The lateral vestibular nucleus is a site for the depressant action of benzodiazepines on the crossed extensor reflex.

The effects of diazepam microinjected into the subcerebellar nuclei, the lateral vestibular nucleus (LVN), the red nucleus and the substantia nigra were investigated using the crossed extensor reflex (CER) model in chloralose-anesthetized rats. Microinjections of diazepam at doses of 1 and 25 ng/animal into the left LVN, ipsilateral to the testing muscle, produced a dose-related CER depression, while injections of diazepam at a dose of up to 100 ng/animal into the right LVN and into other regions tested produced little effect. The CER depression produced by diazepam could be clearly reversed b

Slow postsynaptic potentials in neurones of submucous plexus of guinea-pig caecum and their mimicry by noradrenaline and various peptides.

Intracellular recordings of membrane potential and membrane currents were made from neurones in the submucous plexus of the guinea-pig caecum in vitro. Fast and slow excitatory postsynaptic potentials and slow inhibitory postsynaptic potentials were recorded from the majority of neurones following focal stimulation of presynaptic fibres in the plexus. The slow inhibitory postsynaptic potential was associated with an increase in membrane conductance and reversed its polarity at -90 mV; it was reversibly blocked by yohimbine. The slow excitatory postsynaptic potential and its underlying current

Electrical stimulation of insular cortex elicits cardiac inhibition but insular lesions do not abolish conditioned bradycardia in rabbits.

Conscious rabbits received electrical stimulation of insular and more posterior perirhinal cortex through chronically implanted electrodes. Active sites for cardiovascular responses were found in both anterior and posterior insular cortex as well as more posterior perirhinal regions. Although differential response topographies occurred related to anterior versus posterior insular cortex, all heart rate responses consisted of bradycardia. Pharmacological manipulations revealed that this bradycardia was due to a combination of vagal and sympatho-inhibitory mechanisms. Some posterior sites yielde

[Synaptic processes in smooth muscles].

Junction potentials evoked by intramural stimulation were investigated in smooth muscles of guinea-pig gastrointestinal tract by means of intracellular recording and sucrose-gap methods. It was shown that cholinergic nerve-muscle transmission is only excitatory and more effective in fundic smooth muscles as compared with other part of the gut. Adrenergic nerve control of gastrointestinal motility is carried out both by excitation and inhibition of the smooth muscles. Nonadrenergic inhibition was observed in circular muscle layer of all parts of the gut and was more effective in distal colon. N

Effects of substance P on nociceptive and non-nociceptive trigeminal brain stem neurons.

As little information is available on the chemistry of synaptic transmission in trigeminal brain stem nuclei, an iontophoretic study was done on the effects of glutamate and substance P on single neurons in trigeminal nuclei oralis and caudalis in cats anesthetized with chloralose and paralyzed. The neurons were additionally studied for their responses to natural noxious and innocuous cutaneous and intraoral stimuli as well as to bipolar stimulation of the ipsilateral and contralateral canine tooth pulps, the exposed infraorbital and superior laryngeal nerves and forepaw. Glutamate excited all

Synchronized rhythmic discharges of the secondary olfactory neurons in carp.

(1) Rhythmic and synchronized multiunit bursts of spike discharges, which were superimposed on the peaks of the oscillatory potentials of 4--10 Hz (olfactory tract waves), were induced in the olfactory tract fibers of the carp in response to olfactory stimulation. Suppression of the spike discharges was observed during the interburst periods of discharges. (2) Single-unit analysis revealed three types of discharge patterns in the olfactory tract fibers in response to olfactory stimulation: rhythmic excitation (Type 1), suppression (Type 2) and no response (Type 3). A synchronization of spike d

Inhibition of electromyographic activity in human triceps surae muscles during sinusoidal rotation of the foot.

