Origin and turnover of ECM proteins from the inner limiting membrane and vitreous body.

The inner limiting membrane (ILM) and the vitreous body (VB) are two major extracellular matrix (ECM) structures that are essential for early eye development. The ILM is considered to be the basement membrane of the retinal neuroepithelium, yet in situ hybridization and chick/quail transplant experiments in organ-cultured eyes showed that all components critical for ILM assembly, such as laminin or collagen IV, are not synthesized by the retina. Rather, ILM proteins, with the exception of agrin, originate from the lens or (and) ciliary body and are shed into the vitreous. The VB serves as a re

Reorganization of Burst Initiation Zones as a Signature of Stimulation-Induced Plasticity in In Vitro Cortical Networks.

The spontaneous activity of neuronal cultures is dominated by periods of coherent activity known as network bursts The dynamics of the initiation and propagation of these bursts are believed to depend on the structural connectivity of the neuronal network. Specifically, the spatial loci of burst initiation, referred to as initiation zones (IZs), are shaped by a complex interplay between dynamics and connectivity, with the particular structure of the latter playing a crucial role. Thus, alterations in network structure, such as those following from plasticity mechanisms activated by perturbatio

Consequences of Glycine Mutations in the Fibronectin-binding Sequence of Collagen.

Collagen and fibronectin (Fn) are two key extracellular matrix proteins, which are known to interact and jointly shape matrix structure and function. Most proteins that interact with collagen bind only to the native triple-helical form, whereas Fn is unusual in binding strongly to denatured collagen and more weakly to native collagen. The consequences of replacing a Gly by Ser at each position in the required (Gly-Xaa-Yaa)6 Fn-binding sequence are probed here, using model peptides and a recombinant bacterial collagen system. Fluorescence polarization and solid-state assays indicated that Gly r

New clues for the developing human biliary system at the porta hepatis.

The human biliary system is formed from the hepatic diverticulum, a structure which develops from the embryonic foregut in the fourth week of gestation. The cephalic portion of the hepatic diverticulum lies within the septum transversum, and gives rise to entodermal cells which become the primitive hepatocytes. The caudal part of the hepatic diverticulum is molded by mesenchyme to form the gallbladder, cystic duct, and extrahepatic bile duct. The gallbladder is initially tubular in shape, and undergoes morphological changes to become saccular during the 11th week of gestation. The extrahepatic

Persistent Muscle Fiber Regeneration in Long Term Denervation. Past, Present, Future.

Despite the ravages of long term denervation there is structural and ultrastructural evidence for survival of muscle fibers in mammals, with some fibers surviving at least ten months in rodents and 3-6 years in humans. Further, in rodents there is evidence that muscle fibers may regenerate even after repeated damage in the absence of the nerve, and that this potential is maintained for several months after denervation. While in animal models permanently denervated muscle sooner or later loses the ability to contract, the muscles may maintain their size and ability to function if electrically s

Antenatal sonographic features of persistent extrahepatic vitelline vein aneurysm confused with umbilical vein varix.

Background: The persistent vitelline vein is a portal venous system malformation arising during the embryonic period. These abnormal blood vessels frequently thrombose and can lead superior mesenteric vein obstruction or portal hypertension. Case report: We visualized a fetal intra-abdominal cystic mass with turbulent flow on prenatal ultrasound at 28 weeks' gestation. Initially diagnosed as an umbilical vein varix, it was later determined to be an extrahepatic persistent vitelline vein with an internal thrombus by postnatal ultrasound. It was successfully surgically excised. Conclusion: When

A Paratesticular Multicystic Tumor of the Tunica Vaginalis Testis as Rare Paratesticular Cystadenoma.

The cystadenoma of the testis and paratestis arising from an unequivocal oviduct-like structure, which is morphologically almost identical with those of the ovarian surface epithelium. These are very rare benign tumors of young adults. They present as asymptomatic cystic lesions. Bilateral paratesticular cystadenomas are strongly associated with von Hippel-Lindau syndrome and correlate with infertility. It is a neoplasm with low malignant potential. Most cystadenomas are benign but a few cases of malignant transformation of embryonic remnants have been reported in the appendix testis, includin

A review of cytogenetics in equine reproduction.

