In vivo assessment of a nanofibrous silk tube as nerve guide for sciatic nerve regeneration.

A nanofibrous silk nerve conduit has been evaluated for its efficiency based on the promotion of peripheral nerve regeneration in rats. The designed tubes with or without Schwann cells were implanted into a 10 mm gap in the sciatic nerves of the rats. Four months after the surgery, the regenerated nerves were monitored and evaluated by macroscopic assessments and histology. The results demonstrated that the nanofibrous grafts, especially in the presence of Schwann cells, enabled reconstruction of the rat sciatic nerve trunk with a restoration of nerve continuity and formation of nerve fibres w

Inflammatory profiling of Schwann cells in contact with growing axons distal to nerve injury.

Activated Schwann cells distal to nerve injury upregulate inflammatory mediators, including cytokines. The goal of the present study was to investigate expression of proinflammatory (IL-1β, TNFα) and anti-inflammatory cytokines (IL-4, IL-10) in activated Schwann cells in relation to growing axons distal to crush injury of rat sciatic nerves. Seven days from sciatic nerve crush, transverse cryostat sections were cut 5 mm distal to lesion and incubated for double immunostaining to indicate Schwann cells (GFAP or S100b) and individual investigated cytokines or to demonstrate growing axons (GAP43)

Functional recoveries of sciatic nerve regeneration by combining chitosan-coated conduit and neurosphere cells induced from adipose-derived stem cells.

Suboptimal repair occurs in a peripheral nerve gap, which can be partially restored by bridging the gap with various biosynthetic conduits or cell-based therapy. In this study, we developed a combination of chitosan coating approach to induce neurosphere cells from human adipose-derived stem cells (ASCs) on chitosan-coated plate and then applied these cells to the interior of a chitosan-coated silicone tube to bridge a 10-mm gap in a rat sciatic nerve. Myelin sheath degeneration and glial scar formation were discovered in the nerve bridged by the silicone conduit. By using a single treatment o

Neuropeptide-like intraneuronal profiles in the human VIIIth cranial nerve.

Intraneuronal inclusion bodies corresponding to large dense core vesicles are described in the primary neurons of the cochleovestibular system. Their diameters ranged from 80 to 120 nm. Considering their morphologic characteristics and findings in previous immunohistochemical studies, the vesicles are likely to contain neuropeptides, particularly substance P or enkephalin.

Rapid axoplasmic transport of free leucine.

Axoplasmic transport of free 3H-leucine has been studied in vivo in the pike olfactory nerve following application of labeled leucine to the olfactory mucosa. A considerable amount of free 3H-leucine is transported at constant velocity along the axon in the form of a distinct peak. The maximum transport velocity for free 3H-leucine is the same as for rapidly transported 3H-protein (130 and 135 mm/day, respectively, at 19 degrees C). Microtubule inhibitors block or significantly reduce the amount of free 3H-leucine transported, but do not influence the transport velocity. Disruption of the oxyg

Extracellular matrix remodelling in human diabetic neuropathy.

The extracellular matrix of peripheral nerve plays a vital role in terms of normal nerve fibre function and also in the regenerative response following nerve injury. Nerve fibre loss is a major feature of diabetic neuropathy; however, the regenerative response is limited and this may be associated with changes in the composition of the extracellular matrix. Glycoproteins and collagens are major components of the extracellular matrix and are known to be important in terms of axonal growth. This work has therefore examined whether changes in the expression of two major glycoproteins, laminin and

Sensory nerve endings in the hard palate and papilla incisiva of the rhesus monkey.

The sensory innervation of the hard palate of the rhesus monkey was studied by light and electron microscopy. The mucosa of the hard palate is subject to a particularly heavy mechanical load requiring functional specialisation of the horny epithelium in the form of thickenings - the papilla incisiva and eight pairs of rugae palatinae. A thin layer of firm connective tissue (lamina propria) attaches the mucosa to the periost of the hard palate. Sensory nerve fibres were found most abundantly in the papilla incisiva and first rugae palatinae. Their number decreases in an aboral direction. Five t

An investigation of the digital nerves of the thumb.

With the use of a stereoscopic microscope, observations were made on the course of the proper digital nerve of the thumb on ten upper limbs in adult cadavers. By observation from the dorsal side after removal of the bone, it was possible to clearly see the ramification of the digital nerve from the subcutaneous tissue to the dermis. Ramification from the palm to the level of the interphalangeal joint was segmental. The digital nerve of the thumb trifurcates from the proximal site of the distal palmar thumb crease into the main, medial, and lateral branches. The main branch is thick and extends

Biocompatible 3D-Printed Devices With Adipose Stem Cells in the Regenerative Process of Sciatic Nerve Lesions in Rodent Models: An Experimental Study.