Torque and electromyographic (EMG) responses to sinusoidal rotations of the foot were measured. The frequency range of the movements was 0.5 Hz to 15 Hz at amplitudes ranging between 1 and 10 degrees. At frequencies above 7 Hz, the EMG activity did not follow individual foot rotation cycles. The EMG activity was inhibited whenever the peak torque was large with respect to the first cycle peak torque. Dantrolene sodium reduced the torque developed in triceps surae, allowing the EMG activity to follow individual stretch cycles. As the drug was metabolized, the EMG activity returned to the charac

Visuomotor Activation of Inhibition-Processing in Pediatric Obsessive Compulsive Disorder: A Magnetoencephalography Study.

Background: Response inhibition engages the cortico-striato-thalamo-cortical (CSTC) circuit, which has been implicated in children, and youth with obsessive compulsive disorder (OCD). This study explored whether CSTC engagement during response inhibition, measured using magnetoencephalography (MEG), differed in a sample of medication-naïve youth with OCD, compared to typically developing controls (TDC). Methods: Data was analyzed in 17 medication-naïve children and youth with OCD (11.7 ± 2.2 SD years) and 13 TDC (12.6 ± 2.2 SD years). MEG was used to localize and characterize neural activity d

Mechanisms of action of neurotensin on motility of canine gastric corpus in vitro.

Increasing concentrations of neurotensin produced a biphasic contractile response, inhibition followed by excitation, when added to full-thickness circular strips of canine gastric corpus muscle cut in a circular orientation and incubated at 37 degrees C in vitro. The inhibitory response (mean effective concentration (EC50) 7 x 10(-9)M) was not altered by addition of concentrations of tetrodotoxin or scorpion venom sufficient to block field-stimulated neural responses nor by any other antagonist tested. Therefore, it would appear that inhibition is due to an excitation of the smooth muscle rec

A Computational Framework for Myelin as a Regulator of Temporal Integration, Neuronal Excitability, and Synaptic Plasticity.

Myelin regulates neural signaling not only by enabling fast conduction but also by dynamically tuning axonal conduction velocity (CV) and thereby shaping spike timing. While activity-dependent myelination is thought to optimize neural communication, how changes in myelin translate into altered single-cell dynamics and plasticity remains unclear. Using a computational modeling approach, we show that disruptions of axonal CV reorganize presynaptic input correlations, leading to systematic changes in postsynaptic excitability, excitation-inhibition balance, and spike-timing-dependent plasticity.

The glutamatergic NTS-PAG/PVN pathway underlies myocardial injury and anxiety-like behavior induced by myocardial ischemia-reperfusion.

For patients with myocardial ischemia-reperfusion injury (MIRI), cardiac dysfunction accompanied by anxiety is an extremely common comorbidity. Although the association between these two conditions has received some attention, the underlying neural mechanisms remain unclear. Here, we functionally characterized two distinct glutamatergic (Glu) neural pathways in the nucleus tractus solitarius (NTS) of male mice, a pathway from the NTS to the periaqueductal gray (PAG) that mediates anxiety-like behaviors in MIRI mice, and a pathway from the NTS to the paraventricular nucleus of the hypothalamus

Mast cell extracellular trap formation underlies vascular and neural injury and hyperalgesia in sickle cell disease.

Sickle cell disease (SCD) is the most common inherited monogenetic disorder. Chronic and acute pain are hallmark features of SCD involving neural and vascular injury and inflammation. Mast cells reside in the vicinity of nerve fibers and vasculature, but how they influence these structures remains unknown. We therefore examined the mechanism of mast cell activation in a sickle microenvironment replete with cell-free heme and inflammation. Mast cells exposed to this environment showed an explosion of nuclear contents with the release of citrullinated histones, suggestive of mast cell extracellu

PDK1 Deficit Impairs the Development of the Dentate Gyrus in Mice.

3-Phosphoinositide-dependent protein kinase-1 (PDK1) is crucial for the development of the dentate gyrus (DG), the first gateway receiving afferent inputs from the entorhinal cortex. However, the role of PDK1 in DG development is unclear. In the present study, by crossing Pdk1fl/fl mice with the Emx1-cre line, we identified that the ablation of PDK1 disrupted the development of DG via decreasing the proliferation, and increasing the differentiation of dentate neural progenitor cells, downregulating AKT activity and upregulating GSK3β signaling. Moreover, PDK1 deletion disrupted the distributio

Visual Attention Deficits in Schizophrenia Can Arise From Inhibitory Dysfunction in Thalamus or Cortex.