The karyotype of the horse consists of 64 chromosomes; 18 pairs have a terminal centromere and 13 pairs a non-terminal centromere. Identification of individual chromosomes is enhanced by the use of band-staining techniques which allow recognition of minor structural rearrangements of chromatin material. Seven previously reported cases of male pseudohermaphroditism with chromosome studies are reviewed. Three were genetic females, one was basically an XX/XY chimaera, one was an XX/XXY chimaera, and one was an XXXY. Also reviewed is an infertile mare with 63 chromosomes and no distinguishable sex

Left internal carotid artery agenesis with trans-sellar collateral and a right aortic arch: Case report.

We present a case of unilateral internal carotid artery (ICA) agenesis with trans-sellar anastomosis associated with a right aortic arch, and discuss the embryonic development and clinical implications of this condition. A 26-year-old woman without significant medical history was referred to our hospital complaining of tinnitus. She denied any other symptoms, and examination did not reveal any other neurological deficits. Radiological examinations showed agenesis of the right ICA with trans-sellar anastomosis, and a right aortic arch. This vascular configuration reflects the anatomical homolog

[Anatomy of the liver. What does the radiologist need to know?].

The liver is the largest unpaired parenchymatous organ in the human body and takes part in almost all important metabolic processes. Many patients show alterations of the whole organ due to vascular/cardiac disorders, metabolic or infectious diseases (congestion, fatty liver disease, fibrosis and cirrhosis). However the liver is also a common site for metastatic lesions of malignant tumors. Additionally, benign focal lesions, such as hemangioma or focal nodular hyperplasia (FNH) occur quite frequently. To describe and diagnose these lesions in terms of dignity and location, knowledge of the ma

[Mesonephric adenocarcinoma of the bladder].

The authors report on the third case in the world of mesonephric adenocarcinoma of the bladder. The tumour which may well derive from embryonic rests of mesonephric origin, developed in the region of the bladder neck and proximal urethra. The symptoms that led to the consultation consisted of dysuria with episodes of complete urinary retention. Clinical examination and IVU confirmed the presence of a tumour which could be seen on endoscopy, thus enabling biopsy. As there were no metastases, irradiation with telecobalt cured this patient. The undesired side-effect was the development of bladder

New bacterial agent of pneumonia isolated from renal-transplant recipients.

A gram-negative, weakly acid-fast bacillus has been isolated in embryonated eggs and in guineapigs from lung tissue of two renal-transplant recipients with acute purulent pneumonia. Culture of infected lung tissue and subculture of the egg isolate on artificial media, including media for legionnaires' disease bacterium (Legionella pneumophila), failed to produce growth. Ultrastructural analysis showed that the organism is a prokaryote with a cell-wall structure typical of a gram-negative bacillus but different from that of L. pneumophila. In both patients serum antibody to both isolates develo

Hedgehog Signaling in Tongue Muscle Development: A Comparative Perspective With Limb Myogenesis.

The mammalian tongue is an intricate skeletal muscle organ. From its initial formation to maturation, tongue muscle development involves precisely coordinated processes during embryonic and fetal phases of myogenesis. Extensive research on the regulatory pathways involved in tongue epithelial taste organ development has shown that the Hedgehog (HH) signaling pathway is vital to the formation and epithelial patterning of the tongue and taste organs. Emerging evidence also points to its involvement in the initial formation and spatial patterning of the tongue muscle. HH signaling is a well-estab

Retroperitoneal cysts with congential, ipsilateral renal agenesis of left kidney: a case report.

INTRODUCTION AND IMPORTANCE: Unilateral renal agenesis is defined as the congenital absence of renal tissue of one side that results from a lack of induction of the metanephric blastema by the ureteral bud. Primary retroperitoneal cysts lying in the retroperitoneal fatty tissue have no apparent connections with any anatomical structure except by loose areolar tissue and are rare. They remain asymptomatic and are diagnosed incidentally. CASE PRESENTATION: A 28-year female presented with complain of epigastric pain for 2 years. UVB radiation exposure modulates mitophagy in embryonic cells of freshwater prawn Macrobrachium olfersii: Exploring a protective organelle quality control mechanism.

