Biotrophic rust fungi secrete effector proteins through haustoria to promote successful colonization. Consequently, the functional analysis of haustorial effector proteins has become central to understanding rust pathogenicity. In previous work, we generated the haustorial transcriptome of Gymnosporangium yamadae, the causal agent of apple rust disease, and predicted numerous candidate effector genes. Here, we systematically characterized the functions of 13 haustorially expressed candidate effectors of G. yamadae using a heterologous system to assess their modulation of plant immunity and sub

Plants acquire inorganic phosphate (Pi) either directly through their roots or through symbiosis with arbuscular mycorrhizal (AM) fungi, and the direct uptake pathway is downregulated when the symbiosis establishes. As atmospheric CO2 concentrations rise, it is critical to understand how increased carbon availability alters plant nutrient acquisition strategies, with implications for crop productivity and carbon sequestration alike. Here, we investigated the interaction between elevated CO2, soil Pi availability, and AM symbiosis in the major cereal crop rice. Elevated CO2 enhanced mycorrhizal

Optimal plant height is critical for yield performance and lodging resistance in rice (Oryza sativa L.). However, genetic resources available for optimizing yield and lodging resistance in breeding programs are currently limited. Here, we report the identification of semi-dwarf rice 1 (SDR1), a semi-dwarf gene encoding a RING-H2 type E3 ubiquitin ligase preferentially expressed in stem internodes. Mechanistically, SDR1 directly targets the key gibberellin (GA)-deactivating enzyme ELONGATED UPPERMOST INTERNODE 1 (EUI1) for degradation via the ubiquitin-proteasome system (UPS), thereby fine-tuni

Newly differentiated leaves of albino ornamental kale turn white as exposed to low temperatures, producing a distinctive and commercially desirable unique color pattern. Here, we show that this low-temperature-induced albinism is caused by a mutation in the kale F-box gene BoWl. Using yeast two-hybrid, pull-down, co-immunoprecipitation, and luciferase complementation assays, we demonstrate that BoWl interacts with the transcription factor BoARF1 to form a functional complex. Complementary yeast one-hybrid, electrophoretic mobility shift, chromatin immunoprecipitation, and luciferase assays rev

Dwarfing rootstocks are crucial for high-density and labor-efficient citrus cultivation, yet the molecular mechanisms underlying plant height regulation remain poorly understood. Using an integrated multiomics analysis of dwarf and normal-height red tangerine (Citrus reticulata) × trifoliate orange (Poncirus trifoliata) hybrid seedlings, we identified the Gretchen Hagen 3.1 gene PtGH3.1, an auxin-conjugating enzyme, as an important regulator of dwarfism. Compared with normal-height hybrid seedlings, PtGH3.1 was significantly upregulated in the shoot tips of dwarf hybrid seedlings, correlating

Purple flower coloration in Michelia crassipes represents a rare pigmentation trait within the genus Michelia. Therefore, investigating the regulatory mechanisms of purple flowers in M. crassipes holds significant breeding value. Here, MYB52, a novel R2R3-MYB transcription factor in the M. crassipes flower, was isolated and functionally characterized. Heterologous expression of McMYB52 in tobacco triggered dark-red petal, which was driven by anthocyanin, flavonoid, and lignin accumulation. Histological comparison confirmed that the morphology of transgenic tobacco petal cells changed from fl

Drought stress can damage crop growth and lead to a decline in yield, thereby affecting food security, especially in regions vulnerable to climate change. SNAC1 (stress-responsive NAC1), the NAC transcription factor family member, plays a crucial role in stomatal movement regulation. Effective regulation of stomatal movement is essential for protecting plants from water loss during adverse conditions. Our hypothesis revolves around altering HvSNAC1 activity by introducing a point mutation in its encoding gene, thereby influencing stomatal dynamics in barley. Two TILLING mutants, each harboring

BACKGROUND: Pinus sylvestris var. mongolica is a naturally susceptible host for the pine wood nematodes (PWNs, Bursaphelenchus xylophilus) and a dominant afforestation tree species in northeastern China. With the continuous northward expansion of PWNs, P. sylvestris var. mongolica may become a critical conduit for their further expansion. To prevent pine wilt disease (PWD) from spreading via P. sylvestris var. mongolica in northeastern China, it is essential to elucidate its defense mechanisms against PWN and enhance its resistance.
RESULTS: Transcriptome analysis of P. sylvestris var. mongol

Background: The Arabidopsis FCS-LIKE ZINC FINGER (FLZ) family proteins play crucial roles in responses to various biotic and abiotic stresses, but the functions of many family members remain uncharacterized. Methods: In this study, we investigated the function of FLZ12, a member of the FLZ family, using a reverse genetic approach. Results: We found that overexpression of FLZ12 impaired root hair development, as evidenced by marked reductions in both root hair length and number under normal growth conditions. However, deprivation of phosphate could partially restore root hair formation, althoug