INTRODUCTION: Peripheral nerve injuries are a significant clinical challenge. The rat sciatic nerve serves as an ideal model for studying nerve regeneration. Extensive research has been conducted to unravel the intricate mechanisms involved in peripheral nerve regeneration, aiming to develop effective therapeutic strategies for nerve injury patients. Research including different types of materials that can be used as nerve guides like synthetic polymers have been investigated for their biocompatibility and molding properties. Among multiple stem cell types, adipose-derived stem cells (ASCs), b

Rat-derived processed nerve allografts support more axon regeneration in rat than human-derived processed nerve xenografts.

Processed nerve allografts are increasingly used as "off the shelf" nerve replacements for surgically bridging nerve gaps. Benchmarking the regenerative capacity of a commercially available human-derived nerve or xenograft in a rat nerve injury model would provide a convenient platform for future studies seeking to modify the processed nerve graft. Human and rat processed nerve grafts were used to bridge a 14 mm defect in a Sprague-Dawley rat sciatic nerve. Reversed autografts served as a positive control group. Twelve weeks following surgery, the distal nerve stumps were retrograde labeled an

Intracranial reconstruction of the facial nerve. Clinical observation.

Nine cases of intracranial facial nerve reconstruction are reviewed in this paper. All patients underwent this procedure for severe injury or disruption of the facial nerve during surgery for acoustic neruroma through the modified extended middle cranial fossa approach (1). Satisfactory recovery of facial function was obtained in 4 patients. Three patients underwent hypoglossal-facial nerve anastomosis 1.3-1.5 years later for no or poor recovery of the facial function. One patient refused any further surgical treatment despite unsatisfactory recovery. The remaining 1 patient, during a telephon

Denervation in the primary olfactory pathway of mice. IV. Biochemical and morphological evidence for neuronal replacement following nerve section.

Unilateral olfactory nerve section was performed in the mouse. Three biochemical markers of the olfactory chemoreceptor neurons: carnosine, carnosine synthetase activity and the olfactory marker protein, were measured in the olfactory bulb and epithelium. Parallel observations were made by light microscopy as well as at the ultrastructural level. The specific biochemical markers decrease rapidly in both bulb and epithelium and reach a minimum by the end of the first week after surgery. They then slowly return to 80% of control values by one month. Carnosinase activity in epithelium was essenti

Magnetic resonance imaging of intratemporal facial nerve lesions in the animal model.

The facial nerve is often injured in head trauma. Computed tomography (CT) is the diagnostic study most frequently used to evaluate temporal bone fractures, but it does not demonstrate all soft tissues well. Recently, magnetic resonance imaging (MRI) has been used to evaluate certain soft tissues, including cranial nerves. Experience with MRI evaluation of temporal bone trauma is limited and consists primarily of anecdotal reports. This study assesses the accuracy of MRI in evaluating experimentally induced acute intratemporal facial nerve lesions. The tympanic segments of the right facial ner

Coaxial Bioprinting of Schwann Cells and Neural Stem Cells in a Three-Dimensional Microenvironment for the Repair of Peripheral Nerve Defects.

Currently, autologous nerve (AN) transplantation remains the gold standard for treating peripheral nerve injuries (PNIs). However, its inherent limitations, including donor site morbidity and immune rejection risks associated with allografts, have prompted the exploration of alternative therapeutic strategies. Among these, tissue engineering approaches have gained significant attention, with nerve conduit design emerging as a particularly promising research direction. Electrospinning technology has been widely adopted for its ability to fabricate nanofibrous scaffolds that closely mimic the na

Quantitative study of the development and maturation of human oesophageal innervation.

By 8 wk gestation, the human fetal oesophagus is identifiable as a hollow epithelium-lined tube with primitive nerve and muscle precursors present. From 8-16 wk gestation, the muscle layers and innervation mature until fetal swallowing commences at 16 wk. This study examines quantitatively the development and maturation of nerve fibres and cell bodies within the oesophagus using histochemistry. Oesophageal samples (n = 35) from 8 wk gestation to 28 months of age and adults (n = 3) were immunostained using antisera for the general nerve marker, protein gene product (PGP 9.5), the glial tissue m

Mechanisms Underlying Abnormal Expression of lncRNA H19 in Neonatal Hypoxic-Ischemic Encephalopathy.

OBJECTIVE: Hypoxic-ischemic (HI)-related brain injury, especially HI encephalopathy (HIE) is a leading cause of morbidity and disability in newborns. Long noncoding RNAs (lncRNAs) are implicated in the progress of HI brain damage. However, the mechanisms underlying the regulatory effects of lncRNA H19 on autophagy in HIE remain unknown. This study was designed to identify the potential mechanisms involving lncRNA H19 in HIE. STUDY DESIGN: We selected three HIE newborns and three healthy newborns for neonatal behavioral neurological assessment and screened the differentially expressed lncRNAs
