A new mechanism regulating microglial NLRP3 inflammasome: FMR1 mediates NLRP3 mRNA stability.

BACKGROUND: The NLRP3 inflammasome drives chronic inflammation and contributes to the pathogenesis of multiple sclerosis (MS). This study aimed to elucidate a novel post-transcriptional regulatory mechanism controlling NLRP3 expression in microglia under lipopolysaccharide (LPS) stimulation. METHODS: The experimental autoimmune encephalomyelitis (EAE) mouse model of MS was established and divided into four groups: Sham, EAE, EAE + Lv-con (control lentivirus), and EAE + Lv-FMR1 (FMR1-overexpressing lentivirus). BV2 microglial cells were stimulated with LPS and adenosine triphosphate (ATP). mRN

Curcumin suppresses NLRP3 inflammasome activation by inducing autophagy to alleviate neuropathic pain in rats.

BACKGROUND: Neuropathic pain profoundly affects the quality of life of patients and requires considerable medical resources. Extensive evidence indicates that neuroinflammation within the spinal cord plays a critical role in modulating neuropathic pain. Curcumin, renowned for its anti-inflammatory properties, has shown potential in alleviating neuropathic pain. However, the mechanisms underlying its effectiveness remains elusive. The aim of this study is to investigate whether curcumin mitigates neuropathic pain by suppressing NLRP3 inflammasome activation through the induction of autophagy.

NEK7 is an essential regulator in NLRP3 inflammasome assembly of common carp (Cyprinus carpio L.).

The NIMA-related kinase 7 (NEK7), a member of the Never in Mitosis Gene A (NIMA) kinase family, participates in the assembly of the NLRP3 inflammasome in mammalian. However, it is currently unclear that the functions of NEK7 in the activation and assembly of NLRP3 inflammasome in teleost. In this research, the cDNA sequence of NEK7 of common carp (CcNEK7) was cloned and its role in the assembly of CcNLRP3 inflammasome was investigated. CcNEK7 was conserved throughout evolution, with its amino acid sequence, three-dimensional structure, and subcellular localization being similar to those in mam

Adipose tissue NLRP3 inflammasome: a therapeutic target in obesity and type 2 diabetes mellitus.

Adipose tissue has been identified as a crucial player in the release of proinflammatory mediators, which can result in a chronic inflammatory state, predisposing individuals to obesity, insulin resistance (IR), and type 2 diabetes mellitus (T2DM). A major approach in tackling these diseases involves targeting molecular sensors that can respond to nutritional cues and metabolic status that trigger the early phases of inflammatory cascades. Among these sensors is the nucleotide oligomerization domain (NOD)-, leucine-rich repeat-, and pyrin domain-containing protein 3 (NLRP3) inflammasome. Thera

α-amanitin induces hepatotoxicity via PPAR-γ inhibition and NLRP3 inflammasome activation.

Mushroom poisoning, predominantly caused by α-amanitin, is a critical food safety concern in worldwide, with severe cases leading to hepatotoxicity and fatalities. This study delves into the hepatotoxic effects of α-amanitin, focusing on the NLRP3 inflammasome and PPAR-γ's regulatory role in inflammation. In vitro studies with L-02 cells showed that α-amanitin reduces cell viability and triggers NLRP3 inflammasome activation, increasing NF-κB phosphorylation and pro-inflammatory cytokines IL-18 and IL-1β. The NLRP3 inhibitor MCC950 mitigated these effects without impacting NF-κB. Conversely, P

Fungal Als proteins hijack host death effector domains to promote inflammasome signaling.

High-damaging Candida albicans strains tend to form hyphae and exacerbate intestinal inflammation in ulcerative colitis patients through IL-1β-dependent mechanisms. Fungal agglutinin-like sequence (Als) proteins worsen DSS-induced colitis in mouse models. FADD and caspase-8 are important regulators of gut homeostasis and inflammation. However, whether they link directly to fungal proteins is not fully understood. Here, we report that Als proteins induce IL-1β release in immune cells. We show that hyphal Als3 is internalized in macrophages and interacts with caspase-8 and the inflammasome adapt

Targeting the NLRP3 inflammasome signalling in acute myeloid leukemia: Mechanisms, therapeutics, and future directions.

