Fusion of viral and cellular membranes, essential for entry of enveloped viruses, is mediated by viral fusion proteins (FPs). These proteins insert hydrophobic elements into target membranes to destabilize bilayers and drive fusion. For class I FPs, sequence and structural requirements of the fusion peptide are well characterized. In contrast, class II FPs, such as the alphavirus E1 glycoprotein, employ compact fusion loops, whose conserved sequence and structural features remain less understood. Here, we examined the chikungunya virus (CHIKV) E1 fusion loop using molecular dynamics (MD) simul

We prepared large unilamellar vesicles (LUVs) with three different stratum corneum lipid compositions: constant amounts of ceramides (55 wt %) and fatty acids (15%) with varying amounts of cholesterol sulfate (0-15%) and cholesterol (15-30%). One of the compositions served as a model for normal stratum corneum, while the second one served as a model for recessive X-linked ichthyosis stratum corneum. The third composition consisted of no cholesterol sulfate. Intervesicle lipid interactions in these LUVs were monitored by fluorescence methods for content leakage, and contents mixing at pH 9, in

Single-cell mass spectrometry enables label-free and high-throughput molecular analysis of individual cells. However, conventional vacuum-based secondary ion mass spectrometry (SIMS) faces challenges in probing metabolism of single living cells under native physiological conditions. Here, we introduce a liquid SIMS platform coupled with a vacuum-compatible cell-culture device, which allows in-situ metabolomic profiling of single living cells in their native culture environment without any pretreatment. This platform uniquely enables direct nanoscale interfacial characterization, and we report

A key characteristic of Alzheimer's disease (AD) is the build-up of amyloid-β (Aβ) fibrillar aggregates, commonly known as plaques, within the brain. Current diagnostic tools, such as MRI, CT, and PET, primarily detect neurodegeneration or plaque burden, limiting their utility to advanced stages of the disorder. Thus, there is a strong need for sensitive approaches that can enable detection at the earliest. Aβ aggregation begins with the formation of small, lipophilic oligomers characterized by antiparallel β-sheet structures. Emerging evidence suggests that these oligomers associated with exo

Non-specific membrane disruption induced by the amyloidogenic aggregation of β-amyloid (Aβ) peptides is considered an underlying molecular mechanism of Alzheimer's disease (AD). Therefore, elucidating the membrane interruptive intermediate states of Aβ aggregates is a crucial step towards the understanding of molecular basis of AD pathology. However, such intermediate states are heterogeneous, low-abundant, and insoluble, bringing challenges for the application of high-resolution techniques. The solid-state nuclear magnetic resonance (ssNMR) spectroscopy remains the most feasible technique to

Minimal cells offer a platform to uncover the fundamental physicochemical principles of cellular life. While genomes, proteomes, and metabolic networks of cells have been elucidated, the lipidome, which determines the physicochemical identity of the membrane, remains only partially characterized. Yet, the membrane is a central determinant of cellular viability, regulating solute permeability, mechanical stability, and environmental cues. Here, we investigate membranes that recapitulate the lipid composition of the minimal cell JCVI-syn3A, which is distinguished by an unusually high cholesterol

The selective permeability of reactive oxygen and nitrogen species (RONS), generated by cold atmospheric plasma (CAP), through plant aquaporins was investigated to identify plasma-derived species capable of intracellular delivery. Using atomistic molecular dynamics and enhanced sampling methods, we quantified the free energy profiles of eight RONS (HNO3, HO2, cis-HNO2, trans-HNO2, N2O4, NO, NO2, and O3) across the PIP2;1 aquaporin channel embedded in a lipid bilayer. Permeation was found to be controlled by two complementary features: the free-energy barrier opposing entry at the aromatic/argi

The rise of infectious diseases driven by antibiotic resistance has become a critical global concern, prompting the search for alternative therapeutic strategies. Among these, antimicrobial peptides (AMPs) have emerged as promising candidates. Spinigerin, a potent AMP, has demonstrated significant antibacterial and antifungal activity. Its mechanism involves disrupting microbial cell membranes by forming transmembrane pores that induce cell lysis. However, the precise mechanisms underlying pore formation remain poorly understood. To explore spinigerin's pore-forming potential, a comprehensive

Diffuse axonal injury (DAI), characterized by widespread damage to axons throughout the brain, represents one of the most devastating and difficult-to-treat forms of traumatic brain injury. Different theories exist about the mechanism of DAI, among which one hypothesis states that membrane poration of the axons initiates DAI. To investigate the hypothesis, molecular models of axonal membranes, incorporating 25 different lipids distributed asymmetrically in the leaflets, were developed using a coarse-grain description and simulated using molecular dynamics techniques. Different protein concentr