Aquatic environments are subject to ultraviolet B (UVB) radiation incidence, and its effects on organisms are dose-dependent. Besides DNA, mitochondria are an important target of this radiation that causes structural damage and impairs its functional dynamics. Here, we hypothesize that mitophagy acts as an organelle quality control mechanism to mitigate UVB impacts in embryonic cells. Then, freshwater prawn Macrobrachium olfersii embryos was used as a model to investigate the effects of UVB on genes (Tomm20, Opa1, Pink, Prkn, Sqstm1, and Map1lc3) and proteins (TOM20, PINK1, p62 and LC3B) invol

The omphalomesenteric duct fibrous band as a rare cause of bowel obstruction: A case report and literature review.

INTRODUCTION AND IMPORTANCE: The omphalomesenteric duct (OMD) is an embryonic structure that normally undergoes obliteration during embryonic development, typically not persisting after birth. Failure of complete or partial obliteration can result in a type of malformation known as OMD remnant. CASE PRESENTATION: We report a case of a 24-year-old male patient diagnosed with bowel obstruction. Abdominal computed tomography (CT) scan revealed the presence of an adhesion band. During surgery, a fibrous band connecting from the umbilicus to the mesentery of terminal ileum was found and resected.

Novel MIP gene mutation causes autosomal-dominant congenital cataract.

AIM: To identify disease-causative mutations in families with congenital cataract. METHODS: Two Chinese families with autosomal-dominant congenital cataract (ADCC) were recruited and underwent comprehensive eye examinations. Gene panel next-generation sequencing of common pathogenic genes of congenital cataract was performed in the proband of each family. Sanger sequencing was used to valid the candidate gene mutations and sequence the other family members for co-segregation analysis. The effect of sequence changes on protein structure and function was predicted through bioinformatics analysi

The circRNA Landscape in Recurrent Pregnacy Loss (RPL): A Comparison of Four Reproductive Tissues.

Recurrent Pregnancy Loss (RPL), also named Recurrent Spontaneous Abortion (RSA), is a common fertility problem that refers to at least two consecutive pregnancy losses and affects 1-2% of couples all over the world. Despite common causes such as genetic abnormalities, uterine anomalies or hormonal and metabolic disorders, there is still a huge challenge in identifying the causes of about 40-60% of RPL patients. Circular RNAs (circRNAs) are endogenous ncRNAs with a unique closed-loop and single-stranded structure. Accumulated evidence indicates the role of circRNAs in embryonic development and

A Spatiotemporal Compartmentalization of Glucose Metabolism Guides Mammalian Gastrulation Progression.

Gastrulation is considered the sine qua non of embryogenesis, establishing a multidimensional structure and the spatial coordinates upon which all later developmental events transpire. At this time, the embryo adopts a heavy reliance on glucose metabolism to support rapidly accelerating changes in morphology, proliferation, and differentiation. However, it is currently unknown how this conserved metabolic shift maps onto the three-dimensional landscape of the growing embryo and whether it is spatially linked to the orchestrated cellular and molecular processes necessary for gastrulation. Here

SEPHS1: Its evolution, function and roles in development and diseases.

Selenophosphate synthetase (SEPHS) was originally discovered in prokaryotes as an enzyme that catalyzes selenophosphate synthesis using inorganic selenium and ATP as substrates. However, in contrast to prokaryotes, two paralogs, SEPHS1 and SEPHS2, occur in many eukaryotes. Prokaryotic SEPHS, also known as SelD, contains either cysteine (Cys) or selenocysteine (Sec) in the catalytic domain. In eukaryotes, only SEPHS2 carries out selenophosphate synthesis and contains Sec at the active site. However, SEPHS1 contains amino acids other than Sec or Cys at the catalytic position.