Acute myeloid leukemia (AML) is one of the most frequent hematological malignancies. It is a complex and aggressive disease of undifferentiated hematopoietic progenitor cells, with high rates of relapse and drug resistance, posing significant clinical challenges. Emerging evidence underscores the critical role of inflammation in AML progression, with inflammasomes-key regulators of the inflammatory response-emerging as pivotal players in disease pathogenesis. Among these, the NOD-, LRR-, and pyrin domain-containing 3 (NLRP3) inflammasome has gained particular attention due to its involvement i…

Nrf2-Keap1 Pathway and NLRP3 Inflammasome in Parkinson's Disease: Mechanistic Crosstalk and Therapeutic Implications.

Neurodegenerative disorders such as Parkinson's disease (PD) are characterized by the progressive degeneration of dopaminergic neurons, which is driven primarily by oxidative stress and chronic neuroinflammation. Central to the cellular antioxidant defense system is the nuclear factor erythroid 2-related factor 2 (Nrf2)-Kelch-like ECH-associated protein 1 (Keap1) pathway, which mitigates oxidative damage and preserves mitochondrial integrity. Concurrently, the NOD-like receptor pyrin domain-containing protein 3 (NLRP3) inflammasome acts as a key mediator of innate immune responses and has been

Microglial NFAT5 aggravates neuroinflammation via mediating NLRP6 inflammasome in experimental ischemic stroke.

Microglial activation triggers the inflammatory cascade and exacerbates brain injury following ischemic stroke. Middle cerebral artery occlusion (MCAO) modeling increased the expression of nuclear factor of activated T cells 5 (NFAT5) in microglia. However, the role of microglial NFAT5 in ischemic stroke remains unclear. Here, our findings indicated that microglial NFAT5 knockdown reduced the expression of pro-inflammatory factors, microglial activation, and neutrophil infiltration, ultimately ameliorating cerebral infarction and neurological deficits in mice following MCAO. Additionally, we t

The ABC transporter A7 modulates neuroinflammation via NLRP3 inflammasome in Alzheimer's disease mice.

BACKGROUND: Specific genetic variants in the ATP-binding cassette transporter A7 locus (ABCA7) are associated with an increased risk of Alzheimer's disease (AD). ABCA7 transports lipids from/across cell membranes, regulates Aβ peptide processing and clearance, and modulates microglial and T-cell functions to maintain immune homeostasis in the brain. During AD pathogenesis, neuroinflammation is one of the key mechanisms involved. Therefore, we wanted to investigate the specific role of ABCA7 in microglial activation via the NLRP3 inflammasome. METHODS: We developed the first humanized, Cre-ind

Broxyquinoline targets NLRP3 to inhibit inflammasome activation and alleviate NLRP3-associated inflammatory diseases.

The NLR family pyrin domain-containing 3 (NLRP3) inflammasome is responsible for various pathogenic and non-pathogenic damage signals and plays a critical role in host defense against pathogens and physiological damage. However, inflammasome activation and its subsequent effects also lead to a variety of inflammatory diseases. In this study, we identified broxyquinoline, an FDA-approved antimicrobial drug, as a effective NLRP3 inflammasome inhibitor. Broxyquinoline suppressed NLRP3 inflammasome-dependent interleukin-1β (IL-1β) release, but did not affect NLRC4 or AIM2 inflammasome activation.

NLRP3 inflammasome constrains liver regeneration through impairing MerTK-mediated macrophage efferocytosis.

The NOD-like receptor protein 3 (NLRP3) inflammasome plays a crucial role in human acute and chronic liver diseases. However, the role and cell-specific contribution of NLRP3 in liver regeneration remains unclear. Here, we found that NLRP3 was highly activated during the early stage of liver regeneration via 70% partial hepatectomy (PHx) mice model and clinical data. Global NLRP3 depletion or pharmacologically blocking NLRP3 significantly enhanced liver regeneration, while NLRP3 overexpression impaired it after PHx. Furthermore, mice with myeloid-specific knockout of Nlrp3 (Nlrp3Δmye), rather
