Insecticidal activity and the mechanism of action of three phenylpropanoids isolated from the roots of Piper sarmentosum Roxb.

Hexane, dichloromethane and methanol extracts of the roots of Piper sarmentosum Roxb. were screened for toxicity towards Sitophilus oryzae (L.), Rhyzopertha dominica (F.), and Plodia interpunctella (Hübner) and the hexane extract exhibited the highest mortality percentage. Bioassay-guided fractionation of the hexane extract resulted in the isolation of asaricin 1, isoasarone 2, and trans-asarone 3. Asaricin 1 and isoasarone 2 were the most toxic compounds to Sitophilus oryzae, Rhyzopertha dominica, and Plodia interpunctella. Sitophilus oryzae and Rhyzopertha dominica exposed to asaricin 1 and

Sublethal effects of the insecticidal fusion protein ω-ACTX-Hv1a/GNA on the parasitoid Eulophus pennicornis via its host Lacanobia oleracea.

BACKGROUND: The neurotoxin peptide ω-ACTX-Hv1a, fused to the carrier molecule GNA, presents potential for insect control as a biopesticide, being orally toxic to insect pests from different orders. However, thorough evaluation is required to assure its safety towards non-target invertebrates. Effects of this novel biopesticide on the parasitoid Eulophus pennicornis via its host Lacanobia oleracea are presented. RESULTS: Hv1a/GNA did not cause mortality when injected or fed to fifth-stage L. oleracea, but caused up to 39% reduction in mean larval weight (P < 0.05) and increased developmental t…

Construction of suppression subtractive hybridization libraries and identification of brown planthopper-induced genes.

A suppression subtractive hybridization technique was used to screen for brown planthopper (BPH)-inducible genes in rice (Oryza sativa). cDNAs from a BPH-resistant rice line (B5) infested by BPH were used as the tester population, and mixed cDNAs from a BPH-sensitive line (MH63) and a control (uninfested B5) as the driver population. After hybridizing and cloning, forward and reverse subtraction cDNA libraries were obtained, containing 5700 clones. These clones were further analyzed by differential gene expression screening, and 154 clones that were clearly induced by BPH were identified. Sequ

Trypanosoma cruzi: comparative studies of infectivity of parasites ingested by Triatoma infestans and those present in their feces.

Artificial feeding of the insects with whole blood containing trypomastigotes resulted in triatome infection; the parasites were present in the host feces after 15--30 days and, when inoculated into mice, elicited parasitemia in 100% of the cases and deaths in 20--70%. This mortality indicates a reduction of the original infective capacity of the bloodstream form, which kills 100% of the mice even when inoculated with a single trypomastigote. When triatomes were fed whole normal mouse blood containing culture forms of low infective capacity (TulL), mice inoculated with feces containing the pro

Efficacy of Biopesticides for Management of the Swede Midge (Diptera: Cecidomyiidae).

Management of the swede midge, Contarinia nasturtii Kieffer, in North American crucifer production relies on crop rotation and the timely application of synthetic insecticides, based on pheromone trap monitoring of local adult populations. Organically acceptable formulations of azadirachtin, pyrethrin, and spinosad, and a commercial biopesticide containing the entomopathogenic fungus, Beauveria bassiana, were evaluated for their effects on larval mortality and oviposition deterrence in the greenhouse, and on damage symptoms in the field. In greenhouse trials, pyrethrin and spinosad treatments

Investigating the Contribution of Peri-domestic Transmission to Risk of Zoonotic Malaria Infection in Humans.

BACKGROUND: In recent years, the primate malaria Plasmodium knowlesi has emerged in human populations throughout South East Asia, with the largest hotspot being in Sabah, Malaysian Borneo. Control efforts are hindered by limited knowledge of where and when people get exposed to mosquito vectors. It is assumed that exposure occurs primarily when people are working in forest areas, but the role of other potential exposure routes (including domestic or peri-domestic transmission) has not been thoroughly investigated. METHODOLOGY/PRINCIPAL FINDINGS: We integrated entomological surveillance within

Two step male release strategy using transgenic mosquito lines to control transmission of vector-borne diseases.

Mosquitoes are responsible for the transmission of pathogens that cause devastating human diseases such as malaria and dengue. The current increase in mean global temperature and changing sea level interfere with precipitation frequency and some other climatic conditions which, in general, influence the rate of development of insects and etiologic agents causing acceleration as the temperature rises. The most common strategy employed to combat target mosquito species is the Integrated Vector Management (IVM), which comprises the use of multiple activities and various approaches to preventing t

Tsetse salivary gland hypertrophy virus: hope or hindrance for tsetse control?

MANY SPECIES OF TSETSE FLIES (DIPTERA: Glossinidae) are infected with a virus that causes salivary gland hypertrophy (SGH), and flies with SGH symptoms have a reduced fecundity and fertility. The prevalence of SGH in wild tsetse populations is usually very low (0.2%-5%), but higher prevalence rates (15.2%) have been observed occasionally. The successful eradication of a Glossina austeni population from Unguja Island (Zanzibar) using an area-wide integrated pest management approach with a sterile insect technique (SIT) component (1994-1997) encouraged several African countries, including Ethiop

Environmental sanitary interventions for preventing active trachoma.