Radiolabeled nanocarriers that can simultaneously deliver therapeutic and diagnostic agents are promising for targeted cancer therapy. In this study, we employed molecular dynamics and metadynamics simulations to investigate how the chemical identity of drug molecules influences the behavior of technetium-99 m (⁹⁹ᵐTc)-loaded carbon nanotube (CNT) nanocarriers at the atomic scale. Four clinically relevant drugs-methotrexate, diclofenac, ketotifen, and piroxicam-were conjugated to a single-walled CNT (SWCNT) via a bifunctional chelating group. Our results revealed that drug structure significant

Nicotinamide adenine dinucleotide-dependent formate dehydrogenase from Candida boidinii was immobilized in a 1,2-dimyristoyl-sn-glycero-3-phosphocholine/cholesterol floating lipid bilayer on the gold surface as a biocatalyst for electrochemical CO2 reduction. We report that, in contrast to common belief, the enzyme can catalyze the electrochemical reduction of CO2 to formate without the cofactor protonated nicotinamide adenine dinucleotide. The electrochemical data indicate that the enzyme-catalyzed reduction of CO2 is diffusion-controlled and is a reversible reaction. The orientation and conf

Targeted delivery of drugs or other therapeutic agents through internal or external triggers has been used to control and accelerate the release from liposomal carriers in a number of studies, but relatively few utilize energy of therapeutic X-rays as a trigger. We have synthesized liposomes that are triggered by ionizing radiation (RTLs) to release their therapeutic payload. These liposomes are composed of natural egg phosphatidylethanolamine (PE), 1,2-distearoyl-sn-glycero-3-phosphocholine (DSPC), cholesterol, and 1,2-disteroyl-sn-glycero-3-phosphoethanolamine-N-[methoxy (polyethylene glycol

The understanding of amyloid β-peptide (Aβ) interactions with cellular membranes is a crucial molecular challenge against Alzheimer's disease. Indeed, Aβ prefibrillar oligomeric intermediates are believed to be the most toxic species, able to induce cellular damages directly by membrane damage. We present a neutron-scattering study on the interaction of large unilamellar vesicles (LUV), as cell membrane models, with both freshly dissolved Aβ and early toxic prefibrillar oligomers, intermediate states in the amyloid pathway. In addition, we explore the effect of coincubating the Aβ-peptide with

Cold atmospheric plasma (CAP) has attracted substantial attention in the field of medical disinfection because its main components, reactive oxygen species (ROS), have a strong destructive effect on various cell components. The cell membrane plays an important role in maintaining proper cellular function by blocking harmful substances such as ROS. In this paper, we used molecular dynamics simulations to study the behaviour of different ROS at the membrane-water interface. The results showed that the cell membrane presented a weak barrier to hydrophobic ROS (O2) but effectively prevented hydrop

Fundamental understanding of ion channel formation by amyloid peptides, which is strongly linked to cell toxicity, is very critical for (pre)clinical treatment of neurodegenerative diseases. Here, we combine atomistic simulations and experiments to demonstrate a broad range of conformational states of hIAPP double channels in lipid membranes. All individual channels display high selectivity for Cl(-) ions over cations, but the co-existence of polymorphic double channels of different conformations and orientations with different populations determines the non-ionic selectivity nature of the cha

Niemann-Pick disease and drug-induced phospholipidosis are examples of lysosomal storage disorders in which serious respiratory infections are brought on by high levels of the phospholipid phosphatidylcholine in the acidic lamellar bodies and lysosomes of pulmonary cells. One approach to developing an effective therapeutic agent could involve the use of a metal to preferentially hydrolyze phospholipid phosphate ester bonds at mildly acidic, lysosomal pH values (~pH 4.8). Towards this end, here we have investigated phosphatidylcholine hydrolysis by twelve metal ion salts at 60°C. Using a malach

Amphotericin B (AmB) is the most widely used antibiotic to treat systemic fungal infections. However, the molecular mechanism of its activity is still not completely understood. In the present work we have used FTIR spectroscopy to investigate the conformational state of the aliphatic chains of DPPC liposomes using the 2850 cm(-1) band, associated with the methylene symmetric stretching mode. The liposomes were either binary mixtures of the lipid with AmB, cholesterol or ergosterol, or ternary systems of these constituents. The two sterols contribute to an ordering of the aliphatic chains of t

The opening and closing of voltage-activated Na+, Ca2+ and K+ (Kv) channels underlies electrical and chemical signalling throughout biology, yet the structural basis of voltage sensing is unknown. Hanatoxin is a tarantula toxin that inhibits Kv channels by binding to voltage-sensor paddles, crucial helix-turn-helix motifs within the voltage-sensing domains that are composed of S3b and S4 helices. The active surface of the toxin is amphipathic, and related toxins have been shown to partition into membranes, raising the possibility that the toxin is concentrated in the membrane and interacts onl