Schizophrenia is associated with diverse cognitive deficits, including disorders of attention-related oculomotor behavior. At the structural level, schizophrenia is associated with abnormal inhibitory control in the circuit linking cortex and thalamus. We developed a spiking neural network model that demonstrates how dysfunctional inhibition can degrade attentive gaze control. Our model revealed that perturbations of two functionally distinct classes of cortical inhibitory neurons, or of the inhibitory thalamic reticular nucleus, disrupted processing vital for sustained attention to a stimulus

Sustained activation of mTOR pathway in embryonic neural stem cells leads to development of tuberous sclerosis complex-associated lesions.

Tuberous Sclerosis Complex (TSC) is a multisystem genetic disorder characterized by hamartomatous neurological lesions that exhibit abnormal cell proliferation and differentiation. Hyperactivation of mTOR pathway by mutations in either the Tsc1 or Tsc2 gene underlies TSC pathogenesis, but involvement of specific neural cell populations in the formation of TSC-associated neurological lesions remains unclear. We deleted Tsc1 in Emx1-expressing embryonic telencephalic neural stem cells (NSCs) and found that mutant mice faithfully recapitulated TSC neuropathological lesions, such as cortical lamin

Neural ensembles that encode nocifensive mechanical and heat pain in mouse spinal cord.

Acute pain is an unpleasant experience caused by noxious stimuli. How the spinal neural circuits attribute differences in quality of noxious information remains unknown. By means of genetic capturing, activity manipulation and single-cell RNA sequencing, we identified distinct neural ensembles in the adult mouse spinal cord encoding mechanical and heat pain. Reactivation or silencing of these ensembles potentiated or stopped, respectively, paw shaking, lifting and licking within but not across the stimuli modalities. Within ensembles, polymodal Gal+ inhibitory neurons with monosynaptic contact

Mutation of CHD7 impairs the output of neuroepithelium transition that is reversed by the inhibition of EZH2.

Haploinsufficiency of CHD7 (Chromo-Helicase-DNA binding protein 7) causes a severe congenital disease CHARGE syndrome. Brain anomaly such as microcephaly and olfactory bulb agenesis seen in CHARGE patients have not been mimicked in previous animal models. Here, we uncover an indispensable function of CHD7 in the neuroepithelium (NE) but not in the neural stem cells (NSCs) after NE transition. Loss of Chd7 in mouse NE resulted in CHARGE-like brain anomalies due to reduced proliferation and differentiation of neural stem and progenitor cells, which were recapitulated in CHD7 KO human forebrain o

HB-EGF activates EGFR to induce reactive neural stem cells in the mouse hippocampus after seizures.

Hippocampal seizures mimicking mesial temporal lobe epilepsy cause a profound disruption of the adult neurogenic niche in mice. Seizures provoke neural stem cells to switch to a reactive phenotype (reactive neural stem cells, React-NSCs) characterized by multibranched hypertrophic morphology, massive activation to enter mitosis, symmetric division, and final differentiation into reactive astrocytes. As a result, neurogenesis is chronically impaired. Here, using a mouse model of mesial temporal lobe epilepsy, we show that the epidermal growth factor receptor (EGFR) signaling pathway is key for

Contributions of cortical neuron firing patterns, synaptic connectivity, and plasticity to task performance.

Neuronal responses during behavior are diverse, ranging from highly reliable 'classical' responses to irregular 'non-classically responsive' firing. While a continuum of response properties is observed across neural systems, little is known about the synaptic origins and contributions of diverse responses to network function, perception, and behavior. To capture the heterogeneous responses measured from auditory cortex of rodents performing a frequency recognition task, we use a novel task-performing spiking recurrent neural network incorporating spike-timing-dependent plasticity. Reliable and
