrolimus is a calcineurin inhibitor developed for the topical therapy of inflammatory skin diseases, particularly atopic dermatitis (AD). Pimecrolimus selectively targets T cells and mast cells. Pimecrolimus inhibits T-cell proliferation, as well as production and release of interleukin-2 (IL-2), IL-4, interferon-gamma and tumour necrosis factor-alpha. Moreover, pimecrolimus inhibits mast cell degranulation. In contrast to tacrolimus, pimecrolimus has no effects on the differentiation, maturation and functions of dendritic cells. In contrast to corticosteroids, pimecrolimus does not affect

24-day-old newborn with tinea capitis: A case report.

Tinea capitis is a common childhood fungal infection that is extremely rare in newborns, with limited data available. Its clinical presentation may closely resemble that of other dermatological conditions such as seborrheic dermatitis, neonatal lupus erythematosus and congenital syphilis. It is essential to make a definite diagnosis, as tinea capitis treatment requires systemic antifungal therapy. However, successful topical monotherapies have been reported. We report a rare case of neonatal tinea capitis diagnosed in a 24-day-old newborn, presenting with multiple scaly plaques on the scalp, d

Chemical, Biochemical, and Structural Similarities and Differences of Dermatological cAMP Phosphodiesterase-IV Inhibitors.

Roflumilast, the third phosphodiesterase-IV (PDE4) inhibitor approved for use in dermatology, is indicated for topical treatment of psoriasis, seborrheic dermatitis, and atopic dermatitis, whereas its 2 predecessors, apremilast and crisaborole, are indicated for oral treatment of psoriasis and topical treatment of atopic dermatitis, respectively. All 3 are rationally designed PDE4 inhibitors, but roflumilast is the most potent and effective among the 3, with in vitro inhibitory constant half-maximal inhibitory concentration value of 0.7 nM (roflumilast), 0.14 μM (apremilast), and 0.24 μM (cris

Burden of skin disease and associated socioeconomic status in Asia: A cross-sectional analysis from the Global Burden of Disease Study 1990-2017.

INTRODUCTION: Skin diseases have a significant global impact on quality of life, mental health, and loss of income. The burden of dermatologic conditions and its relationship with socioeconomic status in Asia is currently not well understood. METHODS: We selected Global Burden of Disease Study datasets to analyze disability-adjusted life years (DALYs) in 50 Asian countries, including Central Asia, northern Asia, eastern Asia, western Asia, southeastern Asia, and southern Asia, between 1990 and 2017. We compared DALYs to the socioeconomic status using the sociodemographic index and gross domes

Human sebum extract induces barrier disruption and cytokine expression in murine epidermis.

BACKGROUND: Previous studies have shown that human sebum may play a role in barrier function but with much debate. OBJECTIVE: To elucidate the effects of human sebum on skin barrier function. METHODS: We used hairless mouse skin to study the functional and morphological alternation of epidermis after the application of human sebum. RESULTS: The results showed a significant increase in transepidermal water loss and erythema value, and a decrease in skin hydration, accompanied by epidermal hyperplasia with parakeratosis following sebum application. Nile red staining together with electron mic

Assigning new roles to topical tacrolimus.

Tacrolimus is an ascomycin macrolactam derivative with immunomodulatory and anti-inflammatory activity that belongs to the class of calcineurin inhibitors. Tacrolimus in its topical formulation has been established as a safe and effective alternative to topical corticosteroids because of its mild side effects and its minimal systemic absorption. Topical tacrolimus has been approved for the treatment of atopic dermatitis in two concentrations, 0.03 and 0.1%. In a thorough research of literature the authors review all of the available data regarding the off-label uses of the medication in other

Identifying potential drug targets for seborrhoeic dermatitis and dry eye syndrome through mendelian randomisation analysis.