Lignin biosynthesis is vital for plant growth and development. In model plants, 4-coumarate: coenzyme A ligase (4CL) and caffeic acid O-methyltransferase (COMT) are key regulators of lignin content and composition. However, their functions in gymnosperms, especially conifers, remain largely unexplored. Here, we cloned and functionally characterized the orthologs of these genes in Chinese fir (Cunninghamia lanceolata), the major timber species in China. Enzymatic assays confirmed that Cl4CL catalyzes the conversion of caffeic and p-coumaric acids into CoA esters, while ClCOMT methylates caffeic

In crop plants, there is often a trade-off between growth and defense; modulating the balance is crucial for effective crop improvement. In rice (Oryza sativa), the DENSE AND ERECT PANICLE 2 (DEP2) mutant shows altered phenotypes such as a more vertical leaf angle, erect panicles, small and round grains, slightly lower thousand-grain weight, and dwarfism. Here, we determined that loss of DEP2 function enhances resistance to the rice blast fungus Magnaporthe oryzae. We established that DEP2 interacts with the E3 ubiquitin ligase OsRING80, which ubiquitinates DEP2 and promotes its degradation vi

Plant viral diseases pose a significant threat to agricultural production, and the availability of effective drugs against viral diseases remains limited. In this study, we discovered that betulinic acid (BA), a pentacyclic triterpenoid derived from plants, delays infection by turnip mosaic virus (TuMV) in Nicotiana benthamiana. Transcriptomic analysis revealed that BA treatment specifically induced the expression of N. benthamiana phytosulfokine 3 (NbPSK3), a plant pentapeptide hormone with diverse functions, while TuMV infection suppressed its expression. Further study demonstrated that NbPS

Little is known about nitric oxide (NO)-mediated cotton plants' response to copper (Cu) stress and the underlying tolerance mechanism. It was hypothesized that NO can alleviate Cu toxicity to cotton roots by regulating the root cell wall composition and the transcription of Cu ion transporting-related genes. Cu stress significantly increased NO synthase (EC 1.14.14.47) activity, leading to elevated endogenous NO content. Cu excess-induced growth inhibition was reversed by sodium nitroprusside (SNP, NO donor) application but exacerbated by cPTIO (NO scavenger) addition. The SNP + Cu treatment p

Orchid mycorrhizal (OM) fungi form mutualistic relationships with orchids, facilitating the uptake of minerals from the soil in exchange for sugars and lipids provided by the plant. In this study, we explored the role of the DoSWEET14 sugar transporter in Dendrobium officinale during OM symbiosis. Transcriptome sequencing revealed significant changes in gene expression in mycorrhizal roots, particularly the upregulation of DoSWEET14. Subcellular localization studies confirmed that DoSWEET14 is localized to the plasma membrane, suggesting its involvement in nutrient exchange between the orchid

Wheat (Triticum aestivum) is a major staple crop worldwide, and its yields are significantly threatened by wheat powdery mildew (Blumeria graminis f. sp. tritici). Enhancing disease resistance in wheat is crucial for meeting global food demand. This study investigated the disease response in wheat, focusing on the bioactive small molecules salicylic acid (SA), pipecolic acid (Pip), and N-hydroxypipecolic acid (NHP), to provide new insights for molecular breeding. We found that endogenous levels of SA, Pip, and NHP significantly increased in infected plants, with Pip and NHP levels rising earli

Most land plants alternate between generations of sexual gametophytes and asexual sporophytes. Unlike seed plants, fern gametophytes are free living and grow independently of their sporophytes. In homosporous ferns such as Ceratopteris, gametophytes derived from genetically identical spores exhibit sexual dimorphism, developing as either males or hermaphrodites. Males lack meristems and promote cell differentiation into sperm-producing antheridia. In contrast, hermaphrodites initiate multicellular meristems that stay undifferentiated, sustain cell division and prothallus expansion, and drive t

Cadmium (Cd) is a toxic heavy metal that poses a serious threat to crop safety and human health. Aspartate aminotransferase (AspAT) is a prime enzyme engaged in amino acid metabolism, contributing essential metabolic substances for plant growth and acclimatization to various stresses. In this study, we identified a novel AspAT9 gene with high responsiveness to Cd stress from poplar 'Nanlin895' and subsequently transformed it into Arabidopsis. The resulting transgenic AspAT9-1 plants (designated A9) and wild-type (WT) Arabidopsis were subjected to Cd treatment (T) or maintained under control co
