BACKGROUND: Quiescent non-union refers to a state where a fracture fails to heal, and the site appears clinically inactive despite the possible presence of a dormant or low-grade infection. This condition often follows open fractures or prior surgical interventions. Although typical signs of infection-pain, swelling, or discharge-are absent, latent infections may reactivate, leading to severe complications. Diagnosing infection in such cases is challenging due to the lack of definitive tools, and clinicians typically rely on inflammatory markers like C-reactive protein (CRP), erythrocyte sedim

Microbial translocation has been suspected to be a major contributing factor in the development of sepsis of unknown origin and multiple organ failure syndrome, but there are currently no tests capable of detecting and quantitating translocation sequentially in humans. The purpose of this study was to develop a sensitive polymerase chain reaction (PCR) test to detect Escherichia coli (E. coli) DNA in the blood of animals after inducing bacterial translocation from the gut. DNA was extracted from blood and primers were used to amplify an 800-bp gene fragment of E. coli by 30-cycle PCR. Detectio

BACKGROUND: Pseudomonas aeruginosa remains a serious threat in clinical settings, especially among patients who are immunocompromised, receiving chemotherapy, or in intensive care units. With the rise of antibiotic resistance, drug repurposing offers a promising alternative strategy. Busulfan, an anticancer alkylating agent that induces DNA cross-linking and cytotoxic effects in cancer cells, may exert similar effects on microorganisms, as reported for other alkylating agents.
METHODS: This study evaluated the antibacterial potential of busulfan against P. aeruginosa. Initially, the antimicro

OBJECTIVES: Delafloxacin is a fourth-generation fluoroquinolone that is currently approved in Europe and the United States for bacterial infections of the skin and skin structures and for community-acquired pneumonia, including cases where it is caused by atypical pathogens. It has demonstrated superior efficacy against a broad spectrum of bacteria and fewer adverse effects compared with other quinolones, such as levofloxacin, a current first-line treatment against Legionnaires' disease. This study aimed to characterize for the first time the intracellular activity of delafloxacin compared wit

Bacterial spores in materials and equipment pose significant biosecurity risks, making effective disinfection crucial. This study evaluated Ortho-phthalaldehyde (OPA) and a quaternary ammonia-glutaraldehyde solution (AG) for inactivating spores of Bacillus thuringiensis (BT), B. cereus (BC), and two strains of B. velezensis (BV1 and BV2). Spores of BV1 and BT were treated with 22.5 mg/m3 OPA by dry fumigation or 1 mg/mL AG by spray for 20 min, according to the manufacturer's recommendation. As no sporicidal effect was observed, OPA was tested at 112.5 mg/m3 for 40 min, showing effectiveness fo

OBJECTIVES: In some countries, third-generation cephalosporins (3GCs) serve as first-line therapy in children with urinary tract infections (UTIs). However, their use may contribute to the emergence of antibiotic resistance, notably among Gram-negative bacteria (GNB). Integrons are bacterial genetic elements involved in antibiotic resistance in GNB. Their absence is associated with >97% susceptibility to trimethoprim/sulfamethoxazole in adults infected with GNB. The objective of this study was to examine the value of integron detection directly from urine samples as a predictive marker of resi

Changes in the interpretive-breakpoints for antifungals against various Candida species have raised the need to examine the significance of the phenomenon of the growth of microcolonies in agar diffusion inhibition zones, which has generally been considered negligible. The objective was to determine the incidence of cases in which microcolonies demonstrate fluconazole resistance according to current interpretive-breakpoints and whether their growth is associated with therapeutic failure. The fluconazole minimum inhibitory concentrations (MICs) of 100 blood culture isolates of Candida were perf

Antibiotic resistance is generally associated with a fitness deficit resulting from the burden of producing and maintaining resistance machinery. This additional cost suggests that resistant bacteria will be outcompeted by susceptible bacteria in conditions without antibiotics. However, in practice, this process is slow in part because of regulation that minimizes expression of these genes in the absence of antibiotics. This suggests that if it were possible to turn on their expression, the cost would increase, thereby accelerating removal of resistant strains. Experimental and theoretical stu

Rapid inactivation of antimicrobial test agents after exact incubation times with microorganisms is required in time-kill assays. Sodium thiosulphate and a combination of methionine and histidine were compared for neutralisation of active halogen compounds. Test oxidants were mixed with surplus sodium thiosulphate (3%-6%) or histidine/methionine (1% each) in phosphate-buffered saline and incubated for different times, followed by addition of Staphylococcus aureus, Escherichia coli, or Pseudomonas aeruginosa at 1000CFU/ml. After further incubation, quantitative cultures were performed. Thiosulp

This study aimed to investigate the effect of sub-minimum inhibitory concentrations (sub-MICs) of three food-grade biocides (benzalkonium chloride -BZK-, trisodium phosphate -TSP-, and sodium hypochlorite -SHY-) on Salmonella biofilms. The structural parameters and bacterial viability of the biofilms formed by a S. Typhimurium isolate from poultry was investigated by means of confocal laser scanning microscopy after staining with SYTO9 and propidium iodide. The MIC values for Salmonella cells before exposure to subinhibitory concentrations of biocides were 8.0 μg/mL (BZK), 18.0 mg/mL (TSP), an

The emergence of drug resistance and limitation of antifungal agents bring enormous challenges to the management of Candida infection. Combination therapy is a useful way to treat the fungal infection, especially for those difficult-to-treat infections. In this research, we investigated the interaction effects between diorcinol D (DD), an antifungal active natural product from lichen endophytic fungus and fluconazole (FLC) against planktonic cells and mature biofilms of Candida albicans in vitro by checkerboard microdilution and time-killing tests. Both fractional inhibitory concentration inde