CLINICAL RELEVANCE: Patients with seborrhoeic dermatitis frequently experience comorbid dry eye syndrome, which complicates clinical management. Identifying shared drug targets is a promising strategy for developing novel therapies that concurrently address both conditionggs. BACKGROUND: Ocular involvement is common in seborrhoeic dermatitis and frequently leads to dry eye syndrome, underscoring the importance of identifying shared drug targets for dual-pathway therapy. METHODS: A two-sample Mendelian Randomisation (MR) analysis was conducted to identify potential drug targets. Sensitivity a

Diverse Colonisation and Disease Associations of the Human Commensal Malassezia: Our Body's Secret Tenant.

A niche in the context of microorganisms defines the specific ecological role or habitat inhabited by microbial species within an ecosystem. For the human commensal Malassezia, the skin surface is considered its primary niche, where it adapts to the skin environment by utilising lipids as its main carbon and energy source. However pathogenic characteristics of Malassezia include the production of allergens, immune modulation and excessive lipid utilisation, which result in several diseases such as pityriasis versicolor, seborrheic dermatitis, Malassezia folliculitis and atopic dermatitis. The effects of alcohol and illicit drug use on the skin.

Early identification of cutaneous manifestations of alcohol and illicit drug use can aid in the diagnosis and management of these uses, as well as their sequelae. In addition, the effects of alcohol and illicit drug use on the skin can result in significant morbidity. Alcohol misuse can present with jaundice, pruritus, pigmentary alterations, urticaria, hair and nail changes, and oral changes. It is also a risk factor for skin cancer and infections. Vascular disturbances associated with alcohol misuse include telangiectasias, palmar erythema, caput medusae, and flushing. Diseases related to al

Approaches for Genetic Discoveries in the Skin Commensal and Pathogenic Malassezia Yeasts.

Malassezia includes yeasts belong to the subphylum Ustilaginomycotina within the Basidiomycota. Malassezia yeasts are commonly found as commensals on human and animal skin. Nevertheless, Malassezia species are also associated with several skin disorders, such as dandruff/seborrheic dermatitis, atopic eczema, pityriasis versicolor, and folliculitis. More recently, associations of Malassezia with Crohn's disease, pancreatic ductal adenocarcinoma, and cystic fibrosis pulmonary exacerbation have been reported. The increasing availability of genomic and molecular tools have played a crucial role in

Unilateral acne after facial palsy.

We describe a case of unilateral acne which appeared after an episode of facial nerve palsy. An 18-year-old female patient presented with papules and pustules predominantly located on the side where the facial paralysis occurred. The patient suffered right facial paralysis, which was treated with prednisone and kinesiotherapy with massages, electrostimulation, and infrared light. Two weeks later, acne lesions appeared in the area affected by the paralysis. As suggested in other cases of paralysis, including cases of Parkinson's disease and spinal cord injury, an increased sebum excretion rate

Association between Malassezia furfur colonization and seborrhoeic dermatitis in AIDS patients.

A total of 180 HIV-positive patients were assessed for seborrhoeic dermatitis (SD) and colonization with Malassezia species. Diseased skin of patients with seborrhoeic dermatitis were sampled selectively for Malassezia. In patients without SD, uninvolved skin was sampled. The prevalence of SD was 19%. Of the 34 SD patients, 16 were positive for Malassezia. Of the 146 patients without SD, only 27 were culture positive for Malassezia. Analysis of the largest HIV-positive patient population studied thus far yielded only a weak correlation between SD and Malassezia colonization.

The control of seborrhoeic dermatitis and dandruff by antipityrosporal drugs.

For many years the exact nature of the pathophysiology of seborrhoeic dermatitis and dandruff was in doubt. Different schools of thought debated whether Pityrosporum yeasts were of primary pathogenic significance or a secondary phenomenon, with epidermal hyperproliferation as the primary pathology. Although effective therapy in seborrhoeic dermatitis and dandruff has for a long time been based on compounds whose only common link was antipityrosporal activity, proof of this relevance was lacking until the introduction of effective antifungal drugs, in particular ketoconazole.