BACKGROUND: Trachoma is a major cause of avoidable blindness. It is responsible for about six million blind people worldwide, mostly in the poor communities of developing countries. One of the major strategies advocated for the control of the disease is the application of various environmental sanitary measures to such communities. OBJECTIVES: To assess the evidence for the effectiveness of environmental sanitary measures on the prevalence of active trachoma in endemic areas. SEARCH STRATEGY: We searched the Cochrane Central Register of Controlled Trials - CENTRAL in The Cochrane Library (Is

Biological control of locusts and grasshoppers.

Control of grasshoppers and locusts has traditionally relied on synthetic insecticides, and for emergency situations this is unlikely to change. However, a growing awareness of the environmental issues associated with acridid control as well as the high costs of emergency control are expanding the demand for biological control. In particular, preventive, integrated control strategies with early interventions will reduce the financial and environmental costs associated with large-scale plague treatments. The recent development of effective oil formulations of Metarhizium anisopliae spores in Af

[Historical study on the moth repellent, "Fujisawa Camphor" (1)].

"Fujisawa Camphor" was introduced for sale as a moth repellent for home use in 1897. As a background that camphor was produced on a commercial basis, the following were studied: (1) Camphor was already used as a moth repellent by some people in the Endo Period. (2) With the Sino-Japanese War as a turning point, armaments were increased and they made military uniforms which were made of wool. It also made Western woolen clothes spread in general. (3) As people were in danger to catch infectious diseases in those days, they had a habit of avoiding bad odors and drifting fragrances like camphor w

An elm EST database for identifying leaf beetle egg-induced defense genes.

BACKGROUND: Plants can defend themselves against herbivorous insects prior to the onset of larval feeding by responding to the eggs laid on their leaves. In the European field elm (Ulmus minor), egg laying by the elm leaf beetle ( Xanthogaleruca luteola) activates the emission of volatiles that attract specialised egg parasitoids, which in turn kill the eggs. Little is known about the transcriptional changes that insect eggs trigger in plants and how such indirect defense mechanisms are orchestrated in the context of other biological processes. RESULTS: Here we present the first large scale s

Targeting sex determination to suppress mosquito populations.

Each year, hundreds of millions of people are infected with arboviruses such as dengue, yellow fever, chikungunya, and Zika, which are all primarily spread by the notorious mosquito Aedes aegypti. Traditional control measures have proven insufficient, necessitating innovations. In response, here we generate a next-generation CRISPR-based precision-guided sterile insect technique (pgSIT) for Ae. aegypti that disrupts genes essential for sex determination and fertility, producing predominantly sterile males that can be deployed at any life stage. Using mathematical models and empirical testing,

Transgenic cotton expressing synthesized scorpion insect toxin AaHIT gene confers enhanced resistance to cotton bollworm (Heliothis armigera) larvae.

A synthetic scorpion Hector Insect Toxin (AaHIT) gene, under the control of a CaMV35S promoter, was cloned into cotton via Agrobacterium tumefaciens-mediated transformation. Southern blot analyses indicated that integration of the transgene varied from one to more than three estimated copies per genome; seven homozygous transgenic lines with one copy of the T-DNA insert were then selected by PCR and Southern blot analysis. AaHIT expression was from 0.02 to 0.43% of total soluble protein determined by western blot. These homozygous transgenic lines killed larvae of cotton bollworm (Heliothis ar

Assessing the combined toxicity of conventional and newer insecticides on the cotton mealybug Phenacoccus solenopsis.

Reduced susceptibility to insecticides often results in failure of insect pest control and repetitive use of broad-spectrum insecticides, which could have detrimental effects on beneficial arthropods and surrounding agro-ecosystems. The cotton mealybug, Phenacoccus solenopsis Tinsley is a pest of worldwide importance that can be effectively controlled using a number of insecticides. This insect has developed resistance due to injudicious use and repeated exposure to insecticides throughout the year. The aim of the present study was to investigate the toxicity of the insecticides chlorpyrifos,

The Association Between Acne Vulgaris, Acne Vulgaris with Nonspecific Facial Dermatitis, and Demodex Mite Presence.

BACKGROUND: Demodex mites can lead to various skin disorders, from non-specific dermatitis to conditions that mimic other diseases, making it challenging to diagnose accurately. Additionally, it has been reported that Demodex mites can cause skin conditions such as perioral dermatitis, pustular folliculitis, pityriasis folliculorum, blepharitis, and rosacea. Due to conflicting studies, there is a debate regarding the link between Demodex mites and acne vulgaris. This study aims to determine the prevalence of Demodex mites on the faces of individuals with acne vulgaris, acne with nonspecific fa
