Postharvest drying affects sorghum seed color, but the physiological and metabolic features associated with color retention remain unclear. We compared natural sun-drying (Dn), 40 °C hot-air drying (D40), and 60 °C hot-air drying (D60) using colorimetric, physiological, biochemical, and untargeted metabolomic analyses. D40 showed the best color retention, whereas Dn caused the greatest deterioration and D60 achieved the fastest dehydration. At the final moisture content, D40 had 38.42% lower malondialdehyde (MDA) content than Dn, while superoxide dismutase, peroxidase, and catalase activities

Aging is associated with the accumulation of oxidative damage. Isoliquiritigenin (ISL), a natural flavonoid, exhibits antioxidant properties, but its effect on lifespan and the underlying mechanisms remains incompletely understood. This study aimed to investigate whether ISL extends lifespan in Caenorhabditis elegans through the transcription factor DAF-16 and the aquaporin AQP-2. We employed lifespan assays, stress resistance tests, intracellular ROS measurement, osmotic water permeability assays in Xenopus oocytes, genetic manipulation (using mutants and RNAi), quantitative real-time PCR, an

Rheumatoid arthritis (RA) is frequently complicated by chronic pain, anxiety and depression, yet the neuroimmune mechanisms linking peripheral inflammation to central affective dysfunction remain elusive. Here we demonstrate that the natural flavonoid dihydroquercetin (DHQ) blocks Trim14 and relieves pain-depression comorbidity in RA. Serum Trim14 is elevated in RA patients and correlates with clinical symptom severity. We identify Trim14 as a contributing factor of JAK1/STAT3 signaling in spinal dorsal horn neurons, promoting both nociceptive sensitization and affective symptoms in collagen-i

Safflower, a traditional Chinese medicine, is renowned for its efficacy in promoting blood circulation and alleviating blood stasis. Its principal bioactive components are flavonoids, which predominantly exist as flavonoid glycosides. Glycosyltransferases, as downstream post-modification enzymes in the biosynthesis of these active glycosides, are of considerable research interest. This study leverages transcriptome data from safflower to identify a glycosyltransferase gene, UGT95A2, which was subjected to comprehensive bioinformatics and enzymatic property analyses. In vitro enzymatic assays d…

Environmental stress triggers a dual response in medicinal plants by inhibiting growth while enhancing secondary metabolite production. However, the regulatory mechanism of this trade-off remains unclear. Here, we identified SbBZR1 as a brassinosteroid (BR)-responsive transcription factor that orchestrates drought resilience and baicalin biosynthesis in Scutellaria baicalensis through integrated multi-omics and metabolic flux analysis. In general, drought stress upregulated BR signaling and SbBZR1 expression. Transgenic Arabidopsis expressing SbBZR1 and its hyperactive mutant (Sbbzr1-1D) exhib

Growing evidence indicates that healthy diets are associated with a slower progression of Alzheimer's disease (AD). Flavonoids are among the most abundant natural products in diets beneficial to AD, such as the Mediterranean diet. However, the effect and mechanism of these dietary flavonoids on AD remains incompletely understood. Here, we found that a representative dietary natural flavonoid, chrysin (Chr), significantly ameliorated cognitive impairment and AD pathology in APP/PS1 mice. Furthermore, mechanistic studies showed that Chr significantly reduced the levels of amyloid-β (Aβ) and phos…

Elevated levels of reactive oxygen species (ROS) are caused by ultraviolet B radiation (UV-B) stress. In response, plants strengthen their cell membranes, impeding photosynthesis. Additionally, UV-B stress initiates oxidative stress within the antioxidant defense system and alters secondary metabolism, particularly by increasing the quantity of UV-absorbing compounds such as flavonoids. The v-myb avian myeloblastosis viral oncogene homolog (MYB) transcription factor (TF) may participate in a plant's response to UV-B damage through its regulation of flavonoid biosynthesis. In this study, we dis…

Baicalin (BA), a natural component found in many traditional Chinese medicines, exerts protective effects against several viruses. Although our previous studies have revealed that the anti-hepatitis B virus (anti-HBV) activity of BA depends on hepatocyte nuclear factor (HNF) signaling, the specific mechanisms remain unclear. The present study explored the potential signaling mechanisms involved in BA-mediated HBV suppression. Transcriptomic analysis suggested that BA significantly modulates the estrogen receptor (ER) and AMPK signaling pathways in HepG2 cells. The ER alpha (ERα) binding affini
