Development of redox-labeled electrochemical immunoassay for polycyclic aromatic hydrocarbons with controlled surface modification and catalytic voltammetric detection.

A redox-labeled direct competitive electrochemical immunoassay for polycyclic aromatic hydrocarbons (PAHs) was developed. A ruthenium tris(bipyridine)-pyrenebutyric acid conjugate was synthesized as the redox-labeled tracer. Its recognition by an anti-PAH monoclonal antibody was confirmed by surface plasmon resonance. In the immunoassay, the antibody was immobilized on (3-glycidoxypropyl)-trimethoxysilane (GPTMS)-modified indium tin oxide (ITO) electrodes. The assay was quantified by measuring the electro-catalytic current of the redox label in an oxalate-containing electrolyte which served as

Role of costimulatory molecules in immune response of patients with cutaneous leishmaniasis.

T cell-mediated immunity is critical in resistance against Leishmania parasites, and T cell activation requires signals provided by costimulatory molecules. Herein we evaluated the role of costimulatory molecules on cytokine production and T cell surface molecule expression by peripheral blood mononuclear cells (PBMC) from cutaneous leishmaniasis (CL) patients. PBMC from CL patients were stimulated with soluble Leishmania antigen (SLA, 10 microg/ml), in the presence or absence of soluble CTLA4-Ig to block CD28-B7 interaction or in the presence or absence of anti-human CD40L to block CD40-CD40L

Assessment of the diagnostic value of 99mTc-radiolabelled specific antibodies in experimental hydatidosis.

We assessed the potential of 99mTc labelled specific polyclonal antibodies (99mTc-PoAb) for the diagnosis of hydatid disease by immunoscintigraphy. Experimentally infected mice and rabbits were used for this purpose. A specific rabbit antibody recognizing total somatic antigen from hydatid membranes (HCMA) was obtained. PoAb biological activity before labelling was checked according to Barbieri et al. 99mTc-PoAb labelling was performed according to Thakur et al.; the radiochemical purity was higher than 90%. The following studies of 99mTc-PoAb were made: post-labelling biological activity; in

Genetic markers in diagnosis and prediction of relapse in Graves' disease.

Hyperthyroidism of Graves' disease takes an unpredictable clinical course in the long-term follow-up. Whereas roughly 30-60% of patients relapse after their first antithyroid drug treatment, the likelihood of remission in the remaining group can not be foreseen. We have analysed-retrospectively-patients with Graves' disease that had been on antithyroid drug treatment for one year and were followed up thereafter. Patients were investigated for a variety of clinical parameters like ophthalmopathy status and relapse or remission as well as gene polymorphisms of the HLA and other regions. Of the 2

Novel L1CAM-antibody drug conjugates exert potent anti-tumor activity in preclinical models of even advanced pancreatic cancer.

Pancreatic ductal adenocarcinoma (PDAC) is a lethal cancer with poor 5-year overall survival. This can be mainly explained by late diagnosis mostly at advanced stages, excluding primary tumor resection as putative curative treatment, and a profound therapy resistance. L1 cell adhesion molecule (L1CAM) is upregulated in PDAC cells and associated with different hallmarks of cancer contributing to tumor progression and therapy resistance. Using L1CAM as tumor associated antigen, two antibody drug conjugates (ADC) targeting L1CAM were developed with different toxins as payload. Both L1CAM-ADC led

Human germline-like monoclonal antibody against 5T4 enables potent ADC and CAR-T therapies for solid tumors.

5T4 is an oncofetal antigen overexpressed in a wide range of solid tumors with minimal presence in normal adult tissues, highlighting its promise as a therapeutic target. In this study, we identified germline-like human monoclonal antibodies targeting human 5T4 with high affinity, among which antibody m603 exhibits superior cell binding activity to various cancer cells including breast, pancreatic, ovarian, lung and liver cancer cell lines. Subsequently, we constructed antibody-drug conjugates (ADCs) and chimeric antigen receptor (CAR)-T cell based on m603. By conjugating the antibody with cyt

Concomitant radiation therapy and trastuzumab deruxtecan in metastatic breast cancer: feasibility, safety, and outcomes from a real-life multicentre international cohort.

BACKGROUND AND PURPOSE: Radiation therapy (RT) remains a cornerstone of management for metastatic breast cancer (mBC), though systemic therapy drives overall disease control. Administering RT concomitantly with systemic treatment may raise concerns regarding toxicity. This multicentre retrospective study evaluated the safety of trastuzumab deruxtecan (T-DXd), an antibody-drug conjugate, when delivered alongside RT. MATERIALS AND METHODS: Data from six European centres were analysed. Concomitancy was defined as RT administered during T-DXd or within 10 days of a treatment cycle. The primary en

Tucatinib in Combination With Trastuzumab Deruxtecan in Patients With Previously Treated HER2+ Unresectable Locally Advanced or Metastatic Breast Cancer: An Open-Label Phase 2 Study.

INTRODUCTION: Recent advances in HER2-directed therapies have improved outcomes for patients with HER2+ advanced/metastatic breast cancer (a/mBC), but disease progression ultimately occurs in most cases. Dual targeting of HER2 with tyrosine kinase inhibitors and antibody-drug conjugates has the potential for non-cross-resistant treatments that improve disease control. METHODS: HER2CLIMB-04, a single-arm, open-label, phase 2 study, evaluated tucatinib plus trastuzumab deruxtecan (T-DXd) in patients with HER2+ a/mBC who experienced disease progression on or were intolerant of previous HER2-dire

Beyond R-CHOP: The rise of antibody-drug conjugates in DLBCL.

Diffuse large B-cell lymphoma (DLBCL) remains a therapeutic challenge, as a substantial proportion of patients experience relapsed or refractory (R/R) disease. Antibody-drug conjugates (ADCs) have emerged as a transformative class of targeted therapy, designed to deliver potent cytotoxic payloads directly to malignant cells via specific surface antigens. This approach enhances antitumor efficacy while minimizing toxicity. Recently, ADCs have expanded the DLBCL therapeutic landscape, with the approvals of CD79b-targeted polatuzumab vedotin and CD19-directed loncastuximab tesirine for R/R and ev

Zongertinib, a Novel HER2 Tyrosine Kinase Inhibitor, Maintains an Anticancer Activity for Trastuzumab Deruxtecan-Resistant Cancers Harboring HER2-Overexpression.

Trastuzumab deruxtecan (T-DXd), an antibody-drug conjugate comprising trastuzumab linked to a payload DXd, has been extensively used to treat various cancers harboring HER2 overexpression. However, resistance development has been a major challenge to T-DXd treatment. To explore treatment strategies for T-DXd-resistant cancers, we examined two T-DXd-resistant cells named DSR32 and DSR4, which were obtained from HER2 gene-amplified H2170 lung cancer and N87 gastric cancer cells, respectively. The uptake of T-DXd and its transport to lysosomes in DSR32 cells were reduced. Subsequently, HER2 gene
