Coleopteran-active cry gene profiling and insecticidal activity of indigenous Bacillus thuringiensis strains against Holotrichia serrata.

White grub (Holotrichia serrata F.) is a destructive soil-dwelling pest causing severe yield losses in several agricultural and horticultural crops. Owing to its subterranean habit, chemical control is often ineffective, necessitating sustainable biological alternatives. This study investigated the distribution of coleopteran-active cry genes among indigenous Bacillus thuringiensis strains and evaluated their insecticidal activity against H. serrata. A total of 100 indigenous B. thuringiensis strains isolated from the Western Ghats of India were screened using gene-specific PCR for 12 coleopte

Formulation-dependent pulmonary disposition and secondary hepatic responses of the household insecticide hydramethylnon following respiratory tract delivery.

Hydramethylnon (HM) is a household-use insecticide whose inhalation hazard remains poorly characterized. We combined whole-body disposition analysis with mechanistic assessment in pulmonary and hepatic systems. Structurally identical 18F-labeled HM enabled dynamic positron emission tomography/computed tomography after intratracheal instillation in mice. Pulmonary clearance was rapid, with predominant redistribution to the liver and a fourfold longer half-life for the polyethylene glycol (PEG)-based than the ethanol-based dispersion (26.92 versus 6.59 min). In an air-liquid interface model, HM

Evaluation of five systemic insecticide baits to control fleas (Siphonaptera: Pulicidae and Ceratophyllidae) on rodents (Rodentia: Muridae and Sciuridae).

We evaluated the efficacy of 5 systemic insecticide compounds, each incorporated into edible baits to control flea (Siphonaptera) vectors of the plague bacterium Yersinia pestis (Lehmann and Neumann), with brown rats (Rattus norvegicus, Berkenhout) and rock squirrels (Otospermophilus variegatus, Erxleben) in a laboratory environment. Rodents were exposed to diets lacking or containing insecticides for 24 h at which point adult Ctenocephalides felis (Bouché), Xenopsylla cheopis (Rothschild), or Oropsylla montana (Baker) were applied to rodents. Fleas were monitored for survival 1 d later and su…

Assessment of Long-Lasting Insecticidal Net (LLIN) ownership, utilization, and associated barriers in malaria-endemic communities of Ethiopia.

BACKGROUND: Malaria remains a critical global health challenge, with over 68% of Ethiopia's population living in at-risk areas. While Long-Lasting Insecticidal Nets (LLINs) are a cornerstone of prevention, their effectiveness depends on consistent use. This study aimed to assess LLIN ownership and utilization patterns and identify socio-behavioral and physical determinants of their effectiveness in endemic communities. METHODS: A community-based, cross-sectional survey was conducted from October 2024 to January 2025 across 11 administrative regions in Ethiopia. Using a two-stage stratified cl

Insecticide-treated nets utilization and determinants in Southeast Asia toward malaria elimination: a systematic review and meta-analysis.

Malaria remains a major public health burden in Southeast Asia, where elimination depends on consistent community-level use of insecticide-treated nets (ITNs) and residual spraying, despite limited regional evidence on ITN utilization. To address this gap, a systematic review and meta-analysis were conducted to assess ITN utilization in Southeast Asia. A comprehensive search was performed across PubMed, Web of Science, and ScienceDirect, followed by PRISMA guidelines. Of 450 records screened, 28 studies published between 2003 and 2023 met the inclusion criteria for qualitative synthesis, with

Studies on the Design, Synthesis, and Insecticidal Activity Evaluation of New Caffeine-Acrylate Derivatives as Potential Ryanodine Receptor Activators.

On the basis of insecticidal leading structures of methylxanthine-acrylates I and II, a series of new caffeine-acrylate derivatives A-F were designed and synthesized via various structural optimization strategies. The structures of new compounds were confirmed through 1H NMR, 19F NMR, 13C NMR, and high-resolution mass spectrometry (HRMS). The bioassays showed that some of the new ester compounds possessed high insecticidal potentials against Plutella xylostellaL. and Mythimna separata Walker with improved insecticidal activity compared to the lead compounds. Notably, new compounds B8, B10, C1,

An organic solvent-free self-assembly strategy for scalable preparation of nanobiopesticides with enhanced insecticidal activity against houseflies.

The abuse of toxic organic solvents has caused great harm to human health and the environment. Therefore, developing an environmentally friendly nano-based pesticide formulation without using harmful solvents is urgent to improve the efficacy of pesticides and minimize environmental and health risks. Herein, by combining aspartic acid (Asp) with spinosad (SSD) as an attractive building unit, a self-assembly and carrier-minimized strategy was applied to construct a nanobiopesticide (Asp-SSD) simultaneously. To further improve the storage stability of the formulation, the biogenic surfactant alk

Contributions of health education and social mobilization in the implementation of a malaria control programme using long-lasting insecticidal nets.

BACKGROUND: Malaria is an infectious disease transmitted by mosquitoes of the genus Anopheles that represents a significant global health challenge, particularly in tropical and subtropical regions. In malaria-endemic areas, malaria control programmes have widely adopted long-lasting insecticidal nets (LLINs) and insecticide-treated nets (ITNs) as primary protection strategies against mosquito bites to interrupt transmission. To achieve maximum efficiency with these tools, proper community orientation through Health Education and Social Mobilization Strategies (HESMS) is necessary. This study

Effectiveness of a transfluthrin emanator and insecticide-treated barrier screen in reducing Anopheles biting in a temporary shelter in Sumatra, Indonesia.

BACKGROUND: The World Health Organization-approved Anopheles interventions target indoor biting and resting behaviour, but are impractical or inapplicable in some settings. In Jambi Province, Sumatra, Indonesia, local indigenous populations sleep under temporary tarpaulin-roofed shelters, complicating the use of bed nets and preventing the application of indoor residual spraying. Two pyrethroid-based interventions were tested alongside a no-intervention control in the field using a Latin-square design. A volatile pyrethroid spatial emanator (SE) offers an easily deployable, simple to use inter

Predictive chemoproteomics and functional validation reveal Coeae6g-mediated insecticide cross-resistance in the malaria vector Anopheles gambiae.

Sustaining malaria control in Africa is imperilled by the rapid evolution of insecticide resistance in the major vector Anopheles gambiae. Although current genomic and transcriptomic approaches map known resistance alleles, they often lack predictive power to anticipate liabilities to new insecticides. We present a predictive functional chemoproteomic framework integrating competitive activity-based protein profiling with functional validation to identify enzyme-mediated resistance mechanisms before they arise in field populations. Applied to a susceptible Anopheles gambiae strain, fluorophosp…

Utilization of insecticide-treated bed nets and key factors among households in resource-limited settings, Northwest Ethiopia.

BACKGROUND: Insecticide-treated mosquito nets (ITNs) act as an essential tool in malaria prevention by serving as a physical barrier against the disease. In sub-Saharan Africa, the use of ITNs remains a cornerstone in efforts to reduce malaria transmission. Despite their importance, there is limited evidence on ITN utilization and the factors influencing their use in Ethiopia. The objective of this study was to assess utilization of insecticidal treated bed nets and its key factors among Households in resource-limited settings, Northwest, Ethiopia, in 2024.
